Abstract:Regioselectivity of lipases played a crucial role in structured lipids synthesis and oil modification.Because of differences between aqueous and non-aqueous systems,regioselectivity analyzed by the conventional hydrolysis method might not always match that in synthesis reactions.In this paper,acidolysis of lauric acid and camellia oil was developed to directly evaluate regioselectivity of lipases in solvent-free system.Fatty acid composition and positional distribution of reaction products were detected by gas chromatography.Through this protocol,Lipozyme RM IM,L02,L03 and L04 were identified as sn-1 (3) regioselectivity,L01 was partially selective,and Novozym 435 was nearly non-selective.Predictability of the model reaction had been verified by replacing substrates of lipases.According to the comparative analysis of results of the acidolysis method and hydrolysis one,regioselectivity of lipases under two situations was generally the same,except selectivity of Novozym 435 which was susceptible to solvent systems.Therefore the present method avoided limits of hydrolysis determination results in synthesis reactions.And moreover it lowered the requirement on purity of reactants,which means it can be used to screen numerous lipases at low cost.
Screening and Identification of High-yield Thermostable Lipase Producing Microorganisms

Abstract:Two strains of microorganisms(J2 and J3)with a high-yield lipase producing ability had been screened from dumplings’ steamer cushions, and identified as two variants ofAureobasidium sp. according to morphological observation and 26S rRNA gene(26S rDNA)sequencing. After being incubated at 30℃, 200 r/min for 3-5 d, the lipase activities of their fermentation supernatant were 10.61 U/mL and 14.43 U/mL, respectively, measured by spectrophotometry with p-nitrophenol palmitate(p-NPP)as a substrate. The investigation of physicochemical properties of the produced lipases(Lip2 and Lip3)showed that the optimum catalyzing temperature of Lip2 is 50℃ and there is no significant activity loss after being incubated at 50℃ for 5 h, while the optimum catalyzing temperature of Lip3 is 60℃ and there is no activity loss after being incubated at 40℃ for 5 h, but only 42.19% of lipase activity remained when incubated at 50℃ for 5 h. The results indicated that the lipases produced by strain J2 and J3 have good thermal stability and high optimal reaction temperature.
Extraction of Candida Rugosa Lipase from Aqueous Solutions into Organic Solvents by Forming an Ion‐paired Complex with AOT

Abstract:Candida rugosa lipase was extracted from aqueous solutions into organic solvents by forming an ion-paired complex with sodium bis(2-ethylhexyl)sulfosuccinate (AOT). The optimal aqueous pH for lipase recovery was 4.5 and the optimal CaCl2 concentration was 10 mmol dm(-3). The lipase recovery decreased with increasing aqueous enzyme concentration but increased with increasing AOT concentration in the organic phase. The presence of polar co-solvents in the aqueous phase did not obviously improve the lipase recovery, which was also little influenced by the type of hydrophobic organic solvent used for solubilising AOT. Surprisingly, no detectable activity of the ion-paired C. rugosa lipase was observed for both the esterification of lauric acid with 1-propanol in isooctane and the hydrolysis of olive oil in isooctane containing an appropriate amount of water. The ion-paired C. rugosa lipase mediated the enantioselective crystallisation of racemic ketoprofen in isooctane, indicating the feasibility of using it as a chiral mediator for the enantioseparation of hydrophobic racemic compounds in organic systems. Abstract:The industrial application of lipases is strictly limited by their properties.And thermostable lipases are favorable biocatalyst due to their wide requirement in food industry,lipid processing and bio-energy.Combined with the enrichment of lipase producing microorganism in lipid-rich environment and directed selection of thermostable lipase in condition of high temperature and moisture,the steamers of Chinese dumpling are considered and chosen as an good resource for screening of lipase producing strains.The strain identification of screened lipase producing bacteria were performed by fatty acid profiling using Gas chromatography and MIS standard systems,assisted with morphological,physiological and enzymatic characteristics.67 strains were isolated,and 21 strains among of them,according to their olive oil hydrolysis activity in the screening medium plates,were identified to produce extracellular lipase including 7 strains of filamentous fungi,6 strains of yeast and 8 strains of bacteria.Strain identification of bacteria has revealed that these 8 strains are classified to Acinetobacter,Pseudomonas,Bacillus and Staphylococcus 4 genera and 5species.In this study,it is proved that Chinese dumpling's steamers are abundant in food grade safe microbiological resource,and this research has provided some basis for exploration of novel thermostable lipase.

Abstract:Organic solvent-tolerant lipases have shown new potential application in biotechnological fields. A novel strain which produced an organic solvent-tolerant lipase was isolated from soils in different areas of China. Based on the morphological, physiological and biochemical features and 16S rDNA sequence of the strain, it was identified asPseudomonas aeruginosaCS-2. The crude lipase had an improved stability in the presence of acetonitrile and it exhibited stability in presence of benzene, chloroform,n-Hexane, petroleum ether and isooctane. The optimized composition of culture medium was as follows: olive oil 1%, peptone 4%, K2HPO40.5%, MgSO4·7H2O 0.1%, gum arabic 0.04%. The optimum condition for lipase production was on the following: temperature 37°C, initial pH 7.5, rotary shaking speed 200 rev/min, culture time 48 h.

Abstract:Lipases are a group of enzymes naturally endowed with the property of performing reactions in aqueous as well as organic solvents. The esterification reactions using lipase(s) could be performed in water-restricted organic media as organic solvent(s) not only improve(s) the solubility of substrate and reactant in reaction mixture but also permit(s) the reaction in the reverse direction, and often it is easy to recover the product in organic phase in two-phase equilibrium systems. The use of organic solvent tolerant lipase in organic media has exhibited many advantages: increased activity and stability, regiospecificity and stereoselectivity, higher solubility of substrate, ease of products recovery, and ability to shift the reaction equilibrium toward synthetic direction. Therefore the search for organic solvent tolerant enzymes has been an extensive area of research. A variety of fatty acid esters are now being produced commercially using immobilized lipase in nonaqueous solvents. This review describes the organic tolerance and industrial application of lipases. The main emphasis is to study the nature of organic solvent tolerant lipases. Also, the potential industrial applications that make lipases the biocatalysts of choice for the present and future have been presented.

Abstract:We found a novel lipase gene in the Paenibacillus pasadenensis CS0611 strain. The lipase gene sequence was cloned into the pET-28a expression vector to construct a recombinant lipase protein containing 6 × His tags at the C- and N-termini, respectively. High-level expression of the lipase in E.coli BL21 (DE3) was obtained upon induction with IPTG at 20 °C. The recombinant lipase activity was approximately 1631-fold higher than the wild type. His-tagged recombinant lipase was purified rapidly and efficiently by using Ni-charged affinity chromatography with 63.5% recovery and a purification factor of 10.78. The purified lipase was stable in a broad range of temperatures and pH values, with the optimal temperature and pH being 50 °C and 7.0, respectively. Its activity was stimulated to different degrees in the presence of metal ions such as Ca2+, Mg2+, and some non-ionic surfactants. In addition, the purified lipase was activated by a series of water-miscible organic solvents such as some short carbon chain alcohols and was highly tolerant to some water-immiscible organic solvents.
Optimization of thermostable organic solvent-tolerant lipase production by thermotolerant Rhizopus sp. using solid-state fermentation of palm kernel cake

Abstract:This study enhanced the production of thermostable organic solvent-tolerant (TS-OST) lipase by locally isolated thermotolerant Rhizopus sp. strain using solid-state fermentation (SSF) of palm kernel cake (PKC). The optimum conditions were achieved using a series of statistical approaches. The cultivation parameters, which include fermentation time, moisture content, temperature, pH, inoculum size, various carbon and nitrogen sources, as well as other supplements, were initially screened by the definitive screening design, and one-factor-at-a-time using PKC as the basal medium. Three significant factors (olive oil concentration, pH, and inoculum size) were further optimized using face-centred central composite design. The results indicated a successful and significant improvement of lipase activity by almost two-fold compared to the initial screening production. The findings showed that the optimal conditions were 2% (v/w) inoculum size, 2% (v/w) olive oil, 0.6% (w/w) peptone, 2% (v/w) ethanol, 70% moisture content at initial pH 10.0 and 45 °C within 72 h of fermentation. Process optimization resulted in maximum lipase activity of 58.63 U/gram dry solids (gds). The analysis of variance showed that the statistical model was significant (p value <0.0001) and reliable with a high value of R2 (0.98) and adjusted R2 (0.96). This indicates a better correlation between the actual and predicted responses of lipase production. By considering this study, the low-cost PKC through SSF appears to be promising in the utilization of agro-industrial waste for TS-OST lipase production. This is because satisfactory enzyme activity could be attained that promises industrial applications.

Abstract:Organic solvent-tolerant lipase-producing microorganisms were isolated from petrol spilled soil. From ten morphologically distinct lipase-producing bacterial isolates, highest amount of lipase-producing isolate UBT1 was identified as <i>Acinetobacter</i> sp. using 16S rRNA gene sequencing (NCBI Accession No: MH879815). An increase in lipase production from 42 U/mL to 243 U/mL was obtained when different deoiled seed cakes were supplemented instead of olive oil in the medium. Further optimization of media components by the statistical approach assisted in discerning the main influencing media components and their optimum concentrations. Nine components glucose, castor seedcake, potassium nitrate, gum arabic, calcium chloride, magnesium sulphate, potassium di-hydrogen phosphate, dipotassium hydrogen phosphate, and ferric chloride were selected for Plackett–Burman design. The optimum concentrations of three significant selected components for the lipase production were found to be 0.025 gm% glucose, 0.002 gm% calcium chloride, and 0.2 gm% potassium di-hydrogen phosphate as determined by Response Surface Methodology. Increase in lipase production with 292.29 U/mL was achieved in the media containing optimized components and 2 gm% deoiled castor seed cake. Purification studies with ammonium sulphate precipitation, dialysis, and gel permeation chromatography resulted in 77.54% recovery with 5.77-fold partially purified lipase. The residual activity of lipase in 50 and 75% concentration of n-hexane among other solvents after 24 h was 105.05 and 90.42%, respectively, indicating its solvent tolerance. The present study reports the isolation of organic solvent-tolerant lipase-producing <i>Acinetobacter</i> sp. UBT1, optimization of the culture media for lipase production using the deoiled castor seed cake, and its partial purification.

Abstract:The study illustrated here aims on an organic solvent tolerant lipase from Staphylococcus capitis (SCL). The gene part, encoding the mature lipase, was cloned and sequenced. The concluded polypeptide sequence, equivalent to the protein, consist of 388 amino acid residues with a molecular mass of about 45 kDa. A structure-based alignment of the SCL amino acid sequence shows high identities with those many staphylococcal lipases. From this alignment of sequences, the catalytic triad (Ser 117, Asp 308 and His 347) of SCL could be identified. The mature part of the SCL was expressed in Escherichia coli and the recombinant lipase (r-SCL) was purified to homogeneity. The purified r-SCL presented a quite interesting stability at low temperatures (< 30 °C) and the enzyme was found to be highly stable in polar organic solvent and at a pH ranging from 3 to 12. After that, we have demonstrated that the recombinant enzyme may be implicated in the biodegradability of oily wastewater from effluents of fast-food restaurants; the maximum conversion yield into fatty acids obtained at 30 °C, was 65%.

Abstract:Microorganisms are important sources of lipolytic enzymes with characteristics for wide promising usages in the specific industrial biotechnology. The cellulolytic myxobacterium Sorangium cellulosum is rich of lipolytic enzymes in the genome, but little has been investigated. Here, we discerned 406 potential lipolytic enzymes in 13 sequenced S. cellulosum genomes. These lipolytic enzymes belonged to 12 families, and most are novel with low identities (14–37%) to those reported. We characterized a new carboxylesterase, LipB, from the alkaline-adaptive So0157-2. This enzyme, belonging to family VIII, hydrolyzed glyceryl tributyrate and p -nitrophenyl esters with short chain fatty acids (≤C12), and exhibited the highest activity against p -nitrophenyl butyrate. It retained over 50% of the activities in a broad temperature range (from 20°C to 60°C), alkaline conditions (pH 8.0–9.5), and the enzymatic activity was stable with methanol, ethanol and isopropanol, and stimulated significantly in the presence of 5 mM Ni 2+ . LipB also exhibited β -lactamase activity on nitrocefin, but not ampicillin, cefotaxime and imipenem. The bioinformatic analysis and specific enzymatic characteristics indicate that S. cellulosum is a promising resource to explore lipolytic enzymes for industrial adaptations.

Abstract:Several methods including microwave, Frenchpress, autoclave, bead-beating, ultrasonication, and osmotic shock were compared to identify the most effective microalgal cell disruption method. Botryococcus braunii LB572 was cultured in 5 L flasks containing JM medium mixed with oceanic sediment extract for 13 days. Among the methods tested, enhanced lipid extraction was achieved through microwave treatment (2450MHz, 1250W at 150°C for 20 min). Oleic (C18:1), linolenic (C18:3), and palmitic acids (C16:0) were found to be the major fatty acids among the C14-C24 acids from extracted lipid. In addition, the optimal conditions of transesterification were as follows: 70 mL of methanol, 6 mL of sulfuric acid, 8 mL of chloroform, and boiling at 100°C for 30 min; 85.4% of C14-C24 FAME and 78.5% of C16-C18 FAME were esterified from transesterifiable lipids.

Screening and Identification of Strains for Producing Cheap Oil Favorite Lipase.

Abstract:Nine strains with transesterification activity were isolated from 96 lipase-producing strains,which have been isolated by previous research in our laboratory.Analysis of the efficiency of transesterification in converting glycerol trioleate into biodiesel through gas chromatography showed that the strains F8 and B10 exhibited higher transesterification activity than other strains,the conversions of methyl ester were 48.64%and 56.54%respectively.Analysis of the substrate preference of lipases from these two strains with different oils as substrates showed that the strain B10 has the highest transesterification activity in converting food waste oils and Jatropha curcas L.seed oil into biodiesel,the efficiency of transesterification reached 62.35%and 58.60%,respectively;the strain F8 has the efficiency of transesterification of 26.82%in converting Jatropha curcas L.seed oil into biodiesel.The biolog and 16S rDNA sequence analysis showed that both of B10 and F8 were belonged to Pseudomonas aeruginosa.
Identification and Characterization of Novel Esterases from a Deep-Sea Sediment Metagenome

Abstract:A deep-sea sediment metagenomic library was constructed and screened for lipolytic enzymes by activity-based approach. Nine novel lipolytic enzymes were identified, and the amino acid sequences shared 56% to 84% identity to other lipolytic enzymes in the database. Phylogenetic analysis showed that these enzymes belonged to family IV lipolytic enzymes. One of the lipolytic enzymes, Est6, was successfully cloned and expressed in Escherichia coli Rosetta in a soluble form. The recombinant protein was purified by Ni-nitrilotriacetic affinity chromatography column and characterized using p-nitrophenyl esters with various chain lengths. The est6 gene consisted of 909 bp that encoded 302 amino acid residues. Est6 was most similar to a lipolytic enzyme from uncultured bacterium (ACL67845, 61% identity) isolated from the South China Sea marine sediment metagenome. The characterization of Est6 revealed that it was a cold-active esterase and exhibited the highest activity toward p-nitrophenyl butyrate (C4) at 20°C and pH 7.5.

Abstract:Organic solvent-stable lipases have pronounced impact on industrial economy as they are involved in synthesis by esterification, interesterification, and transesterification. However, very few of such natural lipases have been isolated till date. A study of the recent past provided few pillars to rely on for this work. The three-dimensional structure, inclusive of the surface and active site, of 29 organic solvent-stable lipases was analyzed by subfamily classification and protein solvent molecular docking based on fast Fourier transform correlation approach. The observations revealed that organic solvent stability of lipases is their intrinsic property and unique with respect to each lipase. In this paper, factors like surface distribution of charged, hydrophobic, and neutral residues, interaction of solvents with catalytically immutable residues, and residues interacting with essential water molecules required for lipase activity, synergistically and by mutualism contribute to render a stable lipase organic solvent. The propensity of surface charge in relation to stability in organic solvents by establishing repulsive forces to exclude solvent molecules from interacting with the surface and prohibiting the same from gaining entry to the protein core, thus stabilizing the active conformation, is a new finding. It was also interesting to note that lipases having equivalent surface-exposed positive and negative residues were stable in a wide range of organic solvents, irrespective of their LogP values.

Abstract:The objective of the present study was the isolation, molecular cloning and biochemical characterization of a thermophilic organic solvent-resistant lipase from Bacillus sp. DR90. The lipase gene was expressed in Escherichia coli BL21(DE3) using pET-28a(+) vector. The purification of recombinant lipase was conducted by nickel affinity chromatography and its biochemical properties were determined. The lipase sequence with an ORF of 639 bp contains the conserved pentapeptide Ala-His-Ser-Met-Gly. His-tagged recombinant lipase had a specific activity of 1,126 U/mg with a molecular mass of 26.8 kDa. The cloned lipase was optimally active at pH 8.0 and 75 °C representing high stability in broad ranges of temperature and pH. High performance liquid chromatography was used to determine the major compounds released during the lipase-catalyzed reaction of p-nitrophenyl derivatives as well as the substrate specificity. The purified lipase showed high compatibility towards various organic solvents, surfactants and commercial solid/liquid detergents; therefore the recombinant DR90 lipase could be considered as a probable candidate for future applications, predominantly in detergent processing industries.
Polar Substitutions on the Surface of a Lipase Substantially Improve Tolerance in Organic Solvents

Abstract:Biocatalysis in organic solvents (OSs) enables more efficient routes to the synthesis of various valuable chemicals. However, OSs often reduce enzymatic activity, which limits the use of enzymes in OSs. Herein, we report a comprehensive understanding of interactions between surface polar substitutions and DMSO by integrating molecular dynamics (MD) simulations of 45 variants from Bacillus subtilis lipase A (BSLA) and substitution landscape into a "BSLA-SSM" library. By systematically analyzing 39 structural-, solvation-, and interaction energy-based observables, we discovered that hydration shell maintenance, DMSO reduction, and decreased local flexibility simultaneously govern the stability of polar variants in OS. Moreover, the fingerprints of 1631 polar-related variants in three OSs demonstrated that substituting aromatic to polar amino acid(s) hold great potential to highly improve OSs resistance. Hence, surface polar engineering is a powerful strategy to generate OS-tolerant lipases and other enzymes, thereby adapting the catalyst to the desired reaction and process with OSs.

A New Cold-Adapted, Organic Solvent Stable Lipase from Mesophilic Staphylococcus epidermidis AT2

Abstract:The gene encoding a cold-adapted, organic solvent stable lipase from a local soil-isolate, mesophilic Staphylococcus epidermidis AT2 was expressed in a prokaryotic system. A two-step purification of AT2 lipase was achieved using butyl sepharose and DEAE sepharose column chromatography. The final recovery and purification fold were 47.09 % and 3.45, respectively. The molecular mass of the purified lipase was estimated to be 43 kDa. AT2 lipase was found to be optimally active at pH 8 and stable at pH 6–9. Interestingly, this enzyme demonstrated remarkable stability at cold temperature (<30 °C) and exhibited optimal activity at a temperature of 25 °C. A significant enhancement of the lipolytic activity was observed in the presence of Ca2+, Tween 60 and Tween 80. Phenylmethylsulfonylfluoride, a well known serine inhibitor did not cause complete inhibition of the enzymatic activity. AT2 lipase exhibited excellent preferences towards long chain triglycerides and natural oils. The lipolytic activity was stimulated by dimethylsulfoxide and diethyl ether, while more than 50 % of its activity was retained in methanol, ethanol, acetone, toluene, and n-hexane. Taken together, AT2 lipase revealed highly attractive biochemical properties especially because of its stability at low temperature and in organic solvents.
