Abstract:OBJECTIVE:To observe the effect of dexamethasone on airway remodeling and to explore the significance of the balance between matrix metalloproteinase and the tissue inhibitor of metalloproteinase. METHODS:Thirty male Wistar rats were randomly divided into asthmatic group (n = 10), dexamethasone group (n = 10) and control group (n = 10). Lung tissues were sliced and stained with H. E. The parameters such as bronchial basement membrane perimeter(Pbm), total bronchial wall area(WAt), inner wall area(WAi) and smooth muscle area(WAm), which reflect the thickness of airway wall, were measured by image analysis system. The expression levels of MMP-2 and TIMP-1 mRNA in the lungs were assessed by semiquantitative reverse transcription-polymerase chain reaction. RESULTS:1. WAt/Pbm, WAi/Pbm and WAm/Pbm in asthmatic group (25.3 +/- 2.1 microns 2/micron, 20.4 +/- 2.3 microns 2/micron, 4.2 +/- 2.0 microns 2/micron, respectively) were significantly higher than those in control group (20.8 +/- 1.3 microns 2/micron, 15.3 +/- 2.1 microns 2/micron, 3.1 +/- 1.1 microns 2/micron) (P < 0.01). The differences between those in control group and those in dexamethasone group (21.3 +/- 2.4 microns 2/micron, 14.2 +/- 2.5 microns 2/micron, 3.2 +/- 1.0 microns 2/micron) were not statistically significant (P > 0.05). 2. MMP-2 and TIMP-1mRNA levels in asthmatic group (0.68 +/- 0.14, 0.56 +/- 0.10) and dexamethasone group (0.37 +/- 0.11, 0.31 +/- 0.10) were significantly higher than those in control group (0.14 +/- 0.03, 0.11 +/- 0.05). The differences between asthmatic group and dexamethasone group were also significant (P < 0.01). 3. There was a significant positive correlation between MMP-2 and TIMP-1 mRNA in control group and dexamethasone group (r = 0.67, 0.58, P < 0.05), but not in asthmatic group (r = 0.24, P > 0.05). CONCLUSION:Dexamethasone could prevent airway remodeling by downregulating the expression of MMP-2 and TIMP-1 and restoring the balance between MMP-2 and TIMP-1.
Carbocistein improves airway remodeling in asthmatic mice

Abstract:Background: The alleviating effects of carbocisteine (S-carboxymethylcysteine, SCMC) have been implicat-ed in chronic obstructive pulmonary disease; however, very little is known about its mechanisms in asthma. In this study, we aimed to investigate the effects of SCMC on airway remodeling in asthmatic mice induced by ovalbumin (OVA). Methods: The asthma mouse model was generated by OVA sensitization and stimulation and subsequently intervened by SCMC or dexamethasone. Bronchoalveolar lavage fluid (BALF) and lung tissues were collected from each group of mice. The TGF-beta 1 levels in BALF were measured by ELISA. Masson's staining was used to detect colla-gen fiber deposition in mouse airway tissues, while immunohistochemistry and RT-qPCR were conducted to examine the protein and mRNA expression of TGF-beta 1 in mouse lung airway tissues, respectively. The correlation between TGF-beta 1 mRNA expression and the area of collagen fiber deposition in airway tissues was analyzed by Pearson's correlation coefficient. Results: The area of collagen fiber deposition in the airway tissues of asthmatic mice was significantly increased, while SCMC alleviated the collagen fiber deposition in the airway tissues. TGF-beta 1 expression was significantly elevated in BALF and airway tissues of asthmatic mice, while SCMC inhibited TGF-beta 1 expression. TGF-beta 1 expression was significantly and positively correlated with collagen fiber deposition in mouse airway tissues. Conclusions: SCMC intervention improves collagen fiber deposition in airway tissues and inhibits TGF-beta 1 expression in asthmatic mice.
Effect of Dexamethasone on Eotaxin Expression in Lung Tissue from Rat Model of Asthma

Abstract:Objective By investigating eotaxin expression in rat′s lung, to discuss the possible mechanism of dexamethasone′s therapeutic effect on asthma.Methods The rat model of asthma was established by the ovalbumin challenge method. The animal models were divided into three groups:control,asthma and dexamethasone treated. Eotaxin protein and mRNA were measured by immuneohistochemical and RT-PCR techniques respectively.Results In the asthmatic group, the protein level and the mRNA expressed of eotaxin(0.32±0.04;0.86±0.21)were significantly elevated compared to those in the control group(0.10±0.03;0.18±0.05), the dexamethasone treated group(0.12±0.02;0.21±0.10) were significantly decreased compared to the asthma group,and similar to those in the control group.Conclusions Eotaxin actively participated in the process of eosinophilic inflammation. Dexamethasone demonstrated anti-inflammatory effects by down-regulation of eotaxin.
Expression of Stem Cell in Lung Tissue of Asthmatic Mice and the Influence of Dexamethasone

Abstract:Objective To observe the expression of stem cell factor (SCF) in the lung tissue of asthmatic and dexamethasone-treated mice to reveal the role of SCF in the pathophysiologic process of asthma. Methods We established asthmatic mice model by OVA intranasal administration, and then determined the expression of SCF protein and mRNA by immunochemistry and semi-quantitive RT-PCR in OVA sensitized mice and dexamethasone-treated OVA sensitized mice respectively. Results The expression of SCF protein was increased in the lung of OVA-sensitized mice in comparing with that of control mice, and dexamethasone treatment down regulated SCF protien. RT-PCR showed that mSCF mRNA increased (0.36±0.02 vs 0.16±0.03, P0.01) whereas sSCF mRNA decreased(0.23±0.02 vs 0.41±0.05,P0.01) in the OVA-sensitized mice. After dexamethasone intervention, mSCF mRNA level dropped to near that of the control (0.18±0.03 vs 0.16±0.03, P0.05), and sSCF mRNA increased (0.87 ±0.04 vs 0.23±0.02, P0.01). Conclusion SCF takes part in the pathophysiology process of asthma, and two forms of SCF may play different roles. Dexamethasone may lessen the airway allergic inflammatory reaction by regulating the expression of SCF.
Regulatory Effects of Inhaled Steroids on Expression of Matrix Metalloproteinase-9 and Its Inhibitor in Asthmatic Rats

Abstract:OBJECTIVE:To investigate the role of matrix metalloproteinase-9 (MMP-9) and its tissue inhibitor (TIMP-1) in the pathogenesis of bronchial asthma and assess the effect of steroid treatment on MMP-9 and TIMP-1 levels. Matrix metalloproteinases are a family of zinc and calcium-dependent endopeptidases. Many MMPs such as MMP-1, MMP-2, MMP-3 are associated with asthma, in which MMP-9 is the key factor in asthma. Tissue inhibitor-1 of metalloproteinases is a specific inhibitor of MMP-9; the MMP-9 and TIMP-1 imbalance could lead to airway inflammation and remodeling in lung disease such as asthma.METHODS:Forty Wistar rats were divided into 4 groups randomly: control, asthma model 7 days (7-day group), asthma model 21 days (21-day group) and steroid treatment groups. Asthma model of rats were established by ovalbumin (OVA) sensitization and challenge with mist inhalation. The expression of MMP-9 and TIMP-1 in lung tissues was detected by immunocytochemistry, RT-PCR and Western blotting.RESULTS:(1) By observing the changes of action, tracing respiratory curves, detecting level of serum IgE level and observing the lung tissues sections, the authors demonstrated that the rat asthmatic models were successfully established. (2) The lung tissue sections of the asthma groups stained with hematoxiline and eosin (HE) showed many inflammatory cell infiltrations around the bronchioli and accompanying arterioles, hyperplasia of caliciform cells, broken bronchial mucous membrane and thickening of submucosal layer. The hyperplasia of airway smooth muscle and basement membrane were more significant in asthma model 21-day group than that in 7-day group. These changes were improved after treatment. (3) The expression of MMP-9 in rat's lung tissues: the expression was 2.71 +/- 0.37 in 7-day group, 1.76 +/- 0.27 in 21-day group, 0.88 +/- 0.18 in the treatment group and 0.52 +/- 0.10 in the control group (F = 151.52, P < 0.01). The expression of TIMP-1 in rat's lung tissues was 1.13 +/- 0.19 in the 7-day group, 1.55 +/- 0.24 in 21-day group, 0.77 +/- 0.15 in the treatment group and 0.47 +/- 0.08 in the control group (F = 69.46, P < 0.01). (4) The results of immunocytochemistry and protein expression were consistent with those of RT-PCR.CONCLUSION:The protein and mRNA expression level of MMP-9 and TIMP-1 was high in asthmatic rat's lung tissues. Down-regulation of the expression of MMP-9 and TIMP-1 by steroids may be one of the mechanisms by which airway inflammation and remodeling are inhibited in asthma.
A Study on the Effects of Cysteinyl Leukotriene Receptor Antagonist and Dexamethasone on the Transcription Level of Arginase - I in Asthmatic Mouse Models

Abstract:Objective To evaluate the intervening effects of montelukast(MK) and dexam-ethasone(DXM) on the mediator of asthma, such as Arginase- I , in asthmatic mice models by investigating the pathological features of asthma, hyperplasia of goblet cells and smooth muscles, subepithelial fibrosis and submucosal vascular proliferation. Methods BALB/c mice, sensitized with ovalbumin conjuncted with Al(OH)3, then challenged with repeated exposure to aerosolized o-valburnin. Mice were randomly divided intothe control, asthmatic, MK, and DXM group. The thickness of the smooth muscle (WAm) of intrapulmonary bronchi was measured by image analysis system, and alcianblue staining to identify airway goblet cells. mRNA levels for arginase- I were determined by RT-PCR. Results Obvious infiltration of inflammatory cells and proliferation of goblet cells and smooth muscles were demonstrated in mouse bronchus. Transcription levels of arginase-I in the lung tissue were significantly higherthan those of the control group. Compared with asthmatic group, there was mild inflammation and collagen deposition in MK-treated and DXM - treated groups while no significant differences between treated groups and control were found . Repeated exposure of allergen induced airway inflammation and remodeling . Arginase-I plays an important role in the process of airway remodeling. DXM intervention could inhibit airway remodeling by way of reducing the levels of arginase- I .

Abstract:The expression of interleukin-17 (IL-17) in lung and peripheral blood of asthmatic rats and the influence of dexamethasone, and the role of IL-17 in the pathogenesis of asthma were investigated. Thirty Sprague-Dawley (SD) adult rats were randomly divided into three groups (n=10 in each group): normal group, asthmatic group, and dexamethasone-interfered group. Rat asthmatic model was established by intraperitoneal (i.p.) injection of 10% ovalbumin (OVA) and challenge with 1% OVA via inhalation. Rats in dexamethasone-interfered group were pretreated with dexamethasone (2 mg/kg, i.p.) 30 min before each challenge. The expression of IL-17 protein in serum and bronchoalveolar lavage fluid (BALF) was detected by ELISA. The expression of IL-17 mRNA in peripheral blood mononuclear cells (PBMC) and BALF cells was semi-quantitatively detected by RT-PCR. The expression of IL-17 protein in serum and BALF of asthmatic rats was significantly elevated as compared with normal rats and dexamethsone-interfered rats (P<0.01), and there was significant difference between normal rats and dexamethsone-interfered rats (P<0.05). The expression of IL-17 mRNA in PBMC and BALF cells of asthmatic rats was markedly increased as compared with normal rats and dexamethsone-interfered rats (P<0.01), and significant difference was found between normal rats and dexamethsone-interfered rats (P<0.05). It was concluded that the expression of IL-17 was increased significantly in asthmatic rats and could be inhibited partly by dexamethasone, suggesting that IL-17 might play an important role in the pathogenesis of asthma as an inflammation regulation factor.

Abstract:Objective To discuss the effect of dexamethasone on CD4+ CD25+ regulatory T cells (Treg) and the expression of IL-4 and IL-10 in asthmatic mice.Methods Thirty male BALB/c mice were randomly assigned to the control group,the asthma group and the dexamethasone group.A mouse model of asthma was prepared by chicken ovalbumin.Flow cytometry was used to detect the percentage of CD4+ CD25+ Treg in CD4+ T cells.The expression of IL-4 from lungs were detected by immunity histochemistry.The levels of serum IL-10 were detected by ELISA.Results The percentage of CD4+ CD25+Treg and IL10 in asthma group decreased significantly (P ＜0.05).The expression of IL-4 in asthma group increased significantly (P ＜ 0.05).Conclusions Dexamethasone may had anti-inflammatory effect through the up regulation of CD4+ CD25+ Treg and Treg subsets imbalance.
The expression of ORMDL3 in murine asthma model and the effect of dexamethasone

Abstract:Objective:To investigate the role of ORMDL3 in development of bronchial asthma and its response to treatment with dexamethasone.Methods:A total of 30 Balb/c female mice were randomly divided into three groups: control group,untreated asthma group,and dexamethasone-treated group,and 10 mice in each group.The body weight was observed every week and HE-stained was used to observe the changes of pulmonary histopathology.The number of eosnophils(EOS) in peripheral blood of three groups was counted under microscope.The concentration of interlukin-4(IL-4) was determined by ELISA and the expression of ORMDL3 mRNA in lungs was detected by semi-quantitative RT-PCR.Results:Weight gain of asthma group was significantly lower than the other two groups(P < 0.05),and dexamethasone intervention alleviated the physical retardation,but still lower than the control group(P < 0.05).There were mass inflammatory cells infiltration in asthmatic group.Dexamethasone ameliorated the above mentioned symptoms significantly.The number of EOS which in treated in untreated asthma group were significantly higher than those in the other two groups(P < 0.05),and group were higher than those in control group(P < 0.05).IL-4 was increased after challenged by OVA compared to the control group(P < 0.05),and it was lower after treated by dexamethasone(P < 0.05).After challenge,the expression of ORMDL3 mRNA was a significantly elevated in untreated asthma group,and was alleviated in dexamethasone-treated group(P > 0.05),which was still higher than the control group.Conclusion:Over-expression of ORMDL3 was demonstrated in the mouse airways with asthma,and dexmathesone may be effective as an anti-inflammatory agent by inhibiting ORMDL3 expression.
Effect of Dexamethasone on Expression of AGR2 Protein in Asthmatic Mice

Abstract:This study examined the expression of the anterior gradient-2 (AGR2) protein and Muc5ac protein in the lung tissues of asthmatic mice and the effect of dexamethasone, with an attempt to explore the role of AGR2 in the over-secretion of mucus in the airway. Eighteen BALB/c mice were divided into asthma group, control group and dexamethasone group. In dexamethasone group, dexamethasone was intraperitoneally administered. Expression of AGR2 protein and Muc5ac protein in the murine lung tissues was immunohistochemically detected. IL-13 level was determined in the bronchoalveolar lavage fluid (BALF) by ELISA. The results exhibited that the expression of AGR2 protein in asthma group (0.522±0.041) was significantly higher than that in normal controls (0.361±0.047) (P<0.01) and bore a positive linear relationship to the expression of Muc5ac protein (r=0.873, P<0.05) and IL-13 level (r=0.828, P<0.05). Expression of AGR2 protein in the dexamethasone group (0.456±0.049) was significantly lower than that in the asthma group. It was concluded that: (1) the expression of AGR2 protein was significantly higher in asthmatic mice as compared with their normal counterparts; (2) the expression was obviously related to the expression of Muc5ac protein and IL-13; (3) dexamethasone could down-regulate the expression of AGR2 protein. Our findings suggested that AGR2 might be involved in the over-secretion of mucus in the airway in asthma.
[The Expression of CD25 and FOXP3 in Mouse Asthma and the Effect of Dexamethasone].

Abstract:OBJECTIVE:To detect the expression of CD25 and FOXP3 in mouse asthma model, and the effect of dexamethasone (DXM) on it. METHODS:BALB/c mice were randomly divided into three groups which were of normal control group, asthmatic group and DXM treatment group. The pathological changes of lungs were detected by HE staining. The expressions of CD25 and FOXP3 from lungs were evaluated by Western blot and RT-PCR. The expression of FOXP3 from bone marrow of asthmatic group and DXM treatment group were detected by RT-PCR. RESULTS:The expression of FOXP3 in asthmatic group and DXM treatment group was significantly higher than that of normal control group (P < 0.05), DXM could promote the expression of FOXP3 (P < 0.05). The expression level of CD25 in asthmatic group and DXM treatment group was higher than that of normal control group (P < 0.05), but no significant difference was observed between asthmatic and DXM treatment groups (P > 0.05). Expressed FOXP3 mRNA in bone marrow were detected in both asthmatic and DXM treatment groups, but higher expression level was observed in DXM treatment group. CONCLUSION:The expression of CD25 and FOXP3 increased in mouse asthma model, DXM could promote the expression. Cells in bone marrow could express FOXP3, DXM maybe promote the expression of FOXP3.
Dexmedetomidine Protects against Airway Inflammation and Airway Remodeling in a Murine Model of Chronic Asthma through TLR4/NF-κB Signaling Pathway

Abstract:Asthma is a common respiratory disease characterized by chronic airway inflammation. Dexmedetomidine (DEX), a highly selective α2 adrenergic receptor agonist, has been shown to participate in regulating inflammatory states and thus exert organ protective actions. However, the potential of DEX in asthma is still unknown. This study is aimed at investigating the role of DEX in a mouse model of house dust mite- (HDM-) induced asthma and exploring its underlying mechanism. Here, we found that DEX treatment significantly ameliorated airway hyperresponsiveness, airway inflammation, and airway remodeling in the asthmatic mice, which were similar to the efficacy of the reference anti-inflammatory drug dexamethasone. In addition, DEX reversed the increased expression of toll-like receptor 4 (TLR4) and its downstream signaling adaptor molecule nuclear factor-κB (NF-κB) in the lung tissue of asthmatic mice. Furthermore, these protective effects of DEX were abolished by yohimbine, an α2 adrenergic receptor antagonist. These results indicate that DEX is capable of ameliorating airway inflammation and remodeling in asthmatic mice, and this protective effect is associated with the inhibition of the TLR4/NF-κB signaling pathway.

Dexamethasone protects against asthma via regulating Hif-1α-glycolysis-lactate axis and protein lactylation

Abstract:Purpose: Asthma can not be eradicated till now and its control primarily relies on the application of corticosteroids. Recently, glycolytic reprogramming has been reportedly contributed to asthma, this study aimed to reveal whether the effect of corticosteroids on asthma control is related to their regulation of glycolysis and glycolysis-dependent protein lactylation. Methods: Ovalbumin (OVA) aeroallergen was used to challenge mice and stimulate human macrophage cell line THP-1 following dexamethasone (DEX) treatment. Airway hyperresponsiveness, airway inflammation, the expressions of key glycolytic enzymes and pyroptosis markers, the level of lactic acid, real-time glycolysis and oxidative phosphorylation (OXPHOS), and protein lactylation were analyzed. Results: DEX significantly attenuated OVA-induced eosinophilic airway inflammation, including airway hyperresponsiveness, leukocyte infiltration, goblet cell hyperplasia, Th2 cytokines production and pyroptosis markers expression. Meanwhile, OVA-induced Hif-1α-glycolysis axis was substantially downregulated by DEX, which resulted in low level of lactic acid. Besides, key glycolytic enzymes in the lungs of asthmatic mice were notably co-localized with F4/80-positive macrophages, indicating metabolic shift to glycolysis in lung macrophages during asthma. This was confirmed in OVA-stimulated THP-1 cells that DEX treatment resulted in reductions in pyroptosis, glycolysis and lactic acid level. Finally, protein lactylation was found significantly increased in the lungs of asthmatic mice and OVA-stimulated THP-1 cells, which were both inhibited by DEX. Conclusion: Our present study revealed that the effect of DEX on asthma control was associated with its suppressing of Hif-1α-glycolysis-lactateaxis and subsequent protein lactylation, which may open new avenues for the therapy of eosinophilic asthma.
Effect of montelukast and dexamethasone on inflammation of asthma

Abstract:Objective To study the effect of montelukast(MK)and dexamethasone(Dex)on inflammation of asthma.Methods Asthma model was established and treated with MK or Dex.Bronchoalveolar lavage fluid(BALF) and histopathologic change were observed,IL-5mRNA of lung and bone marrow cells were detected by in situ hybridization,IL-5 immunoreactive cells by immunohistochemistry,and CD34+ and CD3+ of bone marrow cells by flow cytometry. ResultsCompared with asthma group,the number of total cells and eosinophils in BALF of MK group and Dex group were significantly decreased(P<0.01),and there also existed significant differences between the two treatment groups(P<0.05).It was revealed by pathological examination that there was an extensive airway inflammation in the lung of asthma group accompanied by airway spasm,however,the inflammation of the two treament groups were significantly improved.Data of in situ hybridization suggested that the expression of IL-5mRNA in lung tissue and bone marrow cells of the two treatment groups were significantly decreased than asthma group(P<0.05,P<0.01),and there were also significant differences between the two treament groups(P<0.05).Compared with asthma group,IL-5 immunoreactive cells,CD34+ and CD3+ cells in the bone marrow of treatment groups were significantly decreased(P<0.01,P<0.05),while there was no significant difference between the two treatment groups(P>0.05). Conclusion MK and Dex can well inhibit airway inflammation and expression of IL-5mRNA in lung and bone marrow cells,though MK may be inferior to Dex in some aspects.The combined treatment of leukotriene receptor antagonist and glucocorticosteroid may be a new direction for asthma.
Effects of Dexamethasone on mast cells and tryptase of asthmatic models in guinea pigs

Abstract:Objective: To explore the effects of Dexamethasone on mast cells in lung tissue and tryptase in plasma of asthmatic model in guinea pigs . Methods: This experiment included 3 groups: normal control group, asthmatic model group, Dexamethasone treatment group, 10 guinea pigs involved in each group. Asthmatic models in guinea pigs were constructed by immunization of intraperitoneal injection with ovalbumin(OVA) and challenge of inspiration with nebulized OVA. Mast cells were observed under a microscope and tryptase levels were measured by antibody against tryptase with Uni CAP 100 system. Results: The numbers of mast cells and the percentage of degranulation of mast cells in lung tissue and the plasma levels of tryptase of asthmatic model group in guinea pigs [14.87±4.67, 42.85±5.99%and 119.78±12.50] were significantly higher than those of normal control group[2.38±0.52, 24.91±9.79%and 88.78±4.74], P<0.01. While the above indices in the dexamethasone group[6.63±1.41, 25.33±6.14%and 92.10±7.11]were significantly lower than those of the asthmatic model group(P<0.01). Conclusion: Mast cell tryptase is concerned with asthmatic process ; dexamethasone could resist inflammation, which is achieved by restraining release of mast cell tryptase.

Abstract:Objective To observe the levels of lymphocyte mitochondrial membrane potential(△·ψm) and Fas/FasL change in lymph,and the effect of dexamethasone on it.Methods The establishment of asthma model.After the inspired collection 2 h,6 h,12 h,24 h,48 h lymph,blood,bronchoalveolar lavage fluid(BALF) of lymphocytes by flow cytometry(FCM) detection of lymphocyte mitochondria transmembrane potential(△·ψm),cell immunohistochemistry(SABC method) was used to detect cell surface Fas FasL protein expression.Results The peak of mitochondrial transmembrane potential in the asthma lymph,blood,BALF lymphocytes was lower compared with the control group and treatment group(P0.001).There was no significant difference between the control and treatment groups(P0.05).The Fas,FasL protein optical density on the surface of lymphocytes in the asthma lymph,BALF was obvious lower than that of the control and treatment group,and there was no significant difference between the control group and treatment group.Conclusion The early lymphocyte apoptosis obstacles and the low of Fas / FasL expression were found in asthma rat in lymph,blood and BALF.Particularly,the degree of T lymphocyte apoptosis obstacles and low of Fas / FasL in lymph were remarkable to the blood and BALF in asthma.Dexamethasone may enhance the cell surface through the Fas / FasL expression to accelerate the apoptosis of lymphocytes in asthma.
Effects of Methylprednisolone on Expression of MMP-9 and Airway Inflammation in Murine Models of Asthma

Abstract:Objective To investigate the effects of methylprednisolone(MP) on the expression of matrix metalloproteinase(MMP)-9 and airway inflammation in murine models of asthma. Methods Thirty female BALB/c mice were randomly divided into asthma group,MP group and control group(n=10).Murine models of acute asthma were established by ovalbumin(OVA) via peritoneal injection and intranasal instillation.The pathological changes of lung tissues were observed with HE staining,and cell quantitation was conducted in bronchalveolar lavage fluid(BALF).The expression of MMP-9 protein was determined by immunohistochemistry and gelatin zymogram,and the expression of MMP-9 mRNA was detected by RT-PCR. Results Compared with control group,there were more significant airway spasm and more infiltration of inflammatory cells in histologic examination,and there was higher eosinophil cell quantitation in BALF in asthma group(P0.01).Compared with asthma group,there were less airway spasm and less infiltration of inflammatory cells in histologic examination,and there was lower eosinophil cell quantitation in BALF in MP group(P0.05).It was revealed by immunohistochemistry and gelatin zymogram that the expression of MMP-9 protein was higher in asthma group than that in control group.It was indicated by RT-PCR that the expression of MMP-9 mRNA was increased in asthma group,while there was less expression in MP group than in asthma group. Conclusion The increased expression of MMP-9 in lung tissues of murine models of asthma may be one of the mechanisms for inflammation of asthma.MP may inhibit the inflammation of asthma through suppressing the expression of MMP-9.

Abstract:AIM:To observe the expression of CD4(+) CD25(+) Foxp3(+) Treg, IL-10 and TGF-beta in lymph and blood, as well as the effect of dexamethasone on it and investigate the relationship between them. METHODS:Lymph and blood samples of 0 h, 24 h, 48 h after the last challenge were collected from the rat model of asthma. The percentage of CD4(+) CD25(+) Foxp3(+) Treg were detected by flow cytometer (FCM), while the levels of IL-10 and TGF-beta were determined by ELISA. RESULTS:The percentage of CD4(+) CD25(+) Foxp3(+) Treg in the blood and the levels of IL-10 and TGF-beta in the plasma in cases of asthma group were significantly lower than that of the control and therapy group at different time points (P<0.05); The percentage of CD4(+) CD25(+) Foxp3(+) Treg and the level of IL-10 in lymph were significantly higher than that of blood (P<0.05), however, the level of TGF-beta in lymph were significantly lower than that of blood (P<0.05); There were no significant differences which the percentage of CD4(+) CD25(+) Foxp3(+) Treg in the blood and the levels of IL-10 and TGF-beta in the plasma between control group and therapy group (P>0.05). CONCLUSION:There is a disbalance both quantity and function of CD4(+) CD25(+) Foxp3(+) in the lymph of bronchial asthmatic rat, and the percentage of CD4(+) CD25(+) Foxp3(+) Treg in lymph are significantly higher than that of blood, which dexamethasone may effectively increase the percentage of CD4(+) CD25(+) Foxp3(+) Treg in bronchial asthmatic rat.
VCAM-1 Expression, Eosinophil Infiltration, and Pharmacological Modulation in Rat Allergic Airway Inflammation.

Abstract:AIM:To determine vascular cell adhesion molecule-1 (VCAM-1) expression and eosinophil infiltration in the lungs of the rats with allergic airway inflammation, and their possible modulation by dexamethasone and neurokinin-1 receptor antagonist SR140333. METHODS:Sensitized rats were challenged with inhalation of 1 % ovalbumin aerosol. Protein expression of VCAM-1 in the lungs and eosinophil infiltration around bronchi in different groups were determined 24 h after challenge. SR140333 (0.01 and 0.10 mg/kg) and dexamethasone (0.50 mg/kg) were injected ip, twice a day, for 3 d before challenge. RESULTS:The expression of VCAM-1 was increased in the sensitized rat lungs as compared with the non-sensitized rats (P<0.05). The increment was inhibited by pretreatment with dexamethasone (P<0.05), but not with SR140333 (P>0.05). On the other hand, antigen challenge in sensitized rats evoked eosinophil infiltration (P<0.05) but no further increase in VCAM-1 expression (P>0.05). Furthermore, SR140333 inhibited eosinophil infiltration (P<0.05) but had no effect on VCAM-1 expression (P>0.05), whereas dexamethasone inhibited both responses (P<0.05). CONCLUSION:The expression of VCAM-1 increases during antigen sensitization in rat lungs, and dexamethasone and SR140333 may inhibit allergic airway inflammation in different mechanisms.
