Abstract:Nine known compounds, 5-hydroxy-7-methoxy-2-methyl-4-chromanone (1), 5, 7-dihydroxy-2-methyl-4-chromanone (2), 5-hydroxyl-2-methyl-4-chromanone (3), 1-(2, 6-dihydroxyphenyl)-3-hydroxybutanone(4), 5alpha, 8alpha-epidioxyergosta-6, 22-dien-3beta-ol (5), (22E,24R)-ergost-7, 22-dien-3beta, 5alpha, 6alpha-triol (6), euphorbol (7), beta-sitosterol (8) and 2-(4-hydroxyphenyl) -ethanol (9) were isolated and identified in Xylaria nigripes for the first time, and their structures were mainly determined by MS and NMR methods. Compounds 1 and 2 were isolated as the natural product for the first time.

Abstract:Totally 21 compounds were isolated from the EtOAc extract of fermentation broth of fungus Xylaria polymorpha (Pers.: Fr.) Grer, including two new drimane-type sesquiterpenoids named polymorphines A and B (1 similar to 2) and one new phenyloxolane compound named 2-methyl-2-(4-hydroxymethylphenyl) oxolane (3), together with 18 known compounds 4 similar to 21. The structures of these compounds were elucidated by NMR and single-crystal X-ray diffraction analysis. Compound 2 exhibited anti-acetylcholinesterase and alpha-glucosidase inhibitory activities. Moreover, compound 3 showed moderate inhibitory activitiy against the nematode Panagrellus redivivus with mortality ratio of 59.6% at 2.5 mg/mL.

Abstract:Chemical investigation of a co-culture of two endophytic fungi Aspergillus fumigatus D and Fusarium oxysporum R1 from two traditional medicinal plants, Edgeworthia chrysantha Lindl. and Rumex madaio Makino, led to isolation of a new amide 1 and six known compounds, including neovasinin (2), neovasifuranone B (3), N-(2-phenylethyl)acetamide (4), α-linolenic acid (5), α-elaeostearic acid (6), palmitoleic acid (7). On the basis of extensive spectroscopic analysis including 1D and 2D NMR, HR-ESI-MS and optical rotation measurement as well as comparison of literature data, chemical structure of 1 was unambiguous elucidated as (S, E)-methyl-2-(2,4-dimethylhex-2-enamido)acetate. Bioassay results indicated that none of these compounds exhibited strong inhibitory effect on three human pathogens Escherichia coli, Staphyloccocus aureus and Candida albicans with MIC values of ≥25 μM.

Abstract:A new diphenyl ether (1) as well as 20 other compounds were identified from the fermentative extracts of marine-derived fungi Phoma sp. CZD-F11 (Compounds 1–8) and Aspergillus sp. CZD-F18(Compounds 9–21). Their structures were elucidated on the basis of extensive spectroscopic analysis. The broth extracts of the fungi exhibited very good anticancer activity against H1975 cells with 5.62 and 25.8% viability at concentration of 10 μg/mL for Phoma sp. CZD-F11 and Aspergillus sp. CZD-F18, respectively. The inhibitory activity of all compounds against PC-3 cell lines, BRD4 and aromatase were evaluated. The results showed compound 7 exhibited moderate anticancer activity with 66.1% inhibition against PC-3 cell lines at the concentration of 10 μg/mL. Compound 7 and 8 exhibited favourable BRD4 inhibitory activity with 78.5 and 76.4% inhibition at the concentration of 10 μg/mL.

Results: Ten compounds were isolated and identified as hexadecanoic acid( 1), methyl 3, 4-dihydroxybenzoate ( 2), protocatechuic aldehyde( 3), caffeic acid methyl ester( 4), vanillic acid( 5), 4-hydroxybenzoic acid( 6), caffeic acid ethyl ester( 7), chlorogenic acid( 8), caffeic acid( 9), 3, 4-di-O-caffeoylquinic acid( 10). Abstract:Lacrymarone (1), a novel diterpenoid, together with seven known compounds 2-8 were isolated from the fermented broth and mycelium of Lacrymaria velutina, an edible fungus collected in Taiwan. The structures of 1-8 were elucidated by analyzing their spectroscopic data. Further biological tests revealed that the co-crystal of 7 and 8 exhibited a moderate inducible nitric oxide synthase (iNOS) inhibitory activity, and with an IC50 value of 59.54 +/- 3.74 mu M.

Abstract:Eight compounds were isolated from the acetone extract of artificially cultured mycelium of Armillaria mellea (Vahl. ex Fr.)Quel.(Tricholomataceae). Three of them are protoilludane sesquiterpenoid aromatic esters. Their structures were elucidated by spectral data analysis as the known 4'-methylmelledonal (I)and two new compounds named armillaritin (II) and armillarivin (III). In addition, five known compounds were identified as D-erythritol, D-mannitol, azelaic acid, orsellinic acid and glycerin-alpha-monooleate.

Abstract:This study was designed to isolate and characterise Flammulina velutipes fruiting body chemical constituents and to find marker compound(s) suitable for processed mushroom products quality control. Extracts were prepared by soaking F. velutipes powdered fruiting bodies in 95% ethanol at 25 degrees C for 24 h; repeated three times; followed by concentrating under reduced pressure at 40 degrees C; and purifying using consecutive chromatographic methods. The compound structures were determined using spectral data analysis and comparing with literature values. Nine chemical constituents: D-arabinitol (1), 9(Z) oleic acid (2), 9(Z),12(Z) linoleic acid (3), ergosta-5,7,22-trien-3 beta-ol (4), 5 alpha,8 alpha-epidioxy-ergosta-6,22-dien-3 beta-ol (5), 3 beta,5 alpha,9 alpha-trihydroxy-ergosta-7,22-dien-6-one (6), 5-hydroxymethyl-2-(1-methyl-ethenyl)-1-cyclohexanol (7), 1,3-dilinolein (8) and hemisceramide (9), were isolated and identified. Compound 7 is a new monoterpene not previously reported. For the first reported time, compound 9 was isolated from the fruiting bodies. Abstract:Antioxidant and antitumor activity of nine compounds from fermental mycelium of Xylaria nigripes were studied.The results showed that(2,6-dihydroxyphenyl)-3-hydroxybutan-one had strong DPPH radical scavenging ability and reducing capacity.The study also indicated that its extract and the nine compounds hadn't showed antitumor activity.

Methods The constituents were isolated by solvent extraction, repeated chromatography with silica gel, Sephadex LH-20, and RP-18 columns. The structures were elucidated by spectral analysis. Results Thirteen compounds were isolated and their structures were identified as (-)-syringaresinol (1), syringaresinol-4-O-β-D-glucopyranoside (2), syringaresinol-4,4'-bis-O-β-D-glucopyranoside (3), (±)-catechin (4), catechin-3-O-β-D-glucopyranoside (5), catechin-5-O-β-D-glucopyranoside (6), 1,3-bis-(4-hydroxy-3,5-dimethoxyphenyl)-1,3-propanediol (7),(R)-(+)-chaulmoogric acid (8), friedelin (9), uracile (10), benzoic acid (11), vanillic acid (12), and 4-hydroxybenzoic acid (13). Abstract:Fifteen compounds, namely three new cytochalasans, one new sesquiterpenoid, two new amides, eight known cytochalasans, and one known amide, were isolated from the rice medium fermented by a soil fungus Xylaria sp. Y01. The structures of these compounds were determined through comprehensive analyses of their extensive spectroscopic data, along with quantum chemical calculations. In vitro cytotoxicity assays, we selected three cancer cell lines (HCT116, 4T1, and MHCC97H) and one normal human gastric epithelial GES-1 cell line to explore their potential as anticancer agents. As a result, 6,7-dihydro-7-oxo-cytochalasin C, 5,6-dihydro-7-oxo19,20-epoxycytochalasin C, 19,20-epoxycytochalasin D, and 19-epi-cytochalasin P1 showed cytotoxicity against GES-1, while only 19,20-epoxycytochalasin D also exhibited cytotoxicity against 4T1. Our findings further enrich the structural diversity of secondary metabolites from Xylaria species and provide clues for exploring their anticancer potential.

Abstract:Objective: To obtain structurally novel and bioactive secondary metabolites from the marine fungus Xylariaceae sp. SCSIO41212. Methods: Compounds were purified by comprehensive chromatography methods of silica gel column, Sephadex LH-20, Ostade-cylsilane (ODS) and semi-preparative HPLC, and their structures were elucidated by means of physical and chemical properties, magnefic resonance imaging, spectroscopic analysis, and comparison with the reported literatures. Results: Chemical investigation of the fungus Xylariaceae sp. SCSIO41212 has led to the isolation of two new compounds, xylaolide B (1) and xylaolide C (2), together with nine known compounds (3–11). Their structures were determined on the basis of detailed analysis of the 1D and 2D NMR as well as MS data. Compounds 1–6 were evaluated for their antiviral, and cytotoxic activities, respectively. Compound 1 showed weak cytotoxicities against K562, MCF-7 and SGC7901 respectively. Conclusion: This study enriches the diversity of secondary metabolites of marine-derived fungi and provides an important reference for novel promising drug leads research.
