Abstract:To investigate the prevalence and the mechanism of quinolone-resistant Acinetobacter pittii, 634 Acinetobacter calcoaceticus-Acinetobacter baumannii complex isolates were collected throughout Zhejiang Province. Identification of isolates was conducted by matrix-assisted laser desorption ionization/time of flight mass spectrometry (MALDI-TOF MS), bla(OXA-51)-like gene, and partial RNA polymerase -subunit (rpoB) amplification. Twenty-seven isolates of A. pittii were identified. Among the 634 isolates, A. baumannii, A. pittii, Acinetobacter nosocomialis, and A. calcoaceticus counted for 87.22%, 4.26%, 8.20%, and 0.32%, respectively. Antimicrobial susceptibility of nalidixic acid, ofloxacin, enoxacin, ciprofloxacin, lomefloxacin, levofloxacin, sparfloxacin, moxifloxacin, and gatifloxacin for 27 A. pittii were determined by the agar dilution method. Detection of quinolone-resistant determining regions of gyrA, gyrB, parC, and parE was performed for the A. pittii isolates. In addition, plasmid-mediated quinolone resistance (PMQR) determinants (qnrA, qnrB, qnrS, qnrC, qnrD, aac(6)-Ib-cr, qepA, oqxA, and oqxB) were investigated. All the 27 isolates demonstrated a higher minimum inhibitory concentration (MIC) to old quinolones than the new fluoroquinolones. No mutation in gyrA, gyrB, parC, or parE was detected in 20 ciprofloxacin-susceptible isolates. Seven ciprofloxacin-resistant A. pittii were identified with a Ser83Leu mutation in GyrA. Among them, six isolates with simultaneous Ser83Leu amino acid substitution in GyrA and Ser80Leu in ParC displayed higher MIC values against ciprofloxacin. Additionally, three were identified with a Met370Ile substitution in ParE, and two were detected with a Tyr317His mutation in ParE, which were reported for the first time. No PMQR determinants were identified in the 27 A. pittii isolates. In conclusion, mutations in chromosome play a major role in quinolone resistance in A. pittii, while resistance mechanisms mediated by plasmid have not been found. Ser83Leu substitution in GyrA and Ser80Leu substitution in ParC are associated with quinolone resistance in A. pittii. Whether Met370Ile and Tyr317His substitutions in ParE play a minor role requires further investigation.
Fluoroquinolone susceptibility and quinolone resistance determining region mutations in Streptococcus pneumoniae isolates from Shanghai

Abstract:Objective: To investigate the antimicrobial susceptibility of S. pneumoniae isolates from Shanghai to fluoroquinolones and other antibiotics, and explore the quinolone resistance determining region (QRDR) mutations in fluoroquinolone-susceptible S. pneumoniae. Methods: Susceptibility testing was performed for 176 S. pneumoniae isolates collected in Shanghai during 2004-2005 by agar dilution method as described by Clinical and Laboratory Standards Institute (CLSI). Selected levofloxacin-susceptible S. pneumoniae isolates were analyzed by PCR and sequencing to understand the mutations in QRDR, including parC, parE, gyrA and gyrB genes. Results: Of the 176 clinical S. pneumoniae isolates, 48.9%, 47.1% and 4.0% were identified as penicillin-susceptible (PSSP), penicillin-intermediate (PISP) and penicillin-resistant (PRSP), respectively. All the isolates were susceptible to ciprofloxacin, levofloxacin, gatifloxacin and moxifloxacin. Of the 20 levofloxacin-susceptible isolates with levofloxacin MIC 1-2 mg/L, 19 showed ParE: Ile460 → Val. Some strains had additional substitution of ParE: Asp435→Asn/Asn 477→Lys and/or ParC: Phe 105→Leu/ Asp 136→Tyr. Only point mutation was found in gyrA and gyrB genes. None of the isolates showed amino acid substitution in GyrA or GyrB. Conclusions: The prevalence of penicillin nonsusceptible S. pneumoniae is high in Shanghai. No S. pneumoniae isolate was found resistant to fluoroquinolones, but first-step QRDR mutations (in parE or parC genes) are identified in levofloxacin-susceptible isolates (MIC 1-2 mg/L).
On the Resistance of Escherichia Coli Strains Isolated from Healthy People's Intestines to Fluroquinolones and Its Mechanism

Abstract:Objective To investigate the resistance of 110 strains of Escherichia Coli isolated from healthy people's intestines to 4 kinds of fluroquinolones and its mechanism.Methods Minimal inhibitory concentrations of these 4 kinds of fluroquinolones to Escherichia Coli strains were determined by agar dilution method.gyrA and parC genes of Escherichia Coli were amplified by polymerase chain reaction,of which products were then sequenced to detect mutations and compared with those of patients who didn't take antibacterial in last two weeks.Results Resistance rates of these 4 kinds of fluroquinolones to Escherichia Coli strains ranged from 12.7% to 15.5% in patients without taking antibacterial,and ranged from 20.8% to 25.0% in patients who took antibacterial.No mutations were found in the susceptible strain(No.60).Ser83leu mutation was detected in gryA gene of all 11 resistant strains,Asp87Asn also detected in 8 strains,three mutations including Glu214Gly found in 6 strains.Ser80Ile mutation was found in parC gene of 8 resistant strains;Glu84Gly was also detected in 2 strains.Conclusions Resistance rates of Escherichia Coli strains isolated from healthy people's intestines to these 4 kinds of fluroquinolones range from 12.7% to 15.5%.The resistance is mainly caused by point mutations of gyrA and parC genes,and gyrA gene is the most important target of fluroquinolones.
A study on GyrA and ParC mutant in fluoroquinolone-resistant clinical isolates of Klebsiella pneumoniae

Abstract:Objective To study the relation between the alterations in the DNA gyrase subunit A (GyrA), topoisomerase Ⅳ subunit C (ParC) and fluoroquinolones(FQNL) resistance in Klebsiella pneumoniae . Methods The minimal inhibitory concentrations (MIC) of 31 clinically isolated strains and reference strain (ATCC10031) of Klebsiella pneumoniae to ciproxacin, levofloxacin, enoxacin, and norfloxacin were determined by the two fold agar dilution method.The genes of GyrA (gyrA) and ParC (parC) in the 32 strains were amplified by PCR and their DNA sequences were compared. Results The alterations in GyrA existed in all 19 strains of Klebsiella pneumoniae resistant to FQNL. In this study, the alterations related to FQNL resistance included Ser83 (TCC)→ Phe (TTC), Ile (ATC), Tyr (TAC) and Lys (TGC); Asp87 (GAC)→ Ala(GCC). The alterations of Ser83(TCC)→ Ile (ATC) and Lys (TGC) were new discoveries. Alteration in ParC existed in all 9 strains which were highly resistant to FQNL and one strain was moderately resistant to FQNL, and the alteration related to FQNL resistance was Ser80(AGC)→ Ile (ATC). When alteration occurred in GyrA accompanied by that in ParC, the FQNL resistance of bacteria tended to increase. It was discovered in this study that the alteration related to FQNL resistance was Ser80(AGC)→ Ile(ATC). The ParC gene in ATCC10031 was highly homologous to that in Escherichia coli. Conclusions The alterations in GyrA exist commonly in the strains of Klebsiella pneumoniae resistant to FQNL. The alteration at Ser83(TCC) is common. When alteration occurs in GyrA companied by that in ParC, the FQNL resistance of bacteria tends to intensify.

Abstract:Objective To investigate the resistance-related genes of Shigella sonnei with decreased susceptibility to fluoroquinolones.Methods A total of 131 strains of Shigella sonnei were analyzed for their antimicrobial susceptibility.Mutations within the quinolone resistance determining regions (QRDR) of gyrA and parC were detected by polymerase chain reaction (PCR) and PCR products were then sequenced. Meanwhile, the plasmid-mediated quinolone resistance (PMQR) genes,qnr and aac(6')-Ib-cr were screened by PCR.Results Resistance rates of 131 Shigella sonnei isolates to nalidixic acid,tetracycline,ampicillin and trimethoprim-sulfamethoxazole were 100.0％,93.9％,93.2％ and 92.8％,respectively.All strains were susceptible to norfloxacin,ciprofloxacin,levofloxacin,while 94％ nalidixic acid-resistant Shigella sonnei strains showed reduced susceptibilities to fluoroquinolones.All of nalidixic acid-resistant Shigella sonnei strains presented a single mutation at codon 83 (Ser→Leu) of gyrA genes,but no mutations were detected in parC gene.And PMQR genes qnr and aac (6’)-Ib-cr were not detected.Conclusions The nalidixic acid-resistant Shigella sonnei strains with reduced susceptibility to fluoroquinolones are common in the clinical practice,which may mainly due to a single mutation at codon 83 (Ser→Leu) of gyrA genes.

Abstract:Objective To investigate the mechanism of quinolone resistance in Psendomonas aeruginosa.Methods The minimum inhibitory concentrations (MICs)of ciprofloxacin and levofloxacin with and without carbonylcyainde-m-chlorophenylhydrazone(CCCP)were determined by agar dilution method.Polymerase chain reaction(PCR)and DNA sequencing were used to study the mutations in quinolone resistance-determining region of gyrA and parC genes.The strains were genotyped by enterbacterial repetitive intergenie consensus-PCR(ERIC-PCR).Results Sixteen quinolones-resistant Pseudomonas aeruginosa strains were obtained.The MICs of ciprofloxacin and levofloxacin were not reduced significantly by adding CCCP.Thr-83→Ile of gyrA and Ser-87→Leu of parC were found simultaneously in 16 strains of resistant Pseudomonas aeruginosa.Analysis of ERIC-PCR products indicated that 16 quinolone-resistant strains had an identical band pattern which was different from that seen in the sensitive strains.Conclusion Mutations in gyrA and parC may be the main mechanism of quinolone resistance in clinical isolates of Pseudomonas aeruginosa.

Abstract:Antibiotic resistance is a global concern; however, data on antibioticresistant Ureaplasma spp. and Mycoplasma hominis are limited in comparison to similar data on other microbes. A total of 492 Ureaplasma spp. and 13 M. hominis strains obtained in Hangzhou, China, in 2018 were subjected to antimicrobial susceptibility testing for levofloxacin, moxifloxacin, erythromycin, clindamycin, and doxycycline using the broth microdilution method. The mechanisms underlying quinolone and macrolide resistance were determined. Meanwhile, a model of the topoisomerase IV complex bound to levofloxacin in wild-type Ureaplasma spp. was built to study the quinolone resistance mutations. For Ureaplasma spp., the levofloxacin, moxifloxacin, and erythromycin resistance rates were 84.69%, 51.44%, and 3.59% in U. parvum and 82.43%, 62.16%, and 5.40% in U. urealyticum, respectively. Of the 13 M. hominis strains, 11 were resistant to both levofloxacin and moxifloxacin, and five strains showed clindamycin resistance. ParC S83L was the most prevalent mutation in levofloxacin-resistant Ureaplasma strains, followed by ParE R448K. The two mutations GyrA S153L and ParC S91I were commonly identified in quinolone-resistant M. hominis. A molecular dynamics-refined structure revealed that quinolone resistance-associated mutations inhibited the interaction and reduced affinity with gyrase or topoisomerase IV and quinolones. The novel mutations S21A in the L4 protein and G2654T and T2245C in 23S rRNA and the ermB gene were identified in erythromycin-resistant Ureaplasma spp. As fluoroquinolone resistance in Ureaplasma spp. and Mycoplasma hominis remains high in China, the rational use of antibiotics needs to be further enhanced.
Fluoroquinolone resistance mechanism of clinical isolates and selected mutants of Pasteurella multocida from bovine respiratory disease in China

Abstract:The minimum inhibitory concentrations (MICs), mutation prevention concentrations (MPCs) and contribution of quinolone resistance-determining region (QRDR) mutations to fluoroquinolone (ciprofloxacin, enrofloxacin and orbifloxacin) susceptibility in 23 Pasteurella multocida (Pm) isolates were investigated. Fluoroquinolone-susceptible isolates (MICs ≤0.25 µg/ml, 9 isolates) had no QRDR mutations, and their respective MPCs were low. Fluoroquinolone-intermediate isolates (MICs=0.5 µg/ml, 14 isolates) had QRDR mutations (Asp87 to Asn or Ala84 to Pro in gyrA), and their respective MPCs were high (4-32 µg/ml). First-step mutants (n=5) and laboratory-derived highly resistant fluoroquinolone mutants (n=5) also had QRDR mutations. The MICs of fluoroquinolones for mutant-derived strains were decreased in the presence of efflux inhibitors. The results indicated that the fluoroquinolone resistance of Pm is mainly due to multiple target gene mutations in gyrA and parC and the overexpression of efflux pump genes.
Relationship Between Fluoroquinolone Resistance and Gene Mutation in Streptococcus Suis

Abstract:To detect the relationship between fluoroquinolone resistance and gyrA and parC gene mutation in Streptococcus suis.The MIC of 34 strains of Streptococcus suis were determined by microdilution.The genes encoding the quinolone-resistance determining region(QRDRs) of parC and gyrA in fluoroquinolone-susceptible and-resistant Streptococcus suis clinical isolates were identified and sequenced.Ser to Phe and Arg to Leu mutation at position 79 and 87 of the parC gene was detected in fluoroquinolone resistance S.suis,and Arg to Ser,or Ser to Arg mutations at the position 66 or 81 of gyrA gene were detected in 4 highly resistance strains;no amino acid changes in gyrA or parC were detected for 9 fluoroquinolone-susceptible strains,the mutations in both genes was found in the strains with MIC of fluoroquinolone higher than 32μg·L-1.Mutations in parC gene result in low level resistance against fluoroquinolone,and high level resistance is resulted from the mutations in gyrA and parC both genes.

Abstract:There are many studies on fluoroquinolone (FQ) resistance in the literature, but little is known regarding the molecular characterisation of FQ-resistant Haemophilus parasuis. In this study, a total of 138 H. parasuis isolates were examined, among which 83 (60.1%) were resistant to enrofloxacin (EFX) and 8 (5.8%) were resistant to levofloxacin (LFX) as determined by Etest. Ten point mutations in the quinolone resistance-determining regions of gyrA, gyrB, parC and parE were detected by PCR and DNA sequencing. Interestingly, 100% of the resistant isolates contained mutations at 87D of gyrA, but other mutations occurred less frequently. Furthermore, it was found that there was synergy between 73S→R/I in parC and other point mutations with respect to FQ resistance. To examine the effect of different point mutations on FQ resistance, the minimum inhibitory concentrations of EFX and LFX were determined for strains generated by site-directed mutagenesis, and three point mutations (gyrA 87D→N, parC 73S→R and parE 551T→A) were shown to be involved in FQ resistance.
Mutations in Gyrase and Topoisomerase Genes Associated with Fluoroquinolone Resistance in Salmonella Serovars from Retail Meats

Abstract:Mutations in gyrase and topoisomerase genes (gyrA, gyrB, parC and parE) were examined among 30 multidrug-resistant (MDR) Salmonella isolates recovered from retail meats using PCR and DNA sequencing analysis. Six isolates had a single gyrA mutation (Ser83→Phe or Ser83→Tyr or Asp87→Asn) with ciprofloxacin (Cip) minimum inhibitory concentrations (MICs) ranging from 0.125 to 0.5μg/ml except 1 with 16μg/ml. Three isolates had both a gyrA mutation (Ser83→Tyr or Ser83→Phe) and a parC mutation (Ser80→Arg) (Cip MICs, 8 to 16μg/ml). Fifteen isolates had both double mutations in gyrA (Ser83→Phe and Asp87→Gly or Asp87→Asn) and a single mutation (Ser80→Arg) in parC (Cip MICs, 8 to ≥16μg/ml). Three had double gyrA mutations (Ser83→Phe, Asp87→Gly or Asp87→Asn), one parC mutation (Ser80→Arg), and 1 parE mutation (Lys428→Gln or Gly442→Ser) (Cip MICs, 8 to ≥16μg/ml). One had double gyrA mutations (Ser83→Phe, Asp87→Gly or Asp87→Asn), one parC mutation (Ser80→Arg), and 2 parE mutations (Lys441→Ile and Asp494→Asn) (Cip MICs, 8 to ≥16μg/ml). Isolates with single gyrA mutations were less resistant to quinolone and fluoroquinolones than those together with additional parC and/or parE mutations in Salmonella. Our findings confirmed the importance of point mutations in gyrase and topoisomerase in reduced susceptibility to quinolone and fluoroquinolones. Mutations in gyrA confer low-level quinolone and fluoroquinolone resistance, while additional gyrA mutation(s) together with parC and/or parE mutations increase the resistance to a high level.
Mutations of Gyra and Parc in Fluoroquinolones-Resistant Escherichia Coli Strains Isolated from Animals, Farmers and Environment

Abstract:【Objective】To detect mutations of gyrA and parC in 72 Escherichia coli strains isolated from human feces,animal feces and the environment.【Method】MICs of ciprofloxacin or enrofloxacin for 72 isolates were determined by an agar dilution test with standard strain ATCC25922 as the control.The mutations in the quinolone resistance-determining regions(QRDRs) of the gyrA and parC genes were analyzed by PCR and sequencing.【Result】Amino acid substitutions(number of strains) found in 63 mutants were Ser83→Leu(62)and Asp87→Asn(52),Asp87→Tyr(2),Asp87→His(2)in GyrA and Ser80→Ile(47),Ser80→Arg(2)and Glu84→Val(3),Glu84→Lys(4),Glu84→Gly(5),Glu84→Ala(1)in ParC.No amino acid changes in GyrA or ParC were detected when strains MIC of ciprofloxacin were under 0.125 μg.ml-1.A single substitution in GyrA was found in the strains whose MIC of ciprofloxacin ranged from 0.125 to 0.25μg.ml-1.Double substitutions in GyrA or one in GyrA with one in ParC were found in the strains with MICof ciprofloxacin ranging from 0.5 to 32 μg.ml-1.A double changes in GyrA and a single change in ParC were found in strains with MIC of ciprofloxacin ranging from 4 to 128 μg.ml-1.Double changes in GyrA and double changes in ParC were found in strains with MIC of ciprofloxacin ranging from 16 to 128 μg.ml-1.【Conclusion】A variety of amino acid changes in GyrA and ParC were detected in the fluoroquinolone-resistant E.coli isolates from different origin.A correlation was found between the number of changes in the QRDR of GyrA and ParC and the resistance level to fluoroquinolone in E.coli strains.
Frequent topoisomerase IV mutations associated with fluoroquinolone resistance in Ureaplasma species

Abstract:This study aimed to investigate the role of quinolone resistance-determining regions (ORDRs) of DNA gyrase (encoded by gyrA and gyrB) and topoisomerase IV (encoded by parC and parE) associated with fluoroquinolone resistance. A total of 114 Ureaplasma spp. strains, isolated from clinical female patients with symptomatic infection, were tested for species distribution and susceptibility to four fluoroquinolones. Moreover, we analysed the QRDRs and compared these with 14 ATCC reference strains of Ureaplasma spp. serovars to identify mutations that caused antimicrobial resistance. Our study indicated that moxifloxacin was the most effective fluoroquinolone against Ureaplasma spp. (MIC range: 0.125-32 mu g ml(-1)). However, extremely high MICs were estimated for ciprofloxacin (MIC range: 1-256 mu g ml(-1)) and ofloxacin (MIC range: 0.5-128 mu g ml(-1)), followed by levofloxacin (MIC range: 0.5-64 mu g ml(-1)). Seven amino acid substitutions were discovered in GyrB, ParC and ParE, but not in GyrA. Ser-83 -> Leu/Trp (C248T/G) in ParC and Arg-448 -> Lys (G1343A) in ParE, which were potentially responsible for fluoroquinolone resistance, were observed in 89 (77.2%) and three (2.6%) strains, respectively. Pro-462 -> Ser (C13841), Asn-481 -> Ser (A1442G) and Ala-493 -> Val (014781) in GyrB and Met-105 -> Ile (G315T) in ParC seemed to be neutral polymorphisms, and were observed and occurred along with the amino acid change of Ser-83 -> Leu (C2481) in ParC. Interestingly, two novel mutations of ParC and ParE were independently found in four strains. These observations suggest that amino acid mutation in topoisomerase IV appears to be the leading cause of fluoroquinolone resistance, especially the mutation of Ser-83 -> Leu (02481) in ParC. Moxifloxacin had the best activity against strains with Ser-83 -> Leu mutation.
Mutation Analysis of Induced Fluoroquinolone-Resistant Mutants of Streptococcus Suis in Vitro

Abstract:To perform a systematic analysis of point mutations in the quinolone resistance determining regions(QRDRs)of the DNA gyrase and topoisomerase genes of Streptococcus suis type 2 strain after in vitro exposure to stepwise increasing concentrations of enrofloxacin and ciprofloxacin.Four parent strains of S.suis type 2 were chosen for stepwise exposure to increasing levels of enrofloxacin and ciprofloxacin followed by selection of resistant mutants.The QRDRs of gyrA,gyrB,parC and parE correlating to mutants with increased MICs were analysed for point mutations.Multiple mutants with significantly increased MICs(128 mg·L-1)were generated from each parent strain.Most of the amino acid substitutions identified were Ser79Phe of ParC,Ser81Arg of GyrA,Asp315Asn,Ser285Leu,Glu354Lys of GryB and Pro278Ser of ParE,which were consistent to the mechanisms of resistance reported in clinical isolates of S.pneumoniae.However,it has some mutable points Gln118His of GyrA,Asn297Tyr of ParE as well as the deletions 288-291 amino acids of GyrB and 62 amino acid of ParC,which haven′t been documented and whether these nuleotide mutations associated with fluoroquinolone resistance are not clear.The number of induction/selection cycles required for the emergence of key point mutations in gyrA and parC was variable among strains.The results indicate that stepwise increasing concentration of enrofloxacin and ciprofloxacin can easily select resistant strains of S.suis.
Distribution of Fluoroquinolone Resistance Determinants in Carbapenem-resistant Klebsiella Pneumoniae Clinical Isolates Associated with Bloodstream Infections in China

Abstract:BackgroundThe rate of fluoroquinolone (FQ) resistance among carbapenem-resistant Klebsiella pneumoniae (CRKP) is high. The present study aimed to investigate the distribution of fluoroquinolone resistance determinants in clinical CRKP isolates associated with bloodstream infections (BSIs).ResultsA total of 149 BSI-associated clinical CRKP isolates collected from 11 Chinese teaching hospitals from 2015 to 2018 were investigated for the prevalence of fluoroquinolone resistance determinants, including plasmid-mediated quinolone resistance (PMQR) genes and spontaneous mutations in the quinolone resistance-determining regions (QRDRs) of the gyrA and parC genes. Among these 149 clinical CRKP isolates, 117 (78.5%) exhibited resistance to ciprofloxacin. The GyrA substitutions (Ser83 -> IIe/Phe) and (Asp87 -> Gly/Ala) were found among 112 (75.2%) of 149 isolates, while the substitution (Ser80 -> IIe) of ParC was found in 111 (74.5%) of the 149 isolates. In total, 70.5% (105/149) of the CRKP isolates had at least two mutations within gyrA as well as a third mutation in parC. No mutations in the QRDRs were found in 31 ciprofloxacin susceptible CRKP isolates. Eighty-nine (56.9%) of 149 were found to carry PMQR genes including qnrS1 (43.0%), aac(6)-Ib-cr (16.1%), qnrB4 (6.0%), qnrB2 (2.7%), and qnrB1 (1.3%). Nine isolates contained two or more PMQR genes, with one carrying four [aac(6 ')-Ib-cr, qnr-S1, qnrB2, and qnrB4]. The co-existence rate of PMQR determinants and mutations in the QRDRs of gyrA and parC reached 68.5% (61/89). Seventy-four (83.1%, 74/89) PMQR-positive isolates harbored extended-spectrum beta-lactamase (ESBL)-encoding genes. Multilocus sequence typing (MLST) analysis demonstrated that the ST11 was the most prevalent STs in our study.Conclusions Mutations in the QRDRs of gyrA and parC were the key factors leading to the high prevalence of fluoroquinolone resistance among BSI-associated CRKP. The co-existence of PMQR genes and mutations in the QRDRs can increase the resistance level of CRKP to fluoroquinolones in clinical settings. ST11 CRKP isolates with identical QRDR substitution patterns were found throughout hospitals in China.
Gene Mutation Mechanism of Resistance to Fluoroquinolone in E.coli

Abstract:Chromosome DNAs from 8 strains of Escherichia coli were extracted,among which 4 strains were collected from clinical samples,2 strains were got by induction with different concentration of quinolone and 1 susceptible strain.The fragments of gyrA and parC genes of the 8 strains were amplified by PCR,cloned and sequenced.Mutations were found at residues 83 and 87 in gyrA gene and 80 and 84 in parC gene in all the quinolone-resistant strains,while no mutations were found in the susceptible strain.Only one mutation was found in gyrA gene in 2 low resistant strains,which resulted in Ser83→Leu or Asp87→Asn,and no mutation was found in parC gene.Double mutations were found in both gyrA and parC genes in 5 high resistant strains.Mutations in gyrA gene resulted in Ser83→Leu(n=5) and Asp87→Asn(n=4) or Tyr(n=1),and in parC resulted in Ser83→Ile(n=4) and Glu84→Lys(n=1).The mutations are identical with data from the published references.The results revealed that mutations of gyrA and parC genes are involved in the quinolone-resistant mechanism of Escherichia coli.And the low-resistant Escherichia coli exist only single mutation in gyrA gene,the high resistant Escherichia coli exist mutations in both parC and gyrA genes at the same time.
Study on Fluoroquinolone Susceptibility and Resistance Mechanism in Clinical Isolates of Neisseria Gonorrhoeae

Abstract:[Objective] To evaluate the situation of Fluroquinolone resistance in Neisscria Gonon-hoeae in Changsha,P.R.China and to investigate the molecular mechanisms of Fluoroquinolone resistance in clinical isolates of Neisseria Gonorrhoeae.[Methods] The susceptibilities of 126 clinical isolates of N.gonorrhoeae to 4 Fluoroquinolones were tested by disc diffusion method.The minimum inhibitory concentration (MIC) of Ciprofloxacin was determined by E-test method in 63 strains.The fragments including quinolone resistance-determlning region (QRDR) were amplified by PCR in gyrA and parC genes,and their relative fragments were direcdy sequenced.[Resuits] The percentages of resistance to Ofloxacin,Fleroxacin,Lomefloxacin,and Enoxacin were 70.6%,81.7%,72.2% and 82.5% respectively.Range of ciprofloxacin MIC was 0.004-12.0 μg/mL in 63 strains.Percentage of resistance was 65.1%.Ciprofloxacin-susceptible strains contained no mutation in gyrA and parC genes,while intermediate or resistant strains contained mutations in gyrA gene,and some contained mutations in parC gene simultaneously.All parC gene mutations were found on the basis of mutations in gyrA gene.These amino substitutions included Ser91→Tyr,Ser91→Phe,Asp95 →Gly,Asp95→Ala,Asp95→Asn in GyrA and Gly85→Cys,Asp86→Asn,Ser87→Arg,Ser87→Ile,Ser87→Asn,Ser88→Pro,Glu91→Lys,Glu91→Gly in ParC.Of all the mutations in gyrA gene,the percentage of substitution of Ser91→Phe was the highest.[Conclusions] Fluorcquinolone resistance in N.gonorrhoeae in Changsha,P.R.China is very severe at present.Fluoroquinolone resistance in N.gonorrhoeae has close relationship with mutations in gyrA and parC genes.Substitution of Ser91→Phe in gyrA gene is the pivotal mutation resulting in fluoroquinolone resistance in N.gonorrhoeae.The cooperation of multi-point mutations makes the resistance stronger.
Fluoroquinolone Resistance and Mutation Patterns in Gyra and Parc Genes in Neisseria Gonorrhoeae Isolates from Shanghai, China

Abstract:In order to study the resistance of Neisseria (N.) gonorrhoeae to the fluoroquinolone and detect mutation patterns of quinolone resistance-determining regions (QRDRs) of clinical isolates in Shanghai, China, a total of 80 clinical isolates of N. gonorrhoeae were consecutively collected from Shanghai. The MIC of fluoroquinolone for the isolates was examined by using the agar dilution method and the mutation profiles of the QRDRs of gyrA and parC were analyzed by sequencing and restriction fragment length polymorphism (RFLP). Chi-square test was used for comparison of the mutation patterns. The results showed that: (1) High percentages of the 8 isolates were resistant to ciprofloxacin (95.0%), ofloxacin (95.0%) and lomefloxacin (97.5%), only one strain was susceptible to the ciprofloxacin. (2) Sensitive strains had a substitute of Asp95. Ala in the gyrA, and all isolates that were resistant or intermediated to the ciprofloxacin, had a double mutation in the gyrA (Ser91, Ala 92 and Asp95). Some strains also had a mutation in the parC. (3) The MICs of these isolates were significantly associated with the mutation patterns in the gyrA and parC. A double mutation of gyrA combined with parC87 mutation was a predominant pattern in Shanghai and could mediate high level resistance to ciprofloxacin. It suggests that mutations in the QRDRs of gyrA and parC may be responsible for the fluoroquinolone resistance. And fluoroquinolone could not be used as the first line antibiotics for gonorrhea treatment any more in Shanghai, China.
Investigation of fluoroquinolone resistance mechanism in Mycoplasma hominis isolated from urogenital samples in a Chinese hospital

Abstract:Purpose: Mycoplasma hominis is considered among the causes of urogenital infections and shows increasing resistance to fluoroquinolones. However, data regarding the fluoroquinolone resistance mechanism of M. hominis in Southwest China are limited. This study aimed to investigate gene mutations of quinolone resistance-determining regions (QRDRs) of M. hominis isolated from clinical urogenital samples in a Chinese hospital. Methodology: Strains of M. hominis were identified by 16S rRNA gene sequencing. The minimal inhibitory concentrations (MICs) of fluoroquinolones were determined by the broth microdilution method, following CLSI guidelines. PCR was used to amplify the QRDRs of the genes gyrA, gyrB, parC and parE. Positive products were sequenced, and gene mutations and amino acid substitutions were analysed by DNAMAN software and BLAST. Results: The resistance rates of M. hominis to ciprofloxacin (CIP), levofloxacin (LVX), moxifloxacin (MXF) and gatifloxacin (GAT) were 90.5, 85.7, 73.8 and 71.4 %, respectively. A total of 57 isolates of M. hominis were screened, among which 52 strains demonstrated different resistant phenotypes to fluoroquinolones, 41 harboured amino acid substitutions of GyrA S153L, 51 harboured ParC S91I and 22 harboured ParC K144R. ParE A463S and ParC A154T were recorded for the first time and no amino acid change was detected in GyrB. Conclusion: The resistance of M. hominis to fluoroquinolones in Southwest China is mainly related to mutations in QRDRs of either gyrA or parC. High-level resistance is associated with mutations in both DNA gyrase and topoisomerase IV.

Abstract:The polymorphic mutation frequencies for 154 Staphylococcus aureus isolates from Chinese bovine clinical mastitis cases were investigated. We found that nearly 29% of the isolates presented as weak mutators, while only two (1.3%) strong mutators were detected. Of the 15 weak mutators that exhibited ciprofloxacin resistance phenotypes, only one isolate was found to be mutS deficient. All of the ciprofloxacin-resistant isolates had the classic ciprofloxacin resistance mutations at codon 80 within the ParC subunit of topoisomerase IV and codon 84/88 within the GyrA subunit of DNA gyrase. The proportion of ciprofloxacin-resistant isolates among the weak mutators (34.1%) was significantly higher than that found in the normomutators (11.4%) and hypomutators (0%) (P < 0.001, Fisher's exact test), suggesting a positive correlation between weak mutators and ciprofloxacin resistance.
