Method Twenty four patients with acute myelocytic leukemia and 21 patients with acute lymphocytic leukemia were treated with combination intensive chemotherapy.rhG-CSF (300 μg/d) was subcutaneously injected after chemotherapy-induced white blood cell(WBC)1.0×10 9/L until recovery from chemotherapy-induced myelosuppression(absolute neutrophil counts ANC1.5×10 9/L). Results The average time to ANC1.5×10 9/L was (5.9±5.1) d in study group versus (14.8±5.0) d in control group.The administration of rhG CSF significantly decreased infection rate(20%∶80%). Abstract:OBJECTIVES: To determine the serum IL-2, IL-2Ab, IFN-α and IFN-α antibody in patients with GBS and to explore their role in pathogenesis of the disease. METHODS:Serum IL-2, IL-2Ab, IFN-α and IFN-α antibody were determined by ELISA in 50 patients with GBS and 30 healthy subjects. RESULTS: The levels of serum IL-2, IL-2Ab,IFN-α and IFN-α antibody in case with acute attack stage of GBS were significantly higher than those in controls(P0.05). CONCLUSIONS:Serum IL-2 and IFN-α are involved in pathogenesis of GBS, and IFN-α antibody and IL-2Ab maybe restrict pathogenesis of disease.

Abstract:A study was carried out to investigate the efficacy of therapy with recombinant human granulocyte colony-stimulating factor (rhG-CSF) in 24 patients with granulocytopenia and sepsis who had failed to respond to antibiotics. The mean leukocyte count at the start of the study was 911 +/- 334/microliter. Patients were injected subcutaneously with 75 micrograms rhG-CSF once daily for a mean of 5.2 days. The plasma G-CSF concentration was measured by ELISA. The leukocyte count increased approximately 9-fold after 1 week in 19 patients and the percentage of granulocytes rose from 46.2% to 78.9%. These 19 patients survived, while the 5 patients with no leukocyte response to rhG-CSF died. High plasma G-CSF levels were found in patients with granulocytopenia. Plasma G-CSF levels decreased as levels of granulocyte increased in survivors. A high plasma G-CSF concentration persisted in the 5 non-responding patients resulting in a fatal outcome. This study suggests that rhG-CSF both increased the leukocyte count and was a useful therapeutic manoeuvre for sepsis.

Abstract:OBJECTIVE To investigate the effect of recombinant human granulocyte colony-stimulating factor(rhG-CSF) combined with antifungal drugs in the treatment of malignant hematologial diseases with fungus infection.METHODS Malignant hematologial patients with fungus infection were randomized to receive fluconazole with or without rhG-CSF.(RESULTS) The response rate in patients who received fluconazole combined with rhG-CSF was 89.1% and in(control) patients was 62.8%(P<0.05).The absolute neutrophil count(ANC) in patients who(received)(fluconazole) combined with rhG-CSF recovered faster than that of without rhG-CSF.CONCLUSIONS rhG-CSF can accelerate the recovery of ANC,and enhance the curative effect when combined with antifungal drugs.

Abstract:Objective To investigate prognostic significance of granulocyte-macrophage colonystimulating factor (GM-CSF) and transfroming growth factorβ1(TGF-β1) levels in patients with acute leu-kemia (AL).Method ELISA assay were employed to investigate the levels of GM-CSF and TGF-β1in96 AL patients.Results The levels of TGF-β1were much lower at the time of diagnosis than that of nor-mal controls(P0.05),The levels of GM-CSF were much higher at the time of diagnosis than that of nor-mal controls(P0.05),Line correlation analysis:GM-CSF correlated negatively with TGF-β1.The levelsof GM-CSF and TGF-β1returned to normal levels in those who got completely remission (CR).There wasno difference among non-remission(NR),relapsed and de novo patients.Conclusion Monitoring the lev-els of GM-CSF and TGF-β1may have prognostic value in clinical outcome and is useful marker treatmentof AL patients.

Abstract:Objective:To quantitatively estimate the granulocyte colony-stimulating factor receptor(G-CSFR) expressed on nucleated cells in peripheral blood from healthy individuals.Methods:Twenty two blood samples from healthy individuals were examined by flow cytometry with use of fluorescence-labeled antibody against G-CSFR and a quantitative fluorescence kit.Results:Flow cytometric estimations of antibodies bound per cell(ABC) in neutrophil,monocyte,lymphocyte,T-cell lymphocyte,B-cell lymphocyte were 2 943±417,3 755±2 575,996±369,2 787±946,10 112±6 345(Median±Interquartile Range),respectively.There was no significant difference in ABC values among neutrophil,monocyte and CD3 positive T-cell lymphocyte,while ABC value in lymphocytes was less than in other cells significantly,and ABC value in CD19 positive B-cell lymphocytes was higher than in other cells significantly(P<0.05).Conclusion:This study describes the G-CSFR expressing characteristics of nucleated cells in peripheral blood from healthy individuals,and the values could be referred to in similar study in future.Compared to radioimmunoassay,flow cytometric quantitative estimation of cell surface antigen is much more safe and convenient.
Inflammatory Cytokine Measurement Quickly Discriminates Gram-Negative from Gram-Positive Bacteremia in Pediatric Hematology/oncology Patients with Septic Shock

Abstract:Purpose We performed a prospective study to evaluate the ability of inflammatory cytokines in discriminating gram-negative from gram-positive bacteremia in septic shock. Methods During the study period, the serum inflammatory cytokine levels were measured at the onset of septic shock by flow cytometry in pediatric hematology/oncology patients with septic shock. Results One hundred episodes of septic shock were enrolled. Of 97 episodes of monomicrobial infection, 73.2 % were caused by gram-negative bacteremia and 26.8 % by gram-positive bacteremia. Interleukin (IL)-6, IL-10, and tumor necrosis factor (TNF)-α were closely related to the pediatric index of mortality 2 (PIM2) score and mortality. However, although the PIM2 score and mortality were comparable, the IL-6, IL-10, and TNF-α levels were significantly higher in patients with gram-negative bacteremia (GNB) than those with gram-positive bacteremia (median levels, pg/mL: IL-6: 784.1 vs. 254.4, P = 0.001; IL-10: 192.2 vs. 19.7, P < 0.001; TNF-α: 4.2 vs. 2.0, P < 0.001). Of the three cytokines, IL-10 was the most useful biomarker for GNB prediction in the derivation cohort and a cutoff value of 50 pg/mL showed a sensitivity of 70.8 % and a specificity of 80.0 %, with a positive predictive value of 89.5 %. When this cutoff value was applied to the validation cohort, the sensitivity, specificity, and positive predictive value were 80.9, 75.0, and 90.5 %, respectively. Conclusions Flow cytometry-based inflammatory cytokine measurement is a helpful adjuvant approach for early and quick discrimination of gram-negative from gram-positive bacteremia in pediatric hematology/oncology patients with septic shock which might be useful for evaluating the severity of shock and the selection and/or timely withdrawal or switch of antibiotics.
Investigation on related factors of CD 34+ cells in peripheral blood mobilization effect by G-SCF stimulation

Abstract:Objective To investigate the influence factors of peripheral blood hematopoietic stem cell of healthy donor by granu‐locyte colony‐stimulating‐factors(G‐CSF) mobilization .Methods G‐CSF was subcutaneously injected in 24 cases of healthy donor for mobilizing hematopoietic stem cells .The T lymphocyte subgroup and blood routine data were detected .Results After G‐CSF stimulation ,peripheral blood CD3+ (% ) ,CD3+CD4+ (% ) ,white blood cell (WBC) count and platelet were significantly increased (P<0 .05) .And nucleated cells density of bone ,percentage of CD34+ cells on mobilized 4 ,5 ,6 d had no obvious difference .The correlation analysis showed that gender ,age and weight were negatively correlated with CD 34+cells percentage (P<0 .05) and pos‐itively correlated with WBC count (P<0 .01) .Conclusion Male donor is superior to female donor within a certain range ,the smal‐ler age ,the lighter weight ,the higher WBC count ,the higher the percentage of CD34+ cells in peripheral blood hematopoietic stem cells after G‐CSF mobilization .

Abstract:Abstract Objectives: Neutropenic fever is very common in chilidren with chemotherapy. Due to the fact that an infection is usually suggested, broad antibiotic therapy is usually administered. However, early diagnosis of the infection is usually difficult and the antibiotic therapy may not be alway appropriate. Furthermore, blood culture is usually negative for those heavily treated patients by antibiotics. So a sensitive biomarker for early recognition of inction is desired. Previous studies have shown that procalcitonin (PCT) is a good marker for predicting infection. However, in our experience, the sensitivity of infection recognition by PCT is low. Our previous studies have shown that cytokine profiles presented very high sensitivity and sepecificity for predicting infection in hematology-oncology children at the time of neutropenic fever. However, the diagnositic power between cytokine profiles and PCT in those patients has not been systemically compared. The aim of this study was to evaluate the diagnostic power of the cytokine pattern and serum PCT level for predicting infection in pediatric hematology/oncology patients. Methods: In this retrospective study, a total of 3023 samples (2819 febrile episodes and 204 control samples) derived from 992 (including 164 afebrile patients as control) hospitalized hematology-oncology children. Th1/Th2 cytokines including Interleukin (IL)-2, Il-4, IL-6, IL-10, Tumor necrosis factor (TNF) α and interferon (IFN) γ were measured by a FACScalibur flow cytometer using cytometric bead array (CBA) techniques. According to the etiology, imaging and clinical symptoms, 2819 febrile episodes were divided into 5 groups including blood culture positive sepsis, clinical diagnosed sepsis, non-sepsis infection, viral infection and systemic fungal infection. Results: IL-6 and IL-10 levels as well as positive rates in bacterial infection group were significantly higher than those in viral infection group and control group. In positive blood culture sepsis group, 92.8% (297/320) and 82.2% (263/320) patients had increased IL-6 and IL-10 levels that exceeded the upper limit of the normal range. However, the positive rates of PCT and C-reactive protein (CRP) in bacterial infection group were only 33.8% (108/320) and 73.1%(234/320), respectively, which were significantly lower than those of IL-6 and IL-10 (P<0.01). Based on the receiver operating characteristic (ROC) curves, PCT presented a poorer sensitivity in the diagnosis of severe infection, as compared with IL-6 and IL-10 (p<0.01). As for specificity, IL-6 and IL-10 was also significantly higher than PCT in the diagnosis of Gram-negative bacterial infection. Conclusion: The specific cytokine pattern of highly elevated IL-6 and IL-10 shows higher diagnostic accuracy than PCT for bacterial and severe infection among febrile hematology-oncology children. Abstract:Objective To investigate the pharmacokinetic progerty after repeated administration of recombinant human granulocyte colony stimulating factor( rhG-CSF) in children with acute leukemia and its clinical significance. Methods . Serum G-CSF concentrations were measured in 46 acute leukemic children by enzyme linked immunosorbent assay. Results The serum G-CSF concentration was(5. 335 ±5. 597 ) μg/L in 29 children during rhG-CSF treatment . In children received repeted treatment of rhG-CSF,serum G-CSF levels resumed to normal before next administration. Only few cildren (2/8)who didnf receive any rhG-CSF treatment had higher rhG-CSF concentrations;Tbe serum G-CSF concentrations correlated negatively with absolute neutrophil counts ( r = - 0. 8746, P ＜ 0. 01 ). Conclusion There is a granulocyte - madiating clearing mechanism in rhG-CSF pharmacokinetics, and repeated administration of rhG-CSF was safe without accumuting effect.

Abstract:Objective To investigate the correlation of monocyte counts prior to the collection of recombinant human granulocyte colony-stimulating factor(rhG-CSF)primed bone marrow grafts(G-BM)with the quantities of CD34+ cells in the grafts.Methods From June 2004 to July 2007,fifty-three healthy donors were treated with rhG-CSF[5 μg/(kg·d)]injected subcutaneously for five consecutive days.Bone marrow grafts and peripheral blood grafts were harvested on the 4th and 5th day,respectively.The quantities of CD34+ cells and blood routine prior to the collection of the G-BM were determined by flow cytometry and blood analyzer XL2100,respectively.Results The monocyte counts in peripheral blood(PB)of the 53 healthy donors were 896±424 per microliter.The counts of CD34+ cells per microliter and quantities of total CD34+ cells in the bone marrow grafts were 84±45 and(8.22±4.84)×107,respectively.Pearson correlation analysis indicated that the counts of monocyte in the PB correlated positively with the counts of CD34+ cells per microliter(r=0.573,P<0.01)and quantities of total CD34+ cells(r=0.586,P<0.01)in the bone marrow grafts.A stepwise linear regression model also showed that the counts of monocyte in the PB correlated positively with the counts of CD34+ cells per microliter(β=0.568,P<0.01)and quantities of total CD34+ cells(β=0.462,P<0.01)in the G-BM.Conclusion The monocyte counts prior to the collection of G-BM may be a simple and practical indicator for harvests of CD34+ cells.
Effect of Recombinant Human Granulocyte Colony-Stimulating Factor on T-Lymphocyte Function and the Mechanism of This Effect

Abstract:Whether recombinant human granulocyte colony-stimulating factor (rhG-CSF) affects lymphocyte function directly or indirectly is controversial. In this study, we found that T-cell proliferation was decreased considerably in response to phytohemagglutinin in donors who received rhG-CSF but was partly restored after monocytes were removed. Intracellular cytokine staining revealed that the interferon gamma-interleukin 4 ratio decreased by 5.97-fold in donor CD4(+) cells after rhG-CSF treatment. No effect of rhG-CSF on ex vivo T-cell function was observed. rhG-CSF indirectly induced significant quantitative and qualitative changes on lymphocytes, including a decrease in T-cell proliferation and type 2 helper T-cell polarization of the cytokine profile. Although monocytes suppressed T-cell proliferation, the suppressive activity induced by the quantitative change in monocyte numbers cannot completely account for the hyporesponsiveness of T-lymphocytes. We believe that there must be another mediating factor. In addition, the numbers and mean fluorescence intensities of CD14(+)CD86(+) cells and CD19(+)CD80(+) cells declined significantly in the peripheral blood after rhG-CSF treatment. Suboptimal amounts of stimulatory signals provided by low expression levels of B7 molecules on antigen-presenting cells (monocytes, B-lymphocytes) may help explain the alteration in T-cell proliferation. In addition, the absolute counts of CD3(+)CD4(-)CD8(-) cells in the peripheral blood were markedly increased and enriched in leukapheresis products following G-CSF treatment. These suppressor cells may contribute to T-cell hyporesponsiveness.
Effects of granulocyte-colony-stimulating factor and granulocyte/macrophage-colony-stimulating factor administration on T cell proliferation and phagocyte cell-surface molecules during hematopoietic reconstitution after autologous peripheral blood progenitor cell transplantation

Abstract:Thirty-four ovarian and breast cancer patients received autologous peripheral blood progenitor cell transplantation after high-dose myeloablative chemotherapy and either granulocyte-colony-stimulating factor (G-CSF) or granulocyte/macrophage-colony-stimulating fictor (GM-CSF) in the immediate post-transplant period. The recovery of T cell functionality was monitored by a three-color flow-cytometric approach using carboxyfluorescein diacetate succinimidyl ester, a probe the fluorescence intensity of which halves at each round of cell replication, in conjunction with CD3 and CD25 monoclonal antibodies. There was no significant difference between the two treatments on days 12, 20, and 40, T cell proliferation always being considerably lower than that of control cultures from healthy donors. At day 80, a significantly higher proportion of mitogen-stimulated T cells from GM-CSF-treated patients expressed interleukin-2 receptor, and a higher proportion of these T cells were actively proliferating. This phenomenon did not reflect any difference in the relative proportion of various lymphocyte subsets (T cells, CD4 and CD8+ T cells, CD45RA+ and CD45RO- T cells, and natural killer cells). At the end of follow-up (1-1.5 years) T cell proliferation had returned to values typically observed in healthy individuals in both groups of patients. Soon after transplantation (day 12), neutrophils from G-CSF-treated patients had a more elevated Fcgamma receptor I density and monocytes from GM-CSF-treated patients had a more elevated Fcgamma receptor II and MHC class II molecules density. The up-modulation of Fcgamma receptor II was maintained until day 40. Thus, administering G-CSF and GM-CSF in the post-transplant period affects T lymphocyte proliferation and phagocyte membrane molecules differently.
In Vivo Induction of T-cell Hyporesponsiveness and Alteration of Immunological Cells of Bone Marrow Grafts Using Granulocyte Colony-Stimulating Factor.

Abstract:Background and Objectives. Granulocyte colony-stimulating factor (6-CSF)-primed bone marrow (G-BM) transplantation is associated with a low incidence of graft-versus-host disease (GVHD) and has been used successfully in patients with human leukocyte antigen (HLA) matched/mismatched donors. This study evaluated the function of T cells and the quantities of immunological cells of G-BM.Design and Methods. Bone marrow was obtained from fifteen donors by aspiration. Lymphocyte proliferation ability, interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) secreted by T cells were determined using a monotetrazolium (MTT) assay and sandwich enzyme-linked immunosorbent assay (ELISA), respectively. T-cell subgroups, dendritic cell (DC) subsets, CD4(+)CD25(+) regulatory T cells and the expression of CD28/CD80/CD86 molecules on monocytes, B and T cells were analyzed using flow cytometry.Results. G-CSF treatment decreased the quantities of IFN-gamma secretion dramatically (p=0.007) and IL-4 moderately (p=0.027), leading to higher ratios of IL-4/IFN-gamma (p=0.004). We confirmed T-cell hyporesponsiveness and lower expression of CD28/CD80/CD86 on monocytes, B and T cells. The absolute values of lymphocytes, T cell subgroups, CD3(+)CD4(-)CD8(-) cells, CD8(+)CD28(-) cells and B cells in bone marrow grafts were similar before and after G-CSF treatment. The number of monocytes per microliter was increased 2.13-fold, while the numbers of CD4(+)CD25(+) regulatory T cells were unchanged. DC2 were preferentially increased.Interpretation and Conclusions. Our results suggest that bone marrow T-cell hyporesponsiveness could be induced and that the increase of monocytes and DC2 and the downregulation of CD28/CD80/CD86 co-stimulatory signals were produced by in vivo use of G-CSF; this may be related to the preferential increase of monocytes and DC2 and the downregulation of CD28/CD80/CD86 co-stimulatory signals.

Abstract:Introduction. Contamination of specimens and overuse of broad spectrum antibiotics contribute to false positives and false negatives, respectively. Therefore, useful and applicable biomarkers of bacteremia are still required.Hypothesis/Gap Statement. IL-6 can be used as a serum biomarker to discriminate among bacterial infections and fungal infections in febrile patients with a bloodstream infection.Aim. We aimed to evaluate the diagnostic efficiency of neutrophil/lymphocyte ratio (NLR), procalcitonin (PCT) and interleukin-6 (IL-6) in discriminating Gram-negative (G-) bacteria from Gram-positive (G+) bacteria and fungi in febrile patients.Methodology. A total of 567 patients with fever were evaluated. Serum levels of IL-6, PCT, NLR and CRP were compared among a G- group (n=188), a G+ group (n=168), a fungal group (n=38) and a culture negative group (n=173). Sensitivity, specificity, Yuden's index and area under the Receiver operating characteristic (ROC) curve (AUC) were obtained to analyse the diagnostic abilities of these biomarkers in discriminating bloodstream infection caused by different pathogens.Results. Serum IL-6 and PCT in the G- group increased significantly when compared with both the G+ group and fungal group (P <0.05). AUC of IL-6 (0.767, 95 % CI:0.725-0.805) is higher than AUC of PCT (0.751, 95 % CI:0.708-0.796) in discriminating the G- group from G+ group. When discriminating the G- group from fungal group, the AUC of IL-6 (0.695, 95 % CI:0.651-0.747) with a cut-off value of 464.3 pg ml-1 was also higher than the AUC of PCT (0.630, 95 % CI:0.585-0.688) with a cut-off value of 0.68 ng ml-1. Additionally, AUC of NLR (0.685, 95 % CI:0.646-0.727) in discriminating the fungal group from G+ group at the cut-off value of 9.03, was higher than AUC of IL-6, PCT and CRP.Conclusion. This study suggests that IL-6 could be used as a serum biomarker to discriminate among bacterial infections and fungal infections in febrile patients with a bloodstream infection. In addition, NLR is valuable to discriminate fungal infections from Gram-positive infections in febrile patients with a bloodstream infection.
Close association between clearance of recombinant human granulocyte colony-stimulating factor (G-CSF) and G-CSF receptor on neutrophils in cancer patients

Abstract:Recombinant human granulocyte colony-stimulating factor (rhG-CSF) is used to counter chemotherapy-induced neutropenia. Our previous study showed an inverse correlation between serum rhG-CSF levels and the number of circulating neutrophils in cancer patients (H. Takatani, H. Soda, M. Fukuda, M. Watanabe, A. Kinoshita, T. Nakamura, and M. Oka, Antimicrob. Agents Chemother. 40:988-991, 1996). The aim of this study was to clarify the relationship between rhG-CSF clearance and G-CSF receptors on circulating neutrophils. In five cancer patients receiving chemotherapy, a bolus dose of rhG-CSF (5 microg/kg) was injected intravenously during defined phases of posttreatment neutropenia and neutrophilia. Serum rhG-CSF levels were measured by a chemiluminescence enzyme immunoassay and analyzed by moment analysis. G-CSF receptors on neutrophils were detected by flow cytometry with biotinylated rhG-CSF. rhG-CSF clearance was significantly higher at neutrophilia than at neutropenia (1,497 +/- 132 versus 995 +/- 266 ml/h; P < 0.01). The percentage of G-CSF receptor-positive neutrophils, reflecting the number of G-CSF receptors per cell, was low at neutropenia without rhG-CSF therapy (44.5% +/- 22.1%) and high at neutrophilia with rhG-CSF therapy (73. 0% +/- 11.4%; P < 0.01). rhG-CSF clearance closely correlated with the percentage of G-CSF receptor-positive neutrophils (r2 = 0.91; P < 0.0001) and neutrophil count (r2 = 0.72; P < 0.005). Our results indicate that, in cancer patients receiving chemotherapy, rhG-CSF increases the number of G-CSF receptors per cell as well as circulating neutrophil counts, resulting in modulation of its own clearance.

Abstract:Introduction: Immune cells and molecules are considered as clinical biomarkers and potential targets for immunotherapy. Analyses of the composition of peripheral blood cells hold promise for providing a basis for diagnosing and prognosis lung cancer. In this study, we assessed correlations between immune cell subset profiles in peripheral blood and disease prognosis in patients with lung cancer. Methods: One hundred and thirteen patients with lung cancer and 99 age-matched healthy people were enrolled in this study. The percentage and cell count of monocytes, neutrophils, T cells, B cells, natural killer (NK), and NKT cells in peripheral blood were analyzed by flow cytometry or peripheral blood analyzer. Serum cytokines and colony-stimulating factors were detected by enzyme-linked immunosorbent assay (ELISA). Results: A reduction in antitumor NK cells (p < 0.0001) and an increase in the protumor MDSCs (p < 0.0001) were observed in the lung cancer patients compared with the controls. Monocyte counts were significantly higher in lung cancer patients with histories of smoking (p < 0.05) or drinking (p < 0.01) than in patients with no relevant history or healthy controls. The number of neutrophils and the neutrophil-to-lymphocyte ratio (NLR) were particularly higher in patients with liver metastasis (p < 0.01) compared with no metastasis patients or healthy controls. Levels of the monocyte-derived cytokine interleukin-6 (p < 0.05), granulocyte colony-stimulating factor (G-CSF) (p < 0.0001), and granulocyte-macrophage colony-stimulating factor (GM-CSF) (p < 0.0001) were higher in patients than in controls. G-CSF levels decreased during the remission phase (p < 0.05), and positively correlated with carbohydrate antigen 19-9 (p < 0.05) and gene mutation (p < 0.05). Conclusion: Monocyte and neutrophil counts were higher in peripheral blood in lung cancer patients than in controls, especially when patients had histories of smoking, drinking, and liver metastasis. Serum levels of G-CSF and GM-CSF were higher in lung cancer patients, and G-CSF levels positively correlated with disease severity.

Abstract:Our preliminary results suggest the existence of quantitative and qualitative differences in immune cells and type1 and type2 cytokines between granulocyte colony-stimulating factor (G-CSF) primed bone marrow (G-BM) and G-CSF-mobilized peripheral blood grafts (G-PB). Our findings suggest that lower T-cell hyporesponsiveness and easier polarization of T cells from Th1 to Th2 are found in G-BM.

Abstract:Objective To observe the clinical effect of low dose rhG-CSF(recombinant human granuloxyte colony-stimulating factor injection) in treating leukopenia of the entity tumor patients caused by the chemical treatment.Methods 54 patients were randomly divided into two groups.31 cases of the therapeutic group were treated with low dose rhG-CSF 100μg per day,While 23 cases of the control group were treated with routine treatment.Results The quantity of the white blood cells of the patients in the therapeutic group increased within 48 hours.And after five days the total quantity of their white blood cells was significantly different from that of the control group.Moreover the recovery time of the white blood cells of the therapeutic group was also obviously less than that of the control group.Conclusion In treating leukopenia of the entity tumor patients caused by chemical treatment,the way of using rhG-CSF to increase white blood cells is obviously effective,safe,ecomical and practical.
Combined measurement of growth and differentiation in suspension cultures of purified human CD34-positive cells enables a detailed analysis of myelopoiesis

Abstract:In this study we have made a detailed analysis of growth factor (granulocyte-macrophage colony-stimulating factor [GM-CSF], granulocyte colony-stimulating factor [G-CSF], and macrophage colony-stimulating factor [M-CSF])-induced proliferation and differentiation of highly purified CD34+ committed human myeloid progenitor cells in suspension cultures. The results were compared with colony formation in semisolid medium. Proliferation in suspension cultures was determined by means of incorporation of [3H]thymidine, differentiation by flow cytometric immunophenotyping using a panel of monoclonal antibodies against monomyeloid antigens, and by morphology. A good correlation was found between the number of granulocyte-macrophage colony-forming units (CFU-GM) in semisolid medium and [3H]thymidine incorporation in suspension (r = 0.82), both assessed at day 11. Moreover, the frequency of proliferating cells as determined in suspension cultures by limiting dilution analysis was similar to the frequencies of CFU-GM as measured in semisolid medium. Studies on GM-CSF- and G-CSF-induced cell-growth kinetics revealed distinct proliferation patterns. Immunophenotypically the subsequent induction of the mature granulocytic antigens CD15 and CD67 was observed to be accompanied by a gradual loss of the HLA-DR antigen, whereas little monocytic differentiation was observed. M-CSF, although inducing no colony formation of CD34+ cells and minimal proliferation in suspension, induced monocytic differentiation, demonstrated by the expression of HLA-DR, CD14, and CD36 in the absence of CD15 and CD67. The observed immunophenotypical profiles were confirmed by the results of cytological characterization. Thus, the combined measurement of growth factor-induced proliferation and differentiation of progenitor cells in suspension cultures can be a useful alternative for the CFU-GM assay. Moreover, because small numbers of cells are required, it allows for detailed studies on cell-growth kinetics and developmental stages within the granulocytic and monocytic lineages.
