Abstract:C-phycocyanin (CPC) and Allophycocyanin (APC) are pigment-protein complexes isolated from antenna systems in cyanobacteria. The crystal structure of CPC has recently been solved and APC has a similar structure. CPC and APC have a trimeric structure, monomeric subunits are composed of an (alpha) and (beta) polypeptide chain, each has a tetrapyrrole chromophore chemically bound to position 84. In CPC and APC trimers, the (alpha) 84 and (beta) 84 chromophores in adjacent monomers are in close proximity, forming relatively strong coupled pairs. Calculation of pairwise energy transfer rates using Foerster theory has suggested an extremely fast transfer (> 1 ps-1) between the (alpha) 84 and (beta) 84 pair in CPC. A femtosecond fluorescence up-conversion apparatus was constructed which achieves subhundred femtosecond time resolution. This allows experimental observation of the fast energy transfer process between the (alpha) 84 and (beta) 84 pair in both CPC and APC. There was also a wavelength dependence of the fluorescence depolarization kinetics which is inconsistent with Foerster inductive resonance energy transfer theory.
Fluorescence Spectroscopy, Exciton Dynamics, and Photochemistry of Single Allophycocyanin Trimers

Abstract:We report a study of the allophycocyanin trimer (APC), a light-harvesting protein complex from cyanobacteria, by room-temperature single-molecule measurements of fluorescence spectra, lifetimes, intensity trajectories, and polarization modulation. Emission spectra of individual APC trimers are found to be homogeneous on the time scale of seconds. In contrast, their emission lifetimes are found to be widely distributed because of generation of long-lived exciton traps during the course of measurements. The intensity trajectories and polarization modulation experiments indicate reversible exciton trap formation within the three quasi-independent pairs of strong interacting alpha 84 and beta 84 chromophores in APC, as well as photobleaching of individual chromophores. Comparison experiments under continuous-wave and pulsed excitation reveal a two-photon mechanism for generating exciton traps and/or photobleaching, which involves exciton-exciton annihilation. These single-molecule experiments provide new insights into the spectroscopy, exciton dynamics, and photochemistry of light-harvesting complexes.
Investigations of Ultrafast Exciton Dynamics in Allophycocyanin Trimer

Abstract:The light-harvesting chromoprotein of cyanobacteria, allophycocyanin (APC, is studied both experimentally and theoretically. We observe fast kinetics and quantum beats in the pump-probe time-resolved profiles. Fourier analysis of the quantum beats provides eight vibrational frequencies: 186-215, 273-303, 352, 400, 449-469, 508, 625-664, and 840-879 cm(-1). We also measure anisotropy profiles which exhibit two types of kinetics; one has two fast (<1 ps) decay components, and the other almost shows no decay within 2 ps. To make use of the observed data in terms of a molecular description, the crude Born-Oppenheimer adiabatic approximation is applied to define both the electronic and vibrational states of the APC trimer. On the basis of this approximation, we construct a vibronic model, which describes the six chromophores in the APC trimer. With this molecular description, it is possible to study the steady-state, optical properties and transient dynamics of the system. We analyze the absorption and CD spectra of the APC trimer, as well as oscillatory behaviors appearing in the pump-probe profiles and kinetics observed in the femtosecond anisotropy profile. We obtain a coherent set of properties of the potential energy surfaces of the system. On the basis of the vibronic model and using these molecular properties, we have simultaneously calculated the absorption and CD spectra in the spectral region 600-700 nm of the APC trimer. We have also simulated IC rate constant as a function of the electronic energy gap, electronic coupling constant, and promoting mode. Photorelated biosystems like the APC trimer may achieve an efficient IC by adjusting the electronic energy gap, promoting mode and vibronic coupling constant due to the breakdown of adiabatic approximation.

Abstract:Abstract— The excitation energy transfer processes in the allophycocyanin (APC) monomer and trimer from phycobilisomes of Polysiphonia urceolata were studied using picosecond time‐resolved fluorescence isotropic and anisotropic spectroscopy. Based on our experimental results, conclusions could be drawn as follows: (1) After the processes of exciton localization are finished, the localized excitation energy on any chromophore can be transferred to the other chromophores due to the weak couplings between them, and the processes among three p84‐phycocyaninbilin (PCB) chromophores in the center of the ring shape of the APC trimer are more important than those of between a84‐ and p84‐PCB chromophores in the same monomer. (2) The decay time constants of 95 ± 5 ps and 40 ± 5 ps components, observed by us in this work, were assigned to the excitation energy transfer or redistribution between α84‐ and β84‐PCB chromophores in the same monomer of the APC trimer and among three β84‐PCB chromophores in the center of the ring shape of the APC trimer, respectively. Specifically, the assignment of the decay constants for the 40 ± 5 ps component was different from those of previous results. (3) Based on the model of Debreczeny, and using the fluorescence residual anisotropy r(∞) with a probing wavelength of 650 nm, the angles between the C3 symmetry axis and transition dipoles of α84‐ and ‐PCB chromophores were found to be φa84= 67° and φβ84= 148°, respectively, which are in agreement with the prediction of the X‐ray crystal structure of APC. (4) The results show that anisotropy decays, observed with the APC trimer, did exhibit a strongly probing wavelength dependence that did not show up in the monomer.
Femtosecond Dynamics of Energy Transfer in Native B800-B850 and B800-Released Lh2 Complexes of Rhodobacter Sphaeroides

Abstract:Two kinds of antenna complexes LH2 of Rhodobacter sphaeroides, wild type RS601 and the removal of B800 pigments (B800-released), were used in our experiment. These two LH2 complexes show quite different behaviour in absorption and femtosecond dynamics. By using the femtosecond pump-probe technique, the energy transfer processes occurring in two complexes were studied. Because of removing the B800 pigment from the LH2 in B800-released LH2 complex, the energy transfer between the B800 to B850 pigment was completely eliminated, while the pure internal energy transfer within the exciton states of B850 pigment could be carefully investigated. The results show that, at B800 absorption band, B800-released LH2 obviously shows a dominated transient absorption different from the photobleaching observed in RS601; while at the B850 band, these two complexes show similar photobleaching behaviour.

Abstract:The charge transfer (CT) process in organic semiconductor thin film structures is an important problem for applications such as photoreceptors and light-emitting devices. The operation of a photoreceptor structure is based on a CT process between a donor molecule and an acceptor transport molecule. We have investigated such a structure formed by vacuum-grown thin films of two organic molecules, N,N'-diphenethyl-3,4,9,10-perylenetetracarboxylic-diimide (DPEP) and N,N'-diphenyl-N,N'-(3-methylphenyl)-1,1'- biphenyl-4,4'-diamine (TPD), with femtosecond time-resolved photoemission spectroscopy. By measuring the lifetimes of the excited electron states in the mixtures of these molecules we are able to determine the time-scale for the electron transfer (ET) between the excited states of the TPD and DPEP molecules. We show that the biexponential ET dynamics consist of a short component of less than 100 fs and a long component of several hundreds fs in length.

Abstract:Photodynamics of peripheral antenna complexes, light-harvesting complex (LH2) of Rhodobacter (Rb) Sphaeroides 601, was studied using femtosecond pump-probe technique at different laser wavelengths. The obtained results reveal dramatic dynamical evolutions within B800 and B850 absorption bands of antenna complexes LH2. At excitation wavelength around 835 nm, a sharp photobleaching signal was observed which was assigned to the contribution of the two-exciton state, which was further confirmed by the power dependence measurement. Rate equations with eight-level scheme were used to calculate the population evolution in LH2 and the transient dynamics under femtosecond pulse excitation. The research results prove that not only the transition from ground state to one-exciton state but also that from one-exciton state to two-exciton state contribute to the photodynamics of B850.

Abstract:We report an in-depth photophysical investigation of an asymmetric donor-acceptor-donor' (D-A-D') thermally activated delayed fluorescence (TADF) molecule (4- (9H-carbazol-9-yl)phenyl) (4- (10 H-phenothiazin-10-yl)-phenyl)sulfone and compare its photophysical properties to the parent symmetric D-A-D and D'-A-D' molecules. These D-A-D type small molecules all show strong TADF. The work reveals how the relative orientations of D-A (D'-A) moieties favor reverse intersystem crossing (rISC) by forming stable charge transfer (CT) states. The key requirement for the efficient TADF emitters is to achieve a very small CT-local triplet state energy splitting, which is shown to be complex in the asymmetric molecule. Throughout the investigations, we show that in the asymmetric D-A-D' system, even though E-CT (D-A) > E-CT (D'-A), no evidence of energy transfer from D-A to A-D' is observed, nor from excited D to D'. This is ascribed to the near orthogonality of the D and D' units and the very strong decoupling of the electrons on the D and A in the CT state. In addition, the possibility of aggregation-induced TADF (AI-TADF) is examined and shown to be a manifestation of solvatochromism in these particular molecules.

Abstract:Femtosecond degenerate pump–probe spectroscopy was carried out on thin films of a series of axially substituted phthalocyanines including tBu4PcInCl(tBu4=tetra-(tert-butyl)), tBu4PcTiO, tBu4PcIn(p-TMP) [p-TMP=p-trifluoromethylphenyl], tBu4PcGa(p-CPO) [p-CPO=p-chlorophenoxy], and μ-oxo-bridged phthalocyanine dimers, e.g., [tBu4PcIn]2O, tBu4PcIn–O–GaPctBu4 and [tBu4PcGa]2O. We demonstrate that the long lived state of the dynamics is the triplet state and the behavior of the degenerate pump–probe was determined by exciton–exciton annihilation (E-EA), nonradiative recombination (NR) and intersystem crossing (ISC) processes. The E-EA process is via the long-range dipole–dipole interaction between excitons or motion-limited diffuse other than exciton hopping migrations. The dimers shows much efficient E-EA and the indium-contained samples have larger ISC rate than the others.
Characterizing the Structures, Spectra, and Energy Landscapes Involved in the Excited-State Proton Transfer Process of Red Fluorescent Protein LSSmKate1

Abstract:By applying molecular dynamics (MD) simulations and quantum chemical calculations, we have characterized the states and processes involved in the excited-state proton transfer (ESPT) of LSSmKate1. MD simulations identify two stable structures in the electronic ground state of LSSmKate1, one with a protonated chromophore and the other with a deprotonated chromophore, thus leading to two separate low-energy absorption maxima with a large energy spacing, as observed in the calculated and experimentally measured absorption spectra. Proton transfer is induced by electronic excitation. When LSSmKate1 is excited, the electrons in the chromophore are transferred from the phenol ring to the N-acylimine moiety; the acidity of a phenolic hydroxyl group is thus enhanced. The calculated potential energy curves (PECs) exhibit energetic feasibility in the generation of the fluorescent species in LSSmKate1, and the exact agreement between the calculated and experimentally measured values of the large Stokes shift further provides solid theoretical evidence for the ESPT process taking place in photoexcited LSSmKate1. The molecular environments play a significant role in the geometries and absorption/emission energies of the chromophores. Overall, TD-ωB97X-D/molecular mechanics (MM) provides a better description of the optical properties of LSSmKate1 than TD-B3LYP/MM, although it always overestimates the excitation energies.

Abstract:Ultra time-resolved fluorescence spectra were used to study the energy transfer processes and mechanism of complex PEC/PC/APC at 77 K, which was reconstructed with phycobiliproteins (PEC, PC and APC) of Anabaena variabilis , and has intact light-harvesting system and single terminal emitter. The energy transfer relationships between different chromophores especially between rod and core were also discussed based on fluorescence decay kinetic under different detected wavelengths. As a result, we got the possible energy transfer pathways and transfer time constants to be 29 ps between two PEC trimers, 12 ps between PEC and C-PC, 51 ps between rod and core.

Abstract:We have employed group theory and picosecond time-resolved fluorescence isotropy and anisotropy spectroscopy methods to explore the excitation transfers within an isolated C-phycocyanin (C-PC) hexamer (αβ)6PCL27RC, situated at the end of the rod proximal to the core of the pycobilisome (PBS) in the cyanobacterium Anabaena variabilis. The group-theory results imply that excitation energy transfer between two trimers occurs between the lowest exciton level of each trimer. The excitation energy-transfer process might occur at a rate of 10–20 ps, and it may be described by an exciton hopping-like Förster transfer mechanism. Dynamic components of 45–50 ps are assigned to the excitation transfer from β155-PCB chromophores to the exciton states of dimers, which consist of two neighbouring monomers of the same trimer in an isolated C-PC hexamer.

Abstract:Aggregation-induced emission (AIE) is the long-sought solution to the problem of aggregation-caused quenching that has hampered efficient application of fluorescent organic materials. An important goal on the way to fully understand the working mechanism of the AIE process was, for more than a decade, and still remains obtaining more comprehensive insights into the correlation between the ultrafast excited-state dynamics in tetraphenylethylene (TPE)-based molecules and the AIE effect in them. Here we report a number of TPE-based derivatives with varying structural rigidities and AIE properties. Using a combination of ultrafast time-resolved spectroscopy and computational studies, we observe a direct correlation between the state-dependent coupling motions and inhibited fluorescence, and prove the existence of photocyclized intermediates in them. We demonstrate that the dominant non-radiative relaxation dynamics, i.e. formation of intermediate or rotation around the elongated C[double bond, length as m-dash]C bond, is responsible for the AIE effect, which is strongly structure-dependent but not related to structural rigidity.
Studies on the energy transfer among the rod-core complex from phycobilisome of Anabaena variabilis by time resolved fluorescence emission and anisotropy spectra

Abstract:Picosecond time-resolved fluorescence emission and polarization spectra were recorded for the rod-core complex (alpha beta)(6)L-PC(RC)27(alpha beta)(3)L-AP(C)8.9 from cyanobacterium Anabaena variablis. Fluorescence decay was measured by single-photon timing and resolved to kinetic components by global data analysis with two different excitation wavelengths at 580 nm and 640 nm. Fluorescence anisotropies were recorded and analyzed at different excitation/emission pairs, such as, 590 nm/625 nm, 625 nm/640 nm, 635 nm/652 nm and 650 nm/665 nm. It was expected through this work to investigate the energy transfer processes between C-PC and APC, as well as those in a C-PC hexamer. Our data indicated that the rod-core complex acted as a functional entity with respect to excitation energy transfer. The kinetic component of 18 ps, observed with excitation at 580 nm and 640 nm, was interpreted as the excitation energy transfer from the three beta(84) chromophores of terminal C-PC trimer to an alpha(84) chromophore in the APC core. The 55 ps component, observed with excitation at 580 nm, most likely represent the transfer from beta(155)-PCB to beta(84)-PCB in a C-PC monomer. The component of 110 ps, observed with excitation at 640 nm, may originate from the transfer between alpha(84)- and beta(84)-PCB chromophores in a APC monomer. The 10 ps component was assigned to the energy transfer processes between the identical beta(155)-PCB or alpha(84)-PCB chromophores in two trimers while the 40 +/- 2 ps and 45 ps components to those among the three beta(84)-PCB chromophores in a C-PC trimer and an APC trimer, respectively.

Abstract:We model the spectra and excitation dynamics in the phycobiliprotein antenna complex PE545 isolated from the unicellular photosynthetic cryptophyte algae Rhodomonas CS24. The excitonic couplings between the eight bilins are calculated using the CIS/6-31G method. The site energies are extracted from a simultaneous fit of the absorption, circular dichroism, fluorescence, and excitation anisotropy spectra together with the transient absorption kinetics using the modified Redfield approach. Quantitative fit of the data enables us to assign the eight exciton components of the spectra and build up the energy transfer picture including pathways and timescales of energy relaxation, thus allowing a visualization of excitation dynamics within the complex.

Abstract:Two-photon absorption induced excited state absorption in the solution of a new fluorenyl-based chromophore is investigated by a time-resolved pump-probe technique using femtosecond pulses. With the help of an additional femtosecond open-aperture Z-scan technique, numerical simulations based on a three-energy level model are used to interpret the experimental results, and we determine the nonlinear optical parameters of this new chromophore uniquely. Large two-photon absorption cross section and excited state absorption cross section for singlet excited state are obtained, indicating a good candidate for optical limiting devices. Moreover, the influence of two-beam coupling induced energy transfer in neat N,N'-dimethylformamide solvent is also considered, although this effect is strongly restrained by the instantaneous two-photon absorption.
Conformational Relaxation and Thermally Activated Delayed Fluorescence in Anthraquinone-Based Intramolecular Charge-Transfer Compound

Abstract:A novel donor-pi-acceptor-pi-donor-type (D-pi-A-pi-D-type) chromophore, 2,6-bis[4-(diphenylamino)phenyl]-9,10-anthraquinone (AQ(PhDPA)(2)), has been reported as an efficient red thermally activated delayed fluorescence (TADF) emitter. Molecular structure and conformation, which directly determine the nature of excited states of a TADF emitter, are critical for obtaining efficient reverse intersystem crossing (rISC) and TADF. In this work, a series of excited-state deactivation processes of AQ(PhDPA)(2), from the optical excitation to fluorescence and TADF emitting, have been investigated by theoretical calculations and ultrafast transient absorption (TA) spectroscopy. Theoretical calculations and steady-state spectra suggest that the TADF emitter appears to have conformational twisting in the excited state. Both the relaxed S-0 and S-1 conformations have a small energy difference between the lowest singlet and triplet excited states (Delta E-ST) in favor of rISC, whereas Delta E-ST increases at the relaxed T-1 conformation. Ultrafast TA spectra reveal that the intramolecular charge transfer (ICT) state of AQ(PhDPA)(2) emits efficient fluorescence after a solvation-stabilization process in nonpolar toluene, while the fluorescence from the solvation-induced conformational relaxed ICT state is quenched in polar tetrahydrofuran. Additionally, we further reveal that the suppression of the conformational relaxation in long-lived triplet states contributes to maintaining a small LEST, which is critical for efficient rISC and TADF. These results provide a guidance for understanding the relationship between TADF and conformational relaxation dynamics, as well as for designing and synthesizing advanced TADF emitters.

Abstract:As two-photon absorption (TPA) materials, phthalocyanine molecules have promising application prospects due to their large TPA absorption cross-section, high third-order nonlinear optical susceptibility, and ultrafast response characteristics. In this work, optical properties and the ultrafast response of three modified zinc phthalocyanine molecules (P-HPcZn, Pc-P-Pc, and (DR1)4PcZn) were analyzed. No obvious side-shoulder absorption peaks in the Q-band can be observed from the steady-state absorption spectra of the three molecules, confirming the lack of aggregation products in the solutions of our measurement. Open-aperture Z-scan results show relatively large TPA cross-section values of 136.4 and 55.3 GM for Pc-P-Pc and (DR1)4PcZn, respectively. The nonlinear optical results show that the absorption process observed under the excitation of femtosecond pulses is a reverse saturable absorption (RSA) mechanism. Up-conversion fluorescence spectra of (DR1)4PcZn in THF solution indicate that the fluorescence emission mechanism is TPA. In the study of ultrafast dynamics, the transient absorption spectra were investigated and the decay lifetime of the dynamic traces corresponding to some representative probe wavelengths was obtained through data fitting with a multi-exponential function. Finally, the charge transfer and excited state properties of the modified zinc phthalocyanine molecules were discussed in depth by the DFT method. The energy gaps of P-HPcZn, Pc-P-Pc, and (DR1)4PcZn are 2.16, 1.39, and 2.13 eV, respectively. The results indicate that the Pc-P-Pc of donor-acceptor-donor (D-A-D) structure has the smallest energy gap as well as the best charge transfer properties.

Abstract:In search of new systems with a photoexcited redox pair which exhibits a strong and stable phatomduced absorption band to understand the photophyscial and photochemical properties of electron trander between fidlerenes (C-60/C-70) mid organic donor [N, N, N', N'-tetra (p-methylphenyl)-4,4'-diamino-1, V-diphenyl sulphide (TFDAS)], we studied characteristic absorption spectra in the near-IR region obtained from 532 am nanosecond laser flash photolysis of a mixture of the fullerenes (C-60/C-70) and TPDAS in polar solvents. When fullerenes (C60C70) were photoexcited, the rise of the radical anion of fullerenes (C-60/C-70) with the rapid decay of their excited triplet states were observed in benzonitrile. It can be deduced that the electron trander reaction does take place from TPDAS to excited triplet state of fullerenes (C-60/C-70). The rate constants (k(et)) and quantum yields (Phi(et)) of this proem have been also evaluated.
