Abstract:In this work, aiming at the construction of a disposable, wireless, low-cost and sensitive system for bioassay, we report a closed bipolar electrode electrochemiluminescence (BPE-ECL) sensing platform based on graphite paper as BPE for the first time. Graphite paper is qualified as BPE due to its unique properties such as excellent electrical conductivity, uniform composition and ease of use. This simple BPE-ECL device was applied to the quantitative analysis of oxidant (H2O2) and biomarker (CEA) respectively, according to the principle of BPE sensing-charge balance. For the H2O2 analysis, Pt NPs were electrodeposited onto the cathode through a bipolar electrodeposition approach to promote the sensing performance. As a result, this BPE-ECL device exhibited a wide linear range of 0.001-15mM with a low detection limit of 0.5µM (S/N=3) for H2O2 determination. For the determination of CEA, chitosan-multi-walled carbon nanotubes (CS-MWCNTs) were employed to supply a hydrophilic interface for immobilizing primary antibody (Ab1); and Au@Pt nanostructures were conjugated with secondary antibody (Ab2) as catalysts for H2O2 reduction. Under the optimal conditions, the BPE-ECL immunodevice showed a wide linear range of 0.01-60ngmL-1 with a detection limit of 5.0pgmL-1 for CEA. Furthermore, it also displayed satisfactory selectivity, excellent stability and good reproducibility. The developed method opened a new avenue to clinical bioassay.
Bipolar Electrode Array for Multiplexed Detection of Prostate Cancer Biomarkers

Abstract:Owing to the characteristics of high throughput, high flexibility, and convenient separation of the sensing and reporting reactions, the bipolar electrode (BPE) shows great potential in clinical analysis. However, there are some difficulties in the combination of BPEs and multiplex electrochemiluminescence (ECL) biosensing, such as the need for small sample consumption, multistep operations, and separated sample loading. In this paper, a microfluidic BPE array chip was fabricated toward multiplex detection of cancer biomarkers. With a special channel structure and the difference in flow resistance of channels of different sizes, the direction of liquid flow was successfully controlled. In this way, rapid and automatic multiplex sampling was achieved on the array, which would help improve the sensing efficiency and reduce the reagent consumption. The ECL BPE array chip served as an immunosensor for multiple prostate cancer biomarkers including prostate-specific antigen (PSA), interleukin-6 (IL-6), and prostate-specific membrane antigen (PSMA). The microfluidic BPE chip shows good reproducibility and high sensitivity. The limits of detection for PSA, IL-6, and PSMA are 0.093 ng/mL, 0.061 pg/mL, and 0.059 ng/mL, respectively. It also exhibits excellent performance in real sample analysis. The integrated ECL BPE array shows a good application prospect in clinical sensing of cancer biomarkers, as well as point-of-care testing.

Abstract:We report a bipolar electrode (BPE) sensing platform for the temporal detection of cancer cells. Combining the advantages of anodic dissolution and electrochemiluminescence (ECL), this strategy shows an ultralow detection limit down to 5 cells/cm(2). At the anode working as the reporting pole, Au NPs were assembled through DNA double strand, which served as both catalyzer for the ECL reaction of luminol/H2O2 and seeds for the chemical reduction of Ag, the anodic dissolution probe. The duration of Ag layer dissolution was positively correlated with the amount of Ag but negatively related to the controlled potential and the conductivity of the circuit. Therefore, it was possible to amplify a slight conductivity change through tuning the other two factors. As the formation of Ag@Au completely quenched the ECL emission of luminol, the ECL emission recovery reflected the extent of anodic dissolution. Through monitoring the ECL recovery time before and after the incubation of cells on the cathode, a few number of cells could be quantified due to slight difference of the conductivity. This method shows several merits. First, the combination of anodic dissolution and ECL significantly increases the detection sensitivity of BPE device. In addition, this strategy broadens the application of BPE for the ultrasensitive monitoring of cancer cells, which was applied to investigate the capture efficiencies of antibodies and aptamers toward MCF-7 and A549.

Abstract:We report a multicolor ECL device based on closed bipolar electrode (BPE) for the visualized sensing of prostate-specific antigen (PSA) in human blood serum. As the emission color of concomitant electrochemiluminophores is potential resolved, similar to a three-electrode system, selective excitation of ECL could be achieved by tuning the interfacial potential at the poles of BPE. Via modulating the resistance of BPE, multicolor ECL emission of [Ru(bpy)3]2+ and [Ir(ppy)3] mixture using tripropylamine (TPrA) as the co-reactant was observed at the anode and the principle was elaborated. The concept was utilized to the quantification of clinical biomarkers with the color variation. A PSA concentration dependent silver bridge was constructed in the gap of the BPEs as an electric conductivity modulator. On the basis of a multicolor BPE-ECL device, the cutoff values (4.0 and 10.0 ng/mL) of human PSA could be recognized with naked eyes by the green-yellow-red ECL emission changing. As the first multicolor ECL device in biological analysis, BPE may raise the application of potential-resolved ECL to a new level.

Abstract:This paper develops an efficient electrochemiluminescence (ECL) enhancement strategy on closed bipolar electrode for the detection of prostate specific antigen (PSA). We first synthesized a cyclometalated iridium(III) complex (pq)2Irbza with high ECL efficiency and used as ECL emitter in the anodic cell of BPE. While we introduced a Pt-tipped Au NRs and constructed a sandwich immune structure at the cathodic pole of BPE. Combined the signal amplification strategies of enzyme catalysis and the synergistic catalytic effect of bimetallic structure for the reduction of H2O2, the attached Pt-tipped Au NRs-GOx-Ab2 nanocomplex as both recognition probes and signal amplification units could mediate the ECL signals of (pq)2Irbza/tripropylamine (TPrA) on the anodes of BPE through faradaic reaction due to the charge neutrality of BPE. Therefore, a highly sensitive BPE-ECL sensor for detection of PSA with a detection limit of 0.72 pg/mL and a linear range from 1.0 pg/mL to 10 ng/mL was obtained. This work is expected to broaden the application of iridium complex and bimetallic nanocatalyst in biological detection and could be utilized to detect many other biological molecules.

Abstract:In the past few years, point-of-care testing (POCT) technology has crossed the boundaries of laboratory determination and entered the stage of practical applications. Herein, the latest advances and principal issues in the design and fabrication of paper-based bipolar electrode electrochemiluminescence (BPE-ECL) sensors, which are widely used in the POCT field, are highlighted. After introducing the attractive physical and chemical properties of cellulose paper, various approaches aimed at enhancing the functions of the paper, and their underlying principles are described. The materials typically employed for fabricating paper-based BPE are also discussed in detail. Subsequently, the universal method of enhancing BPE-ECL signal and improving detection accuracy is put forward, and the ECL detector widely used is introduced. Furthermore, the application of paper-based BPE-ECL sensors in biomedical, food, environmental and other fields are displayed. Finally, future opportunities and the remaining challenges are analyzed. It is expected that more design concepts and working principles for paper-based BPE-ECL sensors will be developed in the near future, paving the way for the development and application of paper-based BPE-ECL sensors in the POCT field and providing certain guarantee for the development of human health.

Visual Electrochemiluminescence Detection of Cancer Biomarkers on A Closed Bipolar Electrode Array Chip

Abstract:This paper describes a novel electrochemiluminescence (ECL) imaging platform for simultaneous detection of cancer biomarkers based on a closed bipolar electrode (BPE) array. It consists of two separated channel arrays: detection channel array and sensing channel array, which are connected by a group of parallel ITO BPEs on a glass substrate. Besides, two parallel ITO strips are fabricated at the two sides of BPE array and employed as driving electrodes. After Au films are electrochemically deposited on the cathodes of the BPE array, nanobioprobes including biorecognition elements (aptamer or antibody) and a novel electrochemical tag, which is synthesized by doping thionine in silica nanoparticles (Th@SiO2 NPs), are introduced into the cathodes by immunoreaction or DNA hybridization. The Th@SiO2 coupled nanobioprobes as both recognition probes and signal amplification indicators could mediate the ECL signals of Ru(bpy)3(2+)/tripropylamine (TPA) on the anodes of BPE array through faradaic reaction due to the charge neutrality of BPE. Thus, multiplex detection of cancer biomarkers (adenosine triphosphate (ATP), prostate-specific antigen (PSA), α-fetoprotein (AFP) and thrombin) is realized by forming specific sensing interfaces onto the cathodic poles of BPEs in different sensing channels and reported by the ECL images of the Ru(bpy)3(2+)/TPA system on the anodic poles of BPEs in detection channels. The results demonstrate that this visual ECL platform enables sensitive detection with excellent reproducibility, which may open a new door toward the development of simple, sensitive, cost-effective, and high throughput detection methods on biochips.
Insights into the Mechanism of Bipolar Electrodeposition of Au Films and Its Application in Visual Detection of Prostate Specific Antigens

Abstract:Au particles are commonly used for deposition on the surface of a bipolar electrode (BPE) in order to amplify electrochemical and electrochemiluminescence (ECL) signal because of their excellent conductivity, biocompatibility, and large surface area. In this work, a closed BPE device was fabricated and Au particles were deposited on the two poles of a BPE via bipolar deposition. Results indicated that the electrochemical stability of Au film on the anode part of the BPE and the reduction of AuCl4– to Au on the cathode part of the BPE depended on the conductivity of the solution. The prepared Au–Au BPE exhibited a remarkable amplification effect on the ECL signal. Then, a specific sensing interface was constructed on one pole of the BPE for the visual detection of prostate-specific antigens (PSA) based on sandwich-type immunoreactions between primary PSA antibodies (Ab1) on the electrode surface, PSA, and SiO2 nanoparticles labeled secondary PSA antibodies (SiO2-Ab2). The designed biosensor exhibited a good linear relationship for the ECL detection of PSA in the range of 1 × 10−6 to 1 × 10−10 g/mL with a correlation coefficient of 0.9866; the limit of detection (LOD) was 1.5 × 10−11 g/mL. Additionally, the biosensor can realize the electrochemical imaging of PSA by regulating the electrochemical oxidation of the Au anode with the immunoreactions on the cathode part of BPE. Therefore, the small, portable and highly sensitive biosensors have great potential for on-site detection.

Abstract:A novel disposable paper-based bipolar electrode (BPE) array is fabricated for multiplexed electrochemiluminescence (ECL) detection of pathogenic DNAs. This proposed BPE array device consists of 15 units, each consisting of six sensing cells and two reporting cells patterned using hydrophobic wax. A hairpin structure DNA assembled on the cathodes of BPEs hybridizes with Pt nanoparticles (NPs) labeled probe DNA in the presence of complementary target DNA. The introduction of Pt NPs catalyzes the reduction of dissolved O 2 at cathodes and induces an enhanced ECL signal from Ru(bpy) 3 2+ /tripropylamine (TPrA) at the anodes of BPEs. The dissolved O 2 lost in reduction reaction could be promptly replenished due to the relatively large contact area of the paper-based cells with air, which ensures the stability of ECL signal. This obtained paper-based BPE array sensor showed excellent performances for the multiplexed analysis of the syphilis (Treponema pallidum) gene, the immunodeficiency virus gene (HIV) and hepatitis B virus gene (HBV).
Paper-based Bipolar Electrode-Electrochemiluminescence (BPE-ECL) Device with Battery Energy Supply and Smartphone Read-Out: A Handheld ECL System for Biochemical Analysis at the Point-of-care Level

Abstract:In this work, a handheld paper-based bipolar electrode-electrochemiluminescence (P-BPE-ECL) system has been proposed. In this system, a rechargeable battery is used for power supply, while a smartphone is applied for read-out of ECL signal. For the P-BPE-ECL, the carbon ink-based BPE and driving electrodes are screen-printed on the paper, and the wax-screen-printing is employed to fabricate microfluidic channels on electrode-patterned paper. Moreover, the luminol/H2O2-based ECL reaction is applied to demonstrate the quantitative ability of the P-BPE-ECL system. Under optimized conditions, H2O2 can be determined over the range of 5-5000 mu M, with a detection limit of 1.75 mu M, and relative standard deviations (RSDs) of 7.59%, 3.82% and 4.93% for 10, 100 and 1000 mu M H2O2 (n = 5). Thus, this system has an acceptable sensitivity, dynamic range, stability and reproducibility. Finally, the applicability of the P-BPE-ECL system is demonstrated for detection of glucose in phosphate buffer solution (PBS) and artificial urine (AU) samples, with the detection limits of 0.017 mM and 0.030 mM, respectively. Moreover, the P-BPE-ECL system has the ability to perform high-throughput detection of glucose. Therefore, the developed system can provide a new platform for many applications such as point-of-care testing, health diagnosis and environmental monitoring. (C) 2016 Elsevier By. Abstract:A chemiluminescence excited photoelectrochemical (PEC) competitive immunosensor for sensitive and specific detection of the prostate specific antigen (PSA) is firstly developed by combining a microfluidic paper-based device. Firstly, porous ZnO spheres with large surface area and good biocompatibility are attached onto the Au nanoparticle modified paper working electrode, which serve as an effective matrix for antigens. CdS nanorods (NRs) are selected as the photoactive materials due to their excellent fast and long distance electron transport capability, which allow the binding of the horseradish peroxidase-labeled signal antibody onto CdS NRs (CdS NR-Ab-HRP). After a competitive immunoassay format, the CdS NR-Ab-HRP labels are captured onto the electrode surface. The chemiluminescent excitation is produced from the oxidation of luminol by H2O2 in the presence of HRP. The more antigens in solution can bind to CdS NR-Ab-HRP the less CdS NR-Ab-HRP can bind to antigens immobilized on the electrode, which result in the decrease of chemiluminescence emission and light absorption, leading to the decrease of photocurrent intensity. The PEC response from CdS NR-Ab-HRP successfully fulfilled the sensitive detection of PSA in the linear range from 0.005 to 150 ng mL(-1) with a detection limit of 2.3 pg mL(-1). The proposed immunosensor shows excellent analytical performance with high reproducibility and stability, and can become a promising platform for other protein detection.

Abstract:A novel paper-based open bipolar electrode-electrochemiluminescence (P-o-BPE-ECL) sensing has been developed for visual detection. Wax-screen-printing was employed to make microfluidic channels on filter paper, and the carbon ink-based BPE and driving electrodes were screen-printed on paper. An inexpensive CCD camera was used as a detector for imaging sensing. In this study, two P-o-BPE-ECL imaging sensing platforms were demonstrated. For the first one, a hydrophilic paper channel was used to host a single "band"-shaped BPE. Under optimal conditions, this P-o-BPE-ECL platform successfully fulfilled the imaging sensing of tri-n-propylamine (TPA) with a linear range from 10 to 1000 mu M with a detection limit of 8.7 mu M. In addition, the determination of H2O2 using this platform could be performed in the linear range of 50 mu M to 5000 mu M with a detection limit of 46.6 mu M. For the second platform, an open BPE array configuration has been proposed for P-o-BPE-ECL sensing, where eleven BPEs were stridden across the two legs of the "U"-shaped paper channel. On this prototype, a simple screening application has been performed. These results show that the proposed strategy offers great promise in providing a simple, low-cost, rapid and environment-friendly solution for biochemical applications. (C) 2015 Elsevier B.V. Abstract:Early diagnosis of cancers relies on the sensitive detection of specific biomarkers, but most of the current testing methods are inaccessible to home healthcare due to cumbersome steps, prolonged testing time, and utilization of toxic and hazardous substances. Herein, we developed a portable selfpowered photoelectrochemical (PEC) sensing platform for rapid detection of prostate-specific antigen (PSA, as a model diseaserelated protein) by integrating a self-powered photoelectric signal output system catalyzed with chemiluminescence-functionalized Au nanoparticles (AuNPs) and a phosphomolybdic acid (PMA)-based photochromic visualization platform. TiO2-g-C3N4-PMA photosensitive materials were first synthesized and functionalized on a sensor chip. The sensor consisted of filter paper modified with a photocatalytic material and a regional laser-etched FTO electrode as an alternative to a conventional PEC sensor with a glass-based electrode. The targeting system involved a monoclonal anti-PSA capture antibody-functionalized Fe3O4 magnetic bead (mAb(1)-MB) and a polyclonal anti-PSA antibody (pAb(2))-N-(4-aminobutyl)-N-ethylisoluminol-AuNP (ABEI-AuNP). Based on the signal intensity of the chemiluminescent system, the photochromic device color changed from light yellow to heteropoly blue through the PMA photoelectric materials integrated into the electrode for visualization of the signal output. In addition, the electrical signal in the PEC system was amplified by a sandwich-type capacitor and readout on a handheld digital multimeter. Under optimum conditions, the sensor exhibited high sensitivity relative to PSA in the range of 0.01-50 ng mL(-1) with a low detection limit of 6.25 pg mL(-1). The flow-through chemiluminescence reactor with a semiautomatic injection device and magnetic separation was avoid of unstable light source intensity inherent in the chemiluminescence process. Therefore, our strategy provides a new horizon for point-of-care analysis and rapid cost-effective clinical diagnosis.

Abstract:This paper describes a novel rotational paper-based analytical device (RPAD) to implement multi-step electrochemiluminescence (ECL) immunoassays. The integrated paper-based rotational valves can be easily controlled by rotating paper discs manually and this advantage makes it user-friendly to untrained users to carry out the multi-step assays. In addition, the rotational valves are reusable and the response time can be shortened to several seconds, which promotes the rotational paper-based device to have great advantages in multi-step operations. Under the control of rotational valves, multi-step ECL immunoassays were conducted on the rotational device for the multiplexed detection of carcinoembryonic antigen (CEA) and prostate specific antigen (PSA). The rotational device exhibited excellent analytical performance for CEA and PSA, and they could be detected in the linear ranges of 0.1-100 ngmL(-1) and 0.1-50 ng mL(-1) with detection limits down to 0.07 ng mL(-1) and 0.03 ng mL(-1), respectively, which were within the ranges of clinical concentrations. Abstract:Prostate cancer is one of the most common cancers in the world, and its early diagnosis can effectively reduce mortality. A new label-free photoelectrochemical (PEC) immunosensor on the basis of Bi 2 WO 6 /BiOBr nanocomposite materials has been successfully prepared for the test of prostate-specific antigen (PSA) in human serum in this work. The Ag 2 S-sensitized Bi 2 WO 6 /BiOBr heterojunction was used as a photosensitive material, which effectively improved the photocurrent response. On Bi 2 WO 6 /BiOBr surface, dopamine immobilized PSA antibody by self-polymerizing to form polydopamine membrane. Antigen and antibody are specifically combined to achieve quantitative detection of PSA according to the current changes at different concentrations of antigen. Under the optimal experimental conditions, the PEC immunosensor has an ideal linear relationship between 1 pg/mL - 50 ng/mL, and the detection limit is 0.084 pg/mL. In addition, the prepared immunosensor has good stability, reproducibility and selectivity, providing a new method for the detection of PSA in actual sample analysis.

A Simple and Sensitive Paper-Based Bipolar Electrochemiluminescence Biosensor for Detection of Oxidase-Substrate Biomarkers in Serum

Abstract:Nowadays, the bipolar electrochemiluminescence (BP-ECL) is basically performed in complicated and expensive microfluidic devices, which may limit its wide applications. Here, a paper-based BP-ECL biosensor using "U"-shaped bipolar electrode (BPE) is developed for detection of oxidase-substrate biomarkers in human serum samples, with no need for complicated and expensive microfluidic technologies and peripheral apparatuses. The BP-ECL paper chips (PCs) can be well fabricated by two screen-printing processes: wax-screen-printing for on-paper hydrophilic and hydrophobic patterns, and carbon ink-screen-printing for BPEs and driving electrodes on wax-patterned paper. A low-cost and portable CCD is used to detect ECL signals from the luminol-H2O2 reaction at the BPE anodic pole. Under optimized conditions, the paper-based BP-ECL biosensor can measure the concentrations of H2O2, with a linear range of 5-10000 mu M and a detection limit of 0.424 mu M. Importantly, the biosensor is capable of achieving a linear range of 10-5000 mu M for choline, lactate or cholesterol; and the respectively corresponding detection limits are 0.573 11M, 3.132 mu M and 7.418 mu M. Moreover, it has an acceptable detection range, sensitivity, stability and selectivity. Finally, the developed method is successfully applied for determination of choline, lactate or cholesterol in human serum. (C) 2018 The Electrochemical Society.
Bio-inspired nanozyme with ultra-thin Fe–Bi2O2S nanosheets for in-situ amplified photoelectrochemical immunoassay of cancer-related protein

Abstract:A Fe-loaded Bi2O2S nanosheet photoanode serving as photoelectric biomonitoring platform for the detection of prostate-specific antigen (PSA) using biologically inspired prussian nanoparticle (PB)-catalyzed biocatalytic precipitation strategy was developed. Primarily, the signal probe PB-mAb2 obtained by electrostatic adsorption was immobilized on a microplate in the presence of target PSA, and 4-chloro-1-naphthol (4-CN) was oxidized to benzo-4-chloro-hexadienone (4-CD) with the assistance of exogenous hydrogen peroxide, which was generated by a large number of hydroxyl radicals catalyzed by PB. The generated 4-CD showed strongly low conductivity characteristics to burst the photocurrent of highly photoactive Fe-Bi2O2S photoanode. The split incubation reaction could be suitable for high volume and low-cost rapid detection. A dynamic response range of 0.1-100 ng mL-1 with a limit of detection of 34.2 pg mL-1 was achieved with the sensor based on a photoelectric sensing platform and a biomimetic catalytic precipitation reaction. Equally important, the sensor also showed good potential in the detection of real samples compared to commercially available ELISA kits. In conclusion, this work provides a fresh scheme for the development of sensitive biosensors through a bio-inspired catalytic strategy of versatility and a photoanode coupling with high photoelectric activity.

Abstract:Microfluidic paper-based analytical device (μPAD), originally developed for improving healthcare in developing countries, presents a simple yet powerful platform for performing low-cost and portable diagnostic devices. Here, we reported an enzyme-free μPAD for the detection of two tumor markers. Firstly, a porous structure of gold nanoparticles (AuNPs) modified paper working electrode (Au-PWE), with a feature of all-around conductivity and plenty of active sites favoring for the biological ligands attachment, was firstly fabricated as a sensor substrate. Next cyclodextrin functionalized AuNPs (CD@AuNPs) as dual mimicking enzyme was prepared to load secondary antibodies or peptide. One the one sample zone, in the presence of carcinoembryonic antigen (CEA), CD@AuNPs could be introduced into the Au-PWE through a sandwich immunoreaction, boosting the electrochemical signal of o-phenylenediamine (o-PD) via the trigger of a cascade catalysis reaction towards glucose & o-PD, eventually resulting in a sensitive detection of CEA. On another working zone, with the introduction of another target prostate specific antigen (PSA), the peptide cleavage took place, which further led to the CD@AuNPs released from Au-PWE, and then the variation of electrochemical signals were recorded for detection of PSA. We demonstrated, using the device, the detection of CEA and PSA clinically had high sensitivity, wide linear ranges and low detection limits. We believe that our work provides a promising platform for point-of-care testing, especially in resource-limited regions.

Abstract:Here a novel paper-based electrochemiluminescence (ECL) device was developed for the detection of HL-60 cancer cells. In this approach, porous filter paper as a thin-layer electrochemical cell connected to a sheet of ITO working electrode. Au nanoparticles (Au NPs) and graphene were coated on the surface of ITO to immobilize capture HS-DNA. Then, the aptamer of HL-60 cancer cells, which is labeled with Ru(bpy)(3)(2+)-conjugated silica nanoparticles (RuSi@Ru(bpy)(3)(2+)) is hybridized with the capture DNA to form bioconjugates. In the presence of HL-60 cells, the aptamer would conjugate with glycoprotein at cell surface and RuSi@Ru(bpy)(3)(2+) NP-aptamer would be released. After the dissociation of RuSi@Ru(bpy)(3)(2+) NP-aptamer from the surface of ITO, ECL signal would decline. This simple, portable and disposable testing platform could detect HL-60 cancer cells down to 56 cells/mL (C) 2014 Elsevier B.V. All rights reserved.
