Abstract:Pluripotent tendon cells (PTCs) play important roles in maintaining metabolic balance and the development of tendinopathy. Recently, substance P (SP) was identified as an important factor in the pathogenesis of tendinopathy. A better understanding of SP's effects on PTCs is essential to gain more insight into tendinopathy. This study aims to investigate the effects of SP on PTCs in vitro and in vivo. For the in vitro study, we quantified exogenous SP production by PTCs and determined the effects of SP on the proliferation and differentiation of PTCs. For the in vivo study, we used a rat patella tendon injection model to investigate the biological effects of SP on PTCs and tendons. Our results showed that exogenous SP production by PTCs was 1.80 pg/10(6) cells and SP significantly enhanced the proliferation of PTCs. Reverse transcription polymerase chain reaction (RT-PCR) results showed that SP upregulated the expression of non-tenocyte genes but downregulated the expression of tenocyte-related genes in the differentiation of PTCs. Moreover, Histological examination showed that SP induced disorganizes of tissue and tendinosis-like changes in vivo. Taken together, these findings indicate that SP enhanced PTCs' proliferation, induced non-tenocyte differentiation and plays an important role in the development of tendinopathy.
Substance P reduces TNF-α-induced apoptosis in human tenocytes through NK-1 receptor stimulation

Abstract:Background: It has been hypothesised that an upregulation of the neuropeptide substance P (SP) and its preferred receptor, the neurokinin-1 receptor (NK-1 R), is a causative factor in inducing tenocyte hypercellularity, a characteristic of tendinosis, through both proliferative and antiapoptotic stimuli. We have demonstrated earlier that SP stimulates proliferation of human tenocytes in culture. Aim: The aim of this study was to investigate whether SP can mediate an antiapoptotic effect in tumour necrosis factor-α (TNF-α)-induced apoptosis of human tenocytes in vitro. Results: A majority (approximately 75%) of tenocytes in culture were immunopositive for TNF Receptor-1 and TNF Receptor-2. Exposure of the cells to TNF-α significantly decreased cell viability, as shown with crystal violet staining. TNF-α furthermore significantly increased the amount of caspase-10 and caspase-3 mRNA, as well as both BID and cleaved-poly ADP ribosome polymerase (c-PARP) protein. Incubation of SP together with TNF-α resulted in a decreased amount of BID and c-PARP, and in a reduced lactate dehydrogenase release, as compared to incubation with TNF-α alone. The SP effect was blocked with a NK-1 R inhibitor. Discussion: This study shows that SP, through stimulation of the NK-1 R, has the ability to reduce TNF-α-induced apoptosis of human tenocytes. Considering that SP has previously been shown to stimulate tenocyte proliferation, the study confirms SP as a potent regulator of cell-turnover in tendon tissue, capable of stimulating hypercellularity through different mechanisms. This gives further support for the theory that the upregulated amount of SP seen in tendinosis could contribute to hypercellularity.
Therapeutic Roles of Tendon Stem/Progenitor Cells in Tendinopathy.

Abstract:Tendinopathy is a tendon disorder characterized by activity-related pain, local edema, focal tenderness to palpation, and decreased strength in the affected area. Tendinopathy is prevalent in both athletes and the general population, highlighting the need to elucidate the pathogenesis of this disorder. Current treatments of tendinopathy are both conservative and symptomatic. The discovery of tendon stem/progenitor cells (TSPCs) and erroneous differentiation of TSPCs have provided new insights into the pathogenesis of tendinopathy. In this review, we firstly present the histopathological characteristics of tendinopathy and explore the cellular and molecular cues in the pathogenesis of tendinopathy. Current evidence of the depletion of the stem cell pool and altered TSPCs fate in the pathogenesis of tendinopathy has been presented. The potential regulatory factors for either tenogenic or nontenogenic differentiation of TSPCs are also summarized. The regulation of endogenous TSPCs or supplementation with exogenous TSPCs as therapeutic targets for the treatment of tendinopathy is proposed. Therefore, inhibiting the erroneous differentiation of TSPCs and regulating the differentiation of TSPCs into tendon cells might be important areas of future research and could provide new clinical treatments for tendinopathy. The current evidence suggests that TSPCs are promising therapeutic targets for the management of tendinopathy.
Effects of Non-Specific Inhibition of Spinal Protein Tyrosine Phosphatases on Inflammatory Pain and Its Underlying Mechanisms

Abstract:Aim To investigate the role of spinal protein tyrosine phosphatases(PTPs) in inflammatory pain and its underlying mechanisms.Methods Complete Freund's Adjuvant(CFA) was intradermally injected into the left hindpaw of male Kunming mice to induce inflammatory pain.PTPs inhibitor sodium orthovanadate was intrathecally given one day after CFA injection to observe the alterations of Paw Withdrawal Threshold(PWT) values and of the phosphorylation of NMDA receptor NR2B subunit at Tyrosine 1472(NR2B-Y1472) in ipsilateral dorsal horn.Results Although intrathecal application of sodium orthovanadate for 30 min had no influence on PWT values in intact mice,it dose-dependently alleviated CFA-induced mechanical allodynia by elevating PWT values from(0.24±0.07) g(n=4) of inflamed mice to(0.17±0.06) g(50 ng,P0.05,n=4),(1.07±0.51) g(100 ng,P0.05,n=4) and(1.38±0.47) g(200 ng,P0.05,n=4) respectively.Meanwhile,CFA-induced phosphorylation of NR2B-Y1472 was also repressed by sodium orthovanadate(100 ng).Conclusions PTPs inhibitor sodium orthovanadate reverses NMDA receptor hyperfunction by suppressing NR2B tyrosine phosphorylation,leading to the amelioration of inflammatory pain.
Sustained Potentiation by Substance P of NMDA-activated Current in Rat Primary Sensory Neurons

Abstract:This study aimed to explore the modulatory effect of substance P (SP) on the current response mediated by N-methyl-d-aspartate (NMDA) receptor in rat primary sensory neurons and its time course using whole-cell patch clamp technique. The majority of neurons (179/213, 84.0%) examined were sensitive to NMDA (0.1–1000 μM) with an inward current, and a proportion of the NMDA-sensitive neurons also responded to SP (78/98, 80.0%) with an inward current. Pretreatment with SP potentiated the NMDA-activated current (INMDA) in a non-competitive manner, which is shown in that SP shifted the concentration–response curve for NMDA upwards compared with the control; the maximal value of INMDA increased fourfold, while the EC50 values for both curves were very close (28 vs. 30 μM). Furthermore, this potentiating effect was time-dependent: the amplitude of INMDA reached its maximum 20 min after SP preapplication, and thereafter maintained a steady level of about 2–3 times its control for 2 or even 3 h. This sustained potentiation by SP of INMDA could be blocked by extracellular application of WIN51708, a selective non-peptide antagonist of NK-1 receptor; and abolished by intracellular application of either BAPTA, or H-7, or KN-93. Though NMDA applied alone also induced a short-term (less than 20 min) self-potentiation of INMDA, it could be abolished by intracellular dialysis of BAPTA or KN-93 completely. As is known, the cell body of dorsal root ganglion (DRG) neurons is generally used as an accessible model for studying the characteristics of the membrane of primary afferent terminals in the dorsal horn of spinal cord. Therefore, these results may offer a clue to the explanation of the symptoms of chronic pain.

Abstract:Substance P (SP) is a neuropeptide that is released from sensory nerve endings and is widely present in nerve fibres. It acts on bones and related tissues by binding to receptors, thereby regulating bone metabolism, cartilage metabolism and fracture healing. SP has attracted widespread attention as a signalling substance that can be recognized by both the immune system and the nervous system. Previous studies have shown that bone and chondrocytes can synthesize and secrete sensory neuropeptides and express their receptors, and can promote proliferation, differentiation, apoptosis, matrix synthesis and the degradation of target cells through autocrine/paracrine modes. In this paper, we review the research progress made in this field in recent years in order to provide a reference for further understanding the regulatory mechanism of bone and cartilage physiology and pathological metabolism.
Substance P increases CCN2 dependent on TGF-beta yet Collagen Type I via TGF-beta1 dependent and independent pathways in tenocytes

Abstract:Transforming growth factor beta 1 (TGFbeta-1) and connective tissue growth factor (CCN2) are important mediators of tissue repair and fibrosis, with CCN2 functioning as a downstream mediator of TGFβ-1. Substance P (SP) is also linked to collagen production in tenocytes. A link between SP, TGFbeta-1 and CCN2 has yet to be established in tenocytes or fibrogenic processes. We sought to determine whether SP induces tenocyte proliferation, CCN2, or collagen production via TGFbeta-1 signaling or independently in rat primary tenocytes. Tenocytes were isolated from rat tendons, cultured and stimulated by SP and/or TGFbeta-1. Cultured cells expressed proteins characteristic of tenocytes (vimentin and tenomodulin) and underwent increased proliferation dose dependently after SP and TGFbeta-1 treatments, alone or combined (more than SP alone when combined). SP induced TGFbeta-1 expression in tenocytes in both dose- and time-dependent manners. SP and TGFbeta-1, alone or combined, stimulated CCN2 expression in tenocytes and their supernatants after both 24 and 48 h of stimulation; a response blocked with addition of a TGFbeta-1 receptor inhibitor. In contrast, SP potentiated collagen type I secretion by tenocytes, a response abrogated by the TGFbeta-1 receptor inhibitor after 48 h of stimulation, but not after the shorter 24 h of stimulation. Our findings suggest that both SP and TGFbeta-1 can stimulate tenocyte fibrogenic processes, albeit differently. TGFbeta-1 pathway signaling was involved in CCN2 production at all time points examined, while SP induced collagen type I production independently prior to the onset of signaling through the TGFbeta-1 pathway.

Abstract:Mast cells are immune cells minimally present in normal tendon tissue. The increased abundance of mast cells in tendinopathy biopsies and at the sites of tendon injury suggests an unexplored role of this cell population in overuse tendon injuries. Mast cells are particularly present in tendon biopsies from patients with more chronic symptom duration and a history of intensive mechanical loading. This study, therefore, examined the cross talk between mast cells and human tendon cells in either static or mechanically active conditions in order to explore the potential mechanistic roles of mast cells in overuse tendon injuries. A coculture of isolated human tenocytes and mast cells (HMC-1) combined with Flexcell Tension System for cyclic stretching of tenocytes was used. Additionally, human tenocytes were exposed to agonists and antagonists of substance P (SP) receptors. Mast cell degranulation was assessed by measuring β-hexosaminidase activity. Transwell and cell adhesion assays were used to evaluate mast cell migration and binding to tendon extracellular matrix components (collagen and fibronectin), respectively. Gene expressions were analyzed using real time qRT-PCR. Our results indicate that mechanical stimulation of human tenocytes leads to release of SP which, in turn, activates mast cells through the Mas-related G-protein-coupled receptor X2 (MRGPRX2). The degranulation and migration of mast cells in response to MRGPRX2 activation subsequently cause human tenocytes to increase their expression of inflammatory factors, matrix proteins and matrix metalloproteinase enzymes. These observations may be important in understanding the mechanisms by which tendons become tendinopathic in response to repetitive mechanical stimulation.

Abstract:Background The possible role of substance P (SP) during wound healing has been the primary research focus in recent years, but its effect on the healing process after bile duct injury is little understood. This study aimed to investigate the effects of SP on growth of fibroblast-like cells derived from rabbit bile duct. Methods Fibroblast-like cells derived from rabbit bile duct were identified and divided randomly into control and experimental groups. SP-treated cells at different concentrations of 10 -9 -10 -5 mol/L and control group were incubated, respectively, for 48 hours. After incubating, the effects of SP on cell proliferation were assessed by cell counts and MTT test. Apoptosis rate (AR) of cells was measured by flow cytometry. Results Cultured rabbit bile duct cells were fibroblast-like in morphology, and these cells were stained positively for vimentin and negatively for desmin. After SP was added to nonconfluent cells for 48 hours, cell numbers were significantly increased in experimental groups than in controls ( P <0.05). The maximum stimulation of cell proliferation was achieved at SP of 10 -5 mol/L. Bile duct fibroblast-like cells in the SP group showed a higher proliferating activity and lower AR than those in the control group or in the SP + Spantide group ( P <0.05). Spantide partly inhibited the effects of SP on fibroblastlike cells. Examination under transmission electron microscopy revealed rough endoplasmic reticulum and prominent Golgi complexes after SP treatment. Conclusions SP has a growth regulatory property on cultivated bile duct fibroblast-like cells in vitro , suggesting that SP may involve in wound healing after bile duct injury by promoting wound fibroblast proliferation and inhibiting apoptosis and participate in pathological scar formation.
Effects of Substance P on the Cell Differentiation of Osteoblasts in Spinal Cord Injury Rats

Abstract:Objective To investigate the effects and mechanism of substance P(SP)on the cell differentiation of osteoblasts derived from mesenchymal stem cells(MSCs) in spinal cord injury(SCI)rats.Methods MSCs from bilateral marrow cavities of tibiae and femurs in SCI rats were cultured under control mediums and induced to become osteoblasts.The ALP activity of osteoblasts were observed after treated with different concentrations(10-12,10-10,10-8,10-7,10-6M) of SP and for different time(1,2,3 week),also were also observed after the treatment of SP and Spantide(SP blocking agent),L703606(NK1 receptor blocking agent).Results Compared with the control group,the ALP activity of osteoblasts decreased after treated with different concentrations of SP,and reached the lowest at the concentration of 10-8 M,all P0.05.The ALP activity decreased significantly with the extension of time,P0.05.The decreased cell differentiation induced by SP was inhibited by spantide and L703606 respectively,P0.05.Conclusions NK1 receptors are the target receptors of SP inducing differentiation in osteoblasts.SP can inhibit the differentiation of osteoblasts,decrease the number of mature osteoblasts,and cause osteoporosis.
Substance P Potentiates Thermal Hyperalgesia Induced by Intrathecal Administration of D-serine in Rats.

Abstract:AIM: To investigate the functional interaction between substance P ( SP) and D-serine, agonist for glycine regulatory site of N-methyl-D-aspartate ( NMDA) receptor, in processing spinal nociception. METHODS: Behavior studies, by testing tail-flick latency (TFL) combined with intrathecal application of drugs, were conducted in lightly anesthetized rats. RESULTS: Decrease in TFL was observed 1.5 min after intrathecal injection of D-serine 1000 mnol. Following pretreatment with SP 0.05 nmol 6 min prior to injection of D-serine 10 nmol, D-serine-induced decrease in TFL was greatly enhanced. The potentiation was blocked by co-administration of 7-chlorokynurenic acid I pmol, the selective antagonist for glycine regulatory site of NMDA receptor, or H-7 10 mu mol, the PKC non-selective inhibitor, with SP 0.05 mnol. CONCLUSION: SP potentiates the D-serine-induced thermal hyperalgesia. Glycine regulatory site of NMDA receptor and intracellular protein kinase system may participate in the interaction of SP and NMDA receptor in the spinal cord.

Abstract:Tendinopathy is a disabling musculoskeletal disease, the pathological process of which is tightly associated with inflammation. Spironolactone (SP) has been widely used as a diuretic in clinical practice. Recently, SP has shown anti-inflammatory features in several diseases. Tendon-derived stem cells (TDSCs), a subset cell type from tendon tissue possessing clonogenic capacity, play a vital role in the pathological process of tendinopathy. In the present study, the protective effect of SP on TDSCs was demonstrated under simulated tendinopathy conditions both in vitro and in vivo. SP contributed to the maintenance of TDSC-specific genes or proteins, while suppressing the interleukin- (IL-) 1β-induced overexpression of inflammation-mediated factors. Additionally, IL-1β-induced cellular senescence in TDSCs was inhibited, while autophagy was enhanced, as determined via β-galactosidase activity, western blot (WB), and quantitative real-time polymerase chain reaction analysis. With the aid of several emerging bioinformatics tools, the mitogen-activated protein kinase (MAPK) pathway likely participated in the effect of SP, which was further validated through WB analysis and the use of MAPK agonist. Immunofluorescence analysis and an NF-κB agonist were used to confirm the inhibitory effect of SP on IL-1β-induced activation of the NF-κB pathway. X-ray, immunofluorescence, immunohistochemistry, hematoxylin and eosin staining, histological grades, and Masson staining showed that SP ameliorated tendinopathy in an Achilles tenotomy (AT) rat model in vivo. This work elucidates the protective role of SP on the pathological process of tendinopathy both in vitro and in vivo, indicating a potential therapeutic strategy for tendinopathy treatment.

Abstract:Substance P (SP) is a prominent neuropeptide that function in the active communication between the nervous system and immune response and pathological conditions such as inflammatory disorders and chronic wound healing. This functionality has been attributed to modulation of SP on production of inflammatory mediators, including chemokine and cytokines by binding to tachykinin receptors. In the present study, the effects of SP-induced production of chemokine MCP was examined and the activation of the proinflammatory transcription factor NF-kappa B was investigate as a molecular mechanism by which SP stimulate inflammatory process underlying impaired wound healing. Using genetically diabetic mice skin fibroblasts, we showed that SP up-regulates mRNA and protein level of MCP-1 in a dose-and time-dependent manner via its receptor neurokinin-1. Moreover, this stimulatory effect requires activation of transcription factor, NF-kappa B, which contributed to MCP-1 gene transcription. Furthermore, we found that MG132, a NF-kappa B inhibitor, inhibits the phosphorylation and degradation of I kappa B and MCP-1 production by SP management in diabetic skin fibroblasts. Identification of the functionality exhibited by SP and its molecular mechanisms has important implications for modulation of impaired inflammation in chronic wounds and inflammatory disorders.

Abstract:Bone marrow stromal stem cells (BMSCs) can be used to treat bone defects but BMSCs are damaged under oxidative stress. The neuropeptide substance P (SP) involves various cellular activities. However, SP's role in BMSCs differentiation under oxidative stress is unknown. Rat BMSCs were isolated and assigned into control group; oxidative stress group treated with 200 mu M H2O2; and SP group, in which 10 mM SP was added under oxidative stress followed by analysis of SP secretion by ELISA, cell proliferation by MTT method, Caspase3 activity, Bax and Bcl-2 level by Real time PCR, ALP activity ROS and SOD content as well as NF-kappa B level by Western blot. Under oxidative stress, SP secretion was significantly decreased, BMSCs proliferation was inhibited, Caspase3 activity and Bax expression increased, Bcl-2 and ALP activity was decreased along with increased ROS activity and NF-kappa B level and reduced SOD activity (P<0.05), adding SP to BMSCs under oxidative stress can significantly promote SP secretion and cell proliferation, reduce Caspase3 activity and Bax expression, increase Bcl-2 expression and ALP activity, decreased ROS activity and NF-kappa B level, and elevated SOD activity (P < 0.05). SP secretion from BMSCs cells was reduced under oxidative stress. Up-regulation of SP in BMSCs cells under oxidative stress can inhibit BMSCs apoptosis and promote cell proliferation and osteogenesis by regulating NF-kappa B.

Abstract:Accumulating evidence on bone physiopathology has indicated that the skeleton contains numerous nerve fibers and its metabolism is regulated by the nervous system. Until now, more than 10 neuropeptides have been identified in bone. Substance P (SP) is a neuropeptide released from axons of sensory neurons, belongs to the tachykinin family and plays important roles in many physiological and pathological processes by acting as a neurotransmitter, neuromodulator, or trophic factor. It activates signal transduction cascades by acting on the neurokinin-1 receptor (NK1-R). Previous studies have confirmed that the SP-immunoreactive (IR) axons innervate bone and adjacent tissues, and that their density varies depending on the regions and physiological or pathological conditions. Over the past few decades, it has been found that SP takes part in the stimulation of bone resorption, and its receptors have been demonstrated to be located in osteoclasts. Notably, in studies of skeletal ontogeny, SP-IR axons have been shown to appear at an early stage, mostly coinciding with the sequence of long bone mineralization. These findings, together with data obtained from chemically or surgically targeted nerve deletions, strongly suggest that SP is a potent regulator of skeletal physiology. The specific distribution of SP-IR nerve fibers, the different amount of SP within regions, and the various levels of expression of NK1-R in targeted cells presumably related to and participate in bone metabolism. It can be predicted that the indirect roles of SP through other cytokines are as important as its direct roles in bone metabolism. This new regulating pathway of bone metabolism would have enormous implications in skeletal physiology and the relevant research might present curative potentials to a spectrum of bone diseases.
Substance P Induces Inflammatory Responses Involving NF-κB in Genetically Diabetic Mice Skin Fibroblasts Co-Cultured with Macrophages.

Abstract:Purpose: Delayed wound healing is an intractable complex of diabetes and substance P (SP) is proved to benefit wound healing, whose functioning mechanism remains elusive. This study aims at revealing whether the influence of SP on diabetic wound healing is dependent on inflammatory responses, particularly NF-kappa B. Methods: Skin fibroblasts of genetically diabetic mice were co-cultured with bone marrow-derived macrophages, and treated with SP, SP + L703,606 (a neurokinin-1 receptor antagonist), or SP + MG132 (an inhibitor of NF-kappa B). For macrophages, their migration ability was assessed by Transwell experiments, and their M2 polarization was analyzed by flow cytometry and markers for M2 phenotype. Pro-inflammatory factors in the supernatant were detected by enzymelinked immunosorbent assay. In fibroblasts, the transcription levels of the four pro-inflammatory factors and the protein levels of NF-kappa B regulators like inhibitor of NF-kappa B alpha (I kappa B alpha) and I kappa B kinases (IKKs) were monitored by real-time quantitative PCR and western blot, respectively. Results: SP could significantly induce migration to fibroblasts (P<0.01), M2 polarization (P<0.001) and pro-inflammatory factor concentration (P<0.01) in the co-culture system. It also promotes the transcription process of pro-inflammatory factors in fibroblasts (P<0.01), and induce activation of IKK alpha/beta and phosphorylation of I kappa Ba, which caused NF-kappa B activation. All these effects were reversed if NF-kappa B was inhibited. Conclusion: The promoting effects of SP on diabetic wound healing was dependent on enhanced inflammatory responses, especially the activation of NF-kappa B. This study provided evidence for the potential usage of SP in accelerating diabetic wound healing.
Repeated 100 Hz Tens for the Treatment of Chronic Inflammatory Hyperalgesia and Suppression of Spinal Release of Substance P in Monoarthritic Rats

Abstract:Transcutaneous electrical nerve stimulation (TENS) has been shown to be an effective measure for pain relief The aim of the present study was to determine the optimal intensity and interval of repeated 100 Hz TENS for the treatment of chronic inflammatory hyperalgesia in a monoarthritic pain model of the rat, and to assess the changes of the spinal substance P (SP) release in response to TENS treatment. A reliable, reproducible chronic monoarthritic pain model was produced by intra-articular injection of complete Freund's adjuvant (CFA) at single ankle joint. The efficacy of 100 Hz TENS treatments with different frequencies and intensities was compared. In the acute period (within 3 weeks) of monoarthritis, twice-a-week schedule of TENS reduced the swelling of the inflamed ankle significantly. In the stable period (4-9 weeks), however, once-a-week schedule produced a significantly better therapeutic effect on both inflammation and arthritic hyperalgesia than that of twice- or five-times-a-week schedule. Using three levels of intensity of TENS, we found that the weaker (1-1-2 mA) stimulation produced significantly better therapeutic effects. Repeated TENS produced a reduction of SP content in spinal perfusate in parallel with the progressive reduction of the arthritic pain scores. Our results suggest that (i) consecutive TENS treatments produced cumulative effect for chronic hyperalgesia, (ii) for chronic inflammatory hyperalgesia, a weaker intensity and more sparsely arranged treatment schedule may produce better therapeutic effect and (iii) a decrease in SP release may serve as one of the possible neurochemical mechanisms underlying the therapeutic effects of multiple TENS treatments on chronic inflammatory hyperalgesia.
The effects of extrasynaptic substance P on nociceptive neurons in laminae I and II in rat lumbar spinal dorsal horn.

Abstract:Inflammation of the skin induces release and extrasynaptic spread of neuropeptides such as substance P mainly in spinal laminae I and II and causes changes in discharge properties of nociceptive neurons in spinal dorsal horn. To evaluate the role of extrasynaptic substance P we have superfused the spinal cord at recording segment with artificial cerebrospinal fluid or with substance P. A total of 102 multireceptive neurons responding to both noxious and innocuous skin stimulation were recorded in laminae I or II of lumbar spinal dorsal horn in pentobarbital anaesthetized rats. During superfusion with substance P (10 or 100 microM) significant increases of background activities (from 2.2 +/- 0.6 to 8.4 +/- 3.2 imp./s, mean +/- S.E.M.), enlargement of cutaneous receptive fields (from 359.9 +/- 60.4 to 465.5 +/- 77.3 mm2) and enhanced responses to mechanical (from 89.1 +/- 22.7 to 147.0 +/- 27.5 imp./5 s) but not thermal noxious skin stimuli were observed in the 22 neurons tested. Noxious heat-evoked responses and C-fibre-evoked responses were changed in both directions. In 50 other neurons, the coefficients of dispersion of interspike intervals, which is an indicator of burst-like discharges, were significantly reduced (from 60.4 +/- 5.5 to 52.7 +/- 5.3) after application of substance P. Substance P induced oscillations in background activities in 13 of 40 non-rhythmic neurons and depressed oscillations in 2 of 11 neurons. Cross-correlations of discharges of pairs simultaneously recorded neurons were flat (n = 4), or had a central peak (n = 19) or a central trough (n = 2) and were not changed qualitatively by extrasynaptic substance P. Thus, extrasynaptic substance P can modify not only discharge patterns in the spinal dorsal horn.

Abstract:We have recently explored the role of the tachykinin substance P neuroactive fragment substance P(1-7) in the mediation of anti-inflammatory effects using a blister model in the rat paw (Wiktelius et al., 2006). We observed that this heptapeptide induced a dose-dependent inhibitory effect on the substance P-induced response, which was reversible by the non-selective opioid receptor antagonist naloxone. In the present study, we examined the ability of substance P(1-7) to induce antihyperalgesic effects in streptozotocin-induced diabetic mice. We found that the substance P fragment strongly and dose-dependently produced antihyperalgesia in diabetic mice. This effect was reversed by naloxone but not by the selective opioid receptor antagonist beta-funaltrexamine, naltrindole or nor-binaltorphimine, selective for the micro-, delta- or kappa-opioid receptor, respectively. In addition, the antihyperalgesic effect induced by substance P(1-7) was partly reversed by a sigma(1) receptor agonist (+)-pentazocine, but not a sigma(1) receptor antagonist BD1047 ([2-(3,4-dichlorophenyl)ethyl]-N-methyl-2-(diamino)ethylamine), suggesting that involvement of the naloxone-sensitive sigma-receptor for the action of the SP related heptapeptides. These results suggest that hyperalgesia in diabetic mice may be, in part, due to the enhanced endogenous sigma(1) receptor systems in the spinal cord.

Abstract:Tendinopathy is a common tendon disorder that causes pain and impairs function. It is the most common reason for consultation with musculoskeletal specialists. The available therapies for tendinopathy are limited in number and efficacy and have unclear cellular and molecular mechanisms. Here it is shown that transforming growth factor-beta (TGF-β) activated by integrin αvβ6 promotes tendinopathy in mice. Excessive active TGF-β is found during tendinopathy progression, which led to tenocytes' phenotype transition to chondrocytes. Transgenic expression of active TGF-β in tendons induced spontaneous tendinopathy, whereas systemic injection of a TGF-β neutralizing antibody attenuated tendinopathy. Inducible knockout of the TGF-β type 2 receptor gene (Tgfbr2) in tenocytes inhibited tendinopathy progression in mice. Moreover, it is found that integrin αvβ6 induces TGF-β activation in response to mechanical load in tendons. Conditional knockout of the integrin αv gene in tendons prevented tendinopathy in mice. The study suggests that integrin αvβ6 activation of TGF-β is the mechanism of tendinopathy, and that integrin αvβ6 may be a therapeutic target in tendinopathy.
