Abstract:The base‐ and catalyst‐free reaction of 2‐thiooxoimidazolin‐4‐one with aliphatic, aromatic and heteroaromatic α‐iodomethyl ketones affording monocyclic imidazolidinium iodides was studied for the first time. The interaction of the latter with elemental iodine affords the corresponding triiodides. The cyclization of alkylation products in alcohol is accompanied by alkylation of iminium nitrogen atom with an alkyl radical of the solvent to give imidazo[2,1‐b]thiazolium iodides. The products contain the imidazothiazole structural unit which is a key motif of numerous biologically active compounds. The reaction of 2‐thioxoimidazolidin‐4‐one (thiohydantoin) with aliphatic, aromatic and heteroaromatic α‐iodomethylketones has been investigated in the absence of bases or catalysts. The reaction proceeds as N‐alkylation with subsequent intramolecular dehydrative cyclization. A series of new mono‐ and fused bicyclic iodide and triiodide salts was obtained based on the 5‐oxo‐2‐thioxoimidazolidin‐1‐ium and imidazo[2,1‐b]thiazolium cations. The observed regioselectivity has been rationalized and the mechanism was proposed.
