Effect of Growth Hormone on IGF-I/EGFBP Mrna in Healing of Thermal Wound

Abstract:Objective To investigate the effects of recombinant human growth hormone (rhGH) on insulin like growth factor I (IGF I )/insulin like growth factor binding protein 3 (IGFBP 3) mRNA of hepatic and wound tissue in the process of enhancing thermal wound healing.Methods Deep partial thickness burn was produced in SD rats. The 0.3 U rhGH was administered subcutaneously at 9 A.M. every day (rhGH group) for 10 days. RT PCR was used to detect the expression of IGF I and IGFBP 3 mRNA in hepatic and wound tissue. The serum concentrations of IGF I and IGFBP 3 were assayed by ELISA method. The expression of 1,Ⅲ type collagen was assessed by immunohistochemical stains.Results The expression of IGF I and IGFBP 3 mRNA was increased significantly after rhGH administration. The serum concentrations of IGF I and IGFBP 3 were obviously higher than those before rhGH administration and those in the control group. The expression of I,Ⅲ type collagen was enhanced. The mean healing time of burned wound was 3 days less in rhGH group.Conclusion The application of rhGH in burn rat could increase the expression of IGF I and IGFBP 3 in liver and wound tissue to promote the thermal wound healing.
