Abstract:It has been widely suggested that mammosphere-forming cells from tumor cell lines or primary tumors represent the population of cancer stem cells (CSCs), which is supposed to lead to the failure of routine chemotherapy and the recurrence of the disease. However, it is still difficult to obtain CSCs from primary breast cancer for further investigation. We performed a modified culture system to generate mammosphere-forming cells derived from freshly isolated human breast cancer samples and the breast cancer cell line MCF-7. Cancer stem cell-like phenotypes such as CD44 and CD24 were measured by flow cytometry while alkaline phosphatase (AP) and mammaglobin (MGB1) expression was evaluated immunohistochemically. The expression levels of Klf4, Nanog, Oct4, Sox2 and mdr1 genes were analyzed by quantitative real-time PCR. Resistance to chemotherapeutic drugs was detected through the apoptosis assay upon drug treatments together with the detection of drug-resistant gene mdr1. The results revealed that we successfully obtained mammosphere-forming cells from the primary breast cancer in conditioned medium after 14 days of culture. Mammosphere-forming cells from primary breast cancer displayed a CD44(hi)CD24(lo) phenotype as well as positive AP and MGB1 reactivity. Stem cell-related genes such as Klf4, Nanog and Oct4 were detectably expressed in these cells. These cells formed tumor-like structures in the lymph nodes of nude mice, which were morphologically and histologically similar to breast cancer. Compared to the breast cancer cell line MCF-7 or mammosphere-forming cells from MCF-7 cells, the mammosphere-forming cells from the prithary breast cancer exhibited resistance to three of four first-line chemotherapeutic drugs investigated through the induction of apoptosis, which was largely associated with the increased expression of drug-resistant gene mdr1 upon drug treatment. In conclusion, mammosphere-forming cells generated from the primary breast cancer exhibit CSC-like properties together with multiple drug resistance. Determination of the sensitivity of these primary cancer-derived mammosphere-forming cells to chemotherapeutic drugs may thus provide useful instructions for individualized therapy against the recurrence of breast cancer in the future.
miR-205 Regulates Basal Cell Identity and Stem Cell Regenerative Potential During Mammary Reconstitution.

Abstract:Mammary gland development is fueled by stem cell self-renewal and differentiation. External cues from the microenvironment coupled with internal cues such as post-transcriptional regulation exerted by microRNAs regulate stem cell behavior and fate. Here, we have identified a miR-205 regulatory network required for mammary gland ductal development and stem cell regeneration following transplantation into the cleared mammary fat pad. In the postnatal mammary gland, miR-205 is predominantly expressed in the basal/stem cell enriched population. Conditional deletion of miR-205 in mammary epithelial cells impairs stem cell self-renewal and mammary regenerative potential in the in vitro mammosphere formation assay and in vivo mammary reconstitution. miR-205 null transplants display significant changes in basal cells, basement membrane, and stroma. NKD1 and PTPA, which inhibit the Wnt signaling pathway, and AMOT, which causes YAP cytoplasmic retention and inactivation were identified as miR-205 downstream mediators. These studies also confirmed that miR-205 is a direct ΔNp63 target gene that is critical for the regulation of basal cell identity. Stem Cells 2018;36:1875-15.

Abstract:Breast cancer is a heterogeneous disease. In particular, triple-negative breast cancer (TNBC) comprises various molecular subgroups with unclear identities and currently has few targeted treatment options. Our previous study identified protein C receptor ( Procr ) as a surface marker on mammary stem cells (MaSCs) located in the basal layer of the normal mammary gland. Given the possible connection of TNBC with basal layer stem cells, we conducted comparative analyses of Procr in breast cancers of mouse and human origin. In mouse mammary tumors, we showed that Procr + cells are enriched for cancer stem cells (CSCs) in Wnt1 basal-like tumors, but not in Brca1 basal-like tumors or PyVT luminal tumors. In human cancers, PROCR was robustly expressed in half of TNBC cases. Experiments with patient-derived xenografts (PDXs) revealed that PROCR marks CSCs in this discrete subgroup (referred to as PROCR + TNBC). Interfering with the function of PROCR using an inhibitory nanobody reduced the CSC numbers, arrested tumor growth and prevented rapid tumor recurrence. Our data suggest a key role of MaSC in breast tumorigenesis. Moreover, our work indicates that PROCR can be used as a biomarker to stratify TNBC into clinically relevant subgroups and may provide a novel targeted treatment strategy for this clinically important tumor subtype.

Abstract:Embryonic stem (ES) cell markers are molecules specifically expressed in ES cells. ES cell markers reported previously including transcription factors, receptors, adhesion molecules and so on, are involved in the mechanism of self-renew and differentiation initiation of ES cells. Approaches such as monoclonal antibody technique, transcriptomics technique and proteomics technique have been used to screen for ES cell markers. ES cell markers play a key role in characterization and isolation of ES cells and elucidation for the mechanism of self- renewal in ES cells, which can finally accelerate the clinical application of ES cells. In this review, we discussed the technology of identification of ES cell markers and the possible function of reported markers to date.
Identification and Characterization of Cancer Initiating Cells from BRCA1 Related Mammary Tumors Using Markers for Normal Mammary Stem Cells

Abstract:It is hypothesized that cancer stem cells arise either from normal stem cells or from progenitor cells that have gained the ability to self-renew. Here we determine whether mammary cancer stem cells can be isolated by using antibodies that have been used for the isolation of normal mammary stem cells. We show that BRCA1 mutant cancer cell lines contained a subpopulation of CD24+CD29+ or CD24+CD49f+ cells that exhibited increased proliferation and colony forming ability in vitro, and enhanced tumor-forming ability in vivo. The purified CD24+CD29+ cells could differentiate and reconstitute the heterogeneity found in parental cells when plated as a monolayer. Under low-attachment conditions, we detected “tumorspheres” only in the presence of double positive cells, which maintained their ability to self-renew. Furthermore, CD24+CD29+ cells could form tubular structures reminiscent of the mammary ductal tree when grown in three-dimensional cultures, implying that these cancer cells maintain some of the characteristics of the normal stem cells. Nevertheless, they could still drive tumor formation since as low as 500 double positive cells immediately after sorting from BRCA1 mutant primary tumors were able to form tumors with the same heterogeneity found in the original tumors. These data provide evidence that breast cancer stem cells originate from normal stem cells and advance our understanding of BRCA1-associated tumorigenesis with possible implications for future cancer treatment.
Quantitative Analyses of CD133 Expression Facilitate Researches on Tumor Stem Cells.

Abstract:CD133 is regarded as a marker of tumor initiating cells in many tumors, including colorectal cancer. O'Brien and Ricci et al. have proved that in primary colorectal tumors there are colorectal tumor stem cells (initiating cells) which are marked by CD133 antigen. Using a genetic knockin lacZ reporter mouse model, Shmelkov et al. challenged this increasingly influential viewpoint and drew two important conclusions that challenge former opinions. First, CD133 is widely distributed throughout the full range of tumor epithelial cells in the colon as opposed to being limited to a few cells. Second, CD133 negative cells of colon tumors are also tumorigenic, and are more inclined to metastasize. Based on these two opinions, we hypothesize that the expression of CD133 is different among tumor cells, and that quantitative but not qualitative analyses of CD133 abundance are necessary to determine the relationship between CD133 expression and tumor stem cell characteristics. To verify this hypothesis, colorectal cancer cell line SW620 was cultured and sorted into CD133(Hi), CD133(Mid) and CD133(Low) subgroups using magnetic microbeads to compare their xenograft biological characteristics. The results showed that the CD133(Hi) subgroup of SW620 is more close to the tumor initiating cells in terms of biological characteristics than CD133(Mid) and CD133(low) subgroups, but the CD133(low) subgroup still maintains the ability of tumorigenicity. It supported that tumor initiating cells are more correlated to the abundance of CD133.

Abstract:The cells with mammary repopulating capability can achieve mammary gland morphogenesis in a suitable cellular microenvironment. Using cell surface markers of CD24, CD29 and CD49f, mouse mammary repopulating unit (MRU) has been identified in adult mammary epithelium and late embryonic mammary bud epithelium. However, embryonic MRU remains to be fully characterized at earlier mammary anlagen stage. Here we isolated discrete populations of E14.5 mouse mammary anlagen cells. Only Lin(-)CD24(med)CD29(+) cell population was predicted as E14.5 MRU by examining their capacities of forming mammosphere and repopulating cleared mammary fat pad in vivo. However, when we characterized gene expressions of this E14.5 cell population by comparing with adult mouse MRU (Lin(-)CD24(+)CD29(hi)), the gene profiling of these two cell populations exhibited great differences. Real-time PCR and immunostaining assays uncovered that E14.5 Lin(-)CD24(med)CD29(+) cell population was a heterogeneous stroma-enriched cell population. Then, limiting dilutions and single-cell assays also confirmed that E14.5 Lin(-)CD24(med)CD29(+) cell population possessed low proportion of stem cells. In summary, heterogeneous Lin(-)CD24(med)CD29(+) cell population exhibited mammary repopulating ability in E14.5 mammary anlagen, implying that only suitable mammary stroma could enable mammary gland morphogenesis, which relied on the interaction between rare stem cells and microenvironment.

Abstract:ABSTRACT It is well known that the expansion of the mammary epithelium during the ovarian cycles in female mammals is supported by the transient increase in mammary epithelial stem cells (MaSCs). However, dissecting the molecular mechanisms that govern MaSC function and differentiation is poorly understood due to the lack of standardized methods for their identification and isolation. The development of robust single-cell mRNA sequencing () technologies and the computational methods to analyze them provides us with novel tools to approach the challenge of studying MaSCs in a completely unbiased way without. Here, we have performed the largest scRNA-seq analysis of individual mammary epithelial cells (~70,000 cells). Our study identified a distinct cell population presenting molecular features of MaSCs. Importantly, further purification and additional in-depth single-cell analysis of these cells revealed that they are not a fully homogenous entity. Instead, we identified three subpopulations representing early stages of lineage commitment. By tracking their molecular evolution through single-cell network analysis we found that one of these subpopulations represents bipotent MaSCs from which luminal and basal lineages diverge. Importantly, we also confirmed the presence of these cells in human mammary glands. Finally, through expression and network analysis studies, we have uncovered transcription factors that are activated early during lineage commitment. These data identified E2-2 (Tcf4) and ID3 as a potential molecular switch of mammary epithelial stem cell differentiation.

Abstract:We present an organoid regeneration assay in which freshly isolated human mammary epithelial cells are cultured in adherent or floating collagen gels, corresponding to a rigid or compliant matrix environment. In both conditions, luminal progenitors form spheres, whereas basal cells generate branched ductal structures. In compliant but not rigid collagen gels, branching ducts form alveoli at their tips, express basal and luminal markers at correct positions, and display contractility, which is required for alveologenesis. Thereby, branched structures generated in compliant collagen gels resemble terminal ductal-lobular units (TDLUs), the functional units of the mammary gland. Using the membrane metallo-endopeptidase CD10 as a surface marker enriches for TDLU formation and reveals the presence of stromal cells within the CD49f(hi)/EpCAM(-) population. In summary, we describe a defined in vitro assay system to quantify cells with regenerative potential and systematically investigate their interaction with the physical environment at distinct steps of morphogenesis.

Abstract:The protein C receptor (PROCR) has emerged as a stem cell marker in several normal tissues and has also been implicated in tumor progression. However, the functional role of PROCR and the signaling mechanisms downstream of PROCR remain poorly understood. Here, we dissected the PROCR signaling pathways in breast cancer cells. Combining protein array, knockdown, and overexpression methods, we found that PROCR concomitantly activates multiple pathways. We also noted that PROCR-dependent ERK and PI3k-Akt-mTOR signaling pathways proceed through Src kinase and transactivation of insulin-like growth factor 1 receptor (IGF-1R). These pathway activities led to the accumulation of c-Myc and cyclin D1. On the other hand, PROCR-dependent RhoA-ROCK-p38 signaling relied on coagulation factor II thrombin receptor (F2R). We confirmed these findings in primary cells isolated from triple-negative breast cancer-derived xenografts (PDX) that have high expression of PROCR. To the best our knowledge, this is the first comprehensive study of PROCR signaling in breast cancer cells, and its findings also shed light on the molecular mechanisms of PROCR in stem cells in normal tissue.
Implications of the Cancer Stem Cell Hypothesis for the Early Detection and Prevention of Breast Cancer.

Abstract:PL03-01 Recent research in a variety of tumor systems has given support to the Cancer Stem Cell Hypothesis. Two important components of this hypothesis are that tumors originate in tissue stem or progenitor cells as a result of disregulation of the normally tightly regulated process of self-renewal. As a result of this, tumors contain and are driven by a cellular subcomponent that retains key stem cell properties including self-renewal which drives tumorigenesis and differentiation that contributes to cellular heterogeneity. The Cancer Stem Cell Hypothesis has profound implications for the early detection, prevention, and treatment of cancer. Our laboratory has developed techniques for the isolation and characterization of stem cells from human mammary glands and mammary carcinomas. Mammary stem and progenitor cells can be cultured in vitro as floating spherical colonies which we have termed which are composed of progenitor cells capable of multi-lineage differentiation, as well as stem cells capable of mammosphere formation. These more primitive cells express the stem cell markers OCT-4, Bmi-1, and ALDH-1. Mammosphere initiating cells which are estrogen receptor negative give rise to estrogen receptor positive proliferating progenitor We have utilized these in vitro systems as well as a humanized NOD-SCID mouse model to study the developmental pathways which regulate stem cell self-renewal and lineage specific commitment. Both Notch and Hedgehog signaling are involved in stem cell self-renewal as evidenced by the observation that activation of these pathways increases mammosphere number upon serial passage. These effects are mediated by the polycomb gene Bmi-1. Over expression of the Hedgehog component Gli-2 in mammosphere initiating cells results in the generation of ductal hyperplasias when these cells are implanted into the mammary fat pads of humanized NOD-SCID mice. In contrast, inhibition of Bmi-1 utilizing a siRNA lenti virus inhibits mammosphere formation in vitro and mammary gland development in these mice. These results suggest that disregulation of normal stem cell self-renewal results in expansion of stem cell pools which may then serve as targets for further transformational events. We have previously reported the isolation of a subpopulation of cells within human breast cancers which display stem cell characteristics. These cells characterized as CD44+ CD24 low lin- display activated Hedgehog signaling and increased expression of Bmi-1. In order to define markers which might prove useful for in situ characterization of normal and malignant mammary stem cells, we have examined the expression of aldehyde dehydrogenase (ALDH) an enzyme previously found to be expressed in hematopoietic and neuronal stem ALDH may be assessed by the Aldefluor assay which utilizes a fluorescent probe to detect cells displaying aldehyde dehydrogenase activity. We have found that the Aldefluor positive population of normal mammary epithelium has stem cell characteristics as demonstrated by mammosphere formation and generation of ductal alveolar structures in humanized NOD-SCID mice. Furthermore, in human mammary carcinoma xenografts, Aldeflour positive cells display properties of stem cells. As few as 200 Aldefluor positive cells, which comprise between 1-10% of the tumor cell population, produce tumors in NOD-SCID mice while 2x10 3 Aldefluor negative cells failed to generate tumors. Consistent with the Aldefluor positive population representing cancer stem cells, tumors generated from this population recapitulate the phenotypic heterogeneity of ALDH expression seen in the original tumor. Based on the above studies, we utilized immunochemistry to detect ALDH-1 as a marker of normal and malignant mammary stem/progenitor cells in situ. ALDH-1 antibodies detect a rare cell population in normal human mammary gland and in mammospheres generated from these Expanded ALDH-1 positive populations are found in pre-malignant lesions such as ductal carcinoma in situ (DCIS) and at areas of microinvasion. Furthermore, utilizing tumor tissue microarrays, we have determined that expression of ALDH-1 is associated with a poor clinical outcome. These studies demonstrate that ALDH-1 may be utilized to detect normal and malignant human mammary stem/progenitor cells providing a new tool for the early detection of beast cancer. Furthermore, since these cells may drive carcinogenesis, strategies aimed at their elimination, represents a rational approach for cancer prevention.

Abstract:Abstract We have developed an organoid regeneration assay to quantify the ability of freshly isolated, single human mammary epithelial cells to generate complex branched ductal structures with basal and luminal features. For this purpose, cells are cultured in adherent or floating collagen gels, corresponding to a rigid or compliant matrix. In compliant but not rigid collagen gels, branching ducts in multicellular structures generated by basal cells develop a lumen and alveoli at their tips. Functionally, basal cells in branched structures display cellular contractility, which we reveal to be required for alveologenesis. In conclusion, branched structures generated by single basal cells in compliant collagen gels resemble terminal ductal-lobular units (TDLU), the functional units of the mammary gland. To prospectively isolate basal cells with regenerative potential, we added the membrane metallo-endopeptidase CD10 as a cell surface marker to existing sorting protocols, thereby enriching for TDLU-formation and enabling rigorous quantification of regenerative potential by extreme limiting dilution assay (ELDA). Moreover, the use of CD10 as a cell surface marker reveals the presence of stromal cells with endothelial qualities within the CD49fhi/EpCAM– population, previously labeled basal. Abstract:The mammary gland is an organ comprising two primary lineages, specifically the inner luminal and the outer myoepithelial cell layers. Mammary gland stem cells (MaSCs) are highly dynamic and self-renewing, and can give rise to these mammary gland lineages. The lineages are responsible for gland generation during puberty as well as expansion during pregnancy. In recent years, researchers have focused on understanding how MaSCs are regulated during mammary gland development and transformation of breast cancer. Here, we summarize the identification of MaSCs, and how they are regulated by the signaling transduction pathways, mammary gland microenvironment, and non-coding RNAs (ncRNAs). Moreover, we debate the evidence for their serving as the origin of breast cancer, and discuss the therapeutic perspectives of targeting breast cancer stem cells (BCSCs). In conclusion, a better understanding of the key regulators of MaSCs is crucial for the clinical treatment of breast cancer.

Abstract:Aim:To identify the cause of multi-differentiation potential in mouse bone marrow mesenchymal stem cells.Methods: MSCs were isolated from the femurs and tibiae of the adult mouse and cultured in vitro.Total RNA were extracted from the mouse bone marrow(Day 0) and cultured mouse MSCs at 2,4,6,8 and10 days.The pluripotency marker genes Oct-4 and nanog,pluripotency related transcription factors Klf4 and c-Myc and the marker genes of three embryonal layers,such as CYP51,SM22α and nestin,which are typical genes specific for endoderm,mesoderm and ectoderm respectively,were detected by RT-PCR in order to determine the characteristics of MSCs.Results: The results of RT-PCR showed that mouse MSCs can express mRNA of Oct-4,nanog,Klf4 and c-Myc.Furthermore,in addition to SM22a,RT-PCR revealed the presence of mRNA for CYP51 and nestin during the primary culture of mouse MSCs.Conclusion:Mouse MSCs,which were isolated by adhesion purification,may be consist of pluripotent stern cells and primary cells from the three germ layers.

Global Profiling of Signaling Networks: Study of Breast Cancer Stem Cells and Potential Regulation

Abstract:There is overwhelming evidence that breast cancer may be driven by a small subset of breast cancer stem cells (BCSCs) that display stem/progenitor cell properties. In the present study, we identified the rare population of BCSCs, the so-called side population (SP) cells, using flow cytometry. Then, we used microarray analysis to study the differential gene expression profiles between SP and non-SP cells. Sixty-three probe sets showed a more than fourfold difference. Next, we compared the levels of proteins with Pathway Array using 154 antibodies, focusing on the proteins and phosphorylation sites that differed among SP cells, malignant mammary cells, and breast cancer tissues. Our results revealed that 40 proteins and phosphorylation sites were more than 1.5-fold different in SP cells than in non-SP cells. By comparing SP cells, MCF7 cells, and nontumorigenic MCF10A cells, we found 12 proteins that were significantly upregulated in SP cells; these proteins-cAMP-response element binding protein (CREB), cyclic AMP-dependent transcription factor 1, mesothelin, thyroid transcription factor 1, phosphorylated (p)-focal adhesion kinase, p38, Bad, p-CREB, p-protein kinase C (PKC)δ, Wee1, cell division cycle 42, and Twist-were more likely to play important roles in the signaling regulation of BCSCs. Further, 16 proteins and phosphoproteins showed differential expression in SP cells and tumor tissues. β-catenin, p-PKCα, and p-CREB were upregulated in both SP cells and breast tumors. Finally, we filtered the differential expression proteins, summarized the pathway interactions of these proteins, and rebuilt Path-Net in order to determine molecular mechanisms and core regulators. This process will allow us to identify signature patterns and mechanisms of signaling networks in BCSCs.
Identification of Tumor Stem-Like Cells in a Mouse Myeloma Cell Line.

Abstract:We used colony formation assay in the soft agar media or the serum-free media, the methods of identifying BrdU-label-retaining cells and the SP cells as well as the tumorigenicity test in BALB/c mice, respectively, to analyze tumor stem like cells in the SP2/0 cell line. The results showed that a few SP2/0 cells were capable of forming colonies in the soft agar media, contained BrdU-label-immortal strand in the SP2/0 cell line. The SP2/0 cells in the serum-free media gained higher tumorigenicity in the BALB/c mice than the SP2/0 cells cultivated in the complete media did. Overall, only a few of the SP2/0 cells were found to possess the characteristics of tumor stem-like cells, such as high proliferative potency, more self-renewal and stronger tumorigenesis, or greater similarity to the tumor stem cells (TSCs) traits. The biology of tumor stem-like cells contributes to the identification of molecular targets important for future tumor therapy.

Abstract:The mammary gland undergoes cycles of growth and regeneration throughout reproductive life, a process that requires mammary stem cells (MaSCs). Whilst recent genetic fate-mapping studies using lineage-specific promoters have provided valuable insights into the mammary epithelial hierarchy, the true differentiation potential of adult MaSCs remains unclear. To address this, herein we utilize a stochastic genetic-labelling strategy to indelibly mark a single cell and its progeny in situ, combined with tissue clearing and 3D imaging. Using this approach, clones arising from a single parent cell could be visualized in their entirety. We reveal that clonal progeny contribute exclusively to either luminal or basal lineages and are distributed sporadically to branching ducts or alveoli. Quantitative analyses suggest that pools of unipotent stem/progenitor cells contribute to adult mammary gland development. Our results highlight the utility of tracing a single cell and reveal that progeny of a single proliferative MaSC/progenitor are dispersed throughout the epithelium.

Abstract:Background Sca-1 is controversial as a mammary stem cell marker in the literature, which may be due to the different isolation protocols and culture media used in different laboratories. The object of our study is to establish the Medium to promote the proliferation of mammary stem cell and explore the possibility of Sca-1 as mammary stem cell marker. Methods We used BM medium supplemented with different concentration of 17 MaECM medium which promoted the proliferation of mouse mammary epithelial cells and inhibited the growth of fibroblasts. Flow cytometry was used to isolate Sca-1 + and Sca-1 − cell populations from cultured mammary epithelial cells. Mammary fat pad transplantation and Mammosphere-forming assay were done to confirm the stem cell potential of Sca-1 + cells. Differentiating culture was used to detect the differentiation potential of Sca-1 + cells. Real-time PCR was carried out to analyse the expression of mammary stem cell-related genes in Sca-1+ cells. Results We first selected the medium suitable for mammary stem cell growth. Stem cell medium BM was used to culture mammary organoids, which generated many fibroblasts. We established MaECM medium supplemented with oestrogen and growth hormone (GH), in which oestrogen promoted mammary epithelial cell proliferation and inhibited fibroblast growth, and GH obviously enhanced the effect of oestrogen on mammary epithelial cell proliferation. Flow cytometry showed that 50% of cells were Sca-1 + under the culture of MaECM medium. We confirmed that Sca-1 + cells regenerated mammary outgrowths when transplanted in vivo , formed mammospheres in vitro and differentiated into luminal epithelial cells with milk-secreting function and myoepithelial cells under Matrigel culture. Furthermore, gene expression analysis by Real-time PCR revealed that Sca-1 + cells expressed markedly higher levels of mammary stem cell-related genes in comparison to Sca-1 − cells. Conclusion Our research demonstrates that Sca-1 + mammary stem cells can be more easily isolated when cultured in the presence of oestrogen and GH.

Abstract:Recently, substantial progress has been made in the identification and characterization of stem and progenitor cells in the mouse and human mammary gland. Furthermore, there is increasing evidence that a variety of neoplasms, including breast cancer, may result from transformation of normal stem and progenitor cells. Consistent with this model of carcinogenesis, a breast cancer stem cell population, with the phenotype CD24-CD44 + lineage, was recently identified utilizing flow-cytometry based cell sorting and nonobese diabetic/severe combined immunodeficient (NOD/SCID) mice xenografts. As few as 200 cells of this cancer stem cell population were capable of generating tumors in animals, whereas the bulk of the tumor population was tumorigenic only when implanted in high numbers. Like their normal counterparts, the cancer stem cells have the ability to self-renew, driving tumorigenicity and possibly recurrence and metastasis, and have the ability to differentiate, generating the heterogeneity of the tumors. This stem cell model of carcinogenesis has important implications for understanding the basic biology of breast cancer, as well as other cancers. Furthermore, the concept of cancer as a disease of stem and progenitor cells has profound implications for the development of new strategies for cancer prevention and therapy.

Abstract:Recent studies indicate that tumor maintenance, metastasis and drug-resistance are mainly conducted by a small subset of cancer cells which are termed cancer stem cells (CSCs) or cancer stem-like cells (CSLCs). Successful identification of CSCs/CSLCs might lead to discovery of the novel and effective therapeutic targets for cancers. In our study, lung CSCs/CSLCs were enriched by sphere-forming assay. Screening and selection of specific binding peptides for lung CSCs/CSLCs were performed with bacterial surface display method. Selected peptide named HCBP-1 exhibited highest specific binding capability as examined by flow cytometry and fluorescence microscopy. Drug-resistant lung CSCs/CSLCs might be characterized with HCBP-1 peptide and several microRNAs related to the stem-like properties were discriminatively expressed in HCBP-1+ subpopulation. Moreover, at least two distinct subpopulations in H460 tumor sphere cells could be distinguished by HCBP-1 peptide. Thus, a new method was established to identify lung CSCs/CSLCs, which provided robust approaches for the research of CSCs/CSLCs.
