Abstract:Objective To study the nonflavanoid constituents in the aerial part of Epimedium koreanum.Methods The constituents were isolated through column chromatography on silica gel,Sephadex LH-20,and ODS from the water extract of E.koreanum,and their structures were identified by physicochemical methods and spectroscopic analysis.MTT Method was used to study the effects of the cons-tituents on the proliferation of UMR106 cell in rats.Results Two sesquiterpenes and two 9,10-dihydrophenanthrene derivatives were isolated from the water extract of E.koreanum.Their structures were identified as 3,7,11-trimethyl-2,6-dodecadien-1,10,11-trihydroxyl-10(S)-O-β-D-glucopyranoside(Ⅰ),icariside C1(Ⅱ),icariside A5(Ⅲ),epimedoicarisoside A(Ⅳ).Conclusion Compound Ⅰ is a new compound,named icariside F.Compound Ⅲ is isolated from E.koreanum for the first time.Compounds Ⅱ-Ⅳ show the proliferation-promoting activities on UMR106 cell.

Abstract:This study was to comprehensively evaluate the chemical quality of main species of epimedium planted in China. The contents of 5 marker compounds, epimedin A, epimedin B, epimedin C, icariin and baohuoside I, as well as total flavonoids of 22 samples of 8 officinal species of Epimedium were determined by HPLC and UV, separately. Some physical and chemical tests (H2O, total ash, acid-insoluble ash and EtOH extract) were also carried out to investigate their chemical qualities. There were significant differences in types and contents of prenyl-flavonoid glycosides such as epimedin A, epimedin B, epimedin C, icariin and baohuoside I in different species, meanwhile, the physical and chemical parameters results also showed that there were obvious differences in chemical quality among different species of epimedium herb. The results provide theoretical and experimental basis for the establishment of comprehensive quality assessment system of epimedium in China.

Abstract:Epimedium is a widely used traditional Chinese medicinal plant due to its high content of health-promoting components such as epimedin A, epimedin B, epimedin C, and icariin (hereafter designated as bioactive compounds); however, the biosynthesis of these bioactive compounds in Epimedium remains to be elucidated. In this study, E. sagitattum flavonoid genes, including three copies of chalcone synthase , two copies of dihydroflavonol 4 - reductase , and a single copy of each of chalcone isomerase , flavanone 3 hydroxylase , flavonol synthase , and anthocyanidin synthase , have been isolated. In addition, several transcription factors involved in flavonol, anthocyanin, and proanthocyanin pathways, were identified from an Epimedium 454 EST database. Gene expression profiles and the levels of bioactive compounds in light-stressed E. pubescens leaves in response to phytohormones [abscisic acid (ABA), gibberellic acid (GA 3 ), indole-3-acetic acid (IAA), salicylic acid (SA)] and sucrose were investigated. All bioactive compounds were significantly elevated by phytohormone and sucrose application except for epimedin B in SA-treated samples and epimedin C following GA 3 and sucrose treatment. Our results suggest that phytohormone and sucrose treatments offer a method for promoting the accumulation of bioactive compounds in Epimedium .

Abstract:Herba epimedii (Epimedium), a traditional Chinese medicine, has been widely used as a kidney tonic and antirheumatic medicine for thousands of years. In Epimedium, flavonoids have been demonstrated to be the main bioactive components (BCs). However, the molecular biosynthetic and regulatory mechanisms of flavonoid-derived BCs remain obscure. In this study, we isolated 12 structural genes and two putative transcription factors (TFs) in the flavonoid pathway. Phytochemical analysis showed that the total content of four representative BCs (epimedin A, B, C, and icariin) decreased slightly or dramatically in two lines of Epimedium sagittatum during leaf development. Transcriptional analysis revealed that two R2R3-MYB TFs (EsMYBA1 and EsMYBF1), together with a bHLH TF (EsGL3) and WD40 protein (EsTTG1), were supposed to coordinately regulate the anthocyanin and flavonol-derived BCs biosynthesis in leaves. Overexpression of EsFLS (flavonol synthase) in tobacco resulted in increased flavonols content and decreased anthocyanins content in flowers. Moreover, EsMYB12 negatively correlated with the accumulation of the four BCs, and might act as a transcriptional repressor in the flavonoid pathway. Therefore, the anthocyanin pathway may coordinate with the flavonol-derived BCs pathway in Epimedium leaves. A better understanding of the flavonoid biosynthetic and regulatory mechanisms in E. sagittatum will facilitate functional characterization, metabolic engineering, and molecular breeding studies of Epimedium species.

Abstract:Seven flavonoids named diphylloside A, epimedoside A, epimedin C, icariin, epimedoside C, icarisoside A, desmethylanhydroicaritin, as well as the oleanolic acid, were isolated from the roots of Epimedium wushanense for the first time. These flavonoids manifested significant antioxidant activity in vitro. Scavenging effects of two flavonoids were comparable to that of Vitamin C. Antibacterial experiment has shown that the diphylloside A, icarisoside A and desmethylanhydroicaritin have significant activity towards Pseudomonas aeruginosa.

Abstract:OBJECTIVE:To isolate and determine the chemical constituents from Astilbe chinensis. METHOD:Four compounds were isolated and purified by extraction and column chromatography on silica gel. The chemical structures were determined on the basis of physical-chemical evidence and extensive spectral analysis(HR-EIMS, EIMS, 13C-NMR, 1H-NMR, DEPT, 1H-1H-COSY, HMQC and HMBC). RESULT:Their structures were elucidated as beta-sitosterol palmitate I, daucosterol II beta-sitosterol III and Bergenin IV, respectively. CONCLUSION:Compound I and II were isolated from A. chinensis for the first time.

Abstract:The epimedii herb, a traditional Chinese medicinal plant, has significant pharmacological effects on human health. The bioactive components in the herb (Epimedium sagittatum (Sieb. et Zucc.) Maxim) are mainly prenylated flavonol glycosides, which are end-products of the flavonoid biosynthetic pathway. This has not been clearly elucidated until recently. The genes encoding flavonoid 3′-hydroxylase (F3′H) and flavonoid 3′, 5′-hydroxylase (F3′5′H) involved in the flavonoid biosynthetic pathway, designated as EsF3′H and EsF3′5′H, were isolated from E. sagittatum using a homology-based cloning method and deposited in the GenBank databases (GenBank ID: HM011054 and HM011055), respectively. EsF3′H and EsF3′5′H proteins shared high homology with other plant-specific flavonoid hydroxylases and were clustered into the CYP75B and CYP75A group, respectively. In addition, four conserved cytochrome P450-featured motifs were found in the amino acid sequences of both genes. Transcription levels of both genes were detected in all tissues tested and were high in most of the pigmented tissues. Moreover, the expression levels of both EsF3′H and EsF3′5′H correlated positively with the anthocyanin accumulation pattern in leaves from E. sagittatum. The cloning and molecular characterisation of EsF3′H and EsF3′5′H genes will accelerate progress in the study of the flavonoid biosynthetic pathway to elucidate the molecular mechanisms of the biosynthesis of the bioactive components in E. sagittatum.

Abstract:Phytochemical investigation was carried out for the flowers of Epimedium acuminatum Franchet. by first conducting LC-MS analysis, leading to the identification of 32 compounds. Furthermore, guided by LC-MS profiling, three new 8-prenylated quercetin glycosides (3′-hydroxylikarisoside C, 3′-hydroxylepimedoside E, 3′-hydroxyldiphylloside B), one new anthocyanin (delphinidin-3- O - p -coumaroyl-sophoroside) and six known compounds were isolated from the flowers of E. acuminatum for the first time, and their structures were characterized based on spectroscopic methods including 1D and 2D NMR, and HRESIMS. Combining our discoveries and literature survey, a revised classification of Epimedium flavonols was proposed as Type A (8-prenylated kaempferol based), which was further subdivided into subtype icaritin and subtype demethylicaritin, and Type B (8-prenylated quercetin based), which was further subdivided into subtype 3′-hydroxylicaritin and subtype 3′-hydroxyldemethylicaritin. The structure-activity relationship (SAR) study was carried out by comparing testosterone production-promoting activities of all the new compounds along with nine related Epimedium flavonols, revealing that the new 8-prenylated quercetin glycosides (subtype 3′-hydroxyldemethylicaritin in Type B) exhibited lower testosterone production-promoting activities in rat primary Leydig cells than Epimedium flavonols of subtype demethylicaritin in Type A, but possessed higher activities than the Epimedium flavonols of subtype icaritin in Type A. These results suggested that either methylation at C-4′ position or hydroxylation at C-3′ position of ring B could significantly reduce the testosterone production-promoting activities of Epimedium flavonols.

Abstract:It is usually a tedious task to profile the chemical composition of a given herbal medicine (HM) using high performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) due to the time-consuming sample preparation and laborious post-acquisition data processing procedures. Even worse, some labile compounds may face degradation risks when exposed to organic solvents for a relatively long period. As one of the most popular HMs, the promising therapeutic benefits of Epimedii Herba (Chinese name: Yinyanghuo) are well defined; however, the chemical profile, and in particular those flavonoids that have been claimed to be responsible for the efficacy, remains largely unknown. Attempts are devoted here to achieve direct LC-MS measurement and efficient post-acquisition data processing, and chemome comparison among three original sources of Epimedii Herba, such as Epimedium sagittatum (Esa), E. pubescens (Epu), and E. koreanum (Eko) was employed to illustrate the strategy utility. A home-made online liquid extraction (OLE) module was introduced at the front of the analytical column to comprehensively transfer the compounds from raw materials onto the LC-MS instrument. A mass defect filtering approach was programmed to efficiently mine the massive LC-MS dataset after which a miniature database was built involving all chemical information of flavonoids from the genus Epimedium to draw a pentagonal frame to rapidly capture potential quasi-molecular ions (mainly [M-H](-)). A total of 99 flavonoids (66 in Esa, 84 in Eko, and 66 in Epu) were captured, and structurally annotated by summarizing the mass fragmentation pathways from the mass spectrometric data of authentic compounds and an in-house data library as well. Noteworthily, neutral loss of 144 Da was firstly assigned to the neutral cleavage of rhamnosyl residues. Significant species-differences didn't occur among their chemical patterns. The current study proposed a robust strategy enabling rapid chemical profiling of, but not limited to, HMs.

Abstract:Two new flavonoids, wushanicaritin (1) and wushankaempferol (2), along with 24 known flavonoids were isolated from the whole herb of Epimedium wushanense T.S. Ying (Berberidaceae). On the basis of NMR and ESI-MS spectroscopic analysis, structures of compounds 1 and 2 were elucidated as 8-γ-hydroxy-γ,γ-dimethylpropyl-3,5,7-trihydroxy-4′- methoxyflavone and kaempferol 3-O-α-l-[2,3-di-O-β-d-(6-E-p-coumaroyl) glucopyranosyl]-rhamnopyranosyl-7-O-α-l-rhamnopyranoside, respectively. DPPH radical scavenging activity tests indicated that 1 (IC50 35.3μM) exhibited antioxidant activity comparable to Vitamin C (IC50 32.0μM), while 2 (IC50 443.7μM) showed weak activity.

Abstract:Herba Epimedii is the dried above-ground part of the genus Epimedium. Several herbs belonging to the genus Epimedium have been used in many variations of preparations and formulas for treating a wide range of diseases especially reinforcing Yang and toning the kidney by Chinese physicians for centuries[1,2]. More than 130 chemical constituents have been discovered and classified into many different chemical subgroups. Nearly half of these constituents belong to flavonoid glycosides and have been considered as the major active components in the Epimedium species. Icariin is thought to be the marker constituent of the genus because every Epimedium species contains it. The pharmaceutical effects of Epimedium species have been tested in various biological systems. It has been shown that the single constituent has less activity than that of decoctions, composite preparations or extracts of Epimedium. Therefore, it is necessary for deciphering the ensemble of the active constituents and establishing the fingerprint chromatograms of Herba Epimedii.

Abstract:Epimedii Folium, a famous Chinese materia medica, is botanically originated from Epimedium L. species. Here Epimedium L. species were reviewed in phytochemistry, ethnopharmacology, ADME (absorption, distribution, metabolism, and excretion), and processing. They have been proven to possess the efficacy on anti-osteoporosis, antitumor, and treating sexual dysfunction in traditional Chinese medicines. More than 270 compounds have been isolated from the plants of Epimedium L., some of which could be metabolized into a series of metabolites after oral absorption by the active enzymes and intestinal microflora leading to various bioactivities. Polyglucosides (epimendins A–C) and the coresponding oligoglycosides (icariin and baohuoside I) play important roles in clinical application.
