Abstract:Elevated levels of homocysteine (Hcy) in blood are indicative of many high risk cardiovascular and neurodegenerative diseases. Thus, development of highly efficient and selective dyes for monitoring Hcy levels has attracted much attention. This paper reports the utilization of TPE-Cy, an aggregation-induced-emission active hemicyanine dye, as a probe for the detection of Hcy. More interestingly, this dye shows high selectivity to Hcy over cysteine, glutathione and other amino acids in weakly basic buffer solution.

Abstract:We demonstrate a concept-proof work of using fluorescence (FL) "turn-on" probes for the discriminatory detection of cysteine (Cys) over homocysteine (Hcy). The fluorogens are provided with aggregation-induced emission (AIE) characteristic and functionalized with two aldehyde-groups (DMTPS-ALD and TPE-ALD). All the detections were carried out in a biocompatible medium (10 mm HEPES buffer and DMSO, pH 7.4). In principle, the formation of thiazinane/thiazolidine through the chemical reaction of aldehydes on the probe molecules and the residue of Cys/Hcy determines the selective recognition of Cys and Hcy over other amino acids and glucose. The FL responses originate from the AIE property of thiazinane/thiazolidine resultants, which have low solubility and precipitate (aggregate) in the detection medium. The discrimination between Cys and Hcy comes from the difference in reaction kinetics of TPE-ALD/DMTPS-ALD with Cys and Hcy, thereby the FL responses show different time courses and intensity enhancement. It is worth noting that TPE-ALD outshined the other two probes in performance with fast response, a high FL enhancement up to 16-fold, high sensitivity, and good specificity and selectivity. Moreover, its FL response threshold at 250 mu m is very close to the lower limit of the normal level of Cys in human plasma, which implies that TPE-ALD could be applied as a potential indicator of Cys deficiency.

Abstract:Glutathione (GSH) as a biothiol is an essential peptide related to various diseases. Although multiple strategies for biothiols detection have been developed, there is increasing demand for sensors that can differentiate GSH from cysteine (Cys) and homocysteine (Hcy), owing to the similar structures and thiol groups in these amino acids. Herein, we report a novel Eu3+/LAPONITE (Lap)-based organic/inorganic hybrid material for selective detection of GSH via an "off-on" process. The fluorescence of Eu(DPA)3@Lap-Tris can be quenched by Cu2+ through photoinduced electron transfer (PET). The addition of GSH into the Eu(DPA)3@Lap-Tris/Cu2+ system induces the removal of Cu2+ from Eu(DPA)3@Lap-Tris and blocks PET, resulting in the recovery of fluorescence. This proposed assay demonstrates higher selectivity toward GSH than Cys and Hcy, and showed a detection limit of 162 nM within a linear range of 0.5-30 μM. Unlike other GSH selective sensors, this platform could be formed into a hydrogel while its sensitivity was maintained. The sensitive response to GSH in serum samples makes this platform an efficient tool for biological applications because of its ease of preparation, high selectivity, good biocompatibility, and low toxicity.

Abstract:In this work, manganese(II)-doped zinc/germanium oxide nanoparticles (Mn@ZGNPs) have been hydrothermally synthesized to equip with appealing time-resolved luminescence (TRL). Interestingly, we reveal that they can be readily quenched ("turn off") via a facile surface coating with bioinspired polydopamine (PDA) polymerized from dopamine (DA), resulting from PDA-triggered TRL resonance energy transfer (TRL-RET). By integrated with the thiol-induced inhibition of PDA formation, an ingenious inorganic-organic hybrid tongue-mimic sensor array is thus unveiled for noninvasive pattern recognition of thiols in biofluids in a TRL-RET-reversed "turn on" format toward healthcare monitoring. The sensing principle is based on the new finding that there are differential inhibitions from thiols against the polymerization of DA with various concentrations. Furthermore, density function theory (DFT) studies excellently prove our sensing principle and experimental results, reinforcing the power of the presented system. More importantly, chiral recognition of varied concentrations and mixtures of cysteine enantiomers using our platform are also been demonstrated, promising its practical usage. This is a novel concept of inorganic-organic hybrid-based pattern and chiral recognition platform for TRL background-free sensing and would sprout more novel relevant strategies toward broader applications.

Abstract:A functional ionic microgel sensor array was developed by using 1-(2-pyridinylazo)-2-naphthaleno (PAN)- and bromothymol blue (BTB)-functionalized ionic microgels, which were designed and synthesized by quaternization reaction and anion exchange reaction, respectively. The PAN microgels (PAN-MG) and BTB microgels (BTB-MG) were spherical in shape with a narrow size distribution and exhibited characteristic colors in aqueous solution in the presence of various trace-metal ions, which could be visually distinguished by the naked eye. Such microgels could be used for the colorimetric detection of various metal ions in aqueous solution at submicromolar levels, which were lower than the U.S. Environmental Protection Agency standard for the safety limit of metal ions in drinking water. A total of 10 species of metal ions in aqueous solution, Ba2+, Cr3+, Mn2+, Pb2+, Fe3+, Co2+, Zn2+, Ni2+ Cu2+, and Al2+, were successfully discriminated by the as-constructed microgel sensor array combined with discriminant analysis, agglomerative hierarchical clustering, and leave-one-out cross-validation analysis.
Dynamic Transmission-Reflection Dichroism Based on Humidity-Responsive Metal-Hydrogel-metal Nanocavities

Abstract:“Lycurgus cup” effect, referring dichroism between reflection and transmission modes of the same structures, is a peculiar phenomenon of multi-faceted display in structural color. Beyond the static dichrotic display, the realization of dynamic dichroism desires active materials and tunable structures, and owns the great demand from smart display, anti-counterfeiting and environmental sensing. We hereby propose a metal-hydrogel-metal (MHM) nanocavity for dynamic dichrotic display. This structure includes thin silver layers to induce the partial transmission with the existing reflection, and a polyvinyl alcohol (PVA) hydrogel layer owning the swelling/deswelling deformability to humidity change. The following experimental measurements and theoretical analysis prove that the reflection and transmission modes exist at distinct wavelengths, and the swelling hydrogel layer by humidity change between 10 and 90% RH can dynamically modulate the dichrotic resonance with the wavelength shift over 100 nm. Such environmental-sensitive and real-time tunable dichroism with hydrogel-based structural color is then verified for multi-color printing, resolution test, and cycling test.

Abstract:A novel absorbance recovery method has been developed for the determination of biothiols with a near-infrared reagent. This method employs a two-reagent system composed of cation heptamethine cyanine (CyL) and Hg2+. The absorbance of CyL, with a maximum peak at 760 nm, was decreased due to addition of Hg2+, but recovered when biothiols were added. Under optimal conditions, the reciprocal extent of recovered absorbance was proportional to the concentration of biothiols. The calibration curves are linear over the range of (0.3–7.0) × 10–6 M for cysteine, (1.0–10.0) × 10–6 M for homocysteine and (1.0–9.0) × 10–6 M for glutathione. Because of the specific affinity of Hg2+for biothiols, there is minimal interference from other amino acids. This method has been successfully applied to the determination of homocysteine in human urine samples with satisfactory results.

Abstract:Cysteine (Cys) plays important roles in many physiological processes of eukaryotic cells and its detection in cells is of fundamental significance. However, glutathione (GSH), homocysteine, N-acetyl-L-cysteine and other thiols greatly hamper the detection of Cys. In particular, GSH strongly interferes with the detection of cellular Cys (30-200 mu M) due to its high intracellular concentration (1-10 mM). In this work, an off on fluorescent probe (HOTA) for the detection of Cys is presented. This probe possesses both excellent sensitivity and satisfactory selectivity for cellular Cys detection: with the addition of 200 mu M Cys, the fluorescence intensity of the probe (10 mu M) enhanced 117-fold and the detection limit was calculated to be 13.47 mu M, which is lower than the cellular Cys concentration; the probe also selectively detected 30-200 mu M cysteine over 1-10 mM glutathione. Consequently, cell imaging experiments were performed with probe HOTA. Furthermore, the results of the thiol-blocking and GSH synthesis inhibiting experiments confirmed that the intracellular emission mainly originates from the interaction between Cys and HOTA.
Tunable metallo-hydrogels: Mechanical properties and characterization of stimuli responsive self-assembling peptide hydrogels

Abstract:Supramolecular self-assembly is employed in this study to design short amphiphilic peptides with specific binding affinity for Zn(II). Two selectively modified peptides (Ac-LIVKHH-NH2, P1 & Fmoc-LIVKHH-NH2, P2) were synthesized using standard Fmoc-/Boc- solid-phase peptide synthesis. When these water-soluble peptides are combined with Zn(II) salt solution, they encapsulate large volumes of water to form metallo-hydrogels. The Zn(II) responsiveness in metallo-hydrogels was investigated in buffer or water (pH 7) with or without Zn(II) salts. The afforded supramolecular architectures (metallo-hydrogels) were characterized by circular dichroism and their micro- and nanostructure with scanning electron microscopy (SEM) and transmission electron microscopy (TEM), respectively. Measurements of rheology and thixotropy were conducted to assess the gelation behavior, the viscoelastic character and the mechanical properties of metallo-hydrogels. We found that Zn(II) significantly influences hydrogel formation, leading to improved mechanical properties. The resulting hydrogels show remarkable thixotropic behavior, indicating their potential in 3D printing and as injectable carriers. Such hydrogels can be also used as primary wound dressings, and their antibacterial activity can be further explored. This research work provides valuable insights into the zinc-responsive behavior of the aforementioned peptides, establishing a foundation for their prospective applications in biomaterials and materials science.

Colorimetric Sensor Array for Thiols Discrimination Based on Urease-Metal Ion Pairs.

Abstract:Thiols play a crucial role in various physiological functions, and the discrimination of thiols is a significant but difficult issue. Herein, we presented a new strategy for strengthening the discrimination of thiols by a facile colorimetric sensor array composed of a series of urease-metal ion pairs. The proposed sensor array was fabricated based on the interactions between thiols and metal ions and the effective activation of urease by thiols. Different thiols exhibited different affinities toward the metal ions, producing differential retentions of urease activity and generating distinct colorimetric response patterns. These response patterns are characteristic for each thiol and can be quantitatively differentiated by linear discriminant analysis (LDA). Cysteine (Cys), glutathione (GSH), and four other kinds of thiols have been well distinguished on the basis of this sensor array at a low concentration (1.0 μM). Remarkably, the practicability of the proposed sensor array was further validated by high accuracy (96.67%) identification of 30 unknown thiol samples. In this strategy, urease and its metal ion inhibitors were adapted to fabricate the sensor array, offering a facile way to develop sensitive array sensing systems based on inexpensive and commercially available enzymes and their inhibitors.

Abstract:A facile, low-cost and label-free liquid crystals (LCs) sensing platform laden with Cu(ClO4)2 was herein established for detection and distinction of sulfhydryl amino acids, namely cysteine (Cys) and homocysteine (Hcy). When introducing Cys or Hcy to the fluid interface with Cu(ClO4)2, the optical appearance of LCs exhibited a bright-to-dark transition, which coupled to the interactions between Cu2+ and the sulphydryl of amino acids. Concomitantly with, the orientation of LCs varied from tilted to homeotropic state. Besides, this sensor system did not respond to other nineteen amino acids that make up human proteins. The detection limit of Cys and Hcy is around 0.01 mg/mL (~82.5 μM) and 0.05 mg/mL (~370.0 μM), respectively. Compared with Cys, the as-prepared LCs sensing platform shows a higher detection limit and longer response time for Hcy, based on which these two sulfhydryl amino acids can be distinguished from each other. This can be attributed to the additional methylene for Hcy, resulting in formation of a five-membered ring Hcy thiolactone which may not chelate with Cu2+. The LCs-based analytical platform constructed in this work can provide a convenient and label-free method for detecting and differentiating sulfhydryl amino acids.

Abstract:Biothiols play critical roles in maintaining physiological and pathological processes. However, effective and affordable tools for rapid and simple identification of biothiols are currently lacking. This work constructed a paper-based fluorescence sensor-array platform for biothiol recognition by grafting three biothiol receptors (TRs) modified carbon quantum dots (CQDs) on paper substrates. A multi-channel rotating paper-based microfluidic device (μPAD) was constructed by inkjet printing technology. The three TRs are self-synthesized Michael receptors, and they can trigger photoinduced electron transfer (PET) effect to quench the fluorescence of CQDs. When biothiols were introduced onto the sensor array, they reacted with the CQDs-TRs hybrids at the sensor array due to their high nucleophilicity and disrupted the PET effect, resulting in partial recovery of the fluorescence. Six different biothiols produced their unique fluorescence response patterns as “fingerprints”, which can be quantitatively differentiated by linear discriminant analysis (LDA) and hierarchical clustering analysis (HCA) in concentration ranges of 5∼100 μM. Practicality of the platform was verified by fast, accurate and sensitive identification of the six biothiols in blind samples and fetal bovine serum. In all, the platform features mini size, easy preparation and high integration, showing great potential for clinical point-of-care and other on-site testing towards multiple analytes.

Nitrogen-Doped Carbon Quantum Dot-Anchored Hydrogels for Visual Recognition of Dual Metal Ions through Reversible Fluorescence Response

Abstract:Carbon quantum dots (CQDs) are novel and promising fluorescent materials that are extensively investigated in sensing, drug delivery, and bioimaging. However, they are easy to agglomerate and difficult to recover, which reduced their sensitivity and reusability. In this work, we innovatively prepared a “smart switch sensor” hydrogel (SWSH) by incorporating nitrogen-doped carbon quantum dots (N-CQDs) into a polyacrylamide (PAM) hydrogel network with functionalities of easy handling, recyclability, and simultaneously recognizing dual metal ions. The prepared SWSH showed reversible fluorescence “off/on” response with Cu2+/Ag+ and l-cysteine (l-cys), and the limit of detection values of the SWSH for these metal ions were 0.813 × 10–3, 0.468 × 10–3, and 0.437 × 10–3 mM, respectively. The N-CQDs in the SWSH would form a N-CQD–metal complex with Cu2+ or Ag+ ions, in which the electrons on the surface of CQDs were easily transferred to the metal, resulting in fluorescence quenching (off). However, only the fluorescence of the N-CQD–Cu2+ complex could be recovered (on) by the addition of l-cys as it could form a more stable Cu2+–S bond with Cu2+, thus achieving recognition of Cu2+ and Ag+ through fluorescence change observed with naked eyes. Therefore, the SWSH avoided the agglomeration of CQDs and facilitated their fluorescence recovery and reusability, which exhibited great potential applications in identifying metal ions.

Abstract:Hydrochromic visualization of a liquid interface shows vital potential applications in liquid displays, reversible writing, and acidic environmental detection, which offers a platform for detection and forewarning due to its intuitive and visual characteristics. Herein, we report a hydrochromic display due to the interfacial effect of liquid metal (LM)-triggered ammonium metatungstate (AMT) with instant dual-mode color switching. The double-electron-transfer reaction of the AMT on the surface of gallium-based LM caused the formation of heteropoly blue in the presence of acidic surroundings, resulting in a reversible color switching from being colorless to blue or blue to colorless. This visual interfacial discoloration phenomenon can be applied to the liquid display on diverse patterns of the LM surface. Furthermore, papers with a functional display were prepared, which can be used for writing up to eight times with dual-mode color switching. In addition, the reactive activity of acid triggering make it a potential candidate for use in visualizing an acidic environment with a detection range of pH = 1 to 0 (0.1-1.5 M). Briefly, this interfacial discoloration phenomenon enriches the interfacial engineering of LM and provides a unique prospective and wide-range platform for the application of LM.

Abstract:Small molecular biothiols, such as cysteine (Cys), homocysteine (Hcy), reduced glutathione (GSH), and hydrogen sulfide (H^2S), play crucial parts in regulating the redox balance of life activities, regulating normal physiological activities and preventing various diseases. Quantitative analysis of these important small molecular substances is very important for revealing their diverse physiological and pathological effects. Although many fluorescent probes have been reported to detect biothiols in cells, it is still not sufficiently advanced to detect biothiols with separated fluorescence emission peak by same wavelength excitation. In our work, we designed a simple conjugate of Nile red and NBD (7-nitro-1,2,3- benzoxadiazole) as long-wavelength fluorescent probe NR-NBD for the simultaneous discrimination of these biothiols at single wavelength excitation. Probe NR-NBD could efficiently discriminate Cys/Hcy, GSH and H^2S by two separated fluorescence emission channels and absorption spectra. Importantly, probe NR-NBD has excellent specificity and sensitivity towards the monitoring of endogenous/exogenous Cys/Hcy and GSH/H2S in living cells and zebrafish.

Abstract:Biothiols, including cysteine (Cys), homocysteine (Hcy), glutathione (GSH), and N -acetylcysteine (NAC), possess similar chemical structures and properties but play crucial, distinct roles in biological cells and blood serum. Imbalances in the concentrations of these biothiols are associated with various diseases, highlighting the importance of precise discrimination, especially between Cys and other biothiols. Owing to the similarity of the chemical properties of Cys, Hcy, GSH, and NAC, developing an effective methodology to differentiate these thiol compounds is challenging. In this study, we designed and synthesized a novel dual-channel fluorescent probe, hereafter referred to as CNTC , by integrating coumarin and acrylonitrile. This probe enabled the simultaneous discrimination of Cys from Hcy, GSH, and NAC, producing distinct fluorescent signals: blue for Cys and green for Hcy, GSH, and NAC. CNTC exhibited rapid response kinetics (within 30 min) and impressive detection limits of 0.31, 0.11, 0.029, and 0.032 μM for Cys, Hcy, GSH, and NAC, respectively. Furthermore, CNTC was successfully applied in the fluorescence imaging of both exogenous and endogenous Cys, Hcy, GSH, and NAC in living cells. The remarkable analytical and bioimaging capabilities of CNTC in vivo establish it as a promising tool for elucidating the pathophysiological roles of biothiols, particularly Cys, Hcy, GSH, and NAC.
