Abstract:A general and facile one-pot protocol for the preparation of a broad range of alkyl and aryl isothiocyanates has been developed from their corresponding primary amines under aqueous conditions. This synthetic process involves an in situ generation of a dithiocarbamate salt from the amine substrate by reacting with CS(2) followed by elimination to form the isothiocyanate product with cyanuric acid as the desulfurylation reagent. The choice of solvent is of decisive importance for the successful formation of the dithiocarbamate salt particularly for highly electron-deficient substrates. This novel and economical method is suitable for scale-up activities.

Abstract:α‐Amino acid derivatives are key components of the molecules of life. The synthesis of α‐amino carbonyl/carboxyl compounds is a contemporary challenge in organic synthesis. Herein, we report a practical method for the preparation of α‐amino acid derivatives via direct hydrative amination of activated alkynes under mild conditions, relying on sulfinamides as the nitrogen source. Abstract:A convenient synthetic approach for the synthesis of alpha-oxo-acetamidines via copper-catalyzed C(sp(3))-H amidination has been developed. This approach allows the direct amidination of three C(sp(3))-H bonds without cyclization.Methyl ketones and primary and secondary amines are tolerant and afford the corresponding products in moderate to good yields. Abstract:Modulating the basicity of alkylamines is a crucial factor in drug design. Consequently, alkylamines with a proximal amide, ester, or ketone have become privileged features in many pharmaceutical candidates. The impact of α-amino carbonyls has made the development of new methods for their preparation a continuous challenge in synthesis. Here, we describe a practical strategy that provides a modular and programmable synthesis of a wide range of α-amino carbonyls. The generality of this process is made possible by an extremely mild method to generate carbamoyl radicals, proceeding via a Lewis acid-visible-light-mediated Norrish type-I fragmentation of a tailored carboxamide reagent and intercepted through addition to in situ generated unbiased imines. Aside from the reaction's broad scope in each component, its capacity to draw on plentiful and diversely populated amine and carbonyl feedstocks is showcased through a two-dimensional array synthesis that is used to construct a library of novel, assay-ready, α-amino amides.

This strategy enables the racemization-free cross-coupling of serine/homoserine- derived iodides with aryl/acyl/alkyl halides. Abstract:The synthesis of amides is a long-term theme in chemistry and related areas. The use of readily available and cost-effective precursors for straightforward amide synthesis with atom economy under mild conditions is attractive yet challenging. Herein, we report the direct synthesis of amides from aldehydes and imines in a 100% atom-economical manner. The redox-neutral C–N bond-forming process was enabled by the dual catalysis of visible light and an N-heterocyclic carbene (NHC) at room temperature. This protocol features the unprecedented umpolung of imines to generate N-centered radicals. Mechanistic investigations reveal this reaction undergoes nontrivial radical–radical coupling between an N-centered radical and a C-centered radical to forge a C–N bond. Abstract:An efficient and general protocol for copper-catalyzed N-arylation of aliphatic amines and amino acids has been developed using aryl iodides under mild conditions (coupling temperature at 25-35 degrees C). For the N-(o-nitrophenyl) amino acid derivatives, subsequent reduction of the nitro group in the presence of tin(II) chloride resulted in 3,4-dihydroquinoxalin-2(1H)-one derivatives in good yields.

Abstract:We describe a new strategy for aminoarylation of alkenes by copper-catalyzed smiles rearrangement using O-benzoylhydroxylamines as the amine reagent. This method affords various β-amino amide derivatives possessing a quaternary carbon center with wide functional group tolerance and high regioselectivity. The mechanistic studies indicate that the transformation can involve aminyl radical intermediates under acid-free condition.

Abstract:Despite the remarkable developments of the Ugi reaction and its variants, the use of ammonia in the Ugi reaction has long been recognized as impractical and unsuccessful. Indeed, the ammonia-Ugi reaction often requires harsh reaction conditions, such as heating and microwave irradiation, and competes with the Passerini reaction, thereby resulting in low yields. This study describes a robust and practical ammonia-Ugi reaction protocol. Using originally prepared ammonium carboxylates in trifluoroethanol, the ammonia-Ugi reaction proceeded at room temperature in high yields and showed a broad substrate scope, thus synthesizing a variety of α,α-disubstituted amino acid derivatives, including unnatural dipeptides. The reaction required no condensing agents and proceeded without racemization of the chiral stereocenter of α-amino acids. Furthermore, using this protocol, we quickly synthesized a novel dipeptide, D-Leu-Aic-NH-CH2Ph(p-F), which exhibited a potent inhibitory activity against α-chymotrypsin with a Ki value of 0.091 μM.

Abstract:Ugi's amine has become one type of privileged chiral skeleton for chiral ligand design bearing central/planar chirality, and such ligands have exhibited tremendous success in various asymmetric catalysis. However, the current access to enantiopure Ugi's amine is quite tedious and relies heavily on optical resolution, which impedes its practical applications, to some extent. Herein, we present a facile asymmetric synthesis of enantioenriched Ugi-type amines bearing a long-carbon chain through Ir-catalyzed cascade allylation/2-aza-Cope rearrangement, followed by amino exchange and Pd/C-catalyzed one-pot hydrogenation/reductive amination. The protocol could be readily scaled up, and it has been conducted in 20-g-scale asymmetric synthesis of (S)-Ugi-type amine from commercially available reagents, in >99% ee and >70% overall yield in four steps with one short silica gel-plug purification. Abstract:Transformations enabling the synthesis of α-alkyl, α'-2-azinyl amines by addition of 2-heteroaryl-based nucleophiles to in situ-generated and non-activated alkyl-substituted iminium ions are extremely rare. Approaches involving classical 2-azinyl organometallics, such as the corresponding Grignard reagents, often fail to produce the desired products. Here, we report an operationally straightforward solution to this problem through the development of a multicomponent coupling process wherein a soft 2-azinyl indium nucleophile, generated in situ from the corresponding 2-iodo heteroarene and indium powder, adds to an iminium ion that is also formed directly in the reaction. This modular carbonyl azinylative amination (CAzA) displays a broad scope and only a metal reductant is needed to generate a reactive 2-azinyl nucleophile. Beyond the addition to iminium ions, the 2-azinyl addition to polyfluoromethyl ketones forms the corresponding tertiary alcohols. Together, the products of these reactions possess a high degree of functionality, are typically challenging to synthesize by other methods, and contain motifs recognized as privileged in the context of pharmaceuticals and agrochemicals.

Abstract:Cu/Ir dual catalysis has been developed for the stereodivergent α-allylation of aldimine esters. The method enables the preparation of a series of nonproteinogenic α-amino acids (α-AAs) bearing two contiguous stereogenic centers in high yield with excellent stereoselectivity. All four product stereoisomers could be obtained from the same set of starting materials via pairwise combination of two chiral catalysts. Notably, one-pot protocol could be successfully applied for the preparation of the bimetallic Cu/Ir complexes to simplify the manipulation of Cu/Ir dual catalysis. This method could be further utilized for the construction of the key intermediate of a bioactive pyrrolidine derivative and the concise synthesis of a plant growth regulator (2S,3S)-2-amino-3-cyclopropylbutanoic acid.

Abstract:An simple, efficient method for the construction of an array of α-ketoamides has been described from readily available O-benzoyl hydroxylamines and diazo compounds as starting materials by the combined use of CuI as a catalyst and H2O as oxygen source. The mechanism research reveals that the O-benzoyl hydroxylamines serve a dual role as both amine source and oxidant, thereby obviating the need for additional oxidants in the transformation process. This methodology could afford a wider range of products in good to excellent yields for most substrates, and provides a new idea for synthesis of α-ketoamide.

Abstract:In the past decade, the oxidative coupling reaction has become a new method to afford C–C bonds from C–H bonds due to it being atom economical and environmentally friendly. Meanwhile, a-amino acids play an important role in natural products and are the key structural motifs of numerous natural products. Therefore, the oxidative coupling reaction of glycine derivatives has gained signicant attention for the synthesis of a series of a-substituted a-amino acid derivatives. For example, Li’s group designed the rst method for an oxidative dehydrogenative coupling reaction to synthesize a-amino glycine derivatives from N-glycine derivatives by direct C–C bond formation. Subsequently, Huang’s group disclosed a crossdehydrogenative coupling reaction with N-arylglycine esters to synthesize a-amino glycine derivatives under the cooperative catalysis of copper salt. Recently, the oxidative coupling of Nglycine derivatives has been well developed under stoichiometric amounts of chemical oxidants such as DTBP, DDQ, TBHP and 2,2,6,6-tetramethylpiperidine-1-oxoammonium tetrauoroborate (TBF4 ). Furthermore, transition metal-catalyzed synthesis of chiral a-amino acid derivatives by direct C–H oxidative cross-coupling has been reported in the past few years. For example, Wang’s group developed a signicant method of chiral Lewis acid controlled enantioselective alkylation of a-Csp3–H bonds to synthesize chiral alkyl a-amino acid derivatives.
