Nedd4l Null Mice Are Defective in Down‐regulating ENaC and Have Salt‐sensitive Hypertension

Abstract:Nedd4l has been proposed to play a critical role in regulating ENaC and thus Na homeostasis and blood pressure. The interaction between Sgk1 and Nedd4l connects the mineralocorticoid aldosterone to Na+ reabsorption and completes this important regulatory circuit. In the present study we generated Nedd4l KO mice and tested the in vivo role of Nedd4l on ENaC regulation. On a normal salt diet Nedd4l −/− mice had significantly reduced urine Na+ excretion and higher BP than Nedd4l +/+mice. High salt diet for two weeks further increased the BP for Nedd4l −/−mice, while Nedd4l +/+mice showed no significant increase. Daily urine Na+ excretion was significantly less for Nedd4l −/− than Nedd4l +/+ mice on high salt diet, indicating Na+ retention. Rectal PD, an indirect assessment of ENaC function in the distal nephron, was not significant different between those two groups of mice on a normal diet. However, from an elevated PD level induced by low salt diet, Nedd4l +/+ mice showed a more than 280-fold reduction of PD after two weeks high salt diet while Nedd4l −/− mice can only manage a 6-fold reduction during the same period of time. Our results indicate that Nedd4l −/− mice have an impaired ability to down-regulate ENaC activity in response to dietary changes and confirm the hypothesis that ENaC function is negatively regulated by Nedd4l.
