Cytosolic Triglyceride Storage In Mouse Enterocytes During Dietary Fat Absorption Visualized By Coherent Anti-Stokes Raman Scattering Microscopy

Abstract:Although triglyceride (TG) storage in cytoplasmic lipid droplets (CLDs) in hepatocytes is well accepted, it remains controversial whether enterocytes store TG in CLDs during normal dietary fat absorption. Herein coherent anti‐Stokes Raman scattering (CARS) microscopy was applied to directly visualize TG accumulation in enterocytes based on the resonant CARS signal from symmetric CH 2 stretch vibration. Small intestinal tissues from C57BL/6J mice fed 0, 50, or 300 μL olive oil were examined. CLDs were observed in CARS images. TG storage as a function of digestion time and location were quantified by image‐J analysis and confirmed with a biochemical assay. Combined CARS and two‐photon‐excited fluorescence imaging of individual isolated enterocytes revealed that CLDs are located adjacent to the Golgi complex towards the apical side. In addition, in two mouse models with perturbation of chylomicron secretion, diacylglycerol acyltransferase 1 deficient and PL81‐treated mice, CARS images of enterocytes confirmed increases in CLDs in enterocytes. Taken together, these results demonstrate storage of TGs in CLDs within enterocytes during dietary fat absorption. Further study of the formation, trafficking, and lipolysis of CLDs in enterocytes would deepen the current understanding of dietary fat absorption and facilitate the development of new therapeutic methods for fighting obesity.
