Abstract:In this paper, a novel method is presented for creating a 3-dimensinal sub-diffraction effective observation volume based on microsphere photonic nanojet and fluorescence depletion effect. Using microsphere to achieve photonic nanojet focusing effect, the proposed method applies the radially polarized plane wave to excite fluorescence and the azimuthally polarized beam to obtain fluorescence depletion field. The effective detection volume of photonic nanojet can reach 0.002μm3 (2aL). Compared with conventional confocal microscopy, this effective detection volume represents a reduction of almost 2 orders of magnitude. With simple configuration based on cost-effective microspheres, the proposed method is theoretically proved to be a potential tool for the fluorescence correlation spectroscopy (FCS) to have large analysis range and to investigate single molecule at high concentrations.

Abstract:An axial-resolution-enhanced two-photon laser scanning microscopy system is presented in this paper. In the proposed method, we use a spatial light modulator (SLM) for the phase modulation of the excitation light. The axially split point spread function (PSF) is generated by loading a 0-π pattern on the SLM. The final quality-enhanced images are acquired by subtracting the two consecutive images acquired by the original PSF and the split PSF. Because of the fluorescence differential processing, the axial elongation of the particles images is suppressed, and the axial resolution is enhanced accordingly. With the axial resolution enhanced, the overlap between layer images is also reduced, which decreases the background noise of the images and enhances the contrast and image quality of the acquired fluorescence images. The capability of axial resolution and contrast enhancement is successfully demonstrated by both theoretical calculation and experimental results.

Abstract:In a two-photon excitation fluorescence volume imaging (TPFVI) system, an axicon is used to generate a Bessel beam and at the same time to collect the generated fluorescence to achieve large depth of field. A slice-by-slice diffraction propagation model in the frame of the angular spectrum method is proposed to simulate the whole imaging process of TPFVI. The simulation reveals that the Bessel beam can penetrate deep in scattering media due to its self-reconstruction ability. The simulation also demonstrates that TPFVI can image a volume of interest in a single raster scan. Two-photon excitation is crucial to eliminate the signals that are generated by the side lobes of Bessel beams; the unwanted signals may be further suppressed by placing a spatial filter in the front of the detector. The simulation method will guide the system design in improving the performance of a TPFVI system.

Abstract:We propose a novel fluorescence emission difference microscopy (FED) system based on focusing cylindrical vector beams. In conventional FED, a Gaussian beam and a 0–2π vortex phase plate are used to generate solid and hollow spots. We focus radially polarized and azimuthally polarized cylindrical vector beams to obtain an expanded solid spot and a shrunken hollow spot, taking advantage of the optical properties of cylindrical vector beams to improve the conventional FED performance. Our novel method enhances FED performance because the hollow spot size determines the FED resolution and an expanded solid spot effectively reduces negative side-lobe emergence during image processing. We demonstrate improved performance theoretically and experimentally using an in-house built FED. Our FED achieved resolution of less than λ/4 in test images of 100nm nanoparticles, better than the confocal image resolution by a factor of approximately 1/3. We also discuss detailed simulation analyses and FED imaging of biological cells.

Methods and Applications in Fluorescence

Abstract:Fluorescent samples typically emit isotropically in all directions. Large lenses and other optical components are needed to capture a significant fraction of the emission, and complex confocal microscopes are required for high resolution focal-plane imaging. It is known that Bessel beams have remarkable properties of being able to travel over long distances, over 1000 times the wavelength, without diverging, and hence are called non-diffracting beams. In previous reports the Bessel beams were formed by an incident light source, typically with plane-wave illumination on a circular aperture. It was not known if Bessel beams could form from fluorescent light sources. We demonstrate transformation of the emission from fluorescent polystyrene spheres (FPS) into non-diverging beams which propagate up to 130 mm (13 cm) along the optical axis with a constant diameter. This is accomplished using a planar metal film, with no nanoscale features in the X-Y plane, using surface plasmon-coupled emission. Using samples which contain many FPS in the field-of-view, we demonstrate that an independent Bessel beam can be generated from any location on the metal film. The extremely long non-diffracted propagation distances, and self-healing properties of Bessel beams, offer new opportunities in fluorescence sensing and imaging.
Strong Confinement of Two-Photon Excitation Field by Photonic Nanojet with Radial Polarization Illumination

Abstract:The fluorescence excitation field by photonic nanojet has been investigated, where a SiO2 microsphere is illuminated by the radial polarized beam. We show that radial polarization illumination can achieve very strong three-dimensional confinement of photonic nanojet below the diffraction limit, especially the longitudinal light field. It is also noted that the location of maximum excitation field intensity spot is not almost change by wavelength, but the volume of excitation field is depended on wavelength. Compared to single-photon excitation, two-photon excitation based on photonic nanojet can obtain the least illumination volume, with lateral FWHM≈λ/4 and axial FWHM<λ/10. This offers a broad range of application in single-molecule detection, ultra-resolution microscopy and nanopatterning.

Abstract:Optical vortices have developed rapidly in recent times owing to their theoretical significance and attractive potential applications in extensive realms. Composite optical vortices using two or more coherently superimposed vortex beams have been widely researched. Here, we propose a novel beam pattern incoherently generated by laser beams with two distinct wavelengths. We apply this beam pattern in fluorescence super-resolution imaging based on the stimulated emission effect, and find that the size of the resulting dark spot in the focal plane lies below the diffraction limit. Crucial factors affecting the sub-diffraction dark spot, such as beam intensities, spatial displacement, pulse width, polarization, and aberration, are studied in detail. We believe that the results are beneficial for experimental studies and will serve as useful references in other circumstances where they may be utilized.

Abstract:Three dimensional distributions of cells can be usually acquired by optical sectioning methods, such as multiphoton excitation and confocal fluorescence laser scanning microscopy. Though the lateral scan rates can reach up to several kHz, the relatively slow axial scan comprises the speed of real-time imaging of a volume. Here we propose a three dimensional imaging method that uses Bessel beams as excitation in multiphoton fluorescence microscopy. The extended focus of the Bessel beam allows recording a volume of cells without scanning the depth. The depth information can be retrieved by recording a pair of parallax views of the same volume. We have demonstrated the stereoscope capability on a homebuilt two-photon fluorescence microscope.

Abstract:We propose a novel physical mechanism for breaking the diffraction barrier in the far field. Termed fluorescence emission difference microscopy (FED), our approach is based on the intensity difference between two differently acquired images. When fluorescence saturation is applied, the resolving ability of FED can be further enhanced. A detailed theoretical analysis and a series of simulation tests are performed. The validity of FED in practical use is demonstrated by experiments on fluorescent nanoparticles and biological cells in which a spatial resolution of <λ/4 is achieved. Featuring the potential to realize a high imaging speed, this approach may be widely applied in nanoscale investigations.

In imaging applications, Bessel beams have been successfully used to provide extended focuses for volumetric imaging and uniformed illumination plane in light-sheet microscopy. Coupled with two-photon excitation, Bessel beams have been successfully used in realizing fluorescence projected volumetric imaging. We demonstrated previously a stereoscopic solution-two-photon fluorescence stereomicroscopy (TPFSM)-for recovering the depth information in volumetric imaging with Bessel beams. In TPFSM, tilted Bessel beams were used to generate stereoscopic images on a laser scanning two-photon fluorescence microscope; upon post image processing we could successfully provide 3D perception of acquired volume images by wearing anaglyph 3D glasses. However, tilted Bessel beams were generated by shifting either an axicon or an objective laterally; the slow imaging speed and severe aberrations made it hard to use in real-time volume imaging. In this article, we report recent improvements of TPFSM with newly designed scanner and imaging software, which allows 3D stereoscopic imaging without moving any of the optical components on the setup. This improvement has dramatically improved focusing qualities and imaging speed so that the TPFSM can be performed potentially in real-time to provide 3D visualization in scattering media without post image processing.

Abstract:Bessel beams have been successfully used in two-photon laser scanning fluorescence microscopy to extend the depth of field (EDF), which makes it possible to observe fast events volumetrically. However, the depth information is lost due to integration of fluorescence signals along the propagation direction. We describe the design and implementation of two-photon lasers scanning stereomicroscopy, which allows viewing dynamic processes in three-dimensional (3D) space stereoscopically in real-time with shutter glasses at the speed of 1.4 volumes per second. The depth information can be appreciated by human visual system or be recovered with correspondence algorithms for some cases.

Abstract:Bessel foci used for two-photon fluorescence excitation have enabled high-speed volumetric imaging. At high numeric aperture, their imaging performances are compromised by substantial side-ring excitation and suffer from reduced image contrast. Here, we describe axially extended Bessel-droplet foci with suppressed side rings and more resistance to optical aberrations. Applying novel phase patterns to generate Bessel-droplet foci of variable NAs at high power throughput, we achieved continuous volume imaging by scanning them interferometrically along the axial direction. With Bessel-droplet foci, we demonstrated high-resolution volumetric imaging of synaptic anatomy and function as well as lymphatic circulations in the mouse brain in vivo.

Effective axial scanning is achieved by incorporating a Fresnel lens in the system, which has large chromatic aberration and can therefore focus the excitation beam to different axial positions depending on its wavelength. Abstract:Bessel beam propagation in scattering media is simulated using the angular spectrum method combined with slice-by-slice propagation model. Generating Bessel beams with a spatial light modulator, which provides a means to adjust flexibly the parameters of the Bessel beam, allows us to validate the simulation results experimentally. The study reveals that the self-reconstructing length changes oppositely with the axicon angle (i.e., the larger the axicon angle, the shorter the self-reconstructing length). The radius of the incident beam has little influence on the self-reconstruction of the Bessel beam central lobe.

Abstract:Nonlinear refraction of a thick sample is investigated using a modified transmittance measurement technique with a phase object. Nonlinear refraction can be investigated using a single-shot beam in this technique, and various optical nonlinearities are investigated. Numerical simulation is obtained by solving the electromagnetic wave equations, and agrees with experimental results.

Abstract:Diffraction imaging of scattered light allows extraction of information on scatterer's morphology. We present a method for accurate simulation of diffraction imaging of single particles by combining rigorous light scattering model with ray-tracing software. The new method has been validated by comparison to measured images of single microspheres. Dependence of fringe patterns on translation of an objective based imager to off-focus positions has been analyzed to clearly understand diffraction imaging with multiple optical elements. The calculated and measured results establish unambiguously that diffraction imaging should be pursued in non-conjugate configurations to ensure accurate sampling of coherent light distribution from the scatterer.

Abstract:We report a dual-contrast method of simultaneously measuring and visualizing the volumetric structural information in live biological samples in three-dimensional (3D) space. By introducing a direct way of deriving the 3D scattering potential of the object from the synthesized angular spectra, we obtain the quantitative subcellular morphology in refractive indices (RIs) side-by-side with its fluorescence signals. The additional contrast in RI complements the fluorescent signal, providing additional information of the targeted zones. The simultaneous dual-contrast 3D mechanism unveiled interesting information inaccessible with previous methods, as we demonstrated in the human immune cell (T cell) experiment. Further validation has been demonstrated using a Monte Carlo model. (C) 2019 Chinese Laser Press.

Abstract:While simultaneous phase-contrast and two-photon fluorescence imaging in microscopy can bring abundant biomedical information, it is difficult to retrieve phase information from conventional two-photon microscopes. To realize low-cost, in situ phase-contrast and two-photon fluorescence imaging, we propose Schlieren two-photon microscopy, a method that implements phase-contrast imaging on two-photon microscopes. This method involves spatially modulated fluorescence plates, which are made of two-photon fluorescence dyes or upconversion nanoparticles. We demonstrate that the fluorescence intensity fluctuation reflects the phase gradients of the specimen via theoretical analysis, simulations, and experiments. The proposed method is fully compatible with commercial two-photon microscopes, thus enabling widespread applications in live tissue imaging. (C) 2018 Optical Society of America
