Abstract:Integrating multiple functionalities into individual nanoscale complexes is of tremendous importance in biomedicine, expanding the capabilities of nanoscale structures to perform multiple parallel tasks. Here, the ability to enhance two different imaging technologies simultaneously-fluorescence optical imaging and magnetic resonance imaging-with antibody targeting and photothermal therapeutic actuation is combined all within the same nanoshell-based complex. The nanocomplexes are constructed by coating a gold nanoshell with a silica epilayer doped with Fe3O4 and the fluorophore ICG, which results in a high T-2 relaxivity (390 mM(-1) s(-1)) and 45X fluorescence enhancement of ICG. Bioconjugate nanocomplexes target HER2+ cells and induce photothermal cell death upon near-IR illumination.
Preparation and Two-Photon Imaging of a Water-Soluble Fluorescent Nanoparticle

Abstract:A water-soluble polymeric fluorescent nanoparticle (PTCN) has been proposed for two-photon fluorescence (TPF) imaging of living cells. PTCN was firstly prepared by chemical crosslinking of polyethyleneimine using a dual carboxyl 1, 8-naphthalimide derivative as the crosslinker. Subsequently, the investigations on its structure, morphology, one- and two-photon fluorescence properties and cytotoxicity were carried out. It was found that PTCN was spherical with the diameters of 5-10 nm. It could emit fluorescence at 536 nm with the excitation wavelengths of 443 nm and 800 nm, respectively. PTCN possessed good pH stability and photostability. Its fluorescence did not change in the pH range of 4.0-9.0. In pH 7.4 buffer solution, when it was detected at lambda(ex)/lambda(em) = 443 nm/536 nm for 12000 times, the decrease in fluorescence intensity was less than 1%. Cytotoxic assays indicated it was low toxic upon interactions with Hela cells for 24 h at a concentration of 15.0 mg/L. Finally, the application of PTCN for imaging live cells was assessed by TPF microscopy technique. After Hela cells were incubated with PTCN for 2 h, the green fluorescence was observed in the cellsap at the excitation wavelength of 800 nm. The proposed nanoparticle is potential in the development of targeting TPF probes.
Novel lanthanide–polymer complexes for dye-free dual modal probes for MRI and fluorescence imaging

Abstract:High-resolution imaging is a powerful technique in theranostics and staging of tumors. Dual-modal imaging using lanthanide hybrid nanoparticles as probes has been attracting increasing attention due to their specific and intrinsic physical properties. The present work reports a novel water-soluble micelle of an amphiphilic block copolymer with diketone pendants chelating gadolinium (Gd3+) and europium (Eu3+) cations as a dual probe for magnetic resonance imaging (MRI) and fluorescence imaging for clinical detection. The Gd3+ and Eu3+ hybrid micelles have homogeneous size distribution and hydrodynamic diameters tunable between 80 and 220 nm. The macromolecular diketone ligand enhances the relaxation rate of Gd3+ and sensitizes Eu3+ luminescence efficiently. Further sensitized by the co-doped Gd3+, the Eu3+ complex exhibits a long-lived emission with a large Stokes shift. A strong red light at a wavelength of 614 nm is observed with an excitation in the range of 350 and 405 nm. Cellular uptake experiments reveal excellent biocompatibility and rapid uptake of the nanoparticles by MCF-7 cells. The robust dual-modal probes are promising in applications for early diagnosis of tumors, presurgical planning, and intraoperative fluorescence-guided surgery.

Abstract:Targeted molecular imaging with two-photon fluorescence microscopy (2PFM) is a powerful technique for chemical biology and, potentially, for noninvasive diagnosis and treatment of a number of diseases. The synthesis, photophysical studies, and bioimaging are reported for a versatile norbornene-based block copolymer multifunctional scaffold containing biocompatible (PEG), two-photon fluorescent dyes (fluorenyl) and targeting (cyclic-RGD peptide) moieties. The two bioconjugates, containing two different fluorenyl dyes and cRGDfK covalently attached to the polymer probe, formed a spherical micelle and self-assembled structure in water, for which size was analyzed by transmission electron microscopy (TEM) and dynamic light scattering (DLS). Cell viability and 2PFM imaging of human epithelial U87MG cell lines that overexpress α(v)β(3) integrin was performed via incubation with the new probes, along with negative control studies using MCF-7 breast cancer cells and blocking experiments. 2PFM microscopy confirmed the high selectivity of the biocompatible probe in the integrin-rich area in the U87MF cells while blocking as well as negative control MCF-7 experiments confirmed the integrin-targeting ability of the new probes.
Ultrabright and Multicolorful Fluorescence of Amphiphilic Polyethyleneimine Polymer Dots for Efficiently Combined Imaging and Therapy.

Abstract:Multifunctional nanoparticles as theranostic tools hold great potential for its unique and efficient way to visualize the process of disease treatment. However, the toxicity of conventional fluorescent labels and difficulty of functionalization limit their widespread use. Recently, a number of amino-rich polymers have demonstrated high luminescent fluorescence but rarely showed potential for in vivo imaging due to their blue fluorescence. Here, a general route has been found to construct polymer-based multifunctional nanoparticles for combined imaging and drug delivering. The weak fluorescent polyethyleneimine (PEI) has been conjugated with hydrophobic polylactide as the amphiphilic PEI for construction of nanoparticles which showed bright and multicolor fluorescence with high drug loading capacity. The paclitaxel-loaded nanoparticles showed significant therapy effect in contrast to the free paclitaxel. Meanwhile, fluorescence imaging of the nanoparticles showed accumulation around tumor. These results demonstrate a new type of polymer-based multifunctional nanoparticles for imaging-guided drug delivery.
Multivalent supramolecular fluorescent probes for accurate disease imaging

Abstract:Optical imaging is a powerful tool for early disease detection and effective treatment planning, but its accuracy is often compromised by the uptake of imaging materials by the mononuclear phagocyte system (MPS). Herein, we leverage multivalent host-guest interactions between cyanine dyes and β-cyclodextrin polymers to develop supramolecular probes with enhanced stability, optical, and transport profiles for accurate in vivo imaging. These multivalent interactions not only ensure the stability of the probes but also enhance fluorescence efficiency by minimizing nonradiative decay. Our self-assembly approach effectively modulates probe size and surface properties, enabling evasion of MPS clearance and promoting prolonged bloodstream circulation, thereby improving the signal-to-background ratio for imaging. The effectiveness of our design is demonstrated by substantial advancements in the early diagnosis of acute kidney injury and by providing high-contrast imaging and precise surgical navigation across various tumor models. Our strategy not only advances optical imaging materials toward clinical translation but also establishes a versatile platform applicable to multiple imaging modalities.

Multicolor Two-Photon Nanosystem for Multiplexed Intracellular Imaging and Targeted Cancer Therapy

Abstract:The novel theranostic nanosystems based on two-photon fluorescence can achieve higher spatial resolution of deep tissue imaging for simultaneous diagnosis and therapy of a variety of cancers. Herein, we have designed and prepared FRET-based two-photon mesoporous silica nanoparticles (MTP-MSNs) for single-excitation multiplexed intracellular imaging and targeted cancer therapy for the first time. This nanosystem includes two constituents, containing (1) multicolor two-photon mesoporous silica nanoparticles and (2) cancer cell-targeting aptamers that act as gatekeepers for MTP-MSNs. After incubation with cancer cells, the Dox-loaded and aptamer-capped MTP-MSNs could be internalized into the cells, opening the pores and releasing the drug. Furthermore, using two-photon multicolor fluorescence, MTP-MSNs could serve as good contrast agents for multicolor two-photon intracellular imaging with increased imaging depth and improved spatial localization of tissue. In sum, these multicolor MTP-MSNs provide a promising system for traceable targeted cancer therapy with further applications in multiplex intracellular imaging and the screening of drug.

Abstract:Intelligent polymeric micelles with fluorescence imaging feature have been emerged as promising tools for theranostics. However, conventional fluorescent dyes are limited by short wavelength excitation, interference of tissue autofluorescence, limited imaging depth and quenched emission in aggregation state. Methods: We synthesized a novel mPEG-SS-Poly (AEMA-co-TBIS) (mPEATss) copolymer to develop multifunctional polymeric micelles with great AIE feature for cancer therapy and AIE active two-photon bioimaging. The stimuli-responsive behavior and AIE active two-photon cell and tissue imaging as well as in vitro and in vivo antitumor ability of DOX-loaded mPEATss were studied. Results: mPEATss micelles showed excellent AIE active two-photon cell imaging ability and deep tissue imaging ability. Antitumor drug DOX could be encapsulated to form a drug-loaded micellar system with a small diameter of 65 nm. The disassembly and charge-conversion of mPEATss micelles could be triggered by acidic environment, resulting in accelerated drug release and great antitumor efficacy. In vivo, ex vivo imaging and in vivo pharmacokinetic study demonstrated that mPEATss micelles could efficiently accumulate in tumor sites, which ensured ideal anticancer effect. Conclusions: This pH and redox dual responsive and AIE active two-photon imaging polymeric micelles would be a promising candidate for theranostics.
Two-Photon Absorption Aggregation-Induced Emission Luminogen/Paclitaxel Nanoparticles for Cancer Theranostics

Abstract:Multifaceted nanoplatforms integrating fluorescence imaging and chemotherapy have garnered acknowledgment for their potential potency in cancer diagnosis and simultaneous in situ therapy. However, some drawbacks remain for traditional organic photosensitizers, such as poor photostability, short excitation wavelength, and shallow penetration depth, which will greatly lower the chemotherapy treatment efficiency. Herein, we present lipid-encapsulated two-photon active aggregation-induced emission (AIE) luminogen and paclitaxel (PTX) nanoparticles (AIE@PTX NPs) with bright red fluorescence emission, excellent photostability, and good biocompatibility. The AIE@PTX NPs exhibit dual functionality as two-photon probes for visualizing blood vessels and tumor structures, achieving penetration depth up to 186 and 120 μm, respectively. Furthermore, the tumor growth of the HeLa-xenograft model can be effectively prohibited after the fluorescence imaging-guided and PTX-induced chemotherapy, which shows great potential in the clinical application of two-photon cell and tumor fluorescence imaging and cancer treatment.

Abstract:Optical imaging in the second near-infrared (NIR-II) windows reduces much more autofluorescence and photon scattering from biological tissues and allows further tissue penetration depth and superior spatial resolution in living bodies. Herein, a fused-ring 2,2' - ( (2Z,2'Z)-( (12,13-bis(2-ethylhexyl)-3,9-diundecyl-12,13-dihydro-[1,2,5] thiadiazolo [3,4-e]thieno [2,'' 3 '' :4',5'] thieno-[2',3' :4,5] pyrrolo [ 3,2-g] thieno [2',3' :4,5] thieno [3,2-b] indole-2,10-diyl) bis(methanylylidene))bis (5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene))dimalononitrile (TPBT) molecule was explored as a multifunctional tumor theranostic reagent for photothermal/photodynamic therapy guided by NIR-II imaging. The TPBT molecule has an electron-deficient core with a ladder-type multi-fused ring and shows a narrow band gap that can enhance the near-infrared absorption. The J-aggregative TPBT NPs were formed by nanoprecipitation with great bathochromic shift in absorption and emission spectra, which endows them with ideal fluorescence imaging ability in the NIR-II region. Moreover, TPBT NPs present both higher photothermal conversion efficiency (similar to 36.5%) and effective ROS generation ability, making them excellent candidate for cancer photothermal/photodynamic therapy. Moreover, the biocompatible TPBT NPs can effectively passively target tumor sites due to their enhanced permeability and retention effect for more precision treatment. Thus, TPBT NPs as a multifunctional phototheranostic agent in the NIR-II region present promising potential in clinical cancer NIR-II imaging-guided phototherapy.

Abstract:Fluorescence and magnetic resonance imaging (FL/MRI) has received much attention due to their complementary characteristics. To tackle this difficulty, we put forward a strategy of aggregation and de-aggregation based on luminogens with aggregation-induced emission (AIE) features. NIR II photosensitizer 4,4 '-(6,7-diphenyl-[1,2,5]thiadiazolo[3,4-g]quinoxaline-4,9-diyl)bis(N,N-diphenylaniline) (TQ-TPA) with type I reactive oxygen species (ROS) generation ability and amphiphilic 2TPE-Gd were synthesized, and both of them showed the AIE feature. During the construction of nanomicelles, hydrophobic TQ-TPA spontaneously aggregated into the core of the nanomicelles formed by 1,2-distearoyl-sn-glycerol-3-phosphoethanolamine-N-methoxy(polyethylene glycol)-2000 (DSPE-PEG). Meanwhile, aggregates of 2TPE (tetraphenylethylene)-Gd in aqueous solution could be de-aggregated and inserted into the interface, and formed TGdTT nanomicelles (NMs). Based on this strategy and the AIE feature, the TGdTT NMs exhibited strong NIR II fluorescence and type I ROS generation ability, and enhanced T-1 relaxivity (r(1)). Moreover, in vitro, in vivo, and pharmacokinetics results demonstrated that these nanomicelles have good biosafety and a long blood circulation time. Finally, they allowed successful realization of complementary MR/NIR II fluorescence dual-modal imaging-guided photodynamic therapy (PDT) to inhibit tumor growth. This work demonstrated that the aggregation and de-aggregation strategy of AIEgens in core-shell nanomicelles is very attractive for constructing multifunctional theranostic probes.
Enhanced luminescence and stability of TFMDSA nanoparticles via polymer-induced aggregation for bioimaging

Abstract:In recent years, fluorescence imaging has occupied a very important position in the life science and biomedical fields. However, achieving nanomaterials for bioimaging with both high fluorescence quantum efficiency and high stability remains a significant challenge. Herein, we synthesized mPEG 5K –PCL 10K @TFMDSA and mPEG 5K –PLLA 10K @TFMDSA nanoparticles using polymer-induced aggregation. This method significantly enhanced the luminescence efficiency of TFMDSA nanoparticles in solution, attributed to improved intermolecular interactions and restricted molecular vibrations. The resulting nanoparticles exhibited exceptional optical stability over a period of seven days and demonstrated low cytotoxicity towards HeLa cells, making them highly suitable for bioimaging applications. Cellular uptake studies indicated that these nanoparticles were more efficiently internalized by HeLa cells compared to their amorphous counterparts, likely due to their unique square morphology. Our findings highlight the potential of polymer-induced aggregation in enhancing the optical properties and stability of TFMDSA nanomaterials, suggesting their promise as biofluorescent probes for cancer diagnosis and other biomedical applications.

Abstract:The multifunctional nano-micelle platform holds great promise to enhance the accuracy and efficiency of cancer diagnosis and therapy. In this work, an amphiphilic poly[(poly(ethylene glycol) methyl ether methacrylate)-co-(3-aminopropyl methacrylate)]-block-poly(methyl methacrylate) (P(PEGMA-co-APMA)-b-PMMA) block copolymer was synthesized by successive RAFT polymerizations and subsequent chemical modification. Then the multifunctional micelles with high solubility in physiological environments were developed by a self-assembly and crosslinking processes. The photosensitizer segment, 5,10,15,20-tetrakis (4-carboxyphenyl) porphyrin (TCPP), serves as a tetra-functional cross-linker, photodynamic agent, fluorescence indicator, as well as magnetic resonance (MR) contrast agent after labelling with manganese ions (Mn(2+)), while IR825 simultaneously locating in the interior of the fabricated micelles contributed to the photoacoustic (PA) imaging ability and the photothermal effect. The prepared nanoparticles show great stability in a physiological environment with uniform morphology and diameters of around 80 nm as disclosed by stability investigation, TEM and DLS analysis. IR825@P(PEGMA-co-APMA)-b-PMMA@TCPP/Mn nanoparticles displayed high in vivo tumor uptake with a long blood circulation half-life (∼3.64 h) by the EPR effect after intravenous (i.v.) injection, as revealed by fluorescence, MR and PA imaging models. In vivo anti-tumor effects were achieved via a combined photothermal and photodynamic therapy without noticeable dark toxicity, and this strategy was able to induce a remarkably improved synergistic therapeutic effect to both superficial and deep regions of tumors under mild conditions compared with either single photothermal or photodynamic mechanisms.
Tumor Microenvironment-Regulating Two-Photon Probe Based on Bimetallic Post-Coordinated MOF Facilitating the Dual-Modal and Deep Imaging-Guided Synergistic Therapies

Abstract:The intricate tumor microenvironment (TME) always brings about unsatisfactory therapeutic effects for treatments, although nanomedicines have been demonstrated to be highly beneficial for synergistic therapies to avoid the side effects caused by the complexity and heterogeneity of cancer. Developing nanotheranostics with the functionalities of both synergistic therapies and TME regulation is a good strategy but is still in its infancy. Herein, an "all-in-one" nanoplatform for integrated diagnosis and treatment, namely, Carrier@ICG@DOX@FA (CIDF), is constructed. Benefiting from the bimetallic coordination of Eu3+-HTHA (4,4,4-trifluoro-1-(9-hexylcarbazol-3-yl)-1,3-butanedione) and Fe3+ with the ligands in UiO-67, CIDF can simultaneously achieve two-photon fluorescence imaging, fluorescent lifetime imaging in deep tumors, and regulation of TME. Owing to its porosity, CIDF can encapsulate indocyanine green as photosensitizers and doxorubicin as chemotherapeutic agent, further realizing light-controlled drug release. Moreover, CIDF exhibited good biocompatibility and tumor targeting by coating with folic-acid-modified polymers. Both in vitro and in vivo experiments demonstrate the excellent therapeutic efficacy of CIDF through dual-modal-imaging-guided synergistic photothermal-, photodynamic-, and chemotherapy. CIDF provides a new paradigm for the construction of TME-regulated synergistic nanotheranostics and realizes the complete elimination of tumors without recurrence.

Abstract:D-Glucosamine (DG) was conjugated to a core-cross linked polymeric micelle (CCPM) system equipped with both a near-infrared fluorophore (NIRF) and a gamma emitter (In-111). The resultant nano-scale tumor-targeting imaging tracer, In-111-DG-NIRF-CCPM, selectively accumulated in a human epithelial carcinoma A-431 xenograft model in mice. At 24 hrs post injection, the tumor uptake was 2.62 +/- 0.80 % of the injected dose per gram of tissue (%ID/g). Tumors were clearly delineated in both single-photon emission computed tomography (SPECT) and optical imaging. The results suggest that the prepared imaging tracer is a promising agent for tumor diagnosis.

Abstract:For photothermal therapy (PTT), the improved targeting can decrease the dosage and promote the therapeutic function of photothermal agents, which would effectively improve the antitumor effect. The tumor microenvironment (TME) and cells are targets in designing intelligent and responsive theranostics. However, most of these schemes have been limited to the traditional visible and first near-infrared (NIR-I) regions, eager to expand to the second near-infrared (NIR-II) window. We designed and synthesized a polyethylene glycol conjugated and disulfide-modified macromolecule fluorophore (MPSS). MPSS could self-assemble into core-shell micelles in an aqueous solution (MPSS-NPS), while the small molecule probes were in a high aggregation arrangement inside the nanoparticle. The pronounced aggregation quenching (ACQ) effect caused them to the "sleeping" state. After entering the tumor cells, the disulfide bonds in MPSS-NPS broke in response to a high concentration of glutathione (GSH) in TME, and the molecule probes were released. The highly aggregated state was effectively alleviated, resulting in distinct absorption enhancement in the near-infrared region. Therefore, the fluorescence signal was recovered, and the photothermal performance was triggered. In vitro and in vivo studies reveal that the Nano-system is efficient for the smart NIR-II fluorescence imaging-guided PTT, even at a low dosage and density of irradiation.
Redox-Activated Light-Up Nanomicelle for Precise Imaging-Guided Cancer Therapy and Real-Time Pharmacokinetic Monitoring

Abstract:Simultaneous tumor imaging, therapy, and pharmacokinetic monitoring can offer a safe and effective strategy for cancer therapy. This work describes the design of a fluorescence light-up nanomicelle that can afford precise imaging-guided drug delivery and pharmacokinetic monitoring in a real-time fashion for cancer chemotherapy. The nanomicelle, which contains a boron dipyrromethene based fluorescent probe as the hydrophobic core and a redox-triggered detachable poly(ethylene glycol) (PEG) shell, can accumulate at the tumor site via enhanced permeation and retention effect. The PEG detachment induced by tumoral and intracellular glutathione can destabilize the nanomicelle, leading to fluorescence light up and simultaneous drug release. Importantly, the fluorescence intensities generated by the nanomicelles in different organs are well-correlated with released drug concentrations in both temporal and spatial manners, suggesting its precise role for imaging-guided drug delivery and pharmacokinetic monitoring in vivo. The tumor growth can be effectively inhibited by the docetaxel-loaded nanomicelle formulation, and the nanomicelles are monitored to be excreted via hepatobiliary routes. This nanomicelle for precise imaging-guided chemotherapy provides a safe and robust theranostic strategy for the evaluation of cancer nanomedicine.
Conjugated Polymer-Based Hybrid Nanoparticles with Two-Photon Excitation and Near-Infrared Emission Features for Fluorescence Bioimaging Within the Biological Window.

Abstract:Hybrid fluorescent nanoparticles (NPs) capable of fluorescing near-infrared (NIR) light (centered ∼730 nm) upon excitation of 800 nm laser light were constructed. A new type of conjugated polymer with two-photon excited fluorescence (TPEF) feature, P-F8-DPSB, was used as the NIR-light harvesting component and the energy donor while a NIR fluorescent dye, DPA-PR-PDI, was used as the energy acceptor and the NIR-light emitting component for the construction of the fluorescent NPs. The hybrid NPs possess δ value up to 2.3 × 10(6) GM per particle upon excitation of 800 nm pulse laser. The excellent two-photon absorption (TPA) property of the conjugated polymer component, together with its high fluorescence quantum yield (ϕ) up to 45% and the efficient energy transfer from the conjugated polymer to NIR-emitting fluorophore with efficiency up to 90%, imparted the hybrid NPs with TPEF-based NIR-input-NIR-output fluorescence imaging ability with penetration depth up to 1200 μm. The practicability of the hybrid NPs for fluorescence imaging in Hela cells was validated.

Abstract:<p>Two-photon supramolecular assembly with near-infrared (NIR) fluorescence emission is constructed from tetraphenylethene derivative possessing methoxyl and vinyl pyridine salt (TPE-2SP), cucurbit[8]uril (CB[8]), and β-cyclodextrin modified hyaluronic acid (HA-CD). The obtained experimental results indicate that the TPE-2SP exhibits a very weak fluorescence emission at 650 nm, and then complexes with cucurbit[7]uril (CB[7]) to form 1:2 supramolecular pseudorotaxane with an enhanced NIR fluorescence emission at 660 nm. Compared with CB[7], CB[8] can assemble with TPE-2SP to be two-axial netlike pseudopolyrotaxane, resulting in close packing to increase TPE-2SP fluorescence emission with a redshift of 30 nm. Interestingly, TPE-2SP/CB[8] continues to assemble with cancer cell targeting agent HA-CD into nanoparticles, leading to assembling-induced further enhancement of NIR emission. Surprisingly, supramolecular nanoparticles have the two-photon character, and are successfully applied to mitochondrial targeting imaging. This supramolecular assembly system, with two-photon absorption and assembly-induced enhanced NIR luminescence properties, opens new way for biological targeted imaging.</p>
