Brain Angiotensin Or Nuclear Factor Kappa B Blockade Attenuates Cytokines, Oxidative Stress And Decreases Sympathoexcitation In Hypertension

Abstract:Inflammatory and oxidative stress induced by angiotensin II (ANG II) contributes to the pathogenesis of hypertension. This study determined the effect of central blockade of angiotensin‐1 receptor (AT1‐R) and cytokine transcriptional factor NFκB on ANG II‐induced inflammatory, oxidative stress and hypertensive response. Rats were infused with ANG II (0.6μg/hr) for 4 weeks. Some rats also received ICV infusion of vehicle or AT1‐R antagonist losartan (LOS, 20μg/hr), or pyrrolidine dithiocarbamate (PDTC, 5μg/hr), an inhibitor of NFκB. ANG II infusion resulted in a significant increase in the mean arterial pressure (MAP), renal sympathetic nerve activity (RSNA), plasma norepinephrine (NE) and cytokines. These animals also showed increased expression of cytokines, AT1‐R and NADPH oxidase gene in the paraventricular nucleus (PVN) of hypothalamus. In contrast, ICV treatment with LOS or PDTC significantly decreased MAP, RSNA, circulating NE and cytokines in ANG II‐induced hypertensive rats. ICV Treatments also reduced the expression of cytokines, AT1‐R and oxidative stress gene in the PVN. These finding suggests that ANG II induces the expression of cytokines and oxidative stress in the PVN. The interaction between angiotensin and NFκB in the PVN contribute to the progression of ANG II‐induced hypertensive response.
