Abstract:Existing contactless solutions on sleep respiration monitoring are either performed in controlled environments, having poor usability in practical scenarios, or only provide coarse-grained respiration rates, being unable to accurately detect abnormal events of patients. In this paper, we propose Respnea, a non-invasive sleep respiration monitoring system using an impulse-radio ultra-wideband (IR-UWB) radar. Particularly, we propose a profiling algorithm, which can locate the sleep positions in non-controlled environments and identify different states of subjects. Further, we construct a deep learning model which adopts a multi-headed self-attention and learn the patterns implicit in the respiration signal so as to distinguish sleep respiration events at a granularity of seconds. We conduct experiments on data collected from patients with sleep disorders and healthy subjects. The experimental results show that Respnea achieves a low error (less than 0.27 bpm) in respiration rate estimation and reaches the accuracy of 88.89% diagnosing the severity of Sleep Apnea-Hypopnea Syndrome.

Abstract:Respiratory rate is a vital physiological signal in laboratory rodents research. This paper presents a noninvasive method based on resonant humidity sensor for monitoring respiratory rate of rats. The sensor is composed of an aluminum nitride (AlN) resonant microcantilever coated with graphene oxide (GO) film. The sensor shows high sensitivity and good repeatability. By combining the sensor with a control circuit, a closed-loop respiration measurement system is realized for fast response. The system can monitor respiratory rate of rats in real time by detecting relative humidity difference between exhaled and inhaled breath. The method has advantages of fast response, small size and low cost. Furthermore, the respiratory rate of rats can be noninvasively monitored by integrating the sensor with a face mask.

Abstract:As a major indicator of Obstructive Sleep Apnea Syndrome(OSAS) in clinical diagnosis, the monitoring of sleep apnea plays an important role in medical treatments of modern society. This paper proposes a portable sleep apnea monitoring system, which is of high-precision and low-power consumption, and capable of performing the long-term monitoring of OSAS patients'multiple physiological parameters in clinical treatments. In the system, the AC modulated detection is adopted, and low amplification ratios are utilized in forestage and a high-resolution AD converter is designed in post-stages. Thus, it is able to acquire, analyze, and process physiological signals in real-time. In addition, ultralow-power chips are used in control system to save the power consumption. The experimental results show that our monitoring system has the strengths of high stability, low-power consumption (peak current <= 90mA), and strong anti-interference ability, which demonstrates the potential in practical applications.
A new ultrasonic method for measuring minute motion activities of rats

Abstract:A new ultrasonic method is presented for measuring the minute motion activities of rats. A pair of low-cost 40 kHz ultrasonic transducers are used to transmit ultrasound toward a rat and receive the ultrasound reflected from the rat. The relative motion of the rat modulates the phase difference between the transmitted and received ultrasound signals. An 8-bit digital phase meter was designed to record the phase difference signal which was used to reconstruct the relative motion waveform of the rat in an 8751 single-chip microcomputer. The reconstructed data are then sent to a PC-AT microcomputer for further processing. This method employs a spectrum analysis for the reconstructed data and can measure three minute motion activities including locomotor activity (LMA), tremor and myoclonia. Finally, the method has been tested with real animal experiments. The main advantages of this new method are that it is non-invasive, non-contact, low cost and high precision. This new method could also be profitably employed for other behavioral studies and offer potential for research in basic medicine.

Meanwhile, wearable technology achieves basic respiratory signal acquisition for low physiological and psychological load; Bluetooth achieves short-distance wireless transmission of respiration data and convenient communication with the handheld terminal such as PDA or smart phone, to ensure the continuous real-time monitoring of the patient.

Abstract:The noninvasive sleep monitoring system was synchronized with the standard electroencephalogram to record sleep time. The results of consistency test proved the feasibility of the noninvasive sleep monitoring system in recording sleep time in different states. Introduction Currently, electroencephalogram (EEG)/electromyogram (EMG) system is widely regarded as the "golden standard" for sleep monitoring. Imperfectly, its invasive monitoring may somehow interfere with the natural state of sleep. Up to now, noninvasive methods for sleep monitoring have developed, which could preserve the undisturbed and naïve sleep state of mice to the greatest extent, but the feasibility of their application under different conditions should be extensive validated. Methods Based on existing research, we verified the feasibility of a sleep monitoring system based on mouse behaviors under different conditions. The experimental mice were exposed to various stresses and placed into a combined device comprising noninvasive sleep monitoring equipment and an EEG/EMG system, and the sleep status was recorded under different physiological, pharmacological, and pathophysiological conditions. The consistency of the parameters obtained from the different systems was calculated using the Bland–Altman statistical method. Results The results demonstrated that the physiological sleep times determined by noninvasive sleep monitoring system were highly consistent with those obtained from the EEG/EMG system, and the coefficients were 94.4% and 95.1% in C57BL/6J and CD‐1 mice, respectively. The noninvasive sleep monitoring system exhibited high sensitivity under the sleep‐promoting effect of diazepam and caffeine‐induced wakefulness, which was indicated by its ability to detect the effect of dosage on sleep times, and accurate determination of the sleep/wakeful status of mice under different pathophysiological conditions. After combining the data obtained from all the mice, the coefficient between the sleep times detected by behavior‐based sleep monitoring system and those obtained from the EEG/EMG equipment was determined to .94. Conclusion The results suggested that behavior‐based sleep monitoring system could accurately evaluate the sleep/wakeful states of mice under different conditions.

Abstract:Active sampling in the olfactory domain is an important aspect of mouse behaviour, and there is increasing evidence that respiration-entrained neural activity outside of the olfactory system sets an important global brain rhythm. It is therefore important to accurately measure breathing during natural behaviours. We develop a new approach to do this in freely moving animals, by implanting a telemetry-based pressure sensor into the right jugular vein, which allows for wireless monitoring of thoracic pressure. After verifying this technique against standard head-fixed respiration measurements, we combined it with EEG and EMG recording and used evolving partial coherence analysis to investigate the relationship between respiration and brain activity across a range of experiments in which the mice could move freely. During voluntary exploration of odours and objects, we found that the association between respiration and cortical delta and theta rhythms decreased, while the association between respiration and cortical alpha rhythm increased. During sleep, however, the presentation of an odour was able to cause a transient increase in sniffing without changing dominant sleep rhythms (delta and theta) in the cortex. Our data align with the emerging idea that the respiration rhythm could act as a synchronising scaffold for specific brain rhythms during wakefulness and exploration, but suggest that respiratory changes are less able to impact brain activity during sleep. Combining wireless respiration monitoring with different types of brain recording across a variety of behaviours will further increase our understanding of the important links between active sampling, passive respiration, and neural activity.

Abstract:The respiration state during overnight sleep is an important indicator of human health. To improve the generalization of the model, we propose a contrastive learning strategy to learn a robust representation of the respiration signals. We deploy our system in hospital and home scenarios and conduct experiments on data from healthy subjects and patients with sleep disorders. The experimental results show that Respnea achieves high temporal coverage and low errors (a median error of 0.27 bpm) in respiration rate estimation and reaches an accuracy of 94.44% on diagnosing the severity of sleep apnea-hypopnea syndrome (SAHS).

Investigation of Beat-to-beat Cardiovascular Activity of Rats by Radio Telemetry.

Abstract:This paper was aimed to apply telemetry technology to investigate the cardiovascular activity of rats. A subminiature radio transducer was implanted in the body of rat to measure beat to beat blood pressure, ECG, body temperature, as well as the activity of rat continuously. Mathematic tools were developed to extract beat to beat cardiovascular parameters. Accordingly, spectral analysis of blood pressure variability and heart rate variability was performed to analyze cardiovascular activity in both normotensive and hypertensive rats. New indices were also established to assess the fluctuation of blood pressure of hypertensive rats and thus the effect of anti-hypertensive medication. The ratio of low frequency power to high frequency power in heart rate variability of normotensive rats was much lower than that of hypertensive rats. In hypertensive rats, the blood pressure variability demonstrated significant increase. Furthermore, the circadian rhythm of blood pressure and heart rate variability also altered in hypertensive rats. After the treatment, the blood pressure of hypertensive rats dropped significantly. Meanwhile, the developed indices, such as 24 h smoothness index were consistent with the behavior of blood pressure satisfactorily. The results suggest that the developed methods are powerful in studying cardiovascular system as well as evaluating the effects of antihypertensive medication objectively.

Abstract:Quality of sleep is an important indicator of health and well being. Recent developments in the field of in-home sleep monitoring have the potential to enhance a person’s sleeping experience and contribute to an overall sense of well being. Existing in-home sleep monitoring devices either fail to provide adequate sleep information or are obtrusive to use. To overcome these obstacles, a noncontact and cost-effective sleep monitoring system, named SleepSense, is proposed for continuous recognition of the sleep status, including on-bed movement, bed exit, and breathing section. SleepSense consists of three parts: a Doppler radar-based sensor, a robust automated radar demodulation module, and a sleep status recognition framework. Herein, several time-domain and frequency-domain features are extracted for the sleep recognition framework. A prototype of SleepSense is presented and evaluated using two sets of experiments. In the short-term controlled experiment, the SleepSense achieves an overall 95.1% accuracy rate in identifying various sleep status. In the 75-minute sleep study, SleepSense demonstrates wide usability in real life. The error rate for breathing rate extraction in this study is only 6.65%. These experimental results indicate that SleepSense is an effective and promising solution for in-home sleep monitoring.

Abstract:Rapid non-invasive monitoring of mouse indicators are important for pharmaceutical and pharmacological research. In this paper, a radio frequency (RF) sensing system that can monitor vital signals and autonomic nervous system (ANS) non-invasively is proposed. The system consists of a software-defined radio (SDR) and a test platform. The operational frequency is 1.1 GHz. We monitored the vital signs of the C57BL/6 mouse which is injected with 1% Pentobarbital sodium. The system obtained its heart rate (HR), breathing rate (BR) and heart rate variability (HRV). And the HRV is further analyzed to represent autonomic nervous system activities.

Abstract:In this study ECG signal of unstrained rat was recorded by telemetry device, and heart rate variability (HRV) was analyzed in order to evaluate 24h autonomic nervous activity. The results demonstrated an obvious circadian rhythm in the autonomic nervous activity: sympathetic activity being dominant during wake phase, and parasympathetic activity, dominant during sleep phase. The ratio of the low frequency to high frequency components in HRV power spectrum (LF/HF) fluctuates with the change in the sleep stages. It is concluded that 24h HRV analyses may reveal plentiful information about the behavior of autonomic nervous system and thus facilitate the investigation of its regulating role in physiological and pathological processes.
RASS: A Portable Real-time Automatic Sleep Scoring System

Abstract:It is a well known fact that the quality of sleep is an important factor in health-related quality of life (HRQoL), and people could prevent potential problems by tracking the quality of their sleep. Unfortunately sleep scoring, which is a systematic way to address the sleep staging as well as the scoring of arousals, respiratory, cardiac, and movement events, is usually conducted with specialized equipment which is expensive and operated by specialists in dedicated sleep centers. Related research studies and products (e.g. ZEO) tried to solve this problem, but they either used multiple probes that cause discomfort to the patient, or could not score in real time. In this paper, we design and implement RASS, a portable Real-time Automatic Sleep Scoring system. RASS only requires one probe, which is inexpensive and, as a result, may be used at home or during travel. RASS accurately scores the sleeping state and detects sleep apnea in real-time based on the sensing results of pulse, blood oxygen, activity, sound and light signals. An alarm will be generated when a severely abnormal sleep state is detected. RASS has been tested with 48 patients, and the test results show that RASS could achieve higher than 84% accuracy.
Simple method for detecting sleep episodes in rats ECoG using machine learning

Abstract:In this paper we propose a new method for the automatic recognition of the state of behavioral sleep (BS) and waking state (WS) in freely moving rats using their electrocorticographic (ECoG) data. Three-channels ECoG signals were recorded from frontal left, frontal right and occipital right cortical areas. We employed a simple artificial neural network (ANN), in which the mean values and standard deviations of ECoG signals from two or three channels were used as inputs for the ANN. Results of wavelet-based recognition of BS/WS in the same data were used to train the ANN and evaluate correctness of our classifier. We tested different combinations of ECoG channels for detecting BS/WS. Our results showed that the accuracy of ANN classification did not depend on ECoG-channel. For any ECoG-channel, networks were trained on one rat and applied to another rat with an accuracy of at least 80~\%. Itis important that we used a very simple network topology to achieve a relatively high accuracy of classification. Our classifier was based on a simple linear combination of input signals with some weights, and these weights could be replaced by the averaged weights of all trained ANNs without decreases in classification accuracy. In all, we introduce a new sleep recognition method that does not require additional network training. It is enough to know the coefficients and the equations suggested in this paper. The proposed method showed very fast performance and simple computations, therefore it could be used in real time experiments. It might be of high demand in preclinical studies in rodents that require vigilance control or monitoring of sleep-wake patterns.

Abstract:Sleep plays a significant role in human health. Along with the emergency of diseases related to sleep, sleep monitoring is becoming a hotspot. Traditionally, Polysomnography (PSG) is considered as the gold standard for sleep monitoring, but it is expensive, time-consuming, and uncomfortable, especially not ideal for long-term sleep studies. Thus, exploring alternative sleep monitoring method has drawn much attention of researchers. Currently most of the sleep monitoring methods require complicated signals and feature extraction process. However, this paper innovatively proposes a method using simple signal of heart rate and respiration rate, and integrating the prior knowledge of experts to simplify the feature extraction process, which can make the subsequent classifier distinguish wake state and sleep state easily. In this paper, we used multi-layer neural network as a classifier, getting a Cohen’s kappa value of 0.739, and the overall accuracy of classifying sleep and wake state reached to 88%. Furthermore, we improved the ability of detecting wake states greatly by oversampling method, and the sensitivity and specificity reached to 91.3% and 82.3% respectively. Nowadays, the development of various bed sensors makes it easy to obtain the heart rate and respiration rate signals of human beings. Therefore, the method proposed in this paper is conducive to people’s long-term sleep monitoring more conveniently and has great significance to people’s health.

Abstract:Wireless sensing technologies have exploited various methods for contactless respiration monitoring. One of the main methods is to use an acoustic beam to cover the chest and abdomen with real-time and high-resolution ranging. The system with high ranging resolution can detect small distance transformations, further affecting the precision of respiratory monitoring. However, the traditional range-based respiration monitoring systems are limited by an insufficient sampling rate and poor synchronization. The ranging resolution is between millimeters and centimeters. In this study, we introduced a novel, high-precision ultrasound-based system for contactless respiration monitoring. First, we developed a convenient and high-performance ultrasonic transceiver platform with universal serial bus audio architecture to eliminate the above limitations. The developed platform provides a foundation for acoustic sensing. Furthermore, based on this platform, we designed a cross correlation sine-frequency modulation continuous wave ranging method and a real-time respiration wave recovery method. The median error in monitoring respiration for 12 participants in three different sleep postures is 0 breaths/min, and the maximum error is 0.58 breaths/min. The proposed system has strong robustness concerning different respiration rates, long-term continuous respiration monitoring, and simulated apnea.

Abstract:Studying the biology of sleep requires accurate and efficient assessment of the sleep stages. However, analysis of sleep-wake cycles in mice and other laboratory animals remains a time-consuming and laborious process. In this study, we developed a Python script and a process for the streamlined analysis of sleep data that includes real-time processing of electroencephalogram (EEG) and electromyogram (EMG) signals that is compatible with commercial sleep-recording software that supports user datagram protocol (UDP) communication. The process consists of EEG/EMG data acquisition, automated threshold calculation for real-time determination of sleep stages, sleep staging and EEG power spectrum analysis. It also allows data storage in the format that facilitates further analysis of the sleep pattern in mice. The described method is aimed at increasing efficiency of sleep stage scoring and analysis in mice thus facilitating sleep research. • A process of EEG/EMG recording and streamline analysis of sleep-wake cycle in real time in mice. • The compatibility with commercial sleep-recording software that can generate a UDP stream. • The capability of further analysis of recorded data by an open-source software.
Unobtrusive Monitoring of Cardiorespiratory Signals During Sleep Based on PVDF Sensor and Singular Spectrum Analysis.

Abstract:Unobtrusive cardiorespiratory monitoring in the period of sleep could facilitate researchers for sleep analysis and sleep disorders diagnosis. With the recent advancement of biomedical health informatics, it is possible to monitor individual's sleep disorders through cardiovascular parameters. This paper proposes a polyvinylidene fluoride ( PVDF) sensor based system for unobtrusive measurements of respiration and heartbeat while sleeping; the sensing unit is deployed on the mattress below the back area of the subject. The PVDF sensor generates the sensory activations due to pulsatile pressure change caused by respiratory and heartbeat movement. To extract and separate the heartbeat and respiration accurately, the method singular spectrum analysis (SSA) was applied while the empirical mode decomposition (EMD) and wavelet analysis methods were used for comparison. Experiments are conducted on two healthy subjects to obtain the respiratory rate and heart rate. To verify and compare the performance of the proposed system, the polysomnography (PSG) system was used to record the thorax effort signal and electrocardiography (ECG) as the gold standard for extracting the respiratory rate and heart rate respectively. The average error rate (AER) of respiratory rate extraction is noted 8.53%, with segmenting the data as 2 mins epochs. Meanwhile, the AERs of the heart rate are 8.01%, 8.80%, 8.55% by utilizing the SSA, EMD and wavelet methods respectively. Overall, the proposed system can obtain accurate respiration and heart rate detection. And the novelty lies in introducing a flexible PVDF sensor which can be combined with the sleep mat for cardiorespiratory signal monitoring in home environment; and new SSA algorithm to extract respiration and heart rates. The experimental results are promising which demonstrated that the proposed system satisfied the basic requirements for cardiorespiratory signal monitoring during sleep.

Abstract:Nocturnal respiratory function assessment is essential for inpatients’ daily healthcare. However, the nasal cannula commonly used in hospitals can affect the patient’s breathing pattern and cause discomfort to the patient. Radar-based contactless detection technique provides a non-contact monitoring way to overcome such limitation. However, to be an alternative to airflow, the radar system requires highly accurate respiratory waveform detection. In this paper, a custom-designed biomedical radar-based internet of things (IoT) system integrated with automatic respiratory variables identification algorithm is developed to provide accurate long-term measurement of respiratory phases and amplitude during sleep. The overnight experimental data from biomedical radar and polysomnography was simultaneously recorded for 10 adults, a total duration of approximately 3851 minutes. Data from adults with suspected sleep apnea were also included. Considering the instability of respiratory activity during REM sleep, we compared the performance of the system’s measurements during REM and non-REM sleep. The results demonstrate the continuous measurement can provide the long-term respiratory variables for sleep analysis. The total experimental measurement reveals a remarkable 97%, 93% and 92% accuracy in respiration-to-respiration interval (RRI), inhale duration (ID) and exhale duration (ED), respectively, which provides accurate and convenient monitoring method and might be a part of medical IoT in the future.

The efficient method to get better raw brain signal on rat anesthetics experiment

Abstract:This paper introduces an efficient methodology for conducting rat anesthesia experiments, aimed at enhancing the quality of raw brain signals obtained. The proposed approach enables the acquisition of animal brain signals during experiments without the confounding influence of muscle noise. Initially, the use of alpha-chloralose (a-c) in conjunction with Isoflurane is introduced to induce anesthesia in rats. Subsequently, Dexdomitor is administered to prevent muscular movements during the collection of brain signals, further refining the signal quality. Experimental outcomes conclusively demonstrate that our anesthesia method produces cleaner raw signals and exhibits improved robustness during data acquisition, outperforming existing methods that rely solely on Isoflurane or the Ketamine-Xylazine combination. Notably, this improved performance is achieved with minimal alterations to vital physiological parameters, including body temperature, respiration, and heart rates. Moreover, the efficacy of a-c in maintaining anesthesia for up to 7 h stands in contrast to the shorter durations achievable with continuous Isoflurane administration or the 30-min window offered by Ketamine-Xylazine, highlighting the practical advantages of our proposed method. Finally, post-experiment observations confirmed that the animals gradually returned to normal behavior without any signs of distress or adverse effects, indicating that our method was both effective and safe.
