Abstract:A technique for encapsulating fluorescent organic probes in a micelle system offers an important alternative method to manufacture watersoluble organic nanoparticles (ONPs) for use in sensing Hg2+. This article reports on a study of a surfactant-free micelle-like ONPs based on a 3,6-di(2-thieny1)-2,5-dihydropyrrolo[3,4-c]pyrrole-1,4-dione (TDPP) amphiphile, (2(2-(2-methoxyethoxy)ethyl)-3,6-di(2-thiophyl)-2,5-dihydropyrrolo [3,4-c] pyrrole-1,4-dione (NDPP) fabricated to monitor Hg2+ in water. NDPP was synthesized through a simple one-step modification of a commercially available dye TDPP with a flexible and hydrophilic alkoxy. This study reports, for the first time, that TDPP dyes can respond reversibly, sensitively, and selectively to Hg2+ through TDPP-Hg-TDPP complexation, similar to the well-known thymine(T) Hg thymine(T) model and the accompanying molecular aggregation. Interestingly, transmission electron microscopy (TEM) and dynamic light scattering (DLS) confirmed that, in water, NDPP forms loose micelle-like fluorescent ONPs with a hydrohobic TDPP portion encapsulated inside. These micelle-like nanoparticles offer an ideal location for TDPP Hg complexation with a modest molecular aggregation, thereby providing both clear visual and spectroscopic signals for Hg2+ sensing. An estimated detection limit of 11 nM for Hg2+ sensing with this NDPP nanoparticle was obtained. In addition, NDPP ONPs show good water solubility and high selectivity to Hg2+ in neutral or alkalescent water. It was superior to most micelle based nanosensors, which require a complicated process in the selection or synthesis of suitable surfactants. The determinations in real samples (river water) were made and satisfactory results were achieved. This study provides a low-cost strategy for fabricating small molecule-based fluorescent nanomaterials for use in sensing Hg2+. Moreover, the NDPP nanoparticles show potential ability in Hg2+ ion adsorption and recognization of cysteine using NDPP-Hg composite particle.

Abstract:Mercury(ii), which is a well-known toxic species, exists in the industrial waste water in many cases. In the present work, CdTe quantum dots (QDs) are studied as a fluorescence probe for Hg2+ detection. Ag ions are induced to QDs to enlarge their detection concentration range. l-cysteine is employed in the QD-based fluorescence probe to connect QDs with Hg2+. X-ray diffraction, transmission electron microscopy and X-ray photoelectron spectroscopy results indicate the formation of zinc blende CdTe QDs with sizes of ∼5 nm and the existence of Ag+ in crystalline CdTe. Photoluminescence (PL) spectra and PL decay spectra were acquired to investigate the emission mechanism of Ag-doped CdTe QDs, revealing multi-emission in QD samples with higher concentrations of Ag+ doping. The highest PL quantum yield of the QD samples was 59.4%. Furthermore, the relationship between the fluorescence intensity and the concentration of Hg2+ has been established. Two linear relationships were obtained for the plot of F/F0 against Hg2+ concentration, enlarging the detection concentration range of Hg2+.

Abstract:Mercury sensing and imaging in the bio-system is essential for comprehending its toxicity and therapies. Based on the merocyanine scaffold, we designed and synthesized two novel near-infrared (NIR) fluorescent probes for detecting Hg 2+ . The release of chloro-substituted merocyanine structure on the probe CyHg-Cl enables fluorescence enhancement rapidly by introducing Hg 2+ . In addition, the probe CyHg-Cl exhibits NIR emission and a low detection limit of 0.59 μM. Finally, the probe CyHg-Cl was used to detect Hg 2+ in live cells and real water samples.

Abstract:Background Carbon dots (CDs) emitting near-infrared fluorescence were recently synthesized from green leaves. However, the Hg2+ detection of CDs was limited because of the insufficient water solubility, low fluorescence and poor stability. Methods Dual fluorescence emission water-soluble CD (Dual-CD) was prepared through a solvothermal method from holly leaves and low toxic PEI1.8k. PEG was further grafted onto the surface to improve the water solubility and stability. Results The Dual-CD solution can emit 487 nm and 676 nm fluorescence under single excitation and exhibit high quantum yield of 16.8%. The fluorescence at 678 nm decreased remarkably while the emission at 470 nm was slightly affected by the addition of Hg2+. The ratiometric Hg2+ detection had a wide linear range of 0–100 μM and low detection limit of 14.0 nM. In A549 cells, there was a good linear relation between F487/F676 and the concentration of Hg2+ in the range of 0–60 μM; the detection limit was 477 nM. Furthermore, Dual-CD showed visual fluorescence change under Hg2+. Conclusion Dual-CD has ratiometric responsiveness to Hg2+ and can be applied for quantitative Hg2+ detection in living cells.

Abstract:A novel thioketal containing fluorescent dye 1OS was elaborately designed for mercury(II) determination, which not only integrated the benefits of the remarkable specificity of Hg2+-promoted deprotection reaction and the robust emission of AIEgen in aggregated state, but also provided a surprising platform for ACQ-to-AIE switching, i.e. Hg2+-triggered ACQ to AIE transformation. Once encountered Hg2+ ions, the deprotection reaction happened immediately within seconds, accompanied with a conversion of 1OS (ACQ) to 1O (AIE). Excitedly, the probe shows remarkable selectivity and specificity towards Hg2+ both in solution, aggregated and solid states with significant fluorescence color change. When added trace Hg2+ ions into 1OS solution, its PL intensity declined completely with emission color changed from bright green to darkness with LOD as 0.36 mu M. Oppositely, a "turn on" signal was achieved in the aggregated state (fw = 98 %), which exhibited a dramatically enhancement of PL intensity along with fluorescent emission altered from darkness to brilliant yellow. Other common metal cations gave no interference to the probe. Meanwhile, the constructed test strip could "turn on" signify Hg2+ in water solution.

Abstract:The development of probes for selective detection of mercury ions (Hg2+) is an important mission to accomplish because of the toxicity and ubiquity of Hg2+. Herein, we designed and synthesized a novel fluorescent probe O-(N-butyl-1,8-naphthalimide)-4-yl-O-phenyl carbonothioate (CBONT) for selective and sensitive detection of Hg2+ by turn-on fluorescence spectroscopy. Probe CBONT exhibited a fast response for Hg2+ with excellent sensitivity (limit of detection = 1.9 nM, 3 sigma/slope), and it might be attributed to the adoption of a new recognition receptor of carbonothioate moiety. Additionally, probe CBONT could serve as a "naked-eye" indicator for Hg2+. Finally, probe CBONT could be successfully applied to detect the concentrations of Hg2+ in real water samples. Our proposed recognition receptor would open up new, exciting opportunities for designing highly selective and ultrasensitive fluorescent probes for the determination of Hg2+ in real water samples.

Abstract:Mercury is highly toxic. In this work, a fluorescent probe BTD‐Hg‐Lyso was designed and developed for the specific detection of Hg2+. BTD‐Hg‐Lyso revealed a large Stokes shift (210 nm). It was successfully applied to detect Hg2+ in lysosomes. In addition, BTD‐Hg‐Lyso showed good fluorescence enhancement on Hg2+ in HeLa cells, zebrafish and tobacco seedlings (rhizomes and stems). Mercury is highly toxic, and appropriate amounts of the mercury‐selenium complex can protect plasma. However, when excessive mercury (II) ions (Hg2+) are exposed to human skin or ingested directly, it can lead to irreversible accumulation in the body. Therefore, detecting the presence of Hg2+ in cells is important. A novel fluorescent probe BTD‐Hg‐Lyso was designed and constructed based on the intramolecular charge transfer mechanism. Thiotaldehyde could specifically recognize Hg2+ so that the probe could produce a fluorescence enhancement effect. In addition, the fluorescent probe BTD‐Hg‐Lyso exhibited the advantages of large Stokes shift (210 nm) and good selectivity. More importantly, the probe BTD‐Hg‐Lyso could be used for the determination of Hg2+ in cellular lysosomes. BTD‐Hg‐Lyso was able to image Hg2+ in HeLa cells, zebrafish, and tobacco seedlings (rhizome and stem) successfully.

Abstract:Mercury ions are highly toxic to living organisms and environment, hence Monitoring and quantification of mercury ions remain great significance. Herein, we have designed and synthesized a novel reaction-based fluorescence probe RANS for detection of Hg2+ by using a xanthene derivate as fluorophore and a thiosemicarbazide moiety as the recognition site. By applying mercury-promoted desulfurization reactions, probe RANS exhibited excellent capability of detecting Hg2+, such as rapid response (120 s), high sensitivity (0.019 μM) and good selectivity. Upon reaction with Hg2+, the thiosemicarbazide group of probe RANS formed 1,3,4-oxadiazole along with a significant color change from colorless to light green. Importantly, RANS has been successfully applied to selectively imaging Hg2+ in living cells with low cytotoxicity and quantitative detection of Hg2+ in real water samples.

Abstract:Hg2+ has a significant hazardous impact on the environment and ecosystem. There is a great demand for new methods with high selectivity and sensitivity to determine mercury in life systems and environments. In this paper, a novel turn-on Hg2+ fluorescent probe has been reported with a naphthalimide group. The Hg2+ fluorescent probe was designed by the inspiration of the well-known specific Hg2+-triggered thioacetal deprotection reaction. A 1,2-dithioalkyl group was chosen as the specific recognition site of Hg2+. The probe showed weak fluorescence without Hg2+, and the color of the solution was light yellow. In the presence of Hg2+, the probe reacted specifically with the mercury ion to produce an aldehyde and emitted strong fluorescence, and the color of the solution also turned light green, thus realizing the monitoring of the mercury ion. The Hg2+ fluorescent probe showed outstanding sensitivity and selectivity toward Hg2+. Furthermore, the Hg2+ fluorescent probe could work in a wide pH range. The linear relationship between the fluorescence intensity at 510 nm and the concentration of Hg2+ was obtained in a range of Hg2+ concentration from 2.5 × 10–7 to 1.0 × 10–5 M. The detection limit was found to be 4.0 × 10–8 M for Hg2+. Furthermore, with little cell toxicity, the probe was successfully applied to the confocal image of Hg2+ in PC-12 cells.This article has not yet been cited by other publications.

Abstract:AIM:To determine whether the use of a mixed polymeric micelle delivery system based on vitamin E succinate (VES)-grafted-chitosan oligosaccharide (CSO)/VES-grafted-chitosan (CS) mixed micelles (VES-g-CSO/VES-g-CS MM) enhances the delivery of C-DMSA, a theranostic fluorescent probe, for Hg2+ detection and detoxification in vitro and in vivo. METHODS:Mixed micelles self-assembled from two polymers, VES-g-CSO and VES-g-CS, were used to load C-DMSA and afforded C-DMSA@VES-g-CSO/VES-g-CS MM for cell and in vivo applications. Fluorescence microscopy was used to assess C-DMSA cellular uptake and Hg2+ detection in L929 cells. C-DMSA@VES-g-CSO/VES-g-CS MM was then administered intravenously. Hg2+ detection was assessed by fluorescence microscopy in terms of bio-distribution while detoxification efficacy in Hg2+-poisoned rat models was evaluated in terms of mercury contents in blood and in liver. RESULTS:The C-DMSA loaded mixed micelles, C-DMSA@VES-g-CSO/VES-g-CS MM, significantly enhanced cellular uptake and detoxification efficacy of C-DMSA in Hg2+ pretreated human L929 cells. Evidence from the reduction of liver coefficient, mercury contents in liver and blood, alanine transaminase and aspartate transaminase activities in Hg2+ poisoned SD rats treated with the mixed micelles strongly supported that the micelles were effective for Hg2+ detoxification in vivo. Furthermore, ex vivo fluorescence imaging experiments also supported enhanced Hg2+ detection in rat liver. CONCLUSION:The mixed polymeric micelle delivery system could significantly enhance cell uptake and efficacy of a theranostic probe for Hg2+ detection and detoxification treatment in vitro and in vivo. Moreover, this nanoparticle drug delivery system could achieve targeted detection and detoxification in liver.

Abstract:Mercury is a highly toxic and non-essential element that is found in every corner of the globe. The small amount of mercury produced by various pathways eventually enters freshwater and marine ecosystems, circulating through the food chain (especially fish) and causing various environmental problems in aspects including plants, animals, and human. There are several traditional quantitative methods developed for mercury ions (II) analysis in water samples. However, due to the complexity of the detection process, high cost and strong technical expertise, it is difficult to detect mercury ions in real-time. Therefore, in recent years, a large number of researchers have developed small-molecule fluorescent probes for Hg ions detection. Fluorimetry has the advantages of convenient detection, short response time, high sensitivity and good selectivity. This review summarized the small-molecule fluorescent probes for mercuric ion detection developed in recent years according to the chemical structural classification, compared their performances and elaborated the mechanism. We hope that the review will help the researches for the designs of metal ions fluorescent probes and their applications with certain reference value.
A Novel Camphor-Based “Turn-on” Fluorescent Probe with High Specificity and Sensitivity for Sensing Mercury(II) in Aqueous Medium and Its Bioimaging Application

Abstract:Precise detection of Hg2+ in an ecological environment and biological system is of great importance. Hence, in this paper, we have constructed a simple fluorescent turn-on probe CP-Hg toward Hg2+ based on natural and renewable camphor. A significantly enhanced green fluorescence was clearly observed only with the participation of Hg2+ due to the Hg2+-mediated deprotection reaction of thioacetal. The probe CP-Hg could also sensitively detect Hg2+ with a detection limit of 19.3 nM and a large Stokes shift of 153 nm in a near-perfect aqueous solution. The reaction mechanism was fully analyzed through 1H NMR (proton nuclear magnetic resonance), ESI-MS (electrospray ionization mass spectrometry), and DFT (density functional theory) calculations. Moreover, the CP-Hg-coated test strips were successfully fabricated for naked-eye detection of Hg2+. Abstract:2-(2',5'-Dihydroxy-phenyl)-4(3H)-quinazolinone (DHPQ), a new fluorescent dye that exhibits excited state intramolecular proton transfer (ESIPT) reaction and possesses good photophysical properties, is synthesised and used as fluorescent probe for detection of Hg2+. Mercuric ions can be detected and quantitated by measuring the fluorescent intensity decrease of the probe. The decrease of fluorescence intensity of DHPQ upon the addition of Hg2+ was attributed to the blocking of ESIPT reactions of DHPQ and quenching its fluorescence. The analytical performance characteristics of the proposed Hg2+ probe were investigated. The probe can be applied to the quantification of Hg2+ with a concentration range covering from 8.0 x 10(-7) to 2.0 x 10(-4) mol L-1, with a working pH range of 5.5-6.5. It shows excellent selectivity for Hg2+ over other transition metal cations. The proposed method was testified for the Hg2+ assay in river water samples with satisfying recoveries.

Abstract:Two novel Hg2+ fluorescent probes W-5 and W-6 have successfully been designed and synthesized based on benzothiazole derivatives as fluorophores and thiocarbonate moiety as recognition group. The structures of W-5 and W-6 were confirmed by NMR, HRMS and X-ray crystallography analysis. The probes W-5 and W-6 showed the ratio (F-525/F-405, large emission shift 120 nm) and "turn-on" fluorescence signals, respectively. Their advantages are excellent selectivity and anti-interference. Moreover, the probes W-5 and W-6 can provide excellent methods for the determination of Hg2+ in BSA and calf serums. (C) 2020 Elsevier Ltd. All rights reserved.

Abstract:Mercury (Hg) is one of the heavy metal pollutants in the environment. Even a very small amount of mercury can cause serious harm to human beings. Herein, we reported a new carbonothioate-based fluorescent probe for the detection of Hg2+ without interference from other metal ions. This probe possessed a very large Stokes shift (192 nm), which could improve the detection sensitivity by minimizing the interferences resulted from self-absorption or auto-fluorescence. With the addition of Hg2+ to the probe solution, considerable fluorescence enhancement was observed. Additionally, the Hg2+ concentration of 0-16 mu M and fluorescence intensity showed a good linear relationship (y = 22106x + 53108, R-2 = 0.9955). Finally, the proposed probe was used to detect Hg2+ in real water samples, and its result was satisfactory. Therefore, our proposed probe would provide a promising method for the determination of Hg2+ in the environment.

Abstract:As a kind of heavy metal ions, mercury ions (Hg2+) have important effects on ecological environment and human health. Since Hg2+ interact with mercaptan molecules in the living body, they gather in the living body and endanger health or even life, so the detection of Hg2+ is indispensable. In this research, taking advantage of thymine moiety as a specific ligand for chelating with Hg2+, a new type of "T-Hg-T" fluorescent probe BC-Hg was designed and synthesized to specially detect Hg2+ in real water samples and living systems. The barbituric acid unit in the probe can react with Hg2+ and then deprotonate to form polymer and precipitate. Probe BC-Hg has a rapid response to the detection of Hg2+ and strong anti-interference ability. It has a good linear detection of Hg2+ in the range of 0-1.5 mu M, so it can quantitatively detect Hg2+ within a certain concentration range. Probe BC-Hg has obvious "naked eye" colorimetric effect and can be used to determine the generation of Hg2+ polymer by the Tyndall effect. In addition, probe BC-Hg can also be used to detect Hg2+ in real water samples, cells, and zebrafish. Therefore, the probe can serve as an effective tool for detecting Hg2+ in environment and organisms.

Abstract:To determine whether the use of a mixed polymeric micelle delivery system based on vitamin E succinate (VES)-grafted-chitosan oligosaccharide (CSO)/VES-grafted-chitosan (CS) mixed micelles (VES-g-CSO/VES-g-CS MM) enhances the delivery of C-DMSA, a theranostic fluorescent probe, for Hg2+ detection and detoxification in vitro and in vivo.Mixed micelles self-assembled from two polymers, VES-g-CSO and VES-g-CS, were used to load C-DMSA and afforded C-DMSA@VES-g-CSO/VES-g-CS MM for cell and in vivo applications. Fluorescence microscopy was used to assess C-DMSA cellular uptake and Hg2+ detection in L929 cells. C-DMSA@VES-g-CSO/VES-g-CS MM was then administered intravenously. Hg2+ detection was assessed by fluorescence microscopy in terms of bio-distribution while detoxification efficacy in Hg2+-poisoned rat models was evaluated in terms of mercury contents in blood and in liver.The C-DMSA loaded mixed micelles, C-DMSA@VES-g-CSO/VES-g-CS MM, significantly enhanced cellular uptake and detoxification efficacy of C-DMSA in Hg2+ pretreated human L929 cells. Evidence from the reduction of liver coefficient, mercury contents in liver and blood, alanine transaminase and aspartate transaminase activities in Hg2+ poisoned SD rats treated with the mixed micelles strongly supported that the micelles were effective for Hg2+ detoxification in vivo. Furthermore, ex vivo fluorescence imaging experiments also supported enhanced Hg2+ detection in rat liver.The mixed polymeric micelle delivery system could significantly enhance cell uptake and efficacy of a theranostic probe for Hg2+ detection and detoxification treatment in vitro and in vivo. Moreover, this nanoparticle drug delivery system could achieve targeted detection and detoxification in liver.

Abstract:Due to the highly toxic nature of mercury ions to living organisms, accurately detecting Hg2+ in water samples and biological systems is of great significance. In this study, we designed and synthesized a novel red-to-near-infrared Aggregation-Induced Emission (AIE) fluorescent probe (named as DS) based Fluorene derivatives on specifically for Hg2+ detection. Probe DS can visually identify Hg2+ through an red-to-near-infrared fluorescence enhancement change, characterized by a large Stokes shift (130 nm) and AIE feature. This probe offers a fast response, high selectivity and sensitivity. The Hg2+-induced deprotection reaction of the thioketal mechanism was thoroughly investigated using nuclear magnetic resonance spectroscopy (NMR), mass spectrometry (MS) and density functional theory (DFT) calculation. Additionly, dynamic light scattering (DLS) results indicated that the aggregation states changes of the molecular play a crucial role in the AIE fluorescence response of probe DS toward Hg2+. The red-to-near-infrared response with AIE feature not only avoids the interference of auto-fluorescence signals in complex environments, but also reduces the fluorescence quenching caused by probe molecular aggregation. This makes probe DS highly suitable for high-quality imaging detection of Hg2+ in aqueous environments. Furthermore, probe DS demonstrates the capability for visual fluorescence detection of Hg2+ concentrations in water sample, plant roots and living cells.
