Abstract:Sir, The article published recently by Sevilla et al. (2016) provides compelling support for MORC2 as a novel pathogenic gene of axonal Charcot–Marie–Tooth (CMT2) disease. The authors identified two MORC2 mutations c.74C > T (p.S25L) and c.568C > T (p.R190W) in a cohort of Spanish CMT2 patients. With the aim to explore how this newly discovered gene mutated in Chinese CMT2 patients, we detected MORC2 mutations in genetically unidentified CMT2 patients, and would like to report two novel MORC2 mutations and to elucidate their clinical features. This study was approved by the ethics committee of the Third Xiangya Hospital of Central South University and informed consents were obtained from all individuals involved in the study. We detected MORC2 genetic variants in 79 genetically unidentified CMT2 probands from a cohort of 115 index CMT2 families of Chinese origin by using polymerase chain reaction and Sanger sequencing. All of the probands had underwent sequence capture array of 35 CMT genes including 19 CMT2 genes ( MFN2 , MPZ , HSPB1 , HSPB8 , RAB7A , TRPV4 , GARS , NEFL , GDAP1 , DNM2 , AARS , DYNC1H1 , LRSAM1 , DHTKD1 , DNAJB2 , MARS , LMNA , MED25 , TRIM2 ) and one CMT1X gene ( GJB1 ), and less than 40% achieved molecular diagnosis. We identified five MORC2 variants c.1013A > G (p.Q338R), c.1210G > A (p.D404N), c.2207A > G (p.H736R), c.303G > T (p.E101D) and c.440A > G (p.N147S). The variants c.1013A > G and c.1210G > A were absent in the dbSNP129, HapMap 8, Exome Aggregation Consortium and 1000 Genome project databases. Amino acid sequences at these two sites were highly conserved among species. They were predicted to be disease-causing by SIFT, PolyPhen-2 or MutationTaster programs.
