Hyperglycemia Induces Cardiomyocyte Hypertrophy In Part Through Pkc Beta 2 Activation In Cultured Neonatal Rat Myocytes

Abstract:Targeted overexpression of PKCβ2 has been shown to induce myocardial hypertrophy. It is unknown, however, whether PKCβ2 activation is necessary in hyperglycemia induced cardiac myocyte hypertrophy (MH). We hypothesized that PKCβ2 activation plays an important role in hyperglycemia induced MH. Cardiomyocytes isolated from neonatal rat hearts were treated, for 48 hours, either with normal (5.5mmol/L, LG) or high concentration of glucose (25.5mmol/L, HG), HG in the presence of the selective PKCβ2 inhibitor CGP53353 (CGP), or LG in the presence of the PKC activator phorbol-12-myristate-13-acetate (PMA) or PMA plus CGP in the cell culture medium. Myocyte cross-sectional area was measured by immunocytochemistrical analysis. Myocyte protein and DNA content were, respectively, determined by incorporation of [3H]-Leucine and [3H]-Thymine into the cultured myocytes. HG induced MH was manifested by increased myocyte cross-sectional area and increased incorporation of [3H]-Leucine, which was significantly attenuated, but not prevented, by CGP. However, CGP prevented PMA induced hypertrophic changes in cells treated with LG. The results suggest that PKCβ2 activation is sufficient in inducing myocyte hypertrophy, but may not be necessary in hyperglycemia mediated myocyte hypertrophy.
