Abstract:Arctium lappa L. is a prevalent medicinal herb and a health supplement that is commonly used in Asia. Over the last few decades, the bioactive component arctigenin has attracted the attention of researchers because of its anti-inflammatory, antioxidant, immunomodulatory, multiple sclerosis fighting, antitumor, and anti-leukemia properties. After summarising the research and literature on arctigenin, this study outlines the current status of research on pharmacological activity, total synthesis, and structural modification of arctigenin. The purpose of this study is to assist academics in obtaining a more comprehensive understanding of the research progress on arctigenin and to provide constructive suggestions for further investigation of this useful molecule.
Fructus arctii: an overview on its traditional uses, pharmacology and phytochemistry

Abstract:Objectives: Fructus arctii (F. arctii) is the dried ripe fruit of Arctium lappa Willd (Asteraceae). It is being used as a traditional medicine in China, Japan, Iran, Europe, Afghanistan, India, etc. for cough, inflammation, clearing the heat, detoxification, cancer and diabetes. This review summarized the botanical description, distribution, ethnopharmacology, bioactive constituents and pharmacological actions of F. arctii including methods to assess its quality. In addition, this review also provides insights into future research directions on F. arctii to further explore its bioactive constituents, mechanism involved in pharmacological activity, and clinical use including the development of new analytical methods for assessing the quality. Key findings: The comprehensive analysis of the literature revealed that F. arctii contains lignans, volatile oil, flavonoids, sesquiterpenoids, triterpenes, phenolic acids, etc. Experimental studies on various extracts and drug formulations showed that it has antioxidant, antimicrobial, hypoglycaemic, lipid-lowering, anti-inflammatory, analgesic, antiviral, anti-tumour activity, etc. Summary: The pharmacological activity of a few major constituents in F. arctii have been identified. However, there are still need more studies and more new technologies to prove the pharmacological activity and the effective mechanism of the other constituents that undergoing uncertain. Except for the animal experiments, clinical studies should be carried out to provide the evidence for clinical application.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Arctium lappa L., is a biennial plant that grows around the Eurasia. Many parts of Arctium lappa L. (roots, leaves and fruits, etc.) are medically used in different countries. Arctium lappa L. fruit, also called Arctii Fructus, is traditionally applied to dispel wind-heat, ventilate lung to promote eruption, remove toxicity substance and relieve sore throat.THE AIM OF THE REVIEW: The review aims to integrate the botany, ethnopharmacology, quality control, phytochemistry, pharmacology, derivatives and toxicity information of Arctii Fructus, so as to facilitate future research and explore the potential of Arctii Fructus as an agent for treating diseases.MATERIALS AND METHODS: Related knowledge about Arctii Fructus were acquired from Science Direct, GeenMedical, PubMed, China National Knowledge Infrastructure (CNKI), Web of Science, Pharmacopoeia of the People's Republic of China, Doctoral and Master's thesis, ancient books, etc. RESULTS: Arctii Fructus as an herb used for medicine and food was pervasively distributed and applicated around the world. It was traditionally used to treat anemopyretic cold, dyspnea and cough, sore throat, etc. To date, more than 200 compounds have been isolated and identified from Arctii Fructus. It contained lignans, phenolic acids and fatty acids, terpenoids, volatile oils and others. Lignans, especially arctigenin and arctiin, had the extensive pharmacological effects such as anti-cancer, antiviral, anti-inflammatory activities. The ester derivatives of arctigenin had the anti-cancer, anti-Alzheimer's disease and immunity enhancing effects. Although Arctii Fructus extract had no toxicity, arctigenin was toxic at a certain dose. The alleviating effects of Arctii Fructus on chronic inflammation and ageing have been demonstrated by clinical studies.CONCLUSION: Arctii Fructus is regarded as a worthy herb with many chemical components and various pharmacological effects. Several traditional applications have been supported by modern pharmacological research. However, their action mechanisms need to be further studied. Although many chemical components were isolated from Arctii Fructus, the current research mainly focused on lignans, especially arctiin and arctigenin. Therefore, it is very important to deeply clarify the pharmacological activities and action mechanism of the compounds and make full medicinal use of the resources of Arctii Fructus.

Abstract:Arctiin (I), arctigenin (II), matairesinol (III), and lappaol F (IV) were isolated and purified from the traditional Chinese medicine Fructus Arctii by high-speed counter-current chromatography (HSCCC). The crude extracts from Fructus Arctii were treated with D101 macroporous resin first and divided into two parts: fraction 1 and fraction 2. Fraction 1 was separated by ethyl acetate-n-butanol-water (4:0.5:5, v/v/v) and yielded 164 mg of I from 250 mg of fraction 1. Fraction 2 was separated by n-hexane-ethyl acetate-methanol-water (2:3:2:3, v/v/v/v) and yielded 27 mg of II, 5 mg of III, and 3 mg of IV from 150 mg of fraction 2. The purities of the four compounds were 99.64%, 98.48%, 96.16%, and 91.41%, respectively, as determined by HPLC-DAD. The chemical structures of the isolated compounds were identified by MS, UV, 1H NMR, and 13C NMR analysis.

Abstract:Arctium lappa, widely recognized as burdock, is a perennial plant that is employed in the realm of traditional Chinese medicine for a wide range of medicinal applications. The herb is rich in bioactive metabolites with therapeutic potential, encompassing polyphenolic antioxidants in its leaves, and flavonoids and fructo-oligosaccharides in its underground parts. Nutraceuticals originating from botanical sources such as Arctium lappa provide supplementary health advantages alongside their nutritional content and have demonstrated effectiveness in the prevention and management of specific ailments. The utilization of Arctium lappa root extract has exhibited encouraging outcomes in addressing hepatotoxicity induced by cadmium, lead, chromium, and acetaminophen, ameliorating liver damage and oxidative stress. Additionally, the root extract displays properties such as antidiabetic, hypolipidemic, aphrodisiac, anti-rheumatic, anti-Alzheimer, and various other pharmacological actions.

Abstract:OBJECTIVE:To study the metabolic chemistry and pharmaco-dynamics characters of ligan from seeds of Arctium lappa.METHOD:HPLC method was used in the study. The analysis was carried out on C18 column. The mobile phase was CH3CN-0.05% H3PO4 (36:64) with flow-rate at 0.6 mL x min(-1) and wave-length of 210 nm. The column temperature was kept at 25 degrees C.RESULT:The results indicated that the ligan was detected in plasma and the main organs 5 min after po. The main metabolic production in plasma was arctigenin. In addition, arctigenin and an unknown product were found in metabolic production in the organs.CONCLUSION:The method was stable,simple and reproducible. It can be used to determine the metabolic product of the ligan. The metabolic chemistry of ligan in plasma was obviously different from that in the main organs.

Abstract:Arctium lappa L. root has high nutritional and medicinal value, and has been identified as one of the health food raw materials by the china's Ministry of Health. In present study, an aqueous two-phase system (ATPS) of polyethylene glycol (PEG)-(NH4)2SO4 was used to extracted Arctium lappa L. polysaccharides (ALPs) from Arctium lappa L. root, the optimal extraction conditions of crude ALPs were optimized by using single-factor experiment and response surface methodology. The optimized extraction parameters for extraction ALPs were as follows: PEG relative molecular weight 6000, quality fraction of PEG 25%, quality fraction of (NH4)2SO4 18%, extraction temperature 80°C. Under these conditions, the extraction rate of ALPs could reach at 28.83%. The structure and composition of ALPs were determined by fourier infrared spectrum (FI-RT), scanning electron microscopy (SEM), and HPLC. The results showed that ALPs were typical acidic heteropolysaccharides, having uneven particle size distribution, irregular shape and a rough surface. ALPs were mainly composed of glucose, rhamnose, arabinose, and galactose with a molar ratio of 70.19: 10.95: 11.16: 6.90. In addition, ALPs had intense antioxidant activity in vitro with IC50 values in ·OH radical (1.732 mg/mL), DPPH radical (0.29 mg/mL), superoxide anion (0.15 mg/mL) scavenging ability.

Extraction and Separation of Arctiin from Fructus Arctii

Abstract:Arctiin, as a precursor of arctigenin which is one of the main functional components in Fructus Arctii, was extracted with ethanol in a microwave field and separated from the extraction liquid with macroporous resin adsorption chromatography. The microwave-assisted extraction procedure of arctiin was optimized using L9(34) orthogonal array design, and the effects of adsorption and desorption conditions on separation of arctiin were discussed. A maximum extraction yield of arctiin of 9.02% was obtained by the 80 s extraction in an 250 W microwave field with a liquid/material ratio of 20:1 (ml/g). Among four types of macroporous resin tested, HPD-700 possessed the best adsorption and desorption performance. The optimum technological parameters for separating arctiin were determined as follows: sample concentration 3.26 mg/ml, flow rate of sample 1 ml/min, and flow rate of 70% ethanol used as an eluant 1 ml/min.

Abstract:As a common traditional Chinese medicine, Fructus Arctii has important clinical medical values. Its main components are lignans, which are difficult to separate and analyze because of the complex composition, similar chemical structures, and close properties. In this study, an off-line two-dimensional supercritical fluid chromatography/reversed-phase liquid chromatography method, as well as an effective sample pretreatment method based on hydrophilic interaction chromatography material, was developed to enrich the minor lignan fractions and obtain high-purity compounds. In total, 12 high-purity compounds were isolated from Fructus Arctii. Their structures were identified by using high-resolution mass spectrometry and nuclear magnetic resonance spectroscopy, which showed that all were lignans and that most of them were isomers. The results demonstrated the effective off-line two-dimensional supercritical fluid chromatography/reversed-phase liquid chromatography method for the purification of lignans from Fructus Arctii. The separation protocol established here will be beneficial for the separation of complex samples from other kinds of natural products.

Abstract:To explore new antioxidant resource from food, a water-soluble polysaccharide (ALP1) was extracted and purified from the roots of Arctium lappa L. (A. lappa L.) through hot water extraction followed by ethanol precipitation, ion-exchange chromatography and gel filtration. The antioxidant activity of ALP1 was then evaluated in vitro and in vivo. ALP1 was characterized as a fructan composed of fructose and glucose in the ratio of 13.0:1.0, with an average molecular weight of 4600 Da. The linkages in ALP1 were →1)-Fruf-(2→, Fruf-(2→ and Glcp-(1→. In vitro antioxidant assays demonstrated that ALP1 possessed moderate ABTS(+) scavenging activity, strong hydroxyl radical scavenging activity and strong ferrous ion chelating activity. In in vivo antioxidant assays, ALP1 administration significantly enhanced antioxidant enzyme activities and total antioxidant capacity, as well as decreased the levels of malondialdehyde (MDA) in both the serum and liver of aging mice. These results suggest that ALP1 has potential as a novel natural antioxidant in food industry and pharmaceuticals.
Immune regulation and inflammation inhibition of Arctium lappa L. polysaccharides by TLR4/NF-κB signaling pathway in cells

Abstract:Arctium lappa L. polysaccharides (ALP) are important active ingredients of burdocks with various bioactivities. In the present study, a crude polysaccharide was extracted from A. lappa L. roots and purified using DEAE-52 and Sephacryl™ S-400 columns to reach 99 % purity. This neutral polysaccharide contained fructose, glucose, galactose and arabinose in a ratio of 0.675:0.265:0.023:0.016 and had a Mw of 4256 Da. The immunomodulatory activity and intestinal inflammation inhibitory effects of ALP were investigated in in vitro models, including lipopolysaccharide-induced macrophage RAW264.7 and interleukin (IL)-1β-induced colon Caco-2 cells. The results revealed that ALP possessed both antioxidant and anti-inflammatory effects by decreasing nuclear factor-E2-related factor 2 mRNA expression and reactive oxygen species. Furthermore, ALP was found to have inhibitory effects on pro-inflammatory cytokines, including IL-8, IL-6, IL-1β, and tumor necrosis factor-α, as well as inflammatory cytokines, such as intercellular adhesion molecule-1, vascular cell adhesion molecule-1, and monocyte chemoattractant protein-1 by down-regulating the Toll-like receptor 4 (TLR4)/NF-κB (nuclear factor-kappa B signaling) pathway. It indicated that A. lappa L. was an ideal source of bioactive polysaccharides having potential to be developed as functional foods or nutraceuticals to improve immune system and prevent/treat intestinal inflammation.
Studies on Insecticidal Activity and Chemical Components of Arctium lappa

Abstract:For the further development of application value of Arctium lappain pesticide,the Arctium lappaextracts were analyzed by anti-artemia activity test,and the insecticidal components were analyzed by GC-MS.The crude extract was dispersed by water and then extracted by petroleum ether,ethyl acetate and n-butanol respectively.The insecticidal activities of four extracts were tested and the results indicated that the values of IC50of four extracts were 0.13、0.17、1.25、0.19mg·mL-1,respectively.In order to figure out the intrinsic link between the insecticidal activity and the chemical contents,GC-MS analysis of the petroleum ether fraction was carried out,and the value of IC50was relatively lower.51compositions were detected and 39of them were identified,and the relative contents were calculated simultaneously.The results showed that ethyl palmitate(27.791%),terpenoid(21.311%)and saturated hydrocarbon(19.637%)were the main contents,which amounted to 68.739%.

Abstract:Fructus arctii is commonly used in Chinese medicine, and arctiin and arctigenin are its main active ingredients. Arctiin has low bioavailability in the human body and needs to be converted into arctigenin by intestinal microbes before it can be absorbed into the blood. Arctigenin has antiviral, anti-inflammatory, and anti-tumour effects and its development has important value. In this study, we used external microbial fermentation with Aspergillus awamori and Trichoderma reesei to process and convert arctiin from F. arctii powder into arctigenin, hence increasing its bioavailability. We developed a fermentation process by optimising the carbon and nitrogen source/ratio, fermentation time, pH, liquid volume, inoculation volume, and substrate solid-liquid ratio. This allowed for an arctiin conversion rate of 99.84%, and the dissolution rate of the final product was 95.74%, with a loss rate as low as 4.26%. After the fermentation of F. arctii powder, the average yield of arctigenin is 19.51 mg/g. Crude fermented F. arctii extract was purified by silica gel column chromatography, and we observed an arctigenin purity of 99.33%. Our technique effectively converts arctiin and extracts arctigenin from F. arctii and provides a solid basis for further development and industrialisation.

Abstract:AIM:To investigate the bioactive compounds from Arctium lappa.METHODS:The compounds were obtained by various chromatographic methods including silica gel and sephadex LH-20 and their structures were elucidated on the basis of physical parameters and spectral data including IR,UV,NMR and MS.RESULT:Eight compounds were determined as matairesinol(Ⅰ),arctigenin(Ⅱ),lappaol A(Ⅲ),lappaol F(Ⅳ),lappaol C(Ⅴ),arctignan E(Ⅵ),arctiin(Ⅶ)and lappaol H(Ⅷ).CONCLUSION:All the compounds from the active fraction of Arctium lappa are lignans which might be associated with its hypoglycemic activities.

Abstract:Arctigenin was extracted from fruits of Arctium lappa L. by supercritical carbon dioxide.The optimal extraction conditions were:pressure 35 MPa,temperature 40 ℃,flow rate of CO2 6.0 L/h and extraction time 50 min.Arctigenin of 99.66% purity was obtained by simple degreasing of the extract under the optimal extraction conditions,and the extraction rate was 0.45% which was 2.75 times that obtained by traditional solvent-column method.So the supercritical carbon dioxide extraction was simple,safe and highly efficient.

Abstract:Arctium lappa L. is widely used for medicinal purposes across China, and significant effort has been directed toward enhancing its quality. Association with microorganisms has been shown to influence both plant growth and metabolites, providing a possible avenue for its quality improvement. In this study, we investigated the microorganism compositions of the root, stem, leaf, fruit and rhizospheric soil of A. lappa through high-throughput Illumina sequencing of 16S rRNA genes and ITS regions. A total of 796,891 16S rRNA and 626,270 ITS reads were obtained from the samples. Analysis of the sequencing data revealed that bacterial and fungal communities were more diverse in the rhizospheric soil sample compared with other samples. Cyanobacteria , Actinobacteria , Proteobacteria , Firmicutes , and Bacteroidetes phyla were found in all samples. Cyanobacteria was particularly enriched in the root, stem, leaf and fruit at 88.59%, 86.15%, 98.31% and 93.57%, respectively; Actinobacteria was the highest in rhizospheric soil, at 37.53%. Ascomycota was the most dominant fungal phylum, representing 69.17%, 58.18%, 87.93%, 90.18% and 80.21% in the root, stem, leaf, fruit, and rhizospheric soil, respectively. Several novel unclassifiable bacterial and fungal species were also detected. In total, we detected about 922 bacterial and 334 fungal species, which include a number of unclassifiable species. Additionally, the root, stem, leaf, fruit and rhizospheric soil of A. lappa were sources for screening new bioactive metabolites.

Abstract:Astragali Radix (AR) is one of the well-known traditional Chinese medicines with a long history of medical use and a wide range of clinical applications. AR contains a variety of chemical constituents which can be classified into the following categories: polysaccharides, saponins, flavonoids, amino acids, and trace elements. There are several techniques to extract these constituents, of which microwave-assisted, enzymatic, aqueous, ultrasonic and reflux extraction are the most used. Several methods such as spectroscopy, capillary electrophoresis and various chromatographic methods have been developed to identify and analyze AR. Meanwhile, this paper also summarizes the biological activities of AR, such as anti-inflammatory, antioxidant, antitumor and antiviral activities. It is expected to provide theoretical support for the better development and utilization of AR.
