Abstract:BACKGROUND: Platelet-rich plasma is the plasma containing rich platelet extracted by special method. Compared with common serum, platelet-rich plasma contained more rich cytokine, such as platelet derived growth factor, transforming growth factor beta and vascular endothelial growth factor. OBJECTIVE: To observe effects of platelet-rich plasma on biological characteristics of human bone marrow mesenchymal stem cells (BMSCs) under condition of osteogenic induction, and to discuss a new method of promoting proliferation of BMSCs and inducing osteoblasts. DESIGN, TIME AND SETTING: The paired sample controlled observation was performed at the Laboratory of Tissue Engineering, Sun Yat-sen University from March 2007 to March 2008. PARTICIPANTS: Eighteen healthy volunteers, 12 males and 6 females, averagely aged 27.5 years old, were randomly assigned into a platelet-rich plasma group, a fetal bovine serum group and a serum-free control group, with 6 subjects for each group. METHODS: Platelet-rich plasma was collected from autologous peripheral vein blood of 18 healthy volunteers. BMSCs were harvested from healthy volunteers by density gradient centrifugation, and separately routinely cultured in 10% AB serum, 10% autologous platelet-rich plasma and serum-free, combined with high glucose DMEM, 50 mg/L ascorbic acid, 10-8 mol/L dexamethasone and 10-3 mol/L β sodium glycerophosphate. MAIN OUTCOME MEASURES: BMSC morphology was observed under an inverted phase contrast microscope and a scanning electron microscope. BMSC proliferation was detected by MTT. Biological characteristics of the cells were detected with alkaline phosphatase staining and osteocalcin staining. RESULTS: After inoculation of human BMSCs for 12-24 hours, cells adhered to wall, presented round and changed into spindle-shape, polygonal, or irregular forms with many processes. After passage, cells grew rapidly in the platelet-rich plasma group and the fetal bovine serum group compared with the serum-free control group (P < 0.05). At the second, fourth and sixth passages, cell proliferation was rapid in the platelet-rich plasma group compared with the fetal bovine serum group. Alkaline phosphatase activity and osteocalcin contents increased over time. At 3, 6, 9 and 12 days, alkaline phosphatase activity decreased in the fetal bovine serum group compared with the platelet-rich plasma group. At 15 days, there were no significant differences. At 3, 6 and 9 days, there were no significant differences in osteocalcin contents between the platelet-rich plasma group and the fetal bovine serum group. At 12 days, osteocalcin contents were higher in the platelet-rich plasma group than in the fetal bovine serum group. Alkaline phosphatase activity and osteocalcin contents had insignificant changes between the serum-free control group and the platelet-rich plasma group. At each time point, alkaline phosphatase activity and osteocalcin contents were lower in the serum-free control group than in the platelet-rich plasma group (P < 0.01). CONCLUSION: Autologous platelet-rich plasma can effectively accelerate proliferation of human BMSCs, and effectively promote osteogenic characteristic expression of human BMSCs.
