Abstract:Bone and teeth are comprised of carbonate-substituted apatites with cationic substitutions, like sodium and potassium. Cations substitute for calcium in the apatite lattice but it is unclear whether they substitute for Ca(1) or Ca(2). Additionally, although we know that anionic substitutions affect the mineral mechanics, it is unclear how cationic substitutions affect mineral stiffness. Here, a combined experimental and theoretical approach using in situ fluid-mediated hydrostatic loading with synchrotron Wide Angle X-ray Scattering (WAXS) and Density Functional Theory (DFT) is used to elucidate the role of CO 3 2− and Na + or K + co-substitutions on the atomic structure and mechanics of biomimetic apatites. Comparison of WAXS and DFT results showed that preferential substitutions at the Ca(1) and Ca(2) sites depended on cationic type and concentration, with a preference for Ca(1) at higher levels of co-substitution. Substitution levels and location of the cationic substitution both significantly affected the modulus of the minerals. This presents a new paradigm for the development of biomimetic apatites with multi-property tunability by considering composition and atomic organization.

Abstract:Apatite is the principal inorganic component of biological hard tissues such as bone and teeth, and thus is essential to the existence of vertebrate life. Because carbonate species and chloride ions are ubiquitous in the fluids of vertebrate life forms, and these species are known to compete with phosphate species, it is critical to understand the influence of these anions on the nucleation of biological apatite. In this study, the important roles of carbonate and chloride ions in mediating inorganic calcium phosphate (CaP) phase nucleation are revealed both theoretically and experimentally. Theoretical investigations suggest that the Ca-O coordination number is an important descriptor for the nucleation of CaP and the interface interaction of the CaP-additive. Positive surface charges on the outermost Ca2+ ions of CaP clusters could attract carbonate species and Cl− ions, promoting the formation of CaP phase. In the presence of carbonate species and Cl− ions, the crystallization of hydroxyapatite (HA) from its precursor brushite (DCPD) has been observed in our experimental studies. The Cl− ion-doped B-type HA is proposed to be the most likely biological apatite based on equilibrium oxygen isotope fractionation and vibrational spectroscopic analysis. The insight gained here may help rationalize the control of biomineralization, and will guide the synthesis of anion-doped biological functional apatite materials.

Abstract:Tooth mineral is constantly exposed to saliva. Based on many factors including diet and chronic disease, salivary composition can vary in pH and potassium (K+) and sodium (Na+) concentrations. Tooth mineral is composed of bioapatite with an ability for ionic exchange between the mineral and the surrounding fluid. Na+ and K+ are known to integrate into biomimetic apatites during crystallization and affect crystallization growth/rate and morphology of calcium phosphates. However, it is unknown how exogenous Na+ and K+ in the solution affect carbonated apatite after formation. Therefore, we investigated the mechanistic differences between Na+ and K+ on biomimetic apatite dissolution/recrystallization. To do so, biomimetic carbonated apatites with 3 or 7 wt% CO3 2− were exposed to NaCl or KCl solutions at various concentrations and pHs seen in saliva. Powder mass, Raman, FTIR, and XRD were used to determine the weight, composition, and structure of the mineral while the solution was characterized for pH and ionic variations. After mineral-solution exposure, significant differences were seen between NaCl and KCl solutions. The apatites exposed to NaCl underwent a classical dissolution/recrystallization mechanism exhibiting more loss in mass and carbonate during dissolution with modifications of A-, B-, and labile CO3 2− amounts during recrystallization which were dependent on the initial apatite CO3 2− content. Meanwhile, apatites exposed to KCl had less mass loss during dissolution and retained the crystal structure, A-, B-, and labile CO3 2− amounts during recrystallization, suggesting that K+ may shield apatites from dissolution. To our knowledge, this is the first study to parse out mechanistic differences between Na+ and K+ on biomimetic carbonated apatite dissolution/recrystallization. Overall, this study will provide insight on how fluctuating Na+ and K+ in saliva may affect tooth mineral composition and structure.

Abstract:Biomineralization is an important process of bone tissue generation. Calcium (Ca) and phosphate (P) ions aggregate and nucleate under the regulation of biomolecules at the initial mineralization stage. Due to the complexity of the physiological environment, the movement behavior and mineralization mechanism of calcium and phosphate ions, as well as the effect of biomolecules on them, are not clear. In this study, computer simulations and experimental verification were applied to investigate the characteristics of the initial biomineralization from the view of ion aggregation and nucleation. The results prove that P ions play a more important role in mineralization than Ca ions. The guanidyl group and surrounding carboxyl terminal groups are a potential excellent nucleation domain on proteins. The interval distribution of acidic/basic residues on protein is more conductive to the formation of large Ca and P ions clusters. The involvement of protein could increase the probability of hydroxyapatite phase precipitation, especially in the presence of a helical conformation. Location of Carbonate IONS in Structure of Biological Apatite

Abstract:Substitution of the carbonate ion into the channel (A) and phosphate group (B) positions of hydroxyapatite (CHAP), fluorapatite (CFAP) and chlorapatite (CCLAP) has been investigated by Fourier transform infrared (FTIR) spectroscopy and single-crystal X-ray structure, using crystals synthesized at high pressure and temperature (0.5-1.0 GPa, 1000-1350 degrees C). Based on similarity of chemical compositions and FTIR spectra, biological apatites appear to have the crystal structure of synthetic Na-bearing type A-B CHAP. Type A carbonate is oriented in the apatite channel with two oxygen atoms close to the c-axis and the third projecting close to the channel wall, whereas type B is located near the sloping faces of the substituted phosphate group. The Na-bearing carbonate apatite crystals have compositions corresponding approximately to the composition series Ca10-yNay[(PO4)(6-y)(CO3)(y)][(OH/Cl)(2-2x)(CO3)(x)], with x approximate to y up to 1.0 for CHAP and 0.4 for CCLAP, and Ca10-1.5yNay (0.5y)[(PO4)(6-2y)(CO3)(2y)][F2-2x(CO3)(x)], with x approximate to y approximate to 0.1 for CFAP, for equivalent conditions of synthesis. The Na+ cation and A and B carbonate ion defects are locally coupled in the structures of all three composition series to minimize the effects of charge compensation and spatial accommodation.
The Carbonate and Sodium Environments in Precipitated and Biomimetic Calcium Hydroxy-Carbonate Apatite Contrasted with Bone Mineral: Structural Insights from Solid-State NMR

Abstract:Bone mineral consists of calcium hydroxy-carbonate apatite (HCA) that incorporates other minor cation substituents, primarily Na+ (0.5-0.8 wt %). We examine the carbonate species in various HCA specimens with variable CO32- contents (4-10 wt %), encompassing phases prepared by precipitation and a biomimetic specimen formed from a bioactive glass inside a simulated body fluid as well as bone tissue from beagle dog. Using magic-angle spinning (MAS) nuclear magnetic resonance (NMR) along with infrared spectroscopy experiments, we identified and quantified carbonate anions replacing either hydroxyl ("A-type" CO32-) or phosphate ("B-type" CO32-) anions in the HCA lattice, along with the carbonate species present in the amorphous surface layer present at all synthetic and biogenic nanocrystalline HCA particles. Advanced C-13-based NMR experimentation enabled the selective detection of the minor (CO32-)-C-13 population of intact bone monoliths, whose C-13 NMR signals are otherwise swamped by those from collagen, unless chemically invasive deproteination procedures are invoked. The CO32- species present in 4 week- and 8 month-old bone of the alveolar process from beagle dog revealed mainly B-type lattice sites and carbonates present in the amorphous surface layer. No tissue aging effects were observed in the local CO32- environments. Likewise, NMR revealed very similar Na-23(+) parameters in HCA, regardless of its synthetic or biogenic origin or degree of structural order. A combination of interatomic-distance-sensitive MAS NMR experiments allowed the identification of the local environments of the various carbonate, phosphate, hydroxyl, and water species present in both the interior and the surface layer of the synthetic HCA particles. We highlight the similarities/differences in the chemical speciation and the spatial distribution of CO32- anions present in carbonate-bearing amorphous calcium phosphate (ACP) relative to the ACP-like surface layer of HCA and discuss the C-13 NMR peak assignments of the up to four coexisting CO32- populations in the synthetic/biogenic HCA phases.

Abstract:The crystal chemistry of carbonate in Ca apatites, including Na-free and Na-bearing hydroxylapatite (CHAP) and Na-bearing fluorapatite (CFAP) and chlorapatite (CCLAP), has been investigated by Fourier transform infrared (FTIR) spectroscopy and single-crystal X-ray structure, using crystals synthesised from carbonate-rich melts at 1 GPa, 1400-1000°C. The carbonate ion substitutes for both the X (OH, F, Cl) anion species in the apatite channel (type A carbonate) and the phosphate group (type B carbonate). The type A carbonate ion is oriented in the channel with two oxygen atoms close to the c-axis, and the type B carbonate ion is located close to the sloping faces of the substituted phosphate tetrahedron, with details varying with composition series. Sodium promotes the uptake of type B carbonate and also has a profound effect on FTIR spectra. Crystal compositions correspond approximately to the substitution formula Ca10-(y+z), Nay" 2[(PO4)6-(y+2z)(CO3) (y+2z)][X2-2x(CO3)x], with × ≈ y up to 1.0 and z ≈ 0.0 for CHAP, × ≈ y ≈ 4z ≈ 0.4 for CCLAP, and × ≈ y ≈ 2z ≈ 0.1 for CFAP, for equivalent conditions of synthesis. The sodium cation and A and B carbonate ion defects are locally coupled in all three composition series to facilitate charge compensation and minimise the effects of spatial accommodation. Synthetic Na-bearing type A-B CHAP with × ≈ y ≈ 0.5 has a similar chemical composition and FTIR spectrum to biological apatite.
Effect of Magnesium Substitution on Structural Features and Properties of Hydroxyapatite

Abstract:Hydroxyapatite (HAP) is the main mineral component of bones and teeth. It is widely used in medicine as a bone filler and coating for implants to promote new bone growth. Ion substitutions into the HAP structure highly affect its properties. One of the most important substituents is magnesium. This paper presents new results obtained using high-precision hybrid density functional theory calculations for Mg/Ca substitutions in HAP in a wide magnesium concentration range within a 2 × 2 × 2 supercell model. Experimental data on the mechanochemical synthesis of HAP-Mg samples with different Mg concentrations are also presented. A comparison between the experiment and the theory showed good agreement: the HAP-Mg unit cell parameters and volume decreased with increasing degree of Mg/Ca substitution. The changes in the distances between the Ca and O, Ca and H, and Mg and O ions upon Mg/Ca substitution in different calcium positions was analyzed. The resulting asymmetry and distortion of the cell parameters were evaluated. It was shown that bulk modulus, energy levels, and band gap depend on the degree of Mg substitutions in the Ca1 and Ca2 positions. The formation energies of Mg/Ca substitutions showed non-monotonic behavior that was different for Ca1 and Ca2 positions. The Ca2 position had a slightly higher probability (~5 meV/f.u.) of substitution than Ca1 position at a Mg concentration x = 0.5. At x = 1, substitution in both positions can coexist. The simulated IR spectra for different Mg/Ca substitutions showed that Mg in the Ca2 position changes the IR spectrum more significantly than Mg in the Ca1 position. Similar changes were recorded in the IR spectra of the synthesized samples. The electronic structure is shown to be sensitive to the number and position of substitutions, which may be used to tweak the optical properties of the HAP-Mg material.
The Mutual Incorporation of Mg2+ and CO32− into Hydroxyapatite: A DFT Study

Abstract:Hydroxyapatite (HA) with a stoichiometry composition of Ca10(PO4)6(OH)2 is widely applied for various biomedical issues, first of all for bone defect substitution, as a catalyst, and as an adsorbent for soil and water purification. The incorporation of foreign ions changes the acid–base relation, microstructure, porosity, and other properties of the HA materials. Here, we report the results of calculations of the density functional theory and analyze the possibility of two foreign ions, CO32− and Mg2+, to be co-localized in the HA structure. The Na+ was taken into account for charge balance preservation. The analysis revealed the favorable incorporation of CO32− and Mg2+ as a complex when they interact with each other. The energy gain over the sole ion incorporation was pronounced when CO32− occupied the A position and Mg2+ was in the Ca(2) position and amounted to -0.31 eV. In the most energy-favorable complex, the distance between Mg2+ and the O atom of carbonate ion decreased compared to Mg...O distances to the surrounding phosphate or hydroxide ions, and amounted to 1.98 Å. The theoretical calculations agree well with the experimental data reported earlier. Understating the structure–properties relationship in HA materials varying in terms of composition, stoichiometry, and morphology paves the way to rational designs of efficient bio-based catalytic systems.

The samples with different concentrations were investigated via X-ray diffraction, Fourier transform infrared, field emission scanning electron microscopy, thermogravimetric analysis and microhardness. In addition, lattice parameters, lattice distortion and crystallite size upon different models were computed. The two ions competed to replace both Ca2+ sites, and it was found that Bi3+ preferred Ca(2), while Sr2+ selected Ca(1). The hardness was enhanced via substitution where the highest value reached 3.1 GPa at the highest concentration of Bi3+ ions. This study has displayed that co-substituted ions into HAP can cause a great influence on its physico-chemical properties. The thorough study of the co-substitution effects is needed to deepen the understanding of synthetic HAP which may contribute in growth its applications particularly in bone tissue engineering.

Abstract:The understanding of interfaces and interaction of organic molecules and inorganic materials are the important issues in biomineralization. Experimentally, it has been found that amino acids (AA) can regulate the morphology of hydroxyapatite (HAP) crystals significantly. In this study, molecular dynamics simulation is employed to investigate the detailed adsorption behavior of polar, ionic, and hydrophobic AA on the {100} face of HAP at the atomic level. The results indicate that various AA are adsorbed on the HAP crystal surface mainly by amino and carboxylate groups at the specific sites. Multiple interaction points are found for polar and ionic AA. The adsorbed AA molecules occupy the Ca and P sites of the HAP surfaces which may inhibit and regulate the HAP growth. The adsorbed amino acid layer can also change the interfacial hydration layer and influence the transportation of ions in and out of HAP, which may be another strategy of biological control in biomineralization.
Effect of borate, fluoride and strontium ions on biomimetic nucleation of calcium phosphate studied using solid-state nuclear magnetic resonance and X-ray diffraction

Abstract:Objectives: Apatite minerals can have various anions and cations in their crystal structure in addition to phosphate ion (PO₄³⁻) and calcium ion (Ca2+). The aim of this study is to investigate effects of the borate, fluoride and strontium ions on biomimetic nucleation of calcium phosphate. Methods: Nano-crystalline hydroxyapatite (H-Ap) was obtained from a supersaturated buffered solution containing 4.12 mM HPO42- and 5.88 mM Ca2+ (H-Ap). Four additives were used in solid solution methods: (i) 0.588 mM F- (F-Ap), (ii) 5.88 mM Sr2+ (Sr-Ap), (iii) 4.12 mM BO33- (BO3-Ap), and (iv) a surface pre-reacted glass ionomer (S-PRG) filler eluate that contained 0.17 mM Sr2+, 0.588 mM F-, 11.1 mM BO33-- (SPRG-Ap). Apatite crystallization was investigated using a solid-state magic-angle spinning NMR spectroscopy and X-ray diffraction (XRD) with the Rietveld analysis. Results: A 2D 1H-31P heteronuclear-correlation NMR showed F- ion incorporation in the apatite structure of the F-Ap and SPRG-Ap. The peaks on the 31P axis of the F-Ap, Sr-Ap, and BO3-Ap were different from that of the H-Ap, and the full width at half maximum increased in the following order: H-Ap∼F-Ap∼BO3-Ap< SPRG-Ap< Sr-Ap, suggesting the incorporation of the F-, Sr2+ and BO33-. The incorporation of F and BO3 was further confirmed by 19F and 11B NMR. The XRD revealed that Sr2+ was preferentially incorporated into the CaII site. Significance: The F-, Sr2+ and BO33-ions might be involved in modifying the crystallization of apatite precipitation, producing a variety of apatite. S-PRG filler that release these ions may have an effect on remineralization, i.e., the reformation of apatite lost due to caries.
Contribution to Knowledge on Bioapatites: Does Mg Level Reflect the Organic Matter and Water Contents of Enamel?

Abstract:The matter constituting the enamels of four types of organisms was studied. The variability of the ions was presented in molar units. It was proven that the changes in water contents of the enamel are significantly positively related to changes in Mg; inversely, there is also a strong connection with changes in Ca and P, the main components of bioapatite. The variability in the organic matter has the same strong and positive characteristics and is also coupled with changes in Mg contents. Amelogenins in organic matter, which synthesize enamel rods, likely have a role in adjusting the amount of Mg, thus establishing the amount of organic matter and water in the whole enamel; this adjustment occurs through an unknown mechanism. Ca, P, Mg, and Cl ions, as well as organic matter and water, participate in the main circulation cycle of bioapatites. The selection of variations in the composition of bioapatite occurs only along particular trajectories, where the energy of transformation linearly depends on the following factors: changes in the crystallographic d parameter; the increase in the volume, V, of the crystallographic cell; the momentum transfer, which is indirectly expressed by ΔsinΘ value. To our knowledge, these findings are novel in the literature. The obtained results indicate the different chemical and crystallographic affinities of the enamels of selected animals to the human ones. This is essential when animal bioapatites are transformed into dentistic or medical substitutes for the hard tissues. Moreover, the role of Mg is shown to control the amount of water in the apatite and in detecting organic matter in the enamels.

Abstract:Biominerals exist in scale,fin and tooth of seawater Lateolabrax Japonicus.In this paper,the crystallographic characteristics of the inorganic mineral phase existing in scale,tooth and bone (fin,sphenoid,backbone) from seawater Lateolabrax Japonicus were investigated by means of X-ray micro diffractometry (XRMD),transmission electron microscopy (TEM),environmental scanning microscopy (ESEM) and energy dispersive spectrometry (EDS).XRMD can well help to obtain valuable structure information by ways of in-situ and non-destruction measurements,and hence it is a more suitable method than powder diffraction in biomineral studies.The results show that the crystal phase in these hard tissues is sole hydroxylapatite (HAP).The chemical composition characteristics of HAP in hard tissues is lack of Ca and rich in P except sphenoid.Refined lattice parameters of HAP show that in scale:a =0.94210.9461nm and c=0.6844 0.6907nm and the closer to the scale base,the value of lattice parameters is closer to that of the scale base,in bone:a=0.9438 0.9492nm and c=0.6873 0.6887nm and in tooth:a=0.9470nm and c=0.6883nm.The coherent domain sizes calculated from the Scherrer equation are Dscale =9.4～14.1nm,Dbone=16.2～18.3nm and Dtooth =15.1nm.From bone to tooth to scale it shows a crystallinity sequence from good to poor.X-ray diffraction patterns and textural index R values indicate that all HAPs in hard tissues of Lateolabrax Japonicus have a preferring orientation along crystallographic c axis.The crystallographic charateristics of biominerals (grain size,crystallinity and preferring orientation) are designed by tissue function and controlled by organic matrix to provide good mechanical performance.

Abstract:A suite of Na-bearing type A–B carbonate hydroxyapatites {Ca10−yNay[(PO4)6−y(CO3)y][(OH)2−2x(CO3)x], x≈y} has been synthesized at 1200°C and 0.5–1.0GPa, and investigated by single-crystal X-ray structure and FTIR spectroscopy. Crystal data for the maximum content of carbonate (11.1 wt%) are a=9.3855(7), c=6.9142(4)Å, space group P63/m, R=0.023, Rw=0.014. Structural accommodation of the substitutions requires local coupling of Na and channel (type A) and phosphate (type B) carbonate ion defects. The type B carbonate ion is located on the sloping faces of the substituted phosphate group, but is inclined at an angle of 53° to the mirror plane. FTIR spectra have minimal ν3 absorption beyond 1500cm−1 and dominant ν2 absorption at 873cm−1. Synthetic Na-bearing type A–B apatites (with a high content of type A carbonate) are thus similar in both chemical composition and infrared spectra to biological apatites. The latter are reinterpreted as Na-bearing type A–B carbonate apatites with channel carbonate up to 50% of total carbonate.

Abstract:The evolution of the valence band, charge states of atoms, and optical and vibrational spectra in compounds Ca10−xMx(PO4)xY2, M = Fe, Ni, Cu, Mg; Y = OH, Cl, F was studied by using XPS, infrared, and optical spectroscopy, with the addition of quantum mechanics calculations. The changes in the bandgap in these compounds were analyzed. Isomorphic substitution of calcium ions in the cationic sublattice of calcium hydroxyapatite by metal ions changes the shape of the curve that represents the occupied part of the valence band only slightly. It retains a pronounced gapped character with different lengths of individual subbands—the upper and lower parts of the valence band. It is shown that the predominant position of rare earth and uranium atoms in the apatite structure is the Ca(2)-position. Isomorphic substitution of calcium atoms by metal atoms (Fe, Ni, Cu, Mg) in the apatite structure in the range of 1%–2% of atoms leads to the narrowing of the energy gap. The most significant narrowing is observed when calcium is substituted by nickel and copper. The theoretically calculated bandgap width in calcium apatites can be well described in terms of the generalized gradient approximation. The design of the structure of calcium apatites via the method of isomorphic substitutions in the cation sublattice makes it possible to control the bandgap width, thus expanding the field of practical application of these compounds.

Abstract:The element-substituted calcium Deficient hydroxyapatite samples were synthesized using liquid-phase precipitation method. Different element ions (Mg+2, Zn+2, and Cu+2) were added at constant ratio (5% wt.). The phase composition of the element-substituted calcium hydroxyapatite samples was investigated by using X-ray diffraction. A comparison with known characteristics of the synthetic ion-substituted forms of hydroxyapatite was conducted. It was determined, that the investigated materials are single-phase solid solutions of the element-substituted hydroxyapatite. By the methods of IR spectroscopy and Raman scattering (RS) it was discovered the dependence of incorporation of the carbonate anion that occupies a positions of phosphorus-oxygen groups in the hydroxyapatite structure as a result of embedding element ions (Zn+2, Cu+2, Mg+2) in the crystal lattice. Besides, It was determined, that the ratio of the intensities for the C03 group mode to the P04 groups mode depended on the nature of the element atom, which replace the calcium in the lattice of HAP, and reduced by substituting the calcium atoms by the atoms of Zn, Cu, and Mg from 0.15 to 0.06. Modeling of the vibration modes of Raman spectroscopy showed that incorporation of carbon atoms to the hydroxyapatite structure correlate with the Zn, Cu and Mg atomic radius, charge, and activity

Abstract:Ionic substituted hydroxyapatites are widely used in bone tissue engineering applications. The present research is aimed at the synthesis of zinc and fluorine co-substituted hydroxyapatite (ZnFHA10, Ca9.0Zn1.0(PO4)6(OH)F) using a sol–gel method. This research investigates the effect of zinc and fluorine co-substitution on the structure of synthesized hydroxyapatite. The as-synthesized nanopowder was heat-treated in the temperature range 800 °C–1200 °C. The physicochemical properties were investigated using X-ray fluorescence spectroscopy, X-ray diffraction, Fourier-transform infrared spectroscopy and transmission electron microscopy. The thermal stability of nanopowder was assessed by the thermogravimetric analysis. Bioactive behavior was evaluated by studying the variation in pH of simulated body fluid on immersion of nanopowders. Peaks of zinc and fluorine in the XRF spectra confirmed the substitution of ions. TEM images of as-synthesized and calcined nanopowders showed rod-like and spherical morphology. X-ray diffraction pattern of as-synthesized nanopowder showed the formation of nanoparticles of low crystallinity. On calcination, the crystallinity and crystallite size increased and the biphasic mixture of hydroxyapatite and tricalcium phosphate was formed. Spectra obtained from Fourier-transform infrared spectroscopy showed the presence of OH…F…OH group, depicting the presence of fluorine in hydroxyapatite. FTIR spectra of nanopowders obtained after immersion in simulated body fluid and change in pH values on immersion of nanopowders in simulated body fluid confirmed the bioactive behavior of nanopowders. The study revealed that the substitution of ions in hydroxyapatite could significantly increase the bioactivity and bioresorbability of hydroxyapatite, thus finding a wide range of clinical applications in orthopedics and dentistry.

Abstract:Rare Earth elements in the lanthanide series are regarded as one of the finest options for the cationic substitution of calcium ions in hydroxyapatite (HA) because of their favorable impact on the biological characteristics of substituted HA. Neodymium and cerium were used to substitute 5% of calcium ions in HA, prepared via the wet precipitation method. Characterization tests for pure and substituted HA were conducted using XRD, FTIR, EDS, and FESEM. The results showed that changing part from calcium ions in hydroxyapatite to Nd and Ce ions altered its structure, composition, and morphology. Regarding the biological tests, the cytotoxicity test revealed a change in IC50 for both normal and cancer cell lines, where substitution part of the Ca ions with rare Earth elements led to increasing antitumor activity in comparison with HA without substitution; in addition, antibacterial and fungicide activity was evident for both HA and Nd-Ce/HA, with a modest increase in antibacterial activity of Nd-Ce/HA against S. epidermidis and E. coli in comparison with HA. These findings may shed light on the process by which Nd and Ce ions improve the biological characteristics of pure HA and the increased potential of these bioceramics.

Abstract:Mesoporous calcium phosphate based-bioceramics with ionic substitution have attracted attention for treatment of bone diseases and bone regeneration. Carbonate apatite (CO 3 Ap) is a promising candidate for bone replacement through bone remodelling, similar to that of autologous bone, owing to the chemical composition of the bone mineral. In this work, we present a dual approach to synthesize mesoporous carbonate apatite (CO 3 Ap) and strontium-substituted CO 3 Ap (Sr-CO 3 Ap) artificial bone nanocrystals, aiming to assess the impact of strontium on CO 3 Ap characteristics and biological performance. Mesoporous CO 3 Ap was created by transforming hard templates composed of CaCO 3 , while Sr-substituted CaCO 3 (Sr-CaCO 3 ) microspheres were formed using an organic complexing agent to induce pore formation. The resulting CO 3 Ap and Sr-CO 3 Ap granules exhibited an irregular shape, with rod-like nanocrystals composing their particles. Adsorption/desorption isotherms confirmed the mesoporous nature of the structures, featuring pore sizes ranging from 10 to 50 nm. Specifically, mesoporous CO 3 Ap displayed slit-like pores, while Sr-CO 3 Ap showed cylinder-like ones. High-resolution transmission electron microscopy images of Sr-CO 3 Ap powder revealed mesopores within the particles, with Sr-CO 3 Ap exhibiting larger pore volume and specific surface area compared to CO 3 Ap. When osteoblast-like MC3T3-E1 cells were cultured on both CO 3 Ap and Sr-CO 3 Ap samples, they demonstrated good cell attachment, proliferation rates, and increased ALP activity, particularly evident at higher strontium concentrations. These findings suggest promising potential for further exploration of mesoporous apatite nanocrystals in bone replacement and drug delivery applications.
