Abstract:Gold nanoclusters (Au NCs) are a type of emerging ECL emitter with molecule-like properties and low toxicity that hold great potential for sensing application. However, the application of Au NCs in ECL sensing is still limited due to their low ECL efficiency. In this work, we provided an effective way to enhance the ECL efficiency of Au NCs. By doping Ag on GSH-protected Au NCs to form bimetallic clusters (GSH-Ag/Au NCs), the ECL efficiency of Au NCs was greatly improved. Based on the enhanced ECL signal of Au NCs, an ultrasensitive ECL sensor was constructed for the detection of dopamine (DA). DA exhibited a prominent ECL quenching effect towards the formed bimetallic GSH-Ag/Au NCs which can be used for DA detection. The proposed ECL sensor exhibited excellent sensitivity, selectivity and stability and had a wide linear range from 10 nM to 1 mM with a low detection limit of 2.3 nM (S/N = 3). More importantly, this work provided a potential method to improve the ECL properties of Au NCs and widen their analytical application.

Abstract:As shown in the TOC, electrochemiluminescence (ECL) of the nanoclusters is introduced as a new signaling mechanism for elucidating essential electrocatalytic parameters and to complement foot of the wave analysis on voltametric current features. ECL provides kinetic insights during catalytic reactions for operando studies. Retrospectively, mechanistic understanding of the multi‐step ECL reactions offer guidance for the evaluation and optimization of ECL property. Atomically precise metal nanoclusters are increasingly exploited in electrocatalysis such as water electrolysis and CO2 reduction. The catalytic effects are generally evaluated by conventional electrochemical methods or end‐product analysis. Herein, near infrared electrochemiluminescence (ECL) of the nanoclusters is introduced as a new signaling mechanism to complement classical voltammetric analysis for elucidating essential electrocatalytic parameters. Coreactant ECL from an aqueous soluble Au22 nanocluster (NCs) enhanced by a common pH buffer materials 4‐(2‐hydroxyethyl)‐1‐piperazineethanesulfonic acid (HEPES) and by a piperazine drug hydroxyzine (HDZ) is studied under oxidative reduction pathways. Foot of the wave analysis on voltametric features and the kinetics profiles in potential‐step experiments are studied in different nanocluster and coreactant concentrations. Benchmark parameters such as rate constants are determined under EC (Electron‐transfer‐Chemical‐reaction) mechanism as feasibility validations. Because the luminescence properties of AuNCs are sensitive to the surface ligands or adsorbents, ECL is proposed as a new signal readout for operando studies providing insights for governing reaction mechanism and kinetics during catalytic reactions. Retrospectively, mechanistic understanding of the complex multi‐step reactions during ECL generation can provide guidance for the evaluation and optimization of ECL property for better quantitative applications.

Abstract:Strong electrogenerated chemiluminescence (ECL) is detected from dithiolate Au nanoclusters (AuNCs) in aqueous solution under ambient conditions. A novel mechanism to drastically enhance the ECL is established by covalent attachment of coreactants N,N-diethylethylenediamine (DEDA) onto lipoic acid stabilized Au (Au-LA) clusters with matching redox activities. The materials design reduces the complication of mass transport between the reactants during the lifetime of radical intermediates involved in conventional ECL generation pathway. The intracluster reactions are highly advantageous for applications by eliminating additional and high excess coreactants otherwise needed. The enhanced ECL efficiency also benefits uniquely from the multiple energy states per Au cluster and multiple DEDA ligands in the monolayer. Potential step and sweeping experiments reveal an onset potential of 0.78 V for oxidative-reduction ECL generation. Multifolds higher efficiency is found for the Au clusters alone in reference to the standard Rubpy with high excess TPrA. The ECL in near-IR region (beyond 700 nm) is highly advantageous with drastically reduced interference signals over visible ones. The features of ECL intensity responsive to electrode potential and solution pH under ambient conditions make Au-LA-DEDA clusters promising ECL reagents for broad applications. The strategy to attach coreactants on Au clusters is generalizable for other nanomaterials.

Abstract:Electrogenerated chemiluminescence (ECL) emission was observed from the water-soluble, bovine serum albumin (BSA)-stabilized Au nanoclusters for the first time. The possible ECL mechanism was discussed according to the presented results and ascribed to the effective electron transfer from the conduction-band of excited indium tin oxide (ITO) to Au nanoclusters (NCs). A simple label-free method for the detection of dopamine has been developed based on the Au NCs ECL in aqueous media The Au NCs could be an effective candidate for new types of ECL biosensors in the future due to their fascinating features, such as good water solubility, low toxicity, ease of labeling, and excellent stability.

Abstract:This study enhances bimetallic Au12Ag13 nanocluster electrochemiluminescence (ECL) via ligand engineering (PPh3 to P(Ph‐OMe)3), achieving a 3.6‐fold ECL efficiency boost, notably with 1 mM tri‐n‐propylamine. The proposed ECL mechanisms rooted in nanocluster activities offer promising avenues for improved metal nanocluster ECL in sensing and detection applications. The pursuit of highly emissive electrochemiluminescence (ECL) luminophores has long been a goal for both understanding mechanism and advancing detection applications. In this study, we employ ligand engineering to boost the ECL performance of bimetallic Au12Ag13 nanoclusters, substituting the PPh3 ligand with P(Ph‐OMe)3. The results demonstrate a 3.6‐fold enhancement in ECL efficiency following ligand exchange when compared to Au12Ag13‐PPh3, particularly in the presence of 1 mM tri‐n‐propylamine (TPrA) as a coreactant – surpassing the performance of the conventional Ru(bpy)32+ luminophore. Voltammetric analysis reveals the presence of both oxidative and reductive ECL signals under sweeping potentials, with corresponding ECL spectra collected. Proposed ECL mechanisms are rooted in the anodic and cathodic activities of Au12Ag13‐P(Ph‐OMe)3 nanoclusters. Notably, this work captures extremely high and transient ECL signals, sequentially activating LUMO and HOMO states, reminiscent of the behavior observed in Au12Ag13‐PPh3. Our findings present a promising strategy to enhance the ECL of metal nanoclusters, holding potential for further applications in sensing and detection.

Abstract:Herein, a novel Au nanoclusters/Cu2O (Au NCs/Cu2O) heterostructure exhibited exceptionally strong electrochemiluminescence (ECL) emission, in which the p-type semiconductor Cu2O was defined as the electrosensitizer to provide the electrogenerated holes for rapidly transferring the electrogenerated hot electrons of Au NCs. Thus, the fast charge transfer of Au NCs/Cu2O was achieved by the electrosensitizer compared to the sluggish one via intramolecular covalent bond charge transfer of traditional Au NCs, resulting in a greatly higher ECL efficiency (63.8%) than that of pure Au NCs (2.7%) versus the standard [Ru(bpy)3]2+. It solved one main challenge of electrochemiluminophore-based metal NCs: high efficiency with energic charge-transport kinetics. As a proof of concept, Au NCs/Cu2O was successfully employed in an ultrasensitive ECL biosensing platform for determining the biological antioxidant glutathione with a limit of detection (LOD) as low as 6.3 pM. The heterostructure as an ECL emitter is a very promising start for guiding the rational design of efficient electrochemiluminophores in intense light-emitting devices and high-definition ECL imaging.

Abstract:To determine the intrinsic effects of body elements on the electrochemiluminescence (ECL) of metal nanoclusters (NCs), herein, a valence-state engineering strategy is developed to adjust the NCs' ECL with bovine serum albumin (BSA)-stabilized AuNCs as a model, in which engineering the valence state of the Au body element, i.e., Au(0) and Au(I), is performed via successively reducing the precursor AuCl4- to Au(I) and Au(0) with BSA. The obtained BSA-AuNCs/N2H4 system leads to three anodic ECL processes at 0.37 (ECL-1), 0.85 (ECL-2), and 1.45 V (ECL-3). ECL-1 is generated from the BSA-Au(0) section of BSA-AuNCs in a surface-defect-involved route and is much stronger and red-shifted compared to ECL-2 and ECL-3, which are generated from the BSA-Au(I) section of BSA-AuNCs in the band-gap-engineered route. Each of the anodic ECL processes can be selectively generated and/or suppressed via adjusting the Au(I)/Au(0) ratio of BSA-AuNCs, tunable ECL generation route, and triggering potential, and the emission intensity and waveband of metal NCs are conveniently achieved in body-element-involved valence-state engineering.
Efficient and Monochromatic Electrochemiluminescence of Aqueous‐Soluble Au Nanoclusters Via Host–Guest Recognition

Abstract:AbstractInspired by the enhanced photoluminescence of Au nanoclusters (AuNCs) with a rigid shell, the formation of rigid host–guest assemblies on AuNC surfaces was employed to screen novel electrochemiluminophores with 6‐aza‐2‐thiothymine(ATT)‐protected AuNCs (ATT‐AuNCs) and l‐arginine (ARG) as models for the first time. The rigid host–guest assemblies formed between ARG and ATT on the ATT‐AuNC surface enabled aqueous‐soluble ARG/ATT‐AuNCs with a dramatically enhanced electrochemiluminescence (ECL) compared to ATT‐AuNCs. This includes one cathodic ECL process (−1.30 V) and three anodic ECL processes (+0.78, 0.90, and 1.05 V) in a so‐called half‐scan experiment without a co‐reactant, as well as a 70‐fold enhanced oxidative‐reduction ECL at +0.78 V with tri‐n‐propylamine as a co‐reactant. Importantly, the ECL of the ARG/ATT‐AuNCs is highly monochromatic with an emission maximum around 532 nm and a full width at half‐maximum of 36 nm, which is of great interest for color‐selective ECL assays.
Plasmon-Enhanced Electrochemiluminescence at the Single-Nanoparticle Level

Abstract:Plasmon-enhanced electrochemiluminescence (ECL) at the single-nanoparticle (NP) level was investigated by ECL microscopy. The Au NPs were assembled into an ordered array, providing a high-throughput platform that can easily locate each NP in sequential characterizations. A strong dependence of ECL intensity on Au NP configurations was observed. We demonstrate for the first time that at the single-particle level, the ECL of Ru(bpy)(3)(2+)-TPrA was majorly quenched by small Au NPs (<40nm), while enhanced by large Au ones (>80nm) due to the localized surface plasmon resonance (LSPR). Notably, the ECL intensity was further increased by the coupling effect of neighboring Au NPs. Finite Difference Time Domain (FDTD) simulations conformed well with the experimental results. This plasmon enhanced ECL microscopy for arrayed single NPs provides a reliable tool for screening electrocatalytic activity at a single particle.
Near Infrared Electrochemiluminescence of Rod-Shape 25-Atom AuAg Nanoclusters That Is Hundreds-Fold Stronger Than That of Ru(bpy) 3 Standard.

Abstract:Near infrared (near-IR) electrogenerated chemiluminescence (ECL) from rod-shape bimetallic Au12Ag13 nanoclusters is reported. With ECL standard tris(bipyridine)-ruthenium(II) complex (Ru(bpy)(3)) as reference, the self-annihilation ECL of the Au12Ag13 nanoclusters is about 10 times higher. The coreactant ECL of Au12Ag13 is about 400 times stronger than that of Ru(bpy)(3) with 1 mM tripropylamine as coreactants. Voltammetric analysis reveals both oxidative and reductive ECLs under scanning electrode potentials. Transient ECL signals (tens of milliseconds) and decay profiles are captured by potential step experiments. An extremely strong and transient self-annihilation ECL is detected by activating LUMO and HOMO states sequentially via electrode reactions. The ECL generation pathways and mechanism are proposed based on the key anodic and cathodic activities arising from the energetics of this unique atomic-precision bimetallic nanocluster. Successes in the generation of the unprecedented strong near-IR ECL strongly support our prediction and choice of this nanocluster based on its record-high 40% quantum efficiency of near-IR photoluminescence. Correlation of the properties to the atomic/electronic structures has been a long-pursued goal particularly in the fast growing atomic-precision nanoclusters field. The mechanistic insights provided in this fundamental study could guide the design and syntheses of other nanoclusters or materials in general to achieve improved properties and further affirm the structure-function correlations. The high ECL signal in the less interfered near-infrared spectrum window offers combined merits of high-signal-low-noise/interference or high contrast for broad analytical sensing and immunoassays and other relevant applications.
Bright Near-Infrared Circularly Polarized Electrochemiluminescence from Au9Ag4 Nanoclusters.

Abstract:Metal nanoclusters are excellent electrochemiluminescent luminophores owing to their rich electrochemical and optical properties. However, the optical activity of their electrochemiluminescence (ECL) is unknown. Herein, we achieved, for the first time, the integration of optical activity and ECL, i.e., circularly polarized electrochemiluminescence (CPECL), in a pair of chiral Au9Ag4 metal nanocluster enantiomers. Chiral ligand induction and alloying were employed to endow the racemic nanoclusters with chirality and photoelectrochemical reactivity. S-Au9Ag4 and R-Au9Ag4 exhibited chirality and bright-red emission (quantum yield = 4.2%) in the ground and excited states. The enantiomers showed mirror-imaged CPECL signals at 805 nm owing to their highly intense and stable ECL emission in the presence of tripropylamine as a co-reactant. The ECL dissymmetry factor of the enantiomers at 805 nm was calculated to be ±3 × 10-3, which is comparable with that obtained from their photoluminescence. The obtained nanocluster CPECL platform shows the discrimination of chiral 2-chloropropionic acid. The integration of optical activity and ECL in metal nanoclusters provides the opportunity to achieve enantiomer discrimination and local chirality detection with high sensitivity and contrast.
Pre‐oxidation of Gold Nanoclusters Results in a 66 % Anodic Electrochemiluminescence Yield and Drives Mechanistic Insights

Abstract:AbstractGold nanoclusters (AuNCs) are attractive electrochemiluminescence (ECL) emitters because of their excellent stability, near IR emission, and biocompatibility. However, their ECL quantum yield is relatively low, and our limited fundamental understanding has hindered rational improvement of this parameter. Herein, we report drastic enhancement of the ECL of ligand‐stabilized AuNCs by on‐electrode pre‐oxidation with triethylamine (TEA) as a co‐reactant. The l‐methionine‐stabilized AuNCs resulted in a record high ECL yield of 66 %. This strategy was successfully extended to other AuNCs, and it is more effective for ligand shells that allow more effective electron transfer. In addition, excitation of the pre‐oxidized ECL required a lower potential than conventional methods, and no additional instrument was required. This work opens avenues for solving a challenging problem of AuNC‐based ECL probes and enriches fundamental understanding, greatly broadening their potential applications.

Abstract:In the past ten years, the development of electrochemiluminescent (ECL) analytical methods based on various types of nanostructures has become a research hotspot. Nanoclusters, an intermediate between molecules and conventional nanoparticles, are renowned for their luminescent features. The first report on ECL for nanoclusters can be traced back to 2009. Here we summarize the main research progresses since 2011. Firstly, the preparation of ECL-related nanoclusters is briefly introduced. Then, the mechanisms and applications of ECL by nanoclusters are described. To improve ECL performances, two main strategies, i.e., nanostructure-based ECL enhancement and biological signal amplification are proposed. Besides, the nanoclusters as the energy transfer receptors in ECL systems are also discussed. In prospect part, the future development of ECL by nanoclusters is considered. We believe that the synthesis of high quality nanoclusters, the reveal of ECL structure-activity relationships, the rationale design and application of near-infrared ECL, and the role of ECL in the interdisciplinary research are the main problems we faced in the future.
Cu Nanoclusters: Novel Electrochemiluminescence Emitters for Bioanalysis

Abstract:Cu nanoclusters (Cu NCs), which emerged as a new class of nontoxic, economic, and excellent phosphors and catalysts, have attracted increasing interest for a wide variety of promising applications in biolabeling and biocatalysis. However, the electrochemiluminescence (ECL) behavior of Cu NCs has never been reported in previous works. Here, anodic and blue ECL emission of Cu NCs was observed for the first time with the efficient coreactant of hydrazine (HZ), and the possible luminescence mechanism of Cu NCs/HZ ECL system was studied in detail. Briefly, HZ was oxidized, and Cu NCs got the energy to generate excited state Cu NCs* for light radiation. Furthermore, a highly sensitive "signal-off" sensing platform for the determination of dopamine has been developed upon effectively quenching of dopamine toward the Cu NCs/HZ-based ECL system. As a result, this proposed method for dopamine detection possesses high selectivity, good stability, and excellent sensitivity with a detection limit down to 3.5 × 10-13 M. This indicates that Cu NCs show potential for applications in ECL bioanalysis as a new type of low-cost and superior luminophore candidates.

Abstract:Copper nanoclusters (CuNCs) are emerging electrochemiluminescence (ECL) emitters with unique molecule-like electronic structures, high abundance, and low cost. However, the synthesis of CuNCs with high ECL efficiency and stability in a scalable manner remains challenging. Here, we report a facile gram-scale approach for preparing self-assembled CuNCs (CuNCsAssy) induced by ligands with exceptionally boosted anodic ECL and stability. Compared to the disordered aggregates that are inactive in ECL, the CuNCsAssy shows a record anodic ECL efficiency for CuNCs (10%, wavelength-corrected, relative to Ru(bpy)3Cl2/tripropylamine). Mechanism studies revealed the unusual dual functions of ligands in simultaneously facilitating electrochemical excitation and radiative transition. Moreover, the assembly addressed the limitation of poor stability of conventional CuNCs. As a proof of concept, an ECL biosensor for alkaline phosphatase detection was successfully constructed with an ultralow limit of detection of 8.1×10-6 U/L.

A self-assembly strategy was adopted to promote the electrochemcial excitation and radiative transition, allowing efficient enhancement of the electrochemiluminescence (ECL).
