Abstract:Aim: To investigate the role of ESR2 polymorphism in incidence and prognosis of patients with hepatocellular carcinoma (HCC). Methods: The DNA from patients with HCC and hepatitis type B virus carriers was used to observe the distribution of ESR2 variations and carry out the clinical correlation analysis. Results: Single nucleotide polymorphisms (SNPs) in ESR2 gene (rs1256049, rs4986938) were significantly correlated with HCC risk. Individuals carrying AA/GA genotype of rs1256049 were associated with lower HCC risk when compared with GG genotype (OR 0.399; 95% CI 0.256-0.621; P<0.001). The opposite result was found in individuals with the rs4986938 AA/GA genotype (OR 2.154; 95% CI 1.370-3.385; P=0.001). The LL genotype (frequency n≤20) of CA repeat allele was associated with moderate-well differentiated degree (p=0.001) and early stage of HCC (p=0.001), and mean OS time was 46.2 (95% CI: 39.3-53.1) months for patients with LL genotype and 29.4 (95% CI: 24.334.5) months for patients with SS (frequency n>20) genotype (P=0.005). Conclusions: Findings indicated SNPs in ESR2 gene (rs1256049, rs4986938) play a different role in HCC risk and SS genotype lead to poor prognosis of HCC patients underwent hepatectomy. It provided a potential reference for the diagnosis and treatment of HCC.

Abstract:RAS protein activator like 2 (RASAL2) has been reported to be dysregulated in various types of cancer. It has previously been demonstrated that RASAL2 is hypomethylated in hepatocellular carcinoma (HCC). However, the expression pattern of RASAL2 and its potential role in HCC remain to be elucidated. The present study demonstrated that the expression of RASAL2 was upregulated in HCC tissues, compared with in normal liver tissues, by using immunohistochemistry. In addition, Cell Counting Kit-8 assay and invasion assay revealed that knockdown of RASAL2 inhibited the growth and invasion of HCC cells. Western blotting results indicated that the inhibition of RASAL2 reduced the levels of phosphorylated-AKT. Notably, RASAL2 was observed to be a direct target of miR-203 in HCC in luciferase activity assays. Furthermore, overexpression of miR-203 exhibited a similar effect to RASAL2 knockdown in HCC cells. These results indicated that RASAL2 serves a tumor oncogenic role in HCC and may be considered a potential target in HCC.
R-spondin 2 Promotes Proliferation and Migration Via the Wnt/β-catenin Pathway in Human Hepatocellular Carcinoma.

Abstract:Hepatocellular carcinoma (HCC) is a leading cause of malignant disease-associated mortality, particularly in China. The RSPO2 (R-spondin 2) gene is evolutionarily conserved in vertebrates and is involved in developmental and physiological processes. Importantly, RSPO2 has been reported to be associated with colon cancer and potentiate the Wnt/beta-catenin signaling pathway. In the present study, enhanced expression of RSPO2 in HCC was observed using tissue microarray. Similarly, the expression level of RSPO2 was higher in HepG2, Huh7 and Hep3B cells but lower in Bel7404 and QGY7703 cells compared with human normal QSG7701 liver cells. Subsequently, gain-of-function studies indicated that RSPO2 promotes the proliferation and migration of QGY7703 cells based on lentivirus-based gene delivery. Furthermore, it was revealed that p21 and leptin, rather than vascular endothelial growth factor-A, are involved in the function of RSPO2 in QGY7703 cells. Particularly, the signal transducer and activator of transcription 3 (STAT3) and Wnt/beta-catenin signaling pathways are involved in this process. Overexpression of RSPO2 resulted in the elevated expression of phosphorylated STAT3, beta-catenin and c-Myc. Therefore, the present study is beneficial to the understanding of RSPO2-involved liver cancer transformation and drug discovery.
Expression of SRF, E-cadherin and β-catenin in hepatocellular carcinoma and its clinical significances

Abstract:Objective: To investigate the expression of serum response factor(SRF),E-cadherin and βcatenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics of the tumor and prognosis.Methods: Tissue microarray and immunohistochemical study(EnVision method) for SRF,E-cadherin and βcatenin was performed on tissue microarray which consisted of 182 cases of hepatocellular carcinoma,92 cases of adjacent non-neoplastic liver tissues.Results: In the hepatocellular carcinoma group 75.8% tumor samples expressed SRF,and the levels is significantly higher than that in surrounding liver tissue(21.7%,P<0.05).The expression of SRF was positively correlated with the TNM stage,tumor metastasis and recurrence(P<0.05).In the hepatocellular carcinoma group,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,and these levels were higher than those in the adjacent non-neoplastic liver tissue group(30.4% and 26.1% P<0.05).Aberrant expression of E-cadherin andβ-catenin was related to vascular cancer embolus,the TNM stage,tumor metastasis and recurrence(P<0.05).The expression of SRF correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion: In hepatocellular carcinoma,the overexpression of SRF and the downregulated expression of E-cadherin and β-catenin may be important mechanisms for promoting the invasion and metastasis of hepatocellular carcinoma and may also play a key role in the occurrence and development of hepatocellular carcinoma.
RASGRP2 is a potential immune-related biomarker and regulates mitochondrial-dependent apoptosis in lung adenocarcinoma

Abstract:Background: Ras guanine nucleotide-releasing protein 2 (RASGRP2), one of the guanine nucleotide exchange factors (GEFs), has attracted much attention in recent years. However, the correlation between RASGRP2 and immune infiltration and malignant features in lung adenocarcinoma (LUAD) has rarely been mentioned. Methods: The Limma package and the LASSO regression model were performed to screen for differentially expressed genes. Data from the TCGA and 5 GEO databases were used to explore the expression level of RASGRP2 in LUAD patients. A weighted co-expression network and LinkFinder module were established to find the related genes of RASGRP2. The ESTIMATE algorithm was used to analyze the correlation between RASGRP2 and immune infiltration in LUAD. Tumor-infiltrating immune cells were sorted and sequenced at the single-cell level to analyze differences in RASGRP2. Real-time PCR and immunohistochemistry were performed in the real-world cohort to verify the expression of RASGRP2 and its correlation with immune-related genes. Clone formation and EdU assays were used to verify the proliferation ability. The proportion of apoptotic cells was analyzed by flow cytometry. Observation of mitochondrial membrane potential (MMP) changes by fluorescence microscopy. Results: Our results suggested that decreased RASGRP2 was associated with worse clinical parameters and prognosis in LUAD patients. And we constructed a FLI1-HSA-miR-1976-RASGRP2 transcriptional network to support the role of RASGRP2. Enrichment analysis revealed that RASGRP2 was involved in lymphocyte activation and leukocyte adhesion. RASGRP2 was found to be positively correlated with the infiltration of most immune cells, immunoregulators, and chemokines in a subsequent study. Meanwhile, the real-world cohort confirmed that the expression levels of PDCD1, CTLA4, CD40LG, CCL14, CXCR5, and CCR7 were higher in the high-RASGRP2 expression group. Cytological experiments proved that RASGRP2 inhibited cell proliferation in LUAD by regulating mitochondrial-dependent apoptosis. Conclusion: RASGRP2 was a potential immune-related biomarker of LUAD. In addition, RASGRP2 was involved in the malignant progression of LUAD through the regulation of mitochondrial-dependent apoptosis.
Over-expression of RRM2 predicts adverse prognosis correlated with immune infiltrates: A potential biomarker for hepatocellular carcinoma

Abstract:Background: Ribonucleotide reductase regulatory subunit M2 (RRM2) has been reported to be an oncogene in some malignant tumors, such as lung adenocarcinoma, oral squamous cell carcinoma, glioblastoma, and breast cancer. However, the clinical significance of RRM2 in hepatocellular carcinoma has been less studied. The aim of this study was to assess the importance of RRM2 in hepatocellular carcinoma (HCC) based on the Cancer Genome Atlas (TCGA) database. Methods: The RRM2 expression levels and clinical features were downloaded from the TCGA database. Immunohistochemistry results between tumor tissues and normal tissues were downloaded from the Proteinatlas database. Meanwhile, the expression levels of RRM2 in tumor and paraneoplastic tissues were further verified by qRT-PCR and Western Blotting. Gene Ontology (GO)/Kyoto Encyclopedia of Genes and Genomes (KEGG) and protein-protein-interactions (PPI) network were constructed to analyze RRM2-related downstream molecules. In addition, RRM2 expression-related pathways performed by gene set enrichment analysis (GSEA). Association analysis of RRM2 gene expression and immune infiltration was performed by single-sample GSEA (ssGSEA). Results: The RRM2 expression level in tumor tissues was higher than normal tissues (P <0.001). The elevated expression of RRM2 in HCC was significantly correlated with T stage (P <0.05), pathologic stage (P <0.05), tumor status (P <0.05), histologic grade (P<0.001), and AFP (P <0.001). HCC with higher RRM2 expression was positively associated with worse OS (overall survival), PFS (progression-free survival), and DSS (disease-specific survival). In the univariate analysis, the expression of RRM2, T stage, M stage, pathologic stage, and tumor status were negatively correlated with OS (P <0.05). Further analysis using multivariate Cox regression showed that tumor status (P<0.01) and RRM2 expression (P<0.05) were independent prognostic factors of OS in HCC. GO/KEGG analysis showed that the critical biological process (chromosome condensation and p53 signaling pathway) might be the possible function mechanism in promoting HCC. Moreover, GSEA showed that several pathways were enriched in RRM2 high-expression samples, including PD-1 signaling, cell cycle, P27 pathway, and T cell receptor signaling pathway. RRM2 was significantly correlated with the infiltration level of CD8 T cells, Cytotoxic cells, DCs, Neutrophils, NK cells, and T helper cells (P <0.05). Conclusion: Over-expression of RRM2 predict adverse prognosis and is correlated with immune infiltrates in HCC. RRM2 may be a significant molecular biomarker for HCC diagnosis and prognosis.

Abstract:Patients with renal cell carcinoma (RCC) often develop resistance to antivascular drugs and eventually succumb to disease. However, the underlying molecular mechanism remains poorly understood. In this study, we demonstrated that RASAL2, a RAS GTPase-activating protein, played a tumor-suppressive role in RCC by targeting tumor angiogenesis. Firstly, we showed that RASAL2 was frequently epigenetically silenced in RCC, and its loss was negatively correlated with overall survival of RCC patients. Furthermore, we discovered that RASAL2 could inhibit RCC angiogenesis in vitro and in vivo. Mechanistically, we identified that RASAL2 could activate GSK3β by reducing Ser9 phosphorylation and subsequently decrease the expression of c-FOS and vascular endothelial growth factor A (VEGFA). Interruption of the p-GSK3β/c-FOS pathway with the specific inhibitor or small interfering RNA could reverse the expression of VEGFA, which may provide a new insight to prevent RCC from resistance to antivascular therapy.

Abstract:Objective: To explore the effect of protein expressions of ribonueleotide reductase (RR) subunits RRM1, RRM2 and RRM2B on the prognosis of hepatocellular carcinoma (HCC) and their potential relationships with the prognosis of HCC patients. Methods: One hundred and ninety-seven primary HCC tissues were subjected to detect the protein expressions of RRM1, RRM2 and RRM2B by immunohistochemistry. Correlations between RRM1, RRM2 and RRM2B protein expressions and clinicopathological parameters were analyzed by Spearman correlation coefficient. Survival rates were estimated by Kaplan-Meier method. Univariate analysis was performed by log-rank test. COX regression model was used to perform multivariate analysis and hazard ratio (HR) calculation. Results: Positive immunohistochemical stainings of RRM1, RRM2 and RRM2B proteins were mainly located in the cytoplasm of HCC cells. The expressions of RRM1 and RRM2 were not associated with prognosis of HCC patients (P > 0.05); however, the Kaplan-Meier survival analysis demonstrated that the survival time of HCC patients with high expression of RRM2B was longer than those with low expression (P < 0.05). Conclusion: Higher RRM2B protein level in HCC tissues may be a potential independent marker of good prognosis of HCC patients. DOI:10.3781/j.issn.1000-7431.2014.05.012
Ras-association Domain Family 10 Acts As a Novel Tumor Suppressor Through Modulating MMP2 in Hepatocarcinoma

Abstract:Ras-Association Domain Family 10 (RASSF10) is the last identified member of the RASSF family. The functional characteristics of this new gene in human cancers remain largely unclear. Here, we examined RASSF10 for the biological functions and related molecular mechanisms in hepatocellular carcinoma (HCC). We found that RASSF10 is expressed in normal human liver tissue, but is silenced or down-regulated in 62.5% (5/8) of HCC cell lines. The mean expression level of RASSF10 was significantly lower in primary HCCs compared with their adjacent normal tissues (P<0.005, n=52). The promoter methylation contributes to the inactivation of RASSF10 as demonstrated by bisulfite genomic sequencing and demethylation treatment analyses. Transgenic expression of RASSF10 in silenced HCC cell lines suppressed cell viability, colony formation and inhibited tumor growth in nude mice (QGY7703, P<0.01; HepG2, P<0.05). Furthermore, RASSF10 was shown to induce the cell accumulation in G1 phase with the increase of p27, as well as the decrease of cyclinD1 and CDK2/CDK4. Over-expression of RASSF10 also inhibited HCC cells migration (P<0.01) or invasion (P<0.05). Adhesion genes array revealed that Matrix Metalloproteinase 2 (MMP2) was a downstream effector of RASSF10. RASSF10 acting as a tumor suppressor to inhibit HCC invasion partially mediated by Focal Adhesion Kinase or p38 MAPK to decrease the accumulation of MMP2. Our study suggests that RASSF10 acts as a tumor suppressor for HCC.
Expression of HSPA2 in Human Hepatocellular Carcinoma and Its Clinical Significance

Abstract:Hepatocellular carcinoma (HCC) remains one of the most common cancers worldwide. HSPA2 has been highlighted as an important marker in many types of cancers. However, little is known about the role of HSPA2 in HCC. The objective of the current study was to investigate the expression pattern and clinicopathological significance of HSPA2 in patients with HCC. Quantitative reverse-transcriptase ploymerase chain reaction (qRT-PCR) was applied to examine HSPA2 messenger RNA (mRNA) expression in 52 pairs of HCC tissues and adjacent noncancerous tissues. Immunohistochemistry (IHC) was performed to examine HSPA2 protein expression in paraffin-embedded tissues from 119 HCC patients. Statistical analyses were applied to evaluate the diagnostic value and associations of HSPA2 expression with clinicopathological characteristics. We identified abnormally elevated mRNA expression of HSPA2 in HCC tissues compared to paired adjacent noncancerous tissues (P < 0.001). Clinicopathological analysis showed that HSPA2 expression was significantly correlated with tumor size (P = 0.013), histological differentiation (P = 0.04), and tumor stage (P = 0.001). Patients with higher HSPA2 expression had shorter overall survival time, whereas those with lower HSPA2 expression had longer survival time. Furthermore, Cox regression analyses showed that HSPA2 expression was an independent predictor of overall survival. In conclusion, our findings provide evidences that positive expression of HSPA2 in HCC may be important in the acquisition of an aggressive phenotype and it is an independent biomarker for poor prognosis of patients with HCC.
Upregulation of SGOL2 Predicts Poor Prognosis and Facilitates Hepatocellular Carcinoma Progression via Dysregulating the Cell Cycle by Directly Activating MAD2

Abstract:Abstract Background: Hepatocellular carcinoma (HCC) is the third leading cause of cancer-related death worldwide. Shugoshin-like protein 2 (SGOL2) is a centromeric protein that ensures the correct and orderly process of mitosis by protecting and maintaining centripetal adhesions during meiosis and mitosis. However, the role of SGOL2 in cancer is not well understood. Methods: The mRNA and protein levels of SGOL2 and survival analysis were conducted in The Cancer Genome Atlas (TCGA) and further validated in 2 independent cohorts. Differential genes correlated with SGOL2 and mitotic arrest deficient 2 like 1 (MAD2) were obtained using LinkedOmics. Subsequently, loss-of-function and rescue assays were carried out in vitro and in vivo to assess the functions of SGOL2 in hepatic tumorigenisis. Findings: We found that SGOL2 was significantly overexpressed in HCC and predicted unfavorable overall survival in HCC patients. Next, we identified 47 differentially expressed genes positively correlated with both SGOL2 and MAD2 to be mainly involved in the cell cycle. In addition, SGOL2 downregulation suppressed the migration, invasion, proliferation, stemness and EMT of HCC cells and inhibited tumorigenesis in vivo. Furthermore, SGOL2 promoted tumor proliferation by activating MAD2-induced cell cycle dysregulation, which could be reversed by the MAD2 inhibitor M2I-1. We also proved that SGOL2 activated MAD2 by directly binding with MAD2. Conclusions: The results of this study showed that SGOL2 acts as an oncogene in HCC cells by directly activating MAD2 and then dysregulating the cell cycle, thereby providing a potential target for HCC patients in the future.

Abstract:Objective:To study the expression of HAS2 in renal carcinoma and discuss the potential clinical significance.Method:qPCR was applied to detect the expression of HAS1 mRNA,HAS2 mRNA and HAS3 mRNA in five ccRCC cell lines(ACHN,Caki-1,OS-RC-2,786-O,SN12PM6) and human renal proximal tubular cell line(HK-2 cells).Additionally,Western blot was taken to examine the expression of the increased HAS protein in above renal cancer cells and ccRCC tissues.Result:HAS2 mRNA expression was obviously higher in the five ccRCC cell lines than that in the HK-2 cells,especially in the SN12PM6 cells(P<0.05).HAS1 mRNA expression reduced distinctly in the five ccRCC cell lines compared with the HK-2 cells(P>0.05).The expression of HAS3 mRNA was also lower in the renal cancer cell lines than that in the HK-2 cells except Caki-1 cells(P>0.05).The expression of HAS2 protein significantly increased in the five ccRCC cell lines.In addition,the expression of HAS2 protein was significant higher in the ccRCC tissue than those in the normal tissue(P<0.05).Conclusion:Hyaluronan synthase 2 was detected to highly express in the five ccRCC cell lines and ccRCC tissue,which shows that hyaluronan synthase 2 may play a crucial role in the develepment of renal carcinoma especially in ccRCC and may serve as a potential molecular marker for clear renal cell carcinoma.
Co-expression of LASS2 and TGF-β1 predicts poor prognosis in hepatocellular carcinoma

Abstract:Longevity assurance homolog 2 of yeast LAG1 (LASS2) has been reported to act as an important tumor suppressor in the development of human cancers. However, little is known about the prognostic value of LASS2 in hepatocellular carcinoma (HCC) . In the present study, we analyzed correlation between LASS2 and TGF-β1 levels, and evaluated their prognostic values in HCC patients. We first analyzed the expression of LASS2 and TGF-β1 in two independent cohorts (test cohort: 184 HCC patients; validation cohort: 118 HCC patients) using immunohistochemistry (IHC). Kaplan-Meier survival and Cox regression analyses were executed to evaluate the prognosis of HCC. The results of IHC analysis revealed a positive correlation between the expression of LASS2 and TGF-β1. HCC Patients with low expression of LASS2 and TGF-β1 had shorter overall survival (OS) and time to recurrence (TTR) than patients with high expression of LASS2 and TGF-β1. Furthermore, combination of LASS2 and TGF-β1 was an independent and significant risk factor for OS and TTR. In conclusion, low expression of LASS2 and TGF-β1 contributes to the aggressiveness and poor prognosis of HCC, and may represent a novel prognostic biomarker for HCC patients.
Genetic variants of SOS2, MAP2K1 and RASGRF2 in the RAS pathway genes predict survival of HBV-related hepatocellular carcinoma patients

Abstract:The RAS pathway participates in the cascade of proliferation and cell division process, and the activated RAS pathway can lead to tumorigenesis including hepatocellular carcinoma (HCC). However, few studies have explored the effects of genetic variants in the RAS pathway-related genes on the survival of patients with HBV-related HCC. In the present study, we assessed the associations between 11,658 single-nucleotide polymorphisms (SNPs) in 62 RAS pathway genes and the overall survival (OS) of 866 HBV-related HCC individuals, which were randomly split (1:1) into discovery and validation datasets. As a result, three potentially functional SNPs were identified, based on multivariable cox proportional hazards regression analyses, in SOS Ras/Rho guanine nucleotide exchange factor 2 ( SOS2, rs4632055 A > G), Ras protein-specific guanine nucleotide releasing factor 2 ( RASGRF2, rs26418A > G) and mitogen-activated protein kinase 1 ( MAP2K1, rs57120695 C > T), which were significantly and independently associated with OS of HBV-related HCC patients [adjusted hazards ratios (HRs) of 1.42, 1.32 and 1.50, respectively; 95% confidence intervals (CI), 1.14 to 1.76, 1.15 to 1.53 and 1.15 to 1.97, respectively; P = 0.001, < 0.001 and 0.003, respectively]. Additionally, the joint effects as the unfavorable genotypes of these three SNPs showed a significant association with the poor survival of HCC (trend test P < 0.001). The expression quantitative trait loci (eQTL) analysis further revealed that the rs4632055 G allele and the rs26418 A allele were associated with lower mRNA expression levels of SOS2 and RASGRF2 , respectively. Collectively, these potentially functional SNPs of RASGRF2 , SOS2 and M2PAK1 may become potential prognostic biomarkers for HBV-related HCC after hepatectomy.

Abstract:Background As a homologous counterpart to the prokaryotic oligonuclease found in the cellular cytoplasm and mitochondrion, REXO2 assumes a pivotal role in the maintenance of mitochondrial homeostasis. Nevertheless, the precise functions and mechanisms by which REXO2 operates within the context of hepatocellular carcinoma (HCC) have hitherto remained unexamined. Methods The expression levels of REXO2 in HCC tissues were evaluated through the utilization of the immunohistochemical (IHC) method, and subsequently, the association between REXO2 expression and the clinicopathological characteristics of HCC patients was scrutinized employing the χ 2 test. A battery of experimental assays, encompassing CCK8 viability assessment, cell colony formation, wound healing, and transwell assays, were conducted with the aim of elucidating the biological role of REXO2 within HCC cells. Complementary bioinformatics analyses were undertaken to discern potential correlations between REXO2 and immune infiltration in tumor tissues. Results Our IHC findings have unveiled a notable up-regulation of REXO2 within HCC tissues, and this heightened expression bears the status of an independent prognostic factor, portending an adverse outcome for HCC patients ( P < 0.05). Upon the attenuation of REXO2 expression, a discernible reduction in the rates of proliferation, invasion and migration of HCC cells ensued ( P < 0.05). Furthermore, transcriptome sequencing analysis has provided insights into the putative influence of REXO2 on the development of HCC through the modulation of TNF and NF-κB signaling pathways. Additionally, our bioinformatics analyses have demonstrated a positive correlation between REXO2 and tumor immune cell infiltration, as well as immune checkpoint CTLA-4. Conclusions In summation, our results posit an association between the up-regulation of REXO2 and adverse prognostic outcomes, alongside the involvement of immune-related signaling pathways and tumor immune infiltration within the realm of HCC.

[Expression of RASGRP2 in Lung Adenocarcinoma and Its Effect on Immune Microenvironment]

Abstract:Background: Lung cancer still has the highest incidence rate and mortality rate nowadays. In recent years, with the emergence of new drugs and the optimization of treatment mode, especially the clinical application of immunotherapy, the prognosis of lung cancer patients has been improved. However, the benefits of immunotherapy are still limited. Therefore, it is necessary to find new biomarkers to predict the prognosis of lung adenocarcinoma patients and explore its impact on the immune microenvironment. Methods: The Cancer Genome Atlas (TCGA) database was used to analyze the gene sequencing and clinical data of patients with lung adenocarcinoma. The distribution of RASGRP2 in lung adenocarcinoma was determined by using the human protein mapping database. The Kaplan-Meier plotter database was used to explore the relationship between the expression of RASGRP2 and the prognosis of patients with lung adenocarcinoma. KEGG and GO gene enrichment analysis was performed in patients with high and low expression of RASGRP2. TCGA database was used to analyze the co-expression genes of RASGRP2 and TIMER database was used to calculate the immune related lymphoid infiltration of RASGRP2 and its coexpression genes. The relationship between RASGRP2 expression and immune checkpoint expression was analyzed by using TIMER 2.0 database. Results: We found that RASGRP2 was low expressed in lung adenocarcinoma, and its expression level was related to the prognosis of patients. The high expression of RASGRP2 was involved in the process of hematopoietic cell formation and cell adhesion, and RASGRP2 played an important role in the process of T cell activation. Through TCGA database analysis, ZAP70, TBC1D10C, RASAL3, FGD2, CD37 and ACAP1 were significantly correlated with RASGRP2. The high expression of these genes leaded to the increase of the proportion of CD8+ T cells, memory CD4+ T cells, and the decrease of the proportion of neutrophils and Treg cells. Finally, we found that the expression of RASGRP2 was significantly correlated with the expression of CD274, CTLA4, LAG3 and TIGIT. Conclusions: RASGRP2 was abnormally expressed in lung adenocarcinoma and correlated with the infiltration level of immune related cells, which might influence the efficacy of immunotherapy.

Abstract:Background: Colorectal cancer (CRC) is a high-indence malignance of the digestive system with a high mortality rate in the world. Aim: The results are desired to provide an important theoretical basis for discovering new therapeutic targets for CRC. Objective: The expression of human endogenous retrovirus-H-long terminal repeat association protein 2 (HHLA2) in human CRC was detected to explore its correlationship with clinicopathological features and prognosis of patients and its potential in treating CRC. Methods: Western blot was employed to detect HHLA2 expression in fresh tissues obtained from 6 CRC patients' excisions, including cancer, paracancer, and normal issues. Immunohistochemical staining was employed to determine HHLA2 expression in paraffin-embedded specimens of 139 patients with colorectal cancer, and its relationship with the clinicopathological profiles and survival was analyzed. Small interfering RNA (siRNA) targeting HHLA2 was used to transfect CRC cells to silent HHLA2. MTT, plate colony formation, cell scratch, and Transwell assay were conducted to observe the proliferation, migration, and invasion of CRC cells. result: HHLA2 protein was expressed in human colorectal cancer tissues, paracancer tissues and normal tissues, and its expression significantly upregulated in cancer tissues (P&lt;0.01). HHLA2 expression level in colorectal cancer tissues showed a close correlationship with the invasion depth of tumor (P=0.000), metastasis of regional lymph nodes (P=0.018), clinical stage (P=0.010) and patient survival (P=0.011). No significant correlation was found with gender (P=0.873), age (P=0.864), location of tumor (P=0.768), degree of tumor differentiation (P=0.569) and distant metastasis (P=0.494). Survival analysis showed that the survival time of colorectal cancer patients with high and low HHLA2 expression group was 43.231 months and 55.649 months, respectively, with statistical difference between the two groups (P=0.001). HHLA2 silencing significantly inhibited the proliferation, migration and invasion of CRC cells. Results: HHLA2 protein was expressed in human colorectal cancer tissues, paracancer tissues and normal tissues, which was significantly upregulated in cancer tissues (P<0.01). HHLA2 expression level in CRC tissues showed a close correlationship with the invasion depth of the tumor (P=0.000), metastasis of regional lymph nodes (P=0.018), clinical stage (P=0.010), and patient survival (P=0.011). Abstract:Recently, long noncoding RNAs (lncRNAs) have been shown to have important regulatory roles in human cancer biology. The aim of this study was to evaluate the expression and biological role of lncRNA CASC2 in non-small cell lung cancer (NSCLC). By bioinformatics analysis, we found that CASC2 was significantly decreased in NSCLC. qRT-PCR was performed to investigate the expression of CASC2 in tumor tissues and corresponding non-tumor NSCLC tissues from 76 patients. The lower expression of CASC2 was remarkably correlated with advanced TNM stage and tumor size. Multivariate analyses found that CASC2 expression served as an independent predictor for overall survival of NSCLC. Moreover, overexpression of CASC2 significantly inhibited NSCLC cell proliferation both in vitro and in vivo. In conclusion, our study demonstrated that CASC2 is involved in the development and progression of NSCLC and shows that CASC2 may be a potential diagnostic and target for new therapies in patients with NSCLC.

Abstract:Hepatocellular carcinoma (HCC) is known to have a poor prognosis. Accumulating evidence indicates that RRM2 plays a critical role in the occurrence and progression of multiple human cancers. However, the knowledge about RRM2 in HCC is still insufficient, and further research is needed. Here, we first analyzed the expression and prognosis of RRM2 using TCGA and GTEx data, and found that RRM2 may play a potential carcinogenic role in HCC. Then, through a series of comprehensive analysis, including expression analysis, correlation analysis or survival analysis, non-coding RNAs (ncRNAs) that regulate RRM2 overexpression were identified. Finally, MIR4435-2HG/CYTOR were observed to be the most promising upstream lncRNAs for the miR-125b-5p/RRM2 axis in HCC. In addition, RRM2 expression was significantly positively related to immune cell infiltration, immune cell biomarker or immune checkpoint expression in HCC. Altogether, the upregulation of RRM2 mediated by ncRNAs correlates with poor prognosis and tumor immune infiltration of HCC.

Significance and Expression of CXC Chemokine Receptor 2 in Human Hepatocellular Carcinoma

Abstract:Objective To examine the expression of CXC chemokine receptor 2 (CXCR2) in human hepatoeellular carcinoma (HCC), and determine its role in tumorigenesis and progression of HCC.Methods CXCR2 mRNA and protein expression in 42 specimens of HCC and adjacent liver tissues,23 of normal liver tissues was examined by using semi-quantitative reverse transcription-PCR and Western blot.Results The expression levels of CXCR2 mRNA and protein in HCC, adjacent and normal liver tissues were 1.18 ± 0.32,0.83 ± 0.23 and 0.72 ± 0.23, and 1.78±0.20,0.98±0.13 and 0.90 ± 0.18 respectively.As compared with adjacent and normal liver tissues, the expression levels of CXCR2 mRNA and protein in HCC tissues were significantly increased (P <0.05).The expression levels of CXCR2 mRNA and protein in groups with intrahepatic metastasis, portal vein tumor thrombus and low differentiation were 1.37 ± 0.26,1.45 ± 0.34 and 1.31 ± 0.28, and 1.87 ± 0.20,1.93 ± 0.13 and 1.86 ± 0.18 respectively, and those in groups without intrahepatic metastasis, portal vein tumor thrombus and high differentiation were 1.08 ± 0.33,1.08 ± 0.27 and 1.06 ± 0.33, and 1.73 ± 0.18,1.73 ± 0.20 and 1.71 ± 0.19, with the difference being significant (P < 0.05).Furthermore, the protein level of CXCR2 was relevant to TNM staging.The protein levels of CXCR2 in stage Ⅲ -Ⅳ and stage Ⅰ - Ⅱ were 1.86 ± 0.20 and 1.72 ± 0.19, with the difference being significant (P < 0.05).Conclusion CXCR2 was highly expressed in HCC tissue.CXCR2 might play a role in the tumorigenesis,invasion and metastasis of HCC.
