Abstract:We outline a novel approach for the plasmonic detection of β-glucuronidase activity by modulating the silver mirror reaction at the nanoscale on gold nanostars. β-glucuronidase catalyzes the hydrolysis of a non-reducing substrate to generate reducing products that trigger the silver mirror reaction on gold nanostars to alter their surface plasmon resonance. By modulating the silver deposition on gold nanostars, the unique plasmonic property of silver-coated gold nanostars enables a significant change in the surface plasmon resonance that allows for a plasmonic readout for detecting the enzymatic activity. This plasmonic nanosensor enables a detection of the β-glucuronidase activity as low as 0.1 U/L, showing great promise as a plasmonic approach for enzyme detection.

Abstract:Localized surface plasmon resonance (LSPR) associated with metal nanostructures has developed into a highly useful sensor technique. Optical LSPR spectroscopy of nanostructures often shows sharp absorption and scattering peaks, which can be used to probe several bio-molecular interactions. Here, we report nanoplasmonic biosensors using LSPR on nanocup arrays (nanoCA) to recognize bio-molecular binding for biochemical detection. These sensors can be modified to quantify binding of small molecules to proteins for odorant and explosive detections. Electrochemical LSPR biosensors can also be designed by coupling electrochemistry and LSPR spectroscopy measurements. Multiple sensing information can be obtained and electrochemical LSPR property can be investigated for biosensors. In some applications, the electrochemical LSPR biosensor can be used to quantify immunoreactions and enzymatic activity. The biosensors exhibit better performance than those of conventional optical LSPR measurements. With multi-transducers, the nanoplasmonic biosensor can provide a promising approach for bio-detection in environmental monitoring, healthcare diagnostics, and food quality control.

Abstract:Surface plasmon resonance (SPR) and localized SPR (LSPR) effects have been shown as the principles of some highlysensitive sensors in recent decades. Due to the advances in nano-fabrication technology, the plasmon nano-array sensors based on SPR and LSPR phenomena have been widely used in chemical and bioloical analysis. Sensing with surface-enhanced field and sensing for refractive index changes are able to identify the analytes quantitatively and qualitatively. With the newly developed ultrasensitive plasmonic biosensors, platforms with excellent performance have been built for various biomedical applications, including point-of-care diagnosis and personalized medicine. In addition, flexible integration of plasmonics nano-arrays and combining them with electrochemical sensing have significantly enlarged the application scenarios of the plasmonic nano-array sensors, as well as improved the sensing accuracy.
A Bio-Inspired Plasmonic Nanosensor for Angiotensin-Converting Enzyme Through Peptide-Mediated Assembly of Gold Nanoparticles.

Abstract:Angiotensin-converting enzyme (ACE) can indicate blood pressure that relates to human health such as the cardiovascular disease. However, current methods are not competent to detect the ACE activity in a rapid and straightforward way. Plasmonic biosensors built on the modulation of metallic nanomaterials have emerged as novel tools for the detection of biomarkers. In this work, we report a bio-inspired strategy for the plasmonic detection of ACE in a rapid, sensitive, and selective way through peptide-mediated assembly of gold nanoparticles (AuNPs). In this biosensor, cysteine-angiotensin I-cysteine can assemble and aggregate AuNPs due to the Au-S bond. The presence of ACE can specifically catalyze the hydrolysis of angiotensin I, thus dissociating the cysteine-cysteine structure of the peptide that results in the disassembly and dispersion of AuNPs. This bio-inspired plasmonic nanosensor enables naked-eyed readout of ACE detection with great selectivity and high sensitivity with a LOD of 0.40 mU/mL. It also allows for the screening of ACE inhibitors and inhibitory peptides for the development of antihypertensive drugs or food. The biosensing technique developed in this work provides a new plasmonic approach that holds great promise as a point-of-care platform for biomedical diagnostics and the food industry.

Abstract:A plasmonic colorimetric strategy was designed for sensitive detection of biomolecules through enzyme guided silver nanoparticles (AgNPs) growth on gold nanostars (AuNS). The growth of AgNPs on AuNS led to a substantial blue shift of the localized surface plasmon resonance (LSPR) peak and the color change of AuNS from blue to dark blue, purple and ultimately orange. Both the LSPR blueshift wavelength and the color of detection solution containing AuNS, Ag+ and ascorbic acid 2-phosphate (AAP) depend on the amount of enzyme that catalyzed the dephosphorylation of AAP to reduce Ag+ on AuNS surface. Thus this strategy could be used for LSPR and naked-eye detections of both the enzyme such as alkaline phosphatase (ALP) and other biomolecules involved in biorecognition events using ALP as a tag. The LSPR detection method for ALP showed a linear range from 1.0pM to 25nM with a detection limit of 0.5pM. Using DNA as a mode target molecule, this technique showed a detection range from 10fM to 50pM DNA with a detection limit of 2.6fM through the convenient combination with hybridization chain reaction amplification. The proposed plasmonic colorimetric strategy could be extended as a general analytical platform for design of immunosensors and aptasensors with ALP as a label.

Abstract:Lipase is a neglected enzyme in the field of gold nanoparticle-based enzyme assays. This paper reports a novel colorimetric probe to rapidly visualize lipase activities by using Tween 20 functioned GNPs (Tween 20-GNPs) as a reporter. The present strategy hence could overcome the limitations caused by the heterogeneous interface in lipase assay. Catalytic hydrolytic cleavage of the ester bond in Tween 20-GNPs by lipase will trigger the rapid aggregation of GNPs at a high salt solution. The color change from red to purple could be used to sense the activity of lipase. The detection limit (3 sigma) is as low as 2.8 x 10(-2) mg/mL. A preliminary enzyme activity screening was carried out for seven commercially purchased lipase samples. It also has been successfully applied to detecting lipase in fermentation broth of Bacillus subtilis without any pretreatment.

Abstract:Alkaline phosphatase (ALP) is of extraordinary importance in living organisms and widely serves as a labeling enzyme in biochemical assays. Conventional methods based on the colorimetric readout of chemicals have issues in the photosensitivity and the difficulty in visibly distinguishing the color intensity. Herein, we developed a plasmonic assay for detecting ALP activity and its inhibitors based on gold nanoparticles (AuNPs) with good stability and naked-eye readout. Ag(I) specifically oxidizes TMB to produce positively charged oxTMB, which electrostatically aggregates negatively charged AuNPs. ALP as an enzyme specifically hydrolyzes a substrate to generate ascorbic acid, which reduces Ag(I) and modulates the aggregation of AuNPs. This plasmonic ALP assay has a linear detection range of 5–50 mU/mL with a limit of detection of 0.39 mU/mL. In combination with a smartphone, it enables monitoring ALP inhibitors including dimethoate and arsenic that are environmental contaminants. This plasmonic sensing strategy with good stability, visible readout, high specificity, low cost, and ease of operation provides a promising platform for enzyme detection and environmental monitoring.

Abstract:Current techniques for plasmonic immunoassay often require the introduction and additional conjugation of enzyme, and thus cannot accommodate conventional immunoassay platforms. Herein, we develop a plasmonic nanosensor that well accommodates conventional immunoassays and dramatically improves their sensitivity and stability. This plasmonic nanosensor directly employs alkaline phosphatase-triggered click chemistry between azide/alkyne functionalized gold nanoparticles as the readout. This straightforward approach broadens the applicability of nanoparticle-based immunoassays and has great potential for applications in resource-constrained settings.

Abstract:Immunoassay is commonly used for the detection of disease biomarkers, but advanced instruments and professional operating are often needed with current techniques. The facile readout strategy for immunoassay is mainly limited to the gold nanoparticles-based colorimetric detection. Here, we show that photothermal nanoparticles can be applied for biosensing and immunoassay with temperature as readout. We develop a plasmonic and photothermal immunoassay that allows straightforward readout by color and temperature based on crystal growth, without advanced equipment. It is demonstrated that alkaline phosphatase-triggered silver deposition on the surface of gold nanostars causes a large blue shift in the localized surface plasmon resonance of the nanosensor, accompanied by photothermal conversion efficiency changes. This approach also allows dual-readout of immunoassays with high sensitivity and great accuracy for the detection of prostate-specific antigen in complex samples. Our strategy provides a promising way for point-of-care testing and may broaden the applicability of programmable nanomaterials for diagnostics.

Abstract:Rapid and sensitive methodologies for the detection of protein are in urgent requirement for clinic diagnostics. Localized surface plasmon resonance (LSPR) of metal nanostructures has the potential to circumvent this problem due to its sensitive optical properties and strong electromagnetic near-field enhancements. In this work, an enzyme mediated plasmonic biosensor on the basis of a dual-functional nanohybrid was developed for the detection of thrombin. By utilizing LSPR-responsive nanohybrid and anaptamer-enzyme conjugated reporting probe, the sensing platform brings enhanced signal, stability as well as simplicity. Enzymatic reaction catalyzed the reduction of Au3+ to Au° in situ, further leading to the rapid crystal growth of gold nanoparticles (AuNPs). The LSPR absorbance band and color changed company with the nanoparticle generation, which can be real-time monitoring by UV–visible spectrophotometer and naked eye. Nanohybrid constructed by gold and magnetic nanoparticles acts as a dual functional plasmonic unit, which not only plays the role of signal production, but also endows the sensor with the function of magnetic separation. Simultaneously, the introduction of enzyme effectively regulates the programming crystal growth of AuNPs. In addition, enzyme also serves as signal amplifier owing to its high catalysis efficiency. The response of the plasmonic sensor varies linearly with the logarithmic thrombin concentration up to 10nM with a limit of detection of 200pM. The as-proposed strategy shows good analytical performance for thrombin determination. This simple, disposable method is promising in developing universal platforms for protein monitoring, drug discovery and point-of-care diagnostics.

Abstract:Recently emerged liposomal photoelectrochemical (PEC) bioanalysis has brought new opportunities for biosensor development. This work presents the new concept of liposome-assisted enzymatic modulation of plasmonic photoelectrochemistry for PEC bioanalysis, which was exemplified by an Au nanoclusters (NCs)-sensitized nanoporous TiO2 nanotubes (Au NC@TiO2 NT) photoelectrode and an alkaline phosphatase (ALP)-loaded liposomal immunoassay of heart-type fatty acid binding protein in a 96-well plate. After sandwich immunorecognition and subsequent lysis treatment, enzymatically generated ascorbic acid by the released ALP was directed to reduce Au3+ into Au nanoparticles using the Au NCs as seeds, leading to the in situ change of the photoelectrochemistry of the electrode and corresponding reduction of the photocurrent. The depressed signal could be correlated with the target concentration with good analytical performance in terms of sensitivity and selectivity. This work features the liposome-assisted enzymatic modulation of plasmonic photoelectrochemistry, which provides a new protocol for general PEC bioanalysis development.
Plasmonic Sensing Via Photoluminescence of Individual Gold Nanorod

Abstract:Label-free plasmonic sensors based on localized surface plasmon resonances of nanostructured noble metal materials usually transduce optical refractive index changes occurring in the vicinity of the nanostructures by optical scattering or by extinction. We demonstrate in experiments that the photoluminescence of plasmonic nanoparticles can also be employed to detect biological molecule binding events efficiently. Photoluminescence probably due to plasmon emission of a single gold nanorod presents a similar resonance peak and resembles the response to a refractive index change observed by scattering. The well-known biotin streptavidin binding assay was detected successfully using the photoluminescence of an individual isolated nanorod. The localized surface plasmon resonances' responses by scattering in situ with the same nanorod and control experiments were also performed to verify the sensing process. The results evidence that a nanoscale plasmonic sensor can also be archived effectively through the photoluminescence of a single plasmonic nanostructure. Furthermore, key parameters to optimize the photoluminescence based label-free plasmonic sensing are discussed in detail. The photoluminescence provides an alternative way for label-free plasmonic sensing. And it is believed that further exploration of this concept could lead to a whole new class of efficient plasmonic sensors with diverse and novel functionalities.
Localized Surface Plasmon Resonance (lspr) of Polyelectrolyte-Functionalized Gold-Nanoparticles for Bio-Sensing

Abstract:Gold-nanoparticles can be used as a robust tool for bio-sensing based on localized surface plasmon resonance (LSPR). Both local refractive index change and self-assembly of the nanoparticles can lead to some spectral shifts of the LSPR extinction peak. Using different mechanisms of LSPR peak shifts, two types of transducers are set up to detect the streptavidin concentration through the specific recognition with biotin moleculars, which are conjugated to gold-nanoparticles. One is based on the self-assembly of gold-nanoparticles in solution, and the other is based on the increase of the local refractive index near the surface of the immobilized gold-nanoparticles after streptavidin–biotin binding. The sensitivity and efficiency of the two transducers are compared. Gold-nanoparticles are functionalized with polyelectrolyte, which can improve significantly the stability of the nanoparticles and reduce the unspecific binding of streptavidin. Polyelectrolyte functionalization also provides a simple way to conjugate gold-nanoparticles with charged molecules such as biotin and BSA–biotin.
Plasmonic Nanozymes: Localized Surface Plasmonic Resonance Regulates Reaction Kinetics and Antibacterial Performance.

Abstract:Among the members of the rapidly growing nanozyme family, plasmonic nanozymes stand out because of their unique localized surface plasmon resonance (LSPR) characteristics and tunable catalytic activity. We prepared a plasmonic nanozyme of Au gold nanoparticles (AuNPs) and Cu metal-organic framework nanosheets (Cu-MOFNs). The Cu-MOFNs have peroxidase-like activity, while AuNPs present unique LSPR characteristics. We found that the as-prepared AuNPs/Cu-MOFNs composite presents 1.6-fold faster reaction kinetics under LSPR excitation compared to that in the dark. Investigations of energy levels, radical capture, and dark-field scattering spectroscopy revealed that LSPR of AuNPs as well as matched energy levels can facilitate efficient hot electron transfer, which could readily cleave the chemical bond of the substrate and accelerate the reaction kinetics. On the basis of these results, we achieved enhanced antibacterial therapy and wound healing using plasmonic AuNPs/Cu-MOFNs. This study spotlights the superiority of plasmonic nanozymes in improving the enzyme-like performance of nanozymes.
Application of Ag Nanoparticle-Modified Fiber Probe for Plasmonic Catalysis Reaction

Abstract:In this study, a localized surface plasmon resonance (LSPR) fiber probe modified with Ag nanoparticles (NPs) was developed. The LSPR fiber probe not only serves as a reaction substrate for plasmonic catalysis, but also detects in situ surface-enhanced Raman spectroscopy (SERS) signals from the reaction product, thereby achieving the integration of the plasmonic catalysis reactions and SERS signal detection. To fabricate the LSPR probe, plasmonic Ag NPs were first self-assembled on the surface of the fiber probe with assistance by the amination and silanization of (3-aminopropyl) trimethoxysilane (APTMS) molecules. p-Aminothiophenol (PATP) was chosen as a model molecule for plasmonic catalytic reaction. By regulating the self-assembly time of the Ag NPs, a uniform distributed monolayer of Ag NPs was formed on the surface of the probe, with which excellent plasmonic catalysis effects and SERS signal collection from the reaction product of 4,4'-dimercaptoazobenzene (DMAB) were achieved. It was found that the characteristic SERS signal of the plasmonic catalytic reaction product DMAB obtained from internal excitation and collection was 12.8 times more intense than that from the external excitation and collection under the same laser intensity conditions, demonstrating that the internal excitation and collection method was advantageous in the plasmonic catalysis and SERS signal detection. The LSPR fiber probe was demonstrably qualified to quantitatively detect the concentrations of PATP solutions in the concentration ranges 10(-4)-10(-8) mol.L-1. Using the LSPR fiber probe, we also realized an in situ kinetics study of the PATP coupling reaction enhanced by plasmonic catalysis. The results showed that the Ag NP-based LSPR fiber probe with internal excitation and collection modes had the advantages of high sensitivity, low cost, facile preparation, and most importantly, applicability to in situ detection in a flexible manner with less damage to the samples. The preliminary study also indicated that it was feasible to combine the LSPR fiber probe with near-field scanning optical microscopy, not only to obtain morphological images of the surface but also to simultaneously perform the plasmonic catalysis reaction and the detection of micro-domains of the surface. This permitted the acquisition of a two-dimensional distributional assessment of surface reactions by the plasmonic catalysis.
Plasmonic Array at the Liquid-Liquid Interface: A Dual-Mode Optical Sensing Platform for Multianalytes

Abstract:Analyte-triggered nanoparticle (NP) assemblies in bulk colloidal suspension have been extensively utilized in various optical sensors. Nevertheless, the assembling process is still limited by the slow diffusion dynamics of NPs and the low concentration of analytes in trace detections, which hinders further improvement of the sensitivity and repeatability of the sensors. In this work, by functionalizing the gold NPs with specific ligands, we constructed a dual-mode optical sensing platform for multianalytes based on the plasmonic NP array at the liquid-liquid interface. Through emulsification, the NP diffusion kinetics are boosted for several orders, and the NPs are condensed from the bulk aqueous phase to the liquid-liquid interface as a plasmonic array. The as-formed metasurface generates major reflectance and surface-enhanced Raman scattering changes in response to analytes, providing two optical sensing modes. As prototypes, cysteine and glucose are selected as the target molecules, achieving the limit of detection as 193 ± 2 and 297 ± 12 pM, respectively.

Modified with macrocyclic supramolecules, noble metal nanoparticles possess improved biocompatibility and highly efficient biomolecule immobilization and recognition via host-guest interaction. In this work, we synthesize high-quality and monodisperse beta-cyclodextrin-capped gold nanoparticles (beta-CD-capped AuNPs) in one-step facile and eco-friendly process, where beta-CD functions as both reducing and capping agent. Integrated with a microfiber, the beta-CD-capped AuNPs deliver promising localized surface plasmon resonance properties and ultralow detection limit of cholesterol molecules of 5 aM. The selective recognition of our proposed biosensor to cholesterol is further verified by interference study and analysis of real human serum samples. This study shows the bright prospects of supramolecular chemistry assisted plasmonics in achieving highly integrated and highly sensitive biosensing with extremely low consumption of clinical samples and small footprint.
Toward Plasmonic Biosensors Functionalized by A Photoinduced Surface Reaction

Abstract:We present a method for efficient coupling of amine nucleophilic molecules of choice to a nanostructured gold surface via photoinduced surface chemistry. The method is based on photoactive self-assembled monolayers and can be used to functionalize localized surface plasmon resonance (LSPR) based biosensors with biorecognition motifs while reducing nonspecific binding via introduction of hydrophilic units. The photoactive linker molecule, 5-bromo-7-nitroindoline, couples nucleophilic molecules such as biotin ethylenediamine to a surface when exposed to UV-light. The specific, noncovalent recognition between biotin and streptavidin is used for demonstration of a simple biorecognition assay based on the LSPR sensing principle. By doing so, one can envision that the binding of any streptavidin fusion protein, being attached to specific spots at the gold surface, is monitored by an LSPR peak shift. Since the surface functionalization is based on a photoinduced reaction, this method can be used to functionalize the surface in a local and site-specific way, and biomedical applications such as drug-screening platforms, microarrays, solid support protein synthesis, and even single molecule experiments can be envisioned.
