Bisphenol a (BPA) induces abnormal androgen accumulation via androgen-androgen receptor (AR) mdiated cyp19a1 transcription inhibition in granulosa cells (GCs)

Abstract:To investigate the level of BPA in intrafollicular microenvironment -follicular fluid (FF), the effects of BPA on GCs and the mechanisms how BPA represses transcription of CYP19A1 in GCs. Detect the level of BPA and testosterone (T) in the FF from Polycystic ovarian syndrome (PCOS) and control patients, examine the function of GCs stimulated by BPA, and deliberate the transcriptional regulation of BPA on CYP19A1. The FF were obtained from 17 PCOS (Rotterdam Consensus) and 17 infertile women with male factor(control). BPA and T were measured by liquid chromatography/mass spectrometry and ELISA, respectively. The expression and activity of CYP19A1 in GCs were analysed by real-time PCR and ELISA. The transcriptional activity of BPA on AR was illustrated by luciferase and ChIP assay. [1]BPA and T levels were significantly higher in PCOS patients’ FF compared with controls (3.27±0.38 vs. 2.10±0.14 ng/ml and 63.24±4.14 vs. 46.04±2.23 ng/ml, P<0.01); there was a correlation between T and BPA (r=0.4184, P<0.05); BPA levels were negatively associated with fertilization rate of IVF cycle. [2]BPA dose-dependently downregulated the expression and activity of CYP19A1. [3]By using luciferase, BPA alone had little effect on AR transcriptional activity, but dose-dependently inhibited T induced transcriptional activity. Verified by ChIP, BPA alone pulled down little more androgen response element (ARE) compared to control, but disturbed T pulling-down ARE in a dose-dependent way. BPA dose-dependently inhibits the expression and activity of CYP19A1 in GCs, thus disturbing conversion of androgen to estrogen. Therefore, higher BPA in FF may be the cause of hyperandrogenism in PCOS patients. The results depicted by luciferase and ChIP assay indicate that BPA inhibited T induced transcription of CYP19A1.
