Abstract:Herein, a general strategy for the regioselective dearomative 1,2-hydrocyanoalkylation of chromium-bound (hetero)arenes with simple alkylnitriles as pronucleophiles was disclosed, providing rapid access to 1,3-cyclohexadienes possessing useful alkylnitrile groups. The versatility of this methodology further enabled a selective dearomative deuteration reaction. Finally, synthetic applications of the method in the formal synthesis of natural products, including erysotramidine, demethoxyerythratidinone, and morphine, were demonstrated.

Abstract:Catalytic difunctionalization with the direct activation of (O)P-H bonds has been recently established as a potentially robust platform to generate valuable organophosphorus compounds. In terms of 1,3-enynes, despite of the various catalytic methods developed for hydrophosphorylation, the radical-mediated hetero-functionalization of two different atoms has been less explored. In this study, we disclosed an electrochemically induced hydroxyphosphorylation of 1,3-enynes for the construction of phosphinyl-substituted propargyl alcohols. The system involves the direct activation of both arylphosphine oxides and oxygen in ambient air with no external metal or additive needed. The use of electrochemistry ensures the regioselective, atom-economic and eco-friendly for the difunctionalization process. This strategy highlights the advantages of mild reaction conditions, readily available starting materials and broad substrate scope, showing its practical synthetic value in organic synthesis.

Abstract:A ligand-controlled cobalt-catalyzed regioselective hydroboration/cyclization of 1,6-enynes with HBPin was developed by switching the size of the coordinated side arm to afford alkenylboronates and alkylboronates, respectively. Gram-scale reactions could be easily conducted, which is beneficial for further derivatizations. Abstract:Different from the reported work focusing on the construction of single P- or C-stereocenter via hydrophosphinylation of unsaturated carbon bonds, the highly diastereo- and enantioselective hydrophosphinylation reaction of allenes, conjugated enynes and 1,3-dienes is achieved via a designed Pd/Co dual catalysis and newly modified masked phosphinylating reagent. A series of allyl motifs bearing both a tertiary C- and P-stereocenter are prepared in generally good yields, >20:1 dr, >20:1 rr and 99% ee. The unprecedented diastereo- and enantioselective hydrophosphinylation of 1,3-enynes is established to generate skeletons containing both a P-stereocenter and a nonadjacent chiral axis. The first stereodivergent hydrophosphinylation reaction is also developed to achieve all four P-containing stereoisomers. The present protocol features the use of only 3-minutes reaction time and 0.1% catalyst, and with the observation of up to 730 TON. A set of mechanistic studies reveal the necessity and roles of two metal catalysts and corroborate the designed synergistic process.

Abstract:Herein we describe a phosphine-catalyzed internal redox [4 + 2] annulation of 1,4-enynoates with electron-deficient alkenes, in which the γ- and φ-C(sp3)-H of the enynoates are formally oxidized for the annulation while the alkynyl moiety is converted to an alkene. The reaction offers an efficient synthesis of highly functionalized cyclohexenes in moderate to good yields with exclusive regioselectivity and high diastereoselectivity under mild conditions.

Abstract:A regiodivergent hydrophosphorylation of enynes with phosphites has been developed using earth-abundant nickel catalyst. The manipulation of regioselectivity can be achieved by regulating the insertion order of alkyne bonds with (RO) 2 P(O)–Ni–H or R 2 P(O)O–Ni–H species, respectively. Under the Ni/Xantphos catalysis, 4,1-hydrophosphorylation is selectively obtained while the adding of acid can promote reactions towards 1,2-addition. By employing an additional Pd–H catalysis, 2,1-hydrophosphorylation is also an accessible task in one-pot reaction. Mechanistic studies and analysis have also been performed to interpret the origin of the regioselective regulation. This work highlights the arts in accessing different regioisomers by diverting common elementary reaction steps.

Abstract:Described herein is a stereo- and regioselective cis-hydrophosphorylation reaction of the internal alkyne of 1,3-enynes that accesses various 1,3-dienes in good isolated yields. Abstract:Herein,an organocatalytic enantioselective desymmetrizing hydroalkoxylationof 1,4- and 1,8-diynes is disclosed, which represents a unique chiralBronsted acid-catalyzed desymmetrization of diynes. This protocolprovides a facile and practical access to enantioenriched 1,3-diaminopropanolderivatives and & gamma;-butyrolactones with a wide substrate scopeand generally high enantioselectivities. Besides, the backbones ofthese products constitute the structural core of numerous bioactivemolecules, and they can serve as valuable precursors for the expeditiousassembly of versatile N- and O-heterocycles. Abstract:A copper-catalyzed alkyne-selective hydroboration of 1,3-enynes is disclosed, providing access to the previously elusive 2-boryl-1,3-dienes. Using CuOAc, Xantphos, and HBpin, Bpin was installed on the internal carbon of a series of symmetric and nonsymmetric 1,3-enynes, affording products with excellent Z:E selectivity. The utility of the 2-boryl-1,3-diene products was demonstrated by transformation to useful functional groups.

Abstract:A highly regio- and stereoselective hydrocyanation of 1,3-enynes was implemented by nickel/diphosphine catalysts. A wide range of highly regio- and stereoselective alkenyl nitriles were efficiently prepared. In this transformation, both the tethered alkene and the ligand play key roles in the reactivity and selectivity. The origin of regioselectivity was understood preliminarily by DFT studies.

Abstract:A phosphine-catalyzed remote 1,7-addition of vinyl allenoates has been developed, providing a series of 1,3-dienes derivatives in high yields (up to 99%) and with good chemo-, regio-, and stereoselectivity. This reaction demonstrated that the introduction of vinyl in allenoates effectively extended reaction types of phosphine-catalyzed nucleophilic addition of allenoates, leading to concise synthesis of diene carboxylates. Notably, the enantioselective variant of this 1,7-addition can also be performed by chiral phosphine catalyst.

Abstract:An iminopyridine cobalt dichloride complex was synthesized and demonstrated as an effective precatalyst for hydrosilylation/cyclization of 1,6-enynes with silanes. Various functional groups such as amine, free aniline, ester, ether, cyano, halide, trifluoromethyl, and heterocycle were tolerated to afford a variety of silicon-containing compounds. The reaction could be scaled up to afford products on the gram scale which could undergo further derivatizations. A primary mechanism was proposed based on analysis of side products and a deuterated experiment.

Abstract:The phosphine-mediated olefination of aldehydes with electron-deficient allenes to afford trisubstituted conjugated dienes in fair to excellent yields with high E-selectivity is described. The reaction represents a new reactivity pattern of allenes with aldehydes and also provides a highly stereoselective synthetic method for preparing conjugated dienes. In the reaction, the phosphine acts as a nucleophilic promoter to generate in situ an active phosphorus ylide which mediates the intermolecular olefination.

Abstract:We established a strategy for the copper‐catalyzed three‐component 1,4‐phosphinoylcyanation of 1,3‐enynes with diarylphosphine oxides and trimethylsilyl cyanide. The reaction produced various tetrasubstituted phosphinoyl‐ and cyano‐containing allenes with high functional group compatibilities in good yields under mild reaction conditions. A preliminary reaction mechanism involving the formation of phosphinoyl radicals and their regioselective addition to 1,3‐enynes was also proposed.

Abstract:A rhodium-catalyzed enantioselective hydroboration/cyclization reaction of 1,6-enynes is achieved by employing a spirosiladiphosphine ligand. The process allows the synthesis of five-membered hetero- and carbocycles bearing a boron handle with high levels of activity and selectivity. Various enynes and organoboranes (HBdan, HBpin, HBmp, and HBamm) have been accommodated, and enynes containing terminal alkynes have been integrated into the process for the first time. The high yields and selectivities of the transformation highlight the synthetic utility of these novel spirosiladiphosphine ligands.

Abstract:Two types of regio- and stereoselective hydrosilylation reactions of 1,3-enynes catalyzed by Co complexes were developed. Dppf-CoCl2 is applicable for 3,4-hydrosilylation of 4-aryl-substituted 1,3-enynes, while Xantphos-CoCl2 is suitable for 4,3-hydrosilylation of 4-alkyl-substituted 1,3-enynes. Various functional groups are tolerated, and the produced 1,3-dienylsilanes can participate in several valuable transformations.

Abstract:A novel strategy to synthesize 1,3-enynes has been successfully developed based on Cu(I)-catalyzed cross-coupling of α-diazo phosphonates and alkynes with a subsequent Horner-Wadsworth-Emmons (HWE) type reaction. This method provides straightforward access to conjugated enynes with high efficiency, good stereoselectivity and excellent functional group compatibility. Copper(I) carbene migratory insertion plays a crucial role in this transformation.

Abstract:A novel photocatalytic decarboxylative [2 + 2 + m] cyclization of 1,7-enynes with alkyl N-hydroxyphthalimide (NHP) esters, using tricyclohexylphosphine and potassium iodide as redox catalysts, is reported for the construction of functional polycyclic compounds. This protocol tolerates primary, secondary, and tertiary alkyl NHP esters through a single reaction via decarbonylation, radical addition, C-H functionalization, and cyclization under mild conditions.

Abstract:A Transition-metal-catalyzed selective hydroarylation of alkynes represents a state-of-the-art approach in organic chemistry. Herein, we report the reaction of symmetrical 1,3-diynes and arylboronic acids, with Pd(OAc)2 as the catalyst and PCy3 as the ligand, affording functionalized enynes in good to excellent yields. Its efficiency is demonstrated by its good functional groups and broad substrate scope. This method offers a general, convenient, and practical strategy for the modular synthesis of multisubstituted enynes. To comprehensively elucidate the mechanism of the Pd-based catalytic system in the hydroarylation of 1,3-diynes, we performed detailed DFT computations for the Pd-catalyzed hydroarylation reaction.
