Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:Heterocyclic compounds bearing an acidic hydrogen atom attached to nitrogen such as carbazole, indole, and phenothiazine can be efficiently alkylated in DMSO or N,N‐DMF under ultrasonic irradiation in the presence of potassium hydroxide as a base. In almost all cases, a dramatic reduction of the reaction time results and a clear yield increase accompanied by an improved quality of the products occurs.

Abstract:Herein, we report the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions, affording 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is exemplified by the transition metal-free mild conditions and the ability to perform the reaction on a gram scale. Meanwhile, the DFT calculation of the catalytic oxidative transformation pathway has also been studied.

Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:An efficient formation of fused tricyclic [1,3]oxazino[3,4‐a]indol‐1‐ones and dihydropyrimido[1,6‐a]indol‐1(2H)‐ones via Rh(III)‐catalyzed [3+3] or [4+2] annulation of N‐methoxy‐1H‐indole‐1‐carboxamides with sulfoxonium ylides has been developed. These selective annulation reactions were carried out by switching the additives and notable features of this protocol were low catalyst loading and a broad substrate scope providing the corresponding products in up to 99% yields.

Abstract:A mild and efficient synthesis of 3,3-disubstituted oxindoles is achieved. The corresponding diazonium salts are generated in situ from aromatic amines before these salts undergo a homolytic bond cleavage to produce the aryl radicals. Various oxindoles are obtained by homolytic aromatic substitution and radical cyclization. This procedure is an alternative method to oxindole synthesis.

Abstract:An oxidant-free and highly efficient synthesis of phenolic quinazolin-4(3H)-ones was achieved by simply stirring a mixture of 2-aminobenzamides, sulfonyl azides, and terminal alkynes. The intermediate N-sulfonylketenimine underwent two nucleophilic additions and the sulfonyl group eliminated through the power of aromatization. The natural product 2-(4-hydroxybenzyl)quinazolin-4(3H)-one can be synthesized on a large scale under mild conditions with this method.

Abstract:A formal (3+2) annulation reaction between isatin-derived N-tosylhydrazones and o-(trimethylsilyl)aryl triflates allows facile synthesis of spiro[indazole-3,3'-indolin]-2'-ones in high to excellent yields. Abstract:The efficient synthesis of 4-hydroxy-3-arylindolin-2-ones via phosphoric acid-mediated annulation of various β-nitrostyrenes and 1,3-cyclohexanedione is described. This annulation reaction gives a practical method for affording a diverse set of oxindoles, having simple experimentation, readily available starting materials, and very good yields. Additionally, substituted 1,3-cyclohexanediones under the same conditions afforded tetrahydrobenzofuran oxime compounds.

Abstract:An efficient and practical methodology for the synthesis of (benzo)thiazoles-containing amides through pyridine-mediated acylation of 2‐amino(benzo)thiazoles under ultrasonic irradiation was developed. The short reaction time (within 1 min), good functional group tolerance, experimental simplicity and ease of scale-up were salient features of the present strategy, which enables straightforward access to the expected products with good to excellent yields. Additionally, further transformations of the resulting amides were also studied.

Abstract:A convenient method for the synthesis of a series of 2-(arylamino)-3 H -phenoxazin-3-ones based on the nucleophilic substitution reaction between sterically crowded 3 H -phenoxazin-3-one and arylamines performed by short-term heating of the melted reactants at 220–250 °C is described, and the compounds were characterized by means of single-crystal X-ray crystallography, NMR, UV–vis, and IR spectroscopy, as well as cyclic voltammetry. The reaction with o -amino-, o -hydroxy-, and o -mercapto-substituted arylamines widened the scope and provided an access to derivatives of N,O- and N,S-heteropentacyclic quinoxalinophenoxazine, triphenodioxazine and oxazinophenothiazine systems. Abstract:A facile method for preparation of 5-methyl-5-[5-(tert-butyldimethylsiloxymethyl)-3-isoxazolyl]-3-aryl-4-(4-methyloxyphenyl)-4,5-dihydro-1,2,4-oxadiazoles (4a similar to 4e) via 1,3-dipolar cycloaddition of Schiff bases derived from 3-acetyl-5-hydroxymethylisoxazoles and nitrile oxides in situ generated from oximes was described. 5-(tert-Butyldimethylsiloxymethyl)-3-(1,5-diaryl-4,5-dihydropyrazol-3-yl)isoxazoles (6a similar to 6i) were also prepared through cycloaddition of aryl hydrazine and alpha,beta-unsaturated ketones (5) derived from 3-acetyl-5-hydroxylmethylisoxazoles. The structures of all new compounds were characterized by H-1 NMR, C-13 NMR, MS, IR and HRMS spectra.

Abstract:An iodine-promoted one-pot cascade oxidative annulation reaction has been developed for the synthesis of chromone-fused-pyrrolo[2,1-a]isoquinolines and indolizino[8,7-b]indoles from o-acetylphenoxyacrylates, tetrahydroisoquinolines, and noreleagnines. This process underwent a logical approach to both chromone-fused-pyrrolo[2,1-a]isoquinolines and chromone-fused-indolizino[8,7-b]indoles isolamellarin derivatives. Manipulations of l-menthol and dl-α-tocopherol demonstrate the applications of this strategy.

Abstract:An electrochemical protocol for the synthesis of 2,3-disubstituted quinolines by reacting benzoxazinones with arylsulfonyl hydrazides was established under simple and mild conditions (room temperature and an undivided cell). The methodology does not require noble catalysts or external oxidants, providing a green and mild pathway for de novo synthesis of functional quinolines with excellent regioselectivity. Moreover, the method tolerated a variety of functional groups. Notably, the scaled-up experiment and follow-up procedures demonstrate the practicality of electrochemical synthesis.
