Abstract:Diblock and miktoarm star-shaped thermoresponsive copolymers composed of single-stranded DNA (ssDNA) and poly(N-isopropylacrylamide) were successfully synthesized with combination of atom transfer radical polymerization (ATRP) and click chemistry. This approach should be generalizable to other DNA-functionalized copolymers. Such copolymers self-assemble into spherical micelles with ssDNA corona in aqueous solution above the lower critical solution temperature. The micellar size can be tuned from the macromolecular architecture. These DNA-encoded micellar particles are able to encapsulate and release hydrophobic guest molecules upon changing temperature.
A Near‐Infrared Light‐Triggered Nanocarrier with Reversible DNA Valves for Intracellular Controlled Release

Abstract:A near-infrared (NIR) light-triggered nanocarrier is developed for intracellular controlled release with good stability, high nuclease resistance, and good biocompatibility. The nanocarrier consists of a gold nanorod core and mesoporous silica shell, capped with reversible single-stranded DNA valves, which are manipulated by switching between the laser on/off states. Upon laser irradiation, the valves of the nanocarrier open and the cargo molecules can be released from the mesopores. When the NIR laser is turned off, the valves close and the nanocarrier stops releasing the cargo molecules. The release amount of the cargo molecules can be controlled precisely by adjusting the irradiation time and the laser on-off cycles. Confocal fluorescence imaging shows that the nanocarrier can be triggered by the laser irradiation and the controlled release can be accomplished in living cells. Moreover, the therapeutic effect toward cancer cells can also be regulated when the chemotherapeutic drug doxorubicin is loaded into the nanocarrier. This novel approach provides an ideal platform for drug delivery by a NIR light-activated mechanism with precise control of area, time, and especially dosage.

Abstract:Novel DNA-gated mesoporous silica nanoparticle (MSN) vehicles functionalized with disulfide-linked acridinamine intercalators are constructed for multi-responsive controlled release. The DNA-gated MSN vehicles release cargo encapsulated in the MSN pores under different stimuli, including disulfide reducing agents, elevated temperature, and deoxyribonuclease I (DNase I), for codelivery of drugs and DNA/genes in different forms. Furthermore, the cascade release of encapsulated and intercalative drugs is controlled by AND logic gates in combination of dual stimuli. The ingeniously designed DNA-gated MSN vehicles integrates multiple responses and AND logic gate operations into a single smart nanodevice not only for codelivery of drugs and DNA/genes but also for cascade release of two drugs and has promising biological applications to meet diverse requirements of controlled release.

Abstract:Stimuli-responsive biocompatible nanocarriers are very promising for controlled drug release. Here, we report redox-and temperature-responsive biocompatible nanocapsules prepared by growing a temperature-responsive biocompatible shell from 3-(trimethoxysilyl) propyl methacrylate-modified silica nanoparticles (SiO2-MPS) via precipitation polymerization, cross-linking the shell with bioreducible chemical linkages, and subsequently removing the silica core by hydrofluoric acid etching. The produced nanocapsules are redox-and temperature-responsive. TEM, FT-IR, TGA, and XPS were used to characterize the nanocapsules. Dynamic light scattering (DLS) was performed to demonstrate the bioreducible and temperature-responsive properties of the nanocapsules. The lower critical solution temperature (LCST) of the nanocapsules can be tuned precisely. Fluorescein isothiocyanate (FITC) was encapsulated into the nanocapsules and used as a model drug molecule to demonstrate the controlled release behavior under external stimuli.

Abstract:Temperature and H2O2 dual-responsive nanoparticles were fabricated from ferrocene modified mesoporous silica (MSN-Fc) and β-cyclodextrin-poly(N-isopropylacrylamide) (β-CD-PNIPAM) star-shaped polymer due to the host-guest interactions for controlled drug release. The formation and structure of β-CD-PNIPAM@MSN-Fc composite nanoparticles was confirmed by FTIR, TGA, TEM and N2 adsorption-desorption isotherms. The size of nanoparticles was about 100-150 nm with well-ordered mesoporous structure and PNIPAM chains coating on the surface as outer shell. The channels of MSNs and hydrophobic cavities of β-CD were all contributed to the high drug loading capacity for nanoparticles. The release of DOX from nanoparticles was enhanced with the increase of temperature above LCST or adding H2O2 in ambient O2. The release kinetics were studied using different models to explain drug release mechanism. Furthermore, the drug loaded composite nanoparticles exhibited excellent anti-cancer activity.

Abstract:Accidental chemotherapy extravasation exacerbates the side effects of anticancer drugs. Therefore, drug-delivery nanocarriers should be designed to avoid persistent drug release at off-target sites and promote burst drug release at on-target. Considering these requirements, poly(allylamine)-co-poly(allylurea) (PAU), a ureido-derivatized temperature responsive polymer with upper critical solution temperature (UCSTs), is an ideal material. This report describes the fabrication, characterization, and in vitro cellular toxicity of PAU polymer-grafted magnetic mesoporous silica nanoparticles as drug-delivery nanocarriers. A UCST of 43 degrees C and an ultranarrow transition temperature range of 39-43 degrees C was realized, ensuring that the nanocarriers suppressed undesirable leakage to below 10 % of the drug loading for 8 h in the absence of a thermal stimulus. A drug release burst of up to 75 % of the drug loading was achieved within 30 min after the stimulus, reducing the viability of the in vitro cancer cells to 12 %. Therefore, the ureido-derivatized polymer is one of the most suitable gatekeepers for drug-delivery nanocarriers.

Abstract:In this study, dual-responsive polymeric nanocapsules, in which the state (swelling or collapse) can be repeatedly controlled by external stimuli (i.e., temperature and light), have been designed and prepared through distillation–precipitation polymerization. Temperature sensitive monomers of N-isopropylacrylamide are cross-linked by photoresponsive bis(methacryloylamino) onto a silica nanospherical template to form a core–shell (SiO2–PNIPAM/Azo) structure. The silica core is then removed by hydrofluoric acid to produce PNIPAM/Azo nanocapsules (P/ANCs) of diameter ∼238 nm at ∼25 °C. The size of the nanocapsule is temperature responsive and, as such, its diameter could be reduced to ∼182 nm on increasing the temperature to 40 °C. In addition, the permeability of nanocapsules can be adjusted by UV irradiation. The cis–trans transformation of modified azobenzene allowed us to perform both the encapsulation and controlled release of molecules. Rhodamine B (RhB) was successfully encapsulated using the photomechanical method. In controlled release experiments, after the majority of RhB (∼45%) was released from the P/ANCs using temperature (∼40 °C) and UV light, a second stage of release could be triggered by lowering the temperature (∼18.4%) and applying UV–visible lighting cycles (∼29.4%), respectively. We found that the diffusion coefficient, D, was 45% larger under alternate irradiation than UV light alone. Our results demonstrate considerable potential for customizable delivery systems for a variety of drugs.

Abstract:The design of an ideal drug delivery system with targeted recognition and zero premature release, especially controlled and specific release that is triggered by an exclusive endogenous stimulus, is a great challenge. A traceable and aptamer-targeted drug nanocarrier has now been developed; the nanocarrier was obtained by capping mesoporous silica-coated quantum dots with a programmable DNA hybrid, and the drug release was controlled by microRNA. Once the nanocarriers had been delivered into HeLa cells by aptamer-mediated recognition and endocytosis, the overexpressed endogenous miR-21 served as an exclusive key to unlock the nanocarriers by competitive hybridization with the DNA hybrid, which led to a sustained lethality of the HeLa cells. If microRNA that is exclusively expressed in specific pathological cell was screened, a combination of chemotherapy and gene therapy should pave the way for a targeted and personalized treatment of human diseases.

Abstract:In this work, we have successfully developed a novel multifunctional near-infrared (NIR)-stimulus controlled drug release system based on gold nanocages as photothermal cores, mesoporous silica shells as supporters to increase the anticancer drug loading and thermally responsive poly(N-isopropylacrylamide) (PNIPAM) as NIR-stimuli gatekeepers (Au-nanocage@mSiO(2)@ PNIPAM). The unique Au-nano-cage@mSiO(2) nanocarrier was elaborately fabricated by utilizing yolk-shell Ag-nanocube@mSiO(2) nanostructure as a template by means of spatially confined galvanic replacement. The Au nanocage cores can effectively absorb and convert light to heat upon irradiation with a NIR laser, resulting in the collapse of the PNIPAM shell covering the exterior of mesoporous silica, and exposes the pores of mesoporous silica shell, realizing the triggered release of entrapped DOX drugs. The in vitro studies have clearly demonstrated the feasibility and advantage of the novel nanocarriers for remote-controlled drug release systems.

Abstract:A method was developed for the preparation of temperature-responsive multifunctional nanoreactor by coating the poly (N-isopropylacrylamide) (PNIPAM) on the external surface of silicallanthanide-POMs/Au spheres. The nanoreactor showed the repeated on/off catalytic activity and switchable luminescence by temperature stimuli. For example, the nanocomplexes showed strong red luminescence in solution and excellent catalytic activity in the reduction of 4-nitrophenol at 25 degrees C. However, when the temperature was 40 degrees C, the red luminescence almost disappeared in solution and catalytic activity was sharply decreased. Moreover, in solutions of K4Fe(CN)(6) containing Na2SO4 at 25 degrees C, it demonstrated the well-defined cyclic voltammograms (CV) peaks with large peak currents, showing the "on" state; at 40 degrees C, the CV response was significantly suppressed, showing the "off" state. The smart nanoreactors have great potential for applications in many fields, such as bio-materials, controlled labeling catalysis, switches and so on. (C) 2017 Elsevier B.V. All rights reserved.

Abstract:Background The coalition of DNA nanotechnology with diversiform inorganic nanoparticles offers powerful tools for the design and construction of stimuli-responsive drug delivery systems with spatiotemporal controllability, but it remains challenging to achieve high-density oligonucleotides modification close to inorganic nanocores for their sensitive responsivity to optical or thermal signals. Results Inspired by Actinia with retractable tentacles, here we design an artificial nano- Actinia consisted of collapsible DNA architectures attached on gold nanoparticle (AuNP) for efficient drug delivery and enhanced photothermal therapy. The collapsible spheroidal architectures are formed by the hybridization of long DNA strand produced in situ through rolling circle amplification with bundling DNA strands, and contain numerous double-helical segments for the intercalative binding of quercetin as the anti-cancer drug. Under 800-nm light irradiation, the photothermal conversion of AuNPs induces intensive localized heating, which unwinds the double helixes and leads to the disassembly of DNA nanospheres on the surface of AuNPs. The consequently released quercetin can inhibit the expression of heat shock protein 27 and decrease the thermal resistance of tumor cells, thus enhancing photothermal therapy efficacy. Conclusions By combining the deformable DNA nanostructures with gold nanocores, this Actinia -mimetic nanocarrier presents a promising tool for the development of DNA-AuNPs complex and opens a new horizon for the stimuli-responsive drug delivery. Abstract:Nanoscale particulate systems have been studied as the delivery vehicle of various drugs and therapeutic agents for decades with promising outcomes. Recently, nano-particulate systems that are responsive to one or more environmental stimuli (such as temperature, pH, and electromagnetic field) are attracting increasing attention because they allow drug delivery and release to be done in a more controllable fashion [1]. The thermally (temperature) responsive nanoparticles are of particular interest to many researchers because the temperature controlled release of the encapsulated drug can be conveniently done with either thermo (using supraphysiologic temperatures) or cryo (using sub-zero temperature) therapies, minimally invasive energy-based surgical techniques that have been widely studied as potential alternatives to radical surgical intervention for the treatment of cancer and other diseases. Moreover, a significantly improved outcome of cancer treatment has been reported by combining thermotherapy (using supraphysiologic temperatures) and anticancer drug encapsulated in thermally responsive nanoparticles [2]. Here, we report thermally responsive nanocapsules that can be combined with cryotherapy for cancer treatment.
Controlled Assemblies of Stimuli-Responsive Mesoporous Silica Drug Delivery Systems for Controlled Release of Drugs

Abstract:There has been an ever increasing interest in developing stimuli-responsive mesoporous silica drug delivery systems to improve therapeutic efficacy and minimize the adverse effects of drugs. This paper reports the works of our research group on valved and gated mesoporous silica drug delivery systems. Biocompatible mesoporous silica nanoparticles (MSNs), as drug carriers, were modified with active group-terminated silanes via self-assembly followed by diverse functionalization, a variety of macrocyclic hosts, proteins, DNA, and quantum dots were bound to the MSN surfaces to develop nanovalves and nanogates, through multiple noncovalent interactions, dynamic covalent bonds, and even strong covalent bonds, for the encapsulation of drugs within MSN pores, thus smart valved and gated MSN drug delivery systems were constructed. Under the stimuli of pH, redox, competitive binding, enzymes, and near infrared lights, controlled release of the encapsulated drugs was realized, because of the destruction of multiple noncovalent interactions, the cleavage of dynamic covalent bonds, and the disassociation of gatekeeping scaffolds. The carboxylate-substituted pillar [6] arene (CPA [6] )-valved dimethylbenzimidazolium or bipyridinium-functionalized MSN drug delivery systems were constructed for acidic pH, competitive binding, and metal chelating-responsive controlled release. The γ -CD-gated MSN delivery system functionalized with disulfide-linked carbamoylphenylboronic acid moieties and amines via dual dynamic covalent bonds with dual drug loading was constructed for simultaneous and cascade release of two drugs. It is a smart strategy to take advantage of the specific structures and properties of cyclodextrins (CDs) for use in the MSN drug delivery systems not only as gatekeepers but also as drug carriers. The γ -CD-gated MSN delivery system provided a smart platform for combination drug therapy, in addition to resistance to serum and normal blood glucose levels. The concanavalin A (Con A)-gated mannose-functionalized MSN drug delivery system via multivalent carbohydrate - protein interactions was constructed for the controlled release of drugs either by acidic pH or by competitive binding of glucose at high concentrations. The long and flexible spacers linked with the mannose ligands played an important role in adjusting the local spatial arrangement of the ligands to favor multivalent protein binding, as did the surface density of the ligands. The MSN drug delivery systems functionalized with N -(3-trimethoxysilylpropyl)ethylenediamine triacetate ligands, in the presence of metal ions with and without myoglobin containing surface-accessible histidine residues, were constructed for pH-responsive controlled release. Both the metal-latching ligands and the metal-chelating proteins played a synergetic role in gating MSNs for high-loading drug delivery and stimuli-responsive controlled release. The DNA-gated MSN drug delivery system functionalized with disulfide-linked acridinamine intercalators was constructed for multi-responsive controlled release under different stimuli, including disulfide reducing agents, elevated temperature, and deoxyribonuclease I. The DNA-gated MSN drug delivery system integrated multiple responses and AND logic gate operations into a single smart nanodevice not only for codelivery of drugs and DNA/genes but also for cascade release of two drugs in combination of dual stimuli. The DNA-gated gold nanorod-embedded MSN delivery system functionalized with titanium(IV)-chelating phosphonates with dual drug loading was constructed for simultaneous and cascade release of two drugs. Coordination chemistry is the first strategy for DNA cappings through multivalent chelating interactions in drug delivery systems not only as gatekeepers but also as drug carriers. The two drugs were simultaneously released upon triggering of endonuclease degradation or photothermal dehybridization and were successively released upon first triggering of basic pH and subsequent triggering of photothermal heating. The combination of NIR light-based thermotherapy and triggered chemotherapy (thermo-chemotherapy) could maximize therapeutic efficacy. In addition, the ZnO quantum dot (QD)-gated hollow mesoporous silica drug delivery system was constructed for pH and redox- responsive controlled release, and the ZnO QD-gated mesoporous carbon nanoparticle (MCN) drug delivery system was for pH-responsive controlled release. These constructed stimuli-responsive MSN drug delivery systems have promising applications in targeted tumor therapy.

Abstract:The fabrication of well-behaved drug delivery systems that can transport drugs to specifically treat cancer cells rather than normal cells is still a tremendous challenge. A novel drug delivery system with two types of tumor-related mRNAs as "keys" to open the multiple valves of the nanocarrier to control drug release was developed. Hollow mesoporous silica nanoparticles were employed as the nanocarrier and dual DNAs targeting two intracellular mRNAs were employed as "multi-locks" to lock up the nanocarrier. When the nanocarrier enters the cancer cells, the overexpressed endogenous mRNA keys hybridize with the DNA multi-locks to open the valves and release the drug. Each single mRNA could not trigger the opening of the locks to release the cargo. Therefore, the nanocarrier can be applied for specific chemotherapy against cancer cells with minor side effects to normal cells. The current strategy could provide an important avenue towards advancing the practical applications of drug delivery systems used for cancer therapy.

Abstract:A controlled drug-delivery system has been developed based on mesoporous silica nanoparticles that deliver anticancer drugs into cancer cells with minimized side effects. The copolymer of two oligo(ethylene glycol) macromonomers cross-linked by the disulfide linker N,N'-bis(acryloyl)cystamine is used to cap hollow mesoporous silica nanoparticles (HMSNs) to form a core/shell structure. The HMSN core is applied as a drug storage unit for its high drug loading capability, whereas the polymer shell is employed as a switch owing to its redox/temperature dual responses. The release behavior in vitro of doxorubicin demonstrated that the loaded drugs could be released rapidly at higher temperature or in the presence of glutathione (GSH). Thus, the dual-stimulus polymer shell exhibiting a volume phase transition temperature higher than 37 °C can effectively avoid drug leakage in the bloodstream owing to the swollen state of the shell. Once internalized into cells, the carriers shed the polymer shell because of cleavage of the disulfide bonds by GSH, which results in the release of the loaded drugs in cytosol. This work may prove to be a significant development in on-demand drug release systems for cancer therapy.

Abstract:Stimuli-responsive nanofl uidic systems in room temperature ionic liquids ( RTILs). The nanofl uidic device can withstand high temperatures up to 200 degrees C, in which conventional water-based smart materials and nanodevices are invalid. The smart nanopores can be "irreversibly" turned off above the transition temperature of ca. 120-150 degrees C, actuated by the conformational change of the chemicallymodifi ed polymer brushes.

Abstract:A novel nanocontainer, which could regulate the release of payloads, has been successfully fabricated by attaching zwitterionic sulfobetaine copolymer onto the mesoporous silica nanoparticles (MSNs). RAFT polymerization is employed to prepare the hybrid poly(2-(dimethylamino)ethyl methacrylate)-coated MSNs (MSN-PDMAEMA). Subsequently, the tertiary amine groups in PDMAEMA are quaternized with 1,3-propanesultone to get poly(DMAEMA-co-3-dimethyl(methacryloyloxyethyl)ammonium propanesulfonate)-coated MSNs [MSN-Poly(DMAEMA-co-DMAPS)]. The zwitterionic PDMAPS component endows the nanocarrier with biocompatibility, and the PDMAEMA component makes the copolymer shell temperature-responsive. Controlled release of loaded rhodamine B has been achieved in the saline solutions.
