BLNK expression is essential for IL-10 production in regulatory B cells that inhibit contact hypersensitivity

Abstract:CD1dhiCD5+ regulatory B cells (Breg) potently inhibit immune responses including contact hypersensitivity (CHS) via IL-10, although the precise signaling mechanisms required for Breg functions have not been elucidated. B cell linker protein (BLNK) is a central component of “B cell signalosome” that critically regulate intracellular signaling pathways. In this study, we demonstrated that CHS was significantly augmented in BLNK-deficient (BLNK-/−) mice. This was considered to result from impaired B cell function in BLNK-/− mice because, while splenic CD1dhiCD5+ B cells were present in BLNK-/− mice, these cells were unable to produce IL-10 in vitro. Adoptive transfer of splenic CD1dhiCD5+ B cells from wild type mice to BLNK-/− mice diminished CHS inflammation. Toll-like receptor signaling is considered to initiate Breg activation. LPS stimulation induced STAT3 phosphorylation specifically in CD1dhiCD5+ B cells, and not in follicular B cells, from wild type mice. Furthermore, inhibition of Btk or STAT3 activities completely abrogated IL-10 production in B cells, suggesting that Btk-mediated STAT3 activation is critical for IL-10 production in Breg. BLNK expression was necessary for STAT3 activation since STAT3 phosphorylation was completely diminished in CD1dhiCD5+ B cells from BLNK-/− mice. Early IL-10 secretion acted in an autocrine manner to increase the numbers of IL-10-producing B cells by activating JAK-STAT3 pathway. These signaling mechanisms appear to determine the induction of LPS-dependent IL-10 production specifically in CD1dhiCD5+ regulatory B cells.
