A Novel Approach To Mobilize Bone Marrow Stem Cells With Cxcr4 Expression For Enhanced Tissue Repair In Rat Cardiac Allograft

Abstract:We tested whether bone marrow stem cells (BMC) overexpressing CXCR4 have higher migration potential. Female rats were lethally irradiated and substituted with male BMC with null/GFP (G1), or BMC overexpressing CXCR4/GFP (G2), or BMC treated with miRNA targeting CXCR4 (G3). After 30 days, donor hearts were transplanted into abdomen of irradiated rats and subjected to LAD occlusion. Following cardiac function was determined; hearts were harvested to analyze CXCR4, Nkx2.5, GATA‐4, GFP and Y‐chromosome positive cells. CXCR4+ cells were mobilized in the circulation in G2. CXCR4 was strongly expressed around the perivascular area in the transplanted hearts in G2. Myocytes and newly formed vessels expressing Y‐chromosome in donor heart were significantly increased in G2. These cells also expressed early cardiac markers in G2. Heart function was significantly improved in G2. Survival of the allografts was markedly increased in G2 (36.8 days) as compared with G1 (19.2 days), G3 (13.5 days). Donor heart function in G3 and upregulation of early cardiac markers were significantly reduced by miRNA. Overexpression of CXCR4 in BMC was extremely effective in mobilizing BMC to the damaged myocardium for myoangiogenesis and improved cardiac function.
