Abstract:Decarboxylative C(sp2)-heteroatom cross-coupling reactions hold extraordinary potential for the sustainable preparation of biologically active scaffolds. Herein, we report a copper sulfate/1,10-phenathroline catalytic system for the decarboxylative intramolecular C(sp2)-O, C(sp2)-S, and C(sp2)-N coupling reactions leading to the construction of a series of benzo[b]furans, benzo[b]thiophenes, and indole derivatives from the corresponding coumarins, thiocoumarins, or quinolones, respectively. Our mechanistic study based on benzo[b]furan formation suggests a three-step process of the transformations, which consists of (i) base-mediated hydrolytic ring opening of coumarin, (ii) copper-oxygen co-initiated radical decarboxylation, and (iii) copper-catalyzed C-heteroatom cross coupling. Application of this method in the total synthesis of egonol, a bioactive natural product, was demonstrated successfully, with an overall yield of 51.7%.

Abstract:Copper(II)-catalyzed synthesis of sulfones from sulfinate salts and aryl halides was investigated by means of a combination of experiment and DFT calculation. Experimental results demonstrated the wide applicability of the title approach. The reaction mechanisms are revealed by in-situ IR and theoretical study. It reveals remarkable ligand effect the bidentate amine plays in the reaction, that is, it initially activats the C-I bond of iodobenzene so as to enhance the whole catalytic process. Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.

Abstract:A copper‐catalyzed olefinic C(sp2)–H activation/carbene insertion/ester hydrolysis/cyclization sequence has been developed for the synthesis of substituted 2(5H)‐furanones employing readily available enaminones and diazo esters as substrates. This method features operationally simple and good functional group tolerance. The transformation could be scaled up to gram quantities and the obtained 2(5H)‐furanone derivatives could be easily converted to a wide range of useful synthetic building blocks, indicative of the applicability of the synthetic strategy. Isotope labeling experiments demonstrated the ester group of diazo esters was hydrolyzed during the reaction.

Abstract:The decarboxylative technique has been tremendously uplifted due to its ability to increase the reactivity of the substrate. Notably, coumarins and sulfoximines are highly proficient scaffolds with great effectiveness and encourage emerging chemists to create their derivatives. Herein, we have been reporting a competent, one-pot, better-yielding and smooth decarboxylative cross-coupling reaction using copper catalyst for coumarin-3-carboxylic acids and substituted sulfoximines, respectively. The strategy was employed efficiently via radical addition and subsequent decarboxylation to form the CN bond. Different substrate scopes were analyzed, and optimization was carried out through various copper catalysts, solvents, oxidants & additives. Products obtained through various substrates were varying in moderate to reasonable amounts.

Abstract:A copper(II)-catalyzed three-component reaction of 2,3-allenoic acids, sulfur dioxide, and aryldiazonium tetrafluoroborates under mild conditions is developed, leading to 4-sulfonylated furan-2(5H)-ones in good yields. Not only sodium metabisulfite but also 1,4-diazabicyclo[2.2.2]octane-sulfur dioxide (DABCO center dot(SO2)(2)) is workable under the conditions. This transformation proceeds through a radical process initiated by the addition of arylsulfonyl radical to the C-central position of 2,3-allenoic acid. A broad substrate scope is demonstrated, and many sensitive functional groups are tolerated.

Abstract:The cooperative Lewis‐ and Brønsted acid catalysis makes convergent synthesis of 3(2H)‐furanones through a three‐component coupling of 1,3‐diynes, alkyl glyoxylates and water. Control experiments support that Lewis acid‐catalyzed highly chemo‐, regio‐ and stereoselective alkyne‐carbonyl metathesis of 1,3‐diynes and alkyl glyoxylates might be the initial step of this multicomponent annulation. Further chemo‐ and regioselective hydration of the alkyne‐carbonyl metathesis product and subsequent oxa‐Michael addition promoted by Brønsted acid results in the formation of two C‐O bonds of the five‐membered oxygen heterocycle.

Abstract:Tetracyclic coumarins are a class of important compounds with diverse and superior pharmacological activities. However, a direct stereoselective method from simple and readily-made coumarins derivatives remains challenging due to the inertness of coumarins as dienophiles. Herein, we develop a decarboxylative asymmetric [4+2] cycloaddition of 3-cyanocoumarins with vinyl benzoxazinones, affording the coumarin-derived condensed rings bearing three continuous stereocenters in high yields with excellent diastereoselectivities (>20/1 d.r.) and enantioselectivities (up to 99% ee ). This direct enantioselective reaction was achieved by a Pd(0)/Cu(I) bimetallic catalytic system. The mechanism studies indicated that the synergistic activation effect, in which chiral Cu(I) as an available Lewis acid catalyst activates 3-cyanocoumarin and chiral Pd(0) complex activates benzoxazinone by the formation of π-allyl-palladium intermediate, plays an important role on the stereoselective control. The current work provides a new activation modes of Cu catalyst in the Pd/Cu bimetallic catalytic system. Abstract:A copper(II)-P,N,N-ligand catalyzed propargylic [3 + 2] cycloaddition approach for the synthesis of optically enriched dihydrofuro[3,2-c]coumarins has been accomplished for the first time. The cycloaddition utilizes propargylic esters as C2-bis-electrophiles and 4-hydroxycoumarin derivatives as C,O-bis-nucleophiles. In addition, this novel strategy was also explored with 4-hydroxy-2-quinolinones and 4-hydroxythiocoumarins.
