Abstract:The camphor tree (Cinnamomum camphora (L.) Presl.) is the representative species of subtropical evergreen broadleaved forests in eastern Asia and an important raw material for essential oil production worldwide. Although MYBs have been comprehensively characterized and their functions have been partially resolved in many plants, it has not been explored in C. camphora. In this study, 121 CcMYBs were identified on 12 chromosomes in the whole genome of C. camphora and found that CcMYBs were mainly expanded by segmental duplication. They were divided into 28 subgroups based on phylogenetic analysis and gene structural characteristics. In the promoter regions, numerous cis-acting elements were related to biological processes. Analysis of RNA sequencing data from seven tissues showed that CcMYBs exhibited different expression profiles, suggesting that they have various roles in camphor tree development. In addition, combined with the correlation analysis of structural genes in the flavonoid synthesis pathway, we identified CcMYBs from three subgroups that might be related to the flavonoid biosynthesis pathway. This study systematically analyzed CcMYBs in C. camphora, which will set the stage for subsequent research on the functions of CcMYBs during their lifetime and provide valuable insights for the genetic improvement of camphor trees.

Abstract:3-hydroxy-3-methyl-glutaryl-CoA reductase (3-hydroxy-3-methyl glutaryl coenzyme A reductase,HMGR) is one of the key rate-limiting enzymes in mevalonic (MVA) pathway of terpenoid biosynthesis.To analysis the function of HMGR gene in terpenoid biosynthesis in Chamaemelum nobile,a HMGR gene (designated as CnHMGR,GenBank accession number KU589282) was cloned from C.nobile using RT-PCR method.The fulllength cDNA of CnHMGR gene was 1 856 bp and contained an open reading frame (ORF) of 1 746 bp,which encodes a 582 amino-acid protein.The theoretical molecular weight and PI of the CnHMGR were 62.5 kDa and 6.80,respectively.Multi-alignment comparison analysis showed the protein sequence of CnHMGR had high similarity with those of HMGR proteins from other plants.Furthermore,CnHMGR has two HMG-CoA-binding motifs:TTEGCLVA,EMPVGYVQIP,and two NADP(H)-binding motifs:TVGGGT,DAMGMMM,suggesting CnHMGR is one of HMGR family mumbers in C.nobile.The results of tissue expression analysis showed that CnHMGR was constituently expressed indifferent tissues of C.nobile,with highest expression level in flowers and lowest expression level in stems.The present study cloned,characterized and analyzed the tissue expression pattern of CnHMGR in C.nobile,providing basic data for further studying the function of CnHMGR in sesquiterpene biosynthesis of C.nobile.

Abstract:In order to study the resistance function of mango of a cinnamoy-CoA reducase ( CCR) gene.CCR was cloned from mango with 3′RACE and 5′RACE method.The characteristics of cDNA sequence was analyzed by software such as DNAMAN ,TMHMM.The bioinformatics analysis on the obtained sequence was showed:CCR gene of mango contains coding region ,3′and 5′untranslated region of length cDNA sequence of 1 292 bp,encoding 305 a-mino acid.The gene without transmembrance by transmembrance domains predicted .The secondary structure of CCR protein had random coil and beta helix .It was found that the gene encoding for the protein had close relation-ship with American aspen wood ,oil-tea camellia ,other distantly relationship with lily ,and rubber tree through phy-logenetic analysis .The CCR gene was cloned from mango and the bioinformatics analysis , laid the groundwork for the follow-up work of transgenic .

Abstract:As a house-keeping gene,Actin has been always being used as an internal standard to normalize mRNA levels between different samples by quantitative real-time PCR, thus plays an important role in gene expression analysis. In this research, through a method of homology cloning, a pair of degenerate primers were designed based on the conserved sequences of Actin genes from other plants submitted to GenBank, and then 4 cDNA fragments from Cinnamomum camphora were obtained using RT-PCR. Molecular biological analysis showed that, each of the 4 cDNA fragments was 998 bp and encoded a putative protein of 332 amino acids. Moreover, according to the homology analysis, the 4 cDNA fragments belonged to Actin subfamily, and they were named CcACTa,CcACTb,CcACTc and CcACTd, and deposited in GenBank (Accession number:KM086736 KM086737 KM086738 and KM086739). Quantitative real-time PCR results revealed that the expression level of CcACTc in different organs such as root, stem, leaf and leaves under low temperature treaments was relatively stable. It could serve as a candidate reference gene.

Abstract:Cinnamoyl CoA reductase (CCR: EC 1.2.1.44), the entry-point enzyme of the lignin specific biosynthetic pathway, catalyzes the conversion of cinnamoyl CoA esters to their corresponding cinnamaldehydes. Multiple sequence alignment showed that the deduced polypeptide shared 70% similarity and 30% sequence identity at the amino acid level with defined CCR genes from other plant species and they all contain the common signature sequences thought to be the catalytic site as well as the putative NADP binding domain. Using a conserved OsCCR cDNA fragment as the probe for library screening, we isolated the genomic DNA that covered the whole coding region of OsCCR with total length of 3045 bp including 4 introns and 5 exons. The open reading frame for our OsCCR gene contains 337 amino acids. Northern blot indicated that OsCCR was expressed in different organs with the highest level found in stems. In situ hybridization results showed that OsCCR mRNA was localized mainly along the vascular bundles in stems and leaves, and also in lateral roots that was differentiating from the tillering node. We conclude that the vascular-localized expression of OsCCR gene may suggest its possible involvement in lignin biosynthesis. Cloning and characterization of OsCCR will help to clarify how lignifications in plants are regulated and will provide a physical basis for creating genetically engineered rice plants with optimal lignin contents.

Transient expression of recombinant plasmidCsbHLH2/PBI221-GFPinArabidopsisprotoplasts showed that CsbHLH2was located in cell nuclei.Quantitative real-time PCR analysis of the expression profiles showed that the CsbHLH2gene wasexpressed in bud, leaf,stem and root.The highest expression level of theCsbHLH2wasfound in the youngleaf.Different hormone treatmentsresults showedthattheCsbHLH2was induced by ETH, MEJA and SA treatment, respectively.

Abstract:In order to elucidate the molecular mechanism of coconut's cold resistance,this study designed primers according to the CBF gene sequence of oil palm,and then amplified the genomic DNA of Cocos nucifera by using PCR method.The PCR products were retrieved,linked,transformed and sequenced;finally,a DNA sequence of Cocos nucifera was obtained.This sequence had the highest identity(99%) with CBF gene of oil palm,and it was confirmed to be CBF gene.Through the clustering analysis of the cloned CBF gene of coconut and CBF genes of graminaceous crops,the high homology was found between the cloned CBF gene and HvCBF1,OsDREB1E,HvCBF11,TaCBF11.

Abstract:Cinnamomum species attract attentions owing to their scents, medicinal properties, and ambiguous relationship in the phylogenetic tree. Here, we report a high-quality genome assembly of Cinnamomum camphora, based on which two whole-genome duplication (WGD) events were detected in the C. camphora genome: one was shared with Magnoliales, and the other was unique to Lauraceae. Phylogenetic analyses illustrated that Lauraceae species formed a compact sister clade to the eudicots. We then performed whole-genome resequencing on 24 Cinnamomum species native to China, and the results showed that the topology of Cinnamomum species was not entirely consistent with morphological classification. The rise and molecular basis of chemodiversity in Cinnamomum were also fascinating issues. In this study, six chemotypes were classified and six main terpenoids were identified as major contributors of chemodiversity in C. camphora by the principal component analysis. Through in vitro assays and subcellular localization analyses, we identified two key terpene synthase (TPS) genes (CcTPS16 and CcTPS54), the products of which were characterized to catalyze the biosynthesis of two uppermost volatiles (i.e. 1,8-cineole and (iso)nerolidol), respectively, and meditate the generation of two chemotypes by transcriptional regulation and compartmentalization. Additionally, the pathway of medium-chain triglyceride (MCT) biosynthesis in Lauraceae was investigated for the first time. Synteny analysis suggested that the divergent synthesis of MCT and long-chain triglyceride (LCT) in Lauraceae kernels was probably controlled by specific medium-chain fatty acyl-ACP thioesterase (FatB), type-B lysophosphatidic acid acyltransferase (type-B LPAAT), and diacylglycerol acyltransferase 2b (DGAT 2b) isoforms during co-evolution with retentions or deletions in the genome.

Abstract:Abstract In this study, two chlorophyll A/B binding protein (CAB) genes ( CsCP1 and CsCP2 ) in tea plant were cloned. The proteins encoded by these genes belong to the external or internal antenna proteins of PS II, respectively. They may be the targets of physiological regulation for tea leaf cell PS II because they all contain multiple functional domains and modifiable sites. The CAB gene family in the tea genome consists of 25 homologous genes. We measured the expression patterns of ten genes in the CsCP1 and CsCP2 subfamily under six different stresses. CsCP1 expression was inhibited in response to 6 kinds of stress; CsCP2 expression was slightly upregulated only after cold stress and ABA treatment. However, the expression levels of CSA016997 and CSA030476 were upregulated significantly in the six stresses. The results suggested that the 10 CAB genes may have different functions in tea leaves. Moreover, changes in the expression of the 10 genes under stress appear to be related to ABA- and MeJA-dependent signalling pathways, and their responses to MeJA treatment is faster than those to ABA. In addition, we introduced our experiences for cloning the genes in the context of complex genomes.

Abstract:Cinnamoyl-CoA reductaseis the first rate limiting emzyme which catalyzes the biosynthesis of lignin and plays an important role in lignin biosynthesis.Two cinnamoyl-CoA reductase encoding genes,CiCCR2 and CiCCR3,were cloned from Caragana intermedia through PCR.The open reading frame(ORF) of CiCCR2 is 897bp which encoding 299 amino acids,and ORF of CiCCR3 is 966bp,which encoding 322 amino acids.Quantification assay and Histochemical staining showed that the lignin content increased by overexpressing of CiCCR2 and CiCCR3 in Arabidopsis compared with the wild type during both seedling and mature stages.At the same time,the fresh and dry weight of overexpressing plants were higher than that of the wild type.

Abstract:【Objective】The aim of this study was to clone full-length cDNA of a key enzyme gene DLCCoAOMT related to lignin metabolism in longan,investigate its sequence characteristics and analyze its expression in different organs under low temperature stress,as well as expression in the prokaryote.【Method】 The DLCCoAOMT gene cDNA sequence was cloned from longan leaf using RT-PCR and RACE techniques.The bioinformatics methods were used to analyze putative amino acid sequence and real-time PCR method were used to study the expression of DLCCoAOMT gene in different tissues.【Result】 The full-length cDNA of DLCCoAOMT(GenBank accession number: JN093023) in longan was cloned.The sequence consists of 993 bp with an open reading frame of 744 bp,encoding a polypeptide of 247 amino acids.Homology analysis showed that the deduced DLCCoAOMT protein was highly homologous to other CCoAOMT proteins from different species.Phylogenetic analysis also indicated that DLCCoAOMT was very closely related to DLCCoAOMT of Betula.Real-time PCR results showed that the DLCCoAOMT expressed in root,stem and leaf,and its expression was different among three organs.The mRNA of DLCCoAOMT was abundant in root and stem,but less in leaf.Furthermore,DLCCoAOMT transcription level was significantly different among root,stem and leaf with time courses of chilling treatment.Prokaryotic expression showed that recombinant plasmid was efficiently expressed in Escherichia coli BL21.【Conclusion】 Gene DLCCoAOMT was firstly isolated and characterized from longan,which may be involved in chilling stress.
[Cloning and Characterization of a Homologous Gene of Plant Class V Chitinase from Balsampear, Momordica Charantia Linn].

Abstract:Balsampear (M. charantia Linn.) is a vegetable crop, highly resistant to pathogens. Chitinases were proposed to play an important role in the defense response of this crop. Based on the N-terminal sequence of a purified balsampear chitinase, a fragment (ChitB), similar to the tobacco class V chitinase gene, was amplified from the leaf RNA using 3'RACE, and the corresponding 5' sequence was further amplified by the Y-RACE method. By joining the two amplified fragments, the full-length cDNA of M. charatica homologous gene of plant class V chitinase (McChi5) was obtained. The 1348 bp cDNA contained an ORF of 1044 bp, which coded for a polypeptide of 347 amino acids. The deduced polypeptide had a predicted molecular weight of 38.3 kD and a pI of 5.77. Homology analysis demonstrated that, McChi5 protein, which contained a conserved domain of family 18 glycosyl hydrolyse, had the sequence similar to tobacco class V chitinases, several putative chitinases and chitinase-like proteins of Arabidopsis thiliania, and some chitinases from mammals, insects and bacteria. Southern blotting suggested that two copies of McChi5 gene and several homologous genes existed in the M. charatica genome. By RNA dot blotting analysis, expression of the McChi5 gene was detected in cotyledons, roots, stems, and leaves, and it was not induced by wounding treatment. The biological functions and the potential applications of Mochi5 gene were discussed.

Abstract:A cDNA fragment encoding a putative protein of elongat ion factor 1 alpha (EF1a) was ifrstly isolated from Cinnamomum camphora by using homology cloning, A pair of degenerate primers was designed based on the conserved sequences of other EF1a genes from the different species from GenBank, and a fragment of 1 297 bp was obtained through Reverse Transcription Polymerase Chain Reaction (RT-PCR) techniques. The results of homologous alignment show that CcEF1a shared over 80%nucleotide sequence identity and over 96%amino acid sequence identity with EF1a genes in other plants that had been submitted to GenBank. The cDNA fragment sequence was named CcEF1a and registered in GenBank with accession number KM086740. According to the results of real-time quantitative PCR, the mRNA level of CcEF1a in leaves under chilling stress was identical. It could be used as an interrnal control in real-time quantitative PCR.

Abstract:AOC (allene oxide cyclase; EC 5.3.99.6), an essential enzyme in jasmonic acid and its methyl ester biosynthesis, was cloned from Camptotheca acuminata (named as CaAOC), a native medicinal plant species in China. CaAOC had significant similarity at the amino-acid level with AOCs from other plant species. Comparison between the sequences of the full-length cDNA and genomic DNA of CaAOC revealed that the genomic DNA of CaAOC contained an 89-bp intron and a 240-bp intron. Southern-blot analysis indicated that CaAOC was a multiple-copy gene, and real-time quantitative PCR analysis showed that CaAOC was expressed constitutively in all organs tested, with the highest expression level in leaves. The results from treatment experiments using different signalling components, including methyl jasmonate, abscisic acid, salicylic acid and H(2)O(2), revealed that expression of CaAOC had a prominent diversity. Heavy metal (copper) significantly enhanced CaAOC expression, whereas wounding (induced by UV-B) was not so effective.

Abstract:A full-length cDNA sequence of a homologous Cab gene was cloned by employing RT-PCR, which was named as Mcab. The accession code in GenBank is FJ907952. The Mcab is 915 bp in length, encoding a protein of 264 amino acids, with an estimated molecular weight and an isoelectric point of 28.19 ku and 5.35 respectively. A comparison of the nucleotide sequences of homologous Cab genes from different species indicated that Mcab gene had a range of 72% to 85% identity in nucleotide sequence with homologues of other plants. The results indicated that Mcab protein had one chlorophyll a/b binding domain between the 63 st to 232 st amino acid and very conservative of the position between woody plants. Protein subcellular localization prediction showed that Mcab protein was located in the chloroplast and the gene was one of important genes in photosynthesis system of green plants. Prediction of the secondary structure of the protein showed that Mcab protein had 8 α helices, 5 β sheets and 21 β turns. The research was beneficial to the further understanding of the molecular mechanism of photosynthesis in mango.

Abstract:We cloned a Cinnamoyl-CoA Reductase gene (BpCCR1) from an apical meristem and first internode of Betula platyphylla and characterized its functions in lignin biosynthesis, wood formation and tree growth through transgenic approaches. We generated overexpression and suppression transgenic lines and analyzed them in comparison with the wild-type in terms of lignin content, anatomical characteristics, height and biomass. We found that BpCCR1 overexpression could increase lignin content up to 14.6%, and its underexpression decreased lignin content by 6.3%. Surprisingly, modification of BpCCR1 expression led to conspicuous changes in wood characteristics, including xylem vessel number and arrangement, and secondary wall thickness. The growth of transgenic trees in terms of height was also significantly influenced by the modification of BpCCR1 genes. We discuss the functions of BpCCR1 in the context of a phylogenetic tree built with CCR genes from multiple species.

Abstract:The cDNA-AFLP was applied to identify genes expressed differentially in cold acclimatized tea plant. A cDNA fragment, TDF11 encoding a CBF (C-repeat binding factor) protein was isolated and identified. The cDNA fragment had 94%、84%、81% homology with CBF genes from Brassica napus, Arabidopsis thaliana, Nicotiana tabacum by BLAST on NCBI. The complete cDNA sequence was cloned by RACE (rapid amplification of cDNA ends), named Camellia sinensis CBF gene. Its full cDNA sequence was 981bp (GenBank accession number EU857638) and contained an open reading frame encoding a polypeptide of 275 amino acid residues. Analysis of the nucleotide sequence and deduced amino acid sequence showed 74%、57%、57% homology with CBF genes from Brassica napus, Nicotiana tabacum, Arabidopsis thaliana. To identify differential expression of CBF gene, RT-PCR experiments were performed in six samples including leaves cold-induced for different treatments. The results showed that tea CBF gene expressed after cold induced for 4 hours and up to a maximum when cold induced for 8 h, and then decreased. It hinted the CBF gene might play a key role during the development of cold resistant molecular mechanism in tea plant.
Cloning,Sequence Analysis and Induced Expression of a Calmodulin Gene MCaM-3 From Mulberry

Abstract:Calmodulin(CaM) is a multifunctional Ca2+-binding protein in eukaryotic cells.It plays important roles in regulating the growth and development,environment adaptation and stress tolerance in plants.A full-length cDNA sequence coding for CaM in mulberry,designated MCaM-3(GenBank accession number GQ303247),was cloned based on expressed sequence tags from mulberry tree.Sequence analysis showed that the MCaM-3 is 951 bp long and contains a 143 bp 5′-UTR(untranslated region) and a 358 bp 3′-UTR.Its open reading frame(ORF) is of 450 bp,encoding 149 amino acids with a predicted molecular weight of 16.85 kD and an isoelectric point of 3.95.Online SMART analysis showed that the protein of MCaM-3 has four Ca2+-binding functional domains,being able to bind calcium ion.Homology analysis revealed that,CaM genes are highly conservative in mulberry and other species including Arabidopsis,poplar,soybean,wheat,rice and maize.Phylogenetic analysis based on MCaM-3 and CaM genes with other 19 species showed that mulberry shows closer relationships with castor oil plant,tobacco,rhubarb and cherry.The results of semi-quantitative RT-PCR analysis showed that the transcriptional level of MCaM-3 mRNA increased significantly under the conditions of low temperature,drought and salt stress respectively compared to the normal growth environment,suggesting its involvement in general response to abiotic stresses in mulberry.
