Abstract:Metabolism is a common biological mechanism in living cells. Acidification plays an important role in cell metabolism, in which the extracellular pH change is used to indicate the vitality of cells. For extracellular detection of cell metabolic substances, light addressable potentiometric sensor (LAPS) has many advantages, such as high sensitivity, easy encapsulation, and convenient integration with microfluidic system for cell experiments. LAPS is a spatially resolved biochemical sensor based on field-effect. In this work, we present a microfluidic system integrated with LAPS for real-time extracellular acidification rate (ECAR) detection. The functions of traditional microphysiometer are achieved with simpler structure devices. Polydimethylsiloxane (PDMS) chamber was manufactured for cell culture, and microfluidic flow paths were used for medium and drug delivery. A bubble trap device was applied to eliminate air bubbles generated in microfluidics. Characteristic tests and cell metabolism experiments were carried out to determine the performance of LAPS by monitoring the pH change of hepatoma HepG2 cells. Glucose and doxorubicin were delivered to the cell chamber to verify the effects of the drug. The pH sensitivity of LAPS is 335.5 nA/pH at the working point in constant voltage mode. The ECAR of HepG2 cells was -48.53 mpH/min in normal glucose medium (25 mM), and it changed to -114.42 mpH/min in high glucose medium (125 mM) and -17.88 mpH/min under the effect of doxorubicin (10 mu M). The results show that the microfluidic LAPS presents good performance in real-time detection of cell acidification and provides a convenient means of assessing cellular specificity of drugs. The modular structure and high expandability make the microfluidic LAPS has good potential in the application of organ-on-chip.

Abstract:We present a novel nanostructured micro cell chip with impedance spectroscopy for monitoring cell morphology change non-invasively, in real time and independent of any fluorescent or radioactive probes. The key strategy is to integrate the nanoporous alumina membrane with silicon based microfluidic devices for the impedance monitoring. In this configuration, the impedance of even single cell can be measured at the low frequency range but not be affected by the electrode polarization. The KYSE30 human oesophageal cancer cells have been successfully cultured on nanoporous alumina membrane. Initial electrochemical experiments with lipid layers have been done to testify the functionality of this device. Further experiments for cancer cells will be explored in near future.

Abstract:Bio-cellular chips have been important tools to detect extracellular physiology in post-gene era,but most of these tools are confronted with the embarrassments of mono-function,unstable parameters,low S/N ratio and difficulty of repetition.However,the integrated chip can transform the electrical,chemical and motile signals of cells into detectable ones by the integration and network of different sensor units with on-line analysis.The research on a novel integrated cellular physiologic sensor is reported in this article,which includes Micro-electrodes Array Sensors(MEAS),Interdigitated Array(IDA) for impedimetric analysis,and Light-addressable Potentiometric sensors(LAPS),etc.to record the extracellular metabolism chemical and biological substances,action potential and livable characteristics of living cells.The cell-sensor interfacial model of each unit was first introduced,the characteristic curves were then measured,and the fundamental electro-physiological cellular analysis was finally tested and analyzed.The average noise level of MEA is about 80 μV after measuring solution was added,the chip is biocompatible for cell culture and suitable for AP measurement;cultured kidney cells on the surface of IDA chip will cause an cellular impedance change of 12%～16% when exposed to 5-Fluororacil;and the sensitivity of H+-LAPS is 50.65 mV/pH.When kidney cells were cultured on LAPS and exposed to high concentration of 5-Fluororacil,the normal metabolism of cells will be affected,which was similar with the IDA results.This research will be interesting and useful for multi-functionalized cellular detecting platform and facilitate a lot to the lab-on-chip research in near future.

Abstract:This paper reviews a novel cell-based biosensor and Bio-MEMS which incorporate living cells as sensing elements that convert a change in immediate environment to signals conducive for processing.It is characterized with high sensitivity,excellent selectivity and fast response and have been implemented for a number of applications ranging from pharmaceutical screening to environmental pollutant detection.This paper also introduces our recent work about Light-Addressable Potentiometric Sensors （LAPS）,Field Effect Transistor （FET）,Micro-Electrode Array Sensors （MEAS） and Bio-MEMS for detecting the changes of concentration of extracellular ions and the action potential of living cell under effect of drugs and environmental parameters.Finely, the paper gives some prospects of cell-based biosensors in the future.

Abstract:A rigorous preselection of identified compounds by in vitro cellular screening is necessary prior to using the drug candidates for the further time consuming and parallel monitoring stage. In this paper we introduced an integrated cell based chip which can detect changes of electrophysiological parameters by using microelectronic chips such as light-addressable potentiometric sensors(LAPS), micro-electrodes array sensors(MEAS) and interdigitated array sensor(IDA) etc. to record the extracellular metabolism chemical and biological substances, action potential and living characteristics of cells representing the different response. In this research we presented novel ideas including an integrating multifunctional cell sensors chip, obtaining the living cells as the source of cell sensors with cardiac myocytes. Further developments will be done by culturing living olfactory and taste cells on surface of the devices. Now, we have completed the fabrication of the sensor and obtained some promising experiments results in different sensor units. This research will be interesting and useful for parallel extracellular monitoring.

Abstract:Development of an efficient sensing platform capable of continual monitoring of biomarkers is needed to assess the functionality of the in vitro organoids and to evaluate their biological responses toward pharmaceutical compounds or chemical species over extended periods of time. Here, a novel label-free microfluidic electrochemical (EC) biosensor with a unique built-in on-chip regeneration capability for continual measurement of cell-secreted soluble biomarkers from an organoid culture in a fully automated manner without attenuating the sensor sensitivity is reported. The microfluidic EC biosensors are integrated with a human liver-on-a-chip platform for continual monitoring of the metabolic activity of the organoids by measuring the levels of secreted biomarkers for up to 7 d, where the metabolic activity of the organoids is altered by a systemically applied drug. The variations in the biomarker levels are successfully measured by the microfluidic regenerative EC biosensors and agree well with cellular viability and enzyme-linked immunosorbent assay analyses, validating the accuracy of the unique sensing platform. It is believed that this versatile and robust microfluidic EC biosensor that is capable of automated and continual detection of soluble biomarkers will find widespread use for long-term monitoring of human organoids during drug toxicity studies or efficacy assessments of in vitro platforms.

Abstract:Organs-on-chips have emerged as viable platforms for drug screening and personalized medicine. While a wide variety of human organ-on-a-chip models have been developed, rarely have there been reports on the inclusion of sensors, which are critical in continually measuring the microenvironmental parameters and the dynamic responses of the microtissues to pharmaceutical compounds over extended periods of time. In addition, automation capacity is strongly desired for chronological monitoring. To overcome this major hurdle, in this protocol we detail the fabrication of electrochemical affinity-based biosensors and their integration with microfluidic chips to achieve in-line microelectrode functionalization, biomarker detection and sensor regeneration, allowing continual, in situ and noninvasive quantification of soluble biomarkers on organ-on-a-chip platforms. This platform is almost universal and can be applied to in-line detection of a majority of biomarkers, can be connected with existing organ-on-a-chip devices and can be multiplexed for simultaneous measurement of multiple biomarkers. Specifically, this protocol begins with fabrication of the electrochemically competent microelectrodes and the associated microfluidic devices (~3 d). The integration of electrochemical biosensors with the chips and their further combination with the rest of the platform takes ~3 h. The functionalization and regeneration of the microelectrodes are subsequently described, which require ~7 h in total. One cycle of sampling and detection of up to three biomarkers accounts for ~1 h.

Abstract:Continual monitoring of secreted biomarkers from organ-on-a-chip models is desired to understand responses to drug exposure in a noninvasive manner. To achieve this goal, analytical methods capable of trace amounts of secreted biomarkers are of particular interest. However, a majority of existing biosensing techniques from limited sensitivity, selectivity, stability, and require large working volumes, especially when cell culture medium is involved, which usually contains a plethora of nonspecific binding proteins and interfering compounds. Hence, novel analytical platforms are needed to provide noninvasive, accurate information on the status of organoids at low working volumes. Here, we report a novel microfluidic aptamer-based electrochemical biosensing platform for monitoring damage to cardiac organoids. The system is scalable, low-cost, and compatible with microfluidic continued.
In situ noninvasive monitoring of cell secretions based on MOFs/AAO hybrid membrane induced asymmetric ion transport

Abstract:Nanofluidic hybrid membranes display distinct ionic current rectification (ICR) properties and provide high surface area for immobilizing probes on the outer surface, exhibiting great potential in detection of biomolecules. Herein, we fabricated MOFs/AAO hybrid membrane with aptamers functionalized on the outer surface for in situ detection of living cells released secretions. TNF-α (a small molecular protein secreted by macrophages) was used as a model. After TNF-α was specifically captured by aptamers on the membrane surface, the asymmetry of surface charge on the hybrid membrane was amplified, the ICR was increased from 3.89 to 18.85. According to the ICR change, TNF-α was sensitively measured with a detection limit of ∼0.49 pM, which was significantly lower than other reported methods. When the hybrid membrane was clamped in the middle of self-made device, PET membrane incubated macrophages was rolled up and inserted into the chamber to mimic cellular microenvironment. Macrophages released TNF-α could be real time monitored with ionic current, macrophages and normal cells could be effectively distinguished according to the released TNF-α level. Thus, we proposed a nanofluidic platform for accurately measuring cell secretions in an engineered cellular microenvironment with a direct manner, without the need for labels or amplification steps.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:Significance of single cell measurements stems from the substantial temporal fluctuations and cell-cell variability possessed by individual cells. A major difficulty in monitoring surface non-adherent cells such as bacteria and yeast is that these cells tend to aggregate into clumps during growth, obstructing the tracking or identification of single-cells over long time periods. Here, we developed a microfluidic platform for long term single-cell tracking and cultivation with continuous media refreshing and dynamic chemical perturbation capability. The design highlights a simple device-assembly process between PDMS microchannel and agar membrane through conformal contact, and can be easily adapted by microbiologists for their routine laboratory use. The device confines cell growth in monolayer between an agar membrane and a glass surface. Efficient nutrient diffusion through the membrane and reliable temperature maintenance provide optimal growth condition for the cells, which exhibited fast exponential growth and constant distribution of cell sizes. More than 24 h of single-cell tracking was demonstrated on a transcription-metabolism integrated synthetic biological model, the gene-metabolic oscillator. Single cell morphology study under alcohol toxicity allowed us to discover and characterize cell filamentation exhibited by different E. coli isobutanol tolerant strains. We believe this novel device will bring new capabilities to quantitative microbiology, providing a versatile platform for single cell dynamic studies.

Abstract:AbstractThere is an increasing interest in developing microfluidic bioreactors and organs-on-a-chip platforms combined with sensing capabilities for continual monitoring of cell-secreted biomarkers. Conventional approaches such as ELISA and mass spectroscopy cannot satisfy the needs of continual monitoring as they are labor-intensive and not easily integrable with low-volume bioreactors. This paper reports on the development of an automated microfluidic bead-based electrochemical immunosensor for in-line measurement of cell-secreted biomarkers. For the operation of the multi-use immunosensor, disposable magnetic microbeads were used to immobilize biomarker-recognition molecules. Microvalves were further integrated in the microfluidic immunosensor chip to achieve programmable operations of the immunoassay including bead loading and unloading, binding, washing, and electrochemical sensing. The platform allowed convenient integration of the immunosensor with liver-on-chips to carry out continual quantification of biomarkers secreted from hepatocytes. Transferrin and albumin productions were monitored during a 5-day hepatotoxicity assessment in which human primary hepatocytes cultured in the bioreactor were treated with acetaminophen. Taken together, our unique microfluidic immunosensor provides a new platform for in-line detection of biomarkers in low volumes and long-term in vitro assessments of cellular functions in microfluidic bioreactors and organs-on-chips.

Abstract:Biosensors are widely used in various applications, from medical diagnostics to environmental monitoring. Their widespread and continuous use necessitates regeneration methods to ensure cost-effectiveness and sustainability. In the realm of advancing human-centric bioelectronics for continuous monitoring, employing these sensors for real-time, in situ detection of biomarkers presents a considerable challenge. This minireview examines the diverse strategies utilized for the regeneration of biosensors, categorizing them based on their underlying mechanisms and discussing representative works. We explore methods ranging from surface engineering/re-functionalization, chemical treatments, allosteric regulation of bioreceptors, to manipulations of electric/magnetic fields, highlighting their working principles and exemplary studies. The advantages of each method, such as simplicity, high regeneration efficiency, and versatility, are discussed alongside their challenges, including degradation over cycles, limited applicability, and potential damage to sensors. As the demand for continuous and real-time biosensing escalates, the development of efficient and reliable regeneration strategies becomes essential. This minireview offers an overview of the current landscape of biosensor regeneration, aiming to guide future research and innovations in this area.

Abstract:Microscale systems that enable measurements of oncological phenomena at the single-cell level have a great capacity to improve therapeutic strategies and diagnostics. Such measurements can reveal unprecedented insights into cellular heterogeneity and its implications into the progression and treatment of complicated cellular disease processes such as those found in cancer. We describe a novel fluid-delivery platform to interface with low-cost microfluidic chips containing arrays of microchambers. Using multiple pairs of needles to aspirate and dispense reagents, the platform enables automated coating of chambers, loading of cells, and treatment with growth media or other agents (e.g., drugs, fixatives, membrane permeabilizers, washes, stains, etc.). The chips can be quantitatively assayed using standard fluorescence-based immunocytochemistry, microscopy, and image analysis tools, to determine, for example, drug response based on differences in protein expression and/or activation of cellular targets on an individual-cell level. In general, automation of fluid and cell handling increases repeatability, eliminates human error, and enables increased throughput, especially for sophisticated, multistep assays such as multiparameter quantitative immunocytochemistry. We report the design of the automated platform and compare several aspects of its performance to manually-loaded microfluidic chips.
From Chips-in-Lab to Point-of-Care Live Cell Device: Development of a Microfluidic Device for On-Site Cell Culture and High-Throughput Drug Screening

Abstract:Live cell assays provide real-time data of cellular responses. In combination with microfluidics, applications such as automated and high-throughput drug screening on live cells can be accomplished in small devices. However, their application in point-of-care testing (POCT) is limited by the requirement for bulky equipment to maintain optimal cell culture conditions. In this study, we propose a POCT device that allows on-site cell culture and high-throughput drug screening on live cells. We first observe that cell viabilities are substantially affected by liquid evaporation within the microfluidic device, which is intrinsic to the polydimethylsiloxane (PDMS) material due to its hydrophobic nature and nanopatterned surface. The unwanted PDMS-liquid-air interface in the cell culture environment can be eliminated by maintaining a persistent humidity of 95-100% or submerging the whole microfluidic device under water. Our results demonstrate that in the POCT device equipped with a water tank, both primary cells and cell lines can be maintained for up to 1 week without the need for external cell culture equipment. Moreover, this device is powered by a standard alkali battery and can automatically screen over 5000 combinatorial drug conditions for regulating neural stem cell differentiation. By monitoring dynamic variations in fluorescent markers, we determine the optimal doses of platelet-derived growth factor and epidermal growth factor to suppress proinflammatory S100A9-induced neuronal toxicities. Overall, this study presents an opportunity to transform lab-on-a-chip technology from a laboratory-based approach to actual point-of-care devices capable of performing complex experimental procedures on-site and offers significant advancements in the fields of personalized medicine and rapid clinical diagnostics.

Abstract:Purpose Recently, powerful instruments for biomedical engineering research studies, including disease modeling, drug designing and nano-drug delivering, have been extremely investigated by researchers. Particularly, investigation in various microfluidics techniques and novel biomedical approaches for microfluidic-based substrate have progressed in recent years, and therefore, various cell culture platforms have been manufactured for these types of approaches. These microinstruments, known as tissue chip platforms, mimic in vivo living tissue and exhibit more physiologically similar vitro models of human tissues. Using lab-on-a-chip technologies in vitro cell culturing quickly caused in optimized systems of tissues compared to static culture. These chipsets prepare cell culture media to mimic physiological reactions and behaviors. Design/methodology/approach The authors used the application of lab chip instruments as a versatile tool for point of health-care (PHC) applications, and the authors applied a current progress in various platforms toward biochip DNA sensors as an alternative to the general bio electrochemical sensors. Basically, optical sensing is related to the intercalation between glass surfaces containing biomolecules with fluorescence and, subsequently, its reflected light that arises from the characteristics of the chemical agents. Recently, various techniques using optical fiber have progressed significantly, and researchers apply highlighted remarks and future perspectives of these kinds of platforms for PHC applications. Findings The authors assembled several microfluidic chips through cell culture and immune-fluorescent, as well as using microscopy measurement and image analysis for RNA sequencing. By this work, several chip assemblies were fabricated, and the application of the fluidic routing mechanism enables us to provide chip-to-chip communication with a variety of tissue-on-a-chip. By lab-on-a-chip techniques, the authors exhibited that coating the cell membrane via poly-dopamine and collagen was the best cell membrane coating due to the monolayer growth and differentiation of the cell types during the differentiation period. The authors found the artificial membrane, through coating with Collagen-A, has improved the growth of mouse podocytes cells-5 compared with the fibronectin-coated membrane. Originality/value The authors could distinguish the differences across the patient cohort when they used a collagen-coated microfluidic chip. For instance, von Willebrand factor, a blood glycoprotein that promotes hemostasis, can be identified and measured through these type-coated microfluidic chips.

Abstract:A continuous rise in the wastes from industrial effluents, bio-waste, and pharmaceuticals has deteriorated surface water and drinking water sources. Standard laboratory tests of total coliform are time-consuming and logistically inefficient for field data generation. Better and portable sensing technologies are needed. This paper reports an electrical impedance spectroscopic technique incorporated in a micro-fluidic chip with interdigitated microelectrodes to monitor the growth of microbial cells. Lag, log, and stationary phases of Escherichia coli cell growth with an integrated electrode are successfully detected, for samples of reverse osmosis water, standard treated tap water, and recycled water respectively. The results indicate that reverse osmosis water has a higher probability of contamination with bacterial pathogens compared to the other two types of water samples when subjected to the same amount of added nutrients. The statistical analysis shows a possible single detection range with higher-order regression, and repeat use of a single chip with the electrode was found to be within an acceptable limit. The interdigitated electrodes exposed to in-situ cell growth conditions and repeated electrical measurements have shown a promise for possible periodic or continuous monitoring. The paper further identifies several complimentary analysis methodologies that are robust towards phase noise in the measured impedance and are suited particularly for early-stage detection of bacterial contamination. The cell adhesion tendencies over the microelectrode due to the electric field need to be further analyzed.

Abstract:Cell models are one of the most widely used basic models in biological research, and a variety of in vitro cell culture techniques and models have been developed recently to simulate the physiological microenvironment in vivo. However, regardless of the technique or model, cell culture is the most fundamental but crucial component. As a result, we have developed a cell culture monitoring system to assess the functional status of cells within a biochip. This article focuses on a mini-microscope made from a readily available camera for in situ continuous observation of cell growth within a biochip and a pH sensor based on optoelectronic sensing for measuring pH. With the aid of this monitoring system, scientists can keep an eye on cell growth in real time and learn how the pH of the culture medium affects it. This study offers a new approach for tracking cells on biochips and serves as a valuable resource for enhancing cell culture conditions.
