Abstract:Objective To construct recombinant lentiviral vector of miRNA let-7c and verify its effect on liver cancer cell proliferation.Methods Recombinant lentiviral vector containing 494 bp let-7c gene was constructed.The titers of Let-7c and the controls were 5.01 × 10E8 and 5.15 × 10E8 TU/ml respectively.Let-7c lentiviral vector was transfected into HepG2,and the expression of let-7c was detected by using real time polymerase chain reaction (PCR).Cell counting Kit-8 (CCK-8) method was used to testify HepG2 cell proliferation after the vector transfection.Results The successful construction of let-7c lentiviral vector was confirmed by plasmid enzyme digestion and DNA sequencing.Seventy-two h after the vector transfection,the expression of mature let-7c was increased by about 312 folds,compared to blank vector transfection.The over-expression of let-7e significantly suppressed HepG2 cell prolifieration (P ＜0.01 ).Conclusion The let-7c recombinant lentiviral vector was successfully constructed.After the vector was transfected to HepG2 cells,mature let-7c was effectively expressed and suppressed the HepG2 liver cancer cell proliferation.

Abstract:Objective To construct a lentivirus vector for RNA interference(RNAi) of BC047440 gene,and detect the silence effect of the vector in human hepatocellular liver carcinoma cell line(HepG2).Methods The targeting sequence of BC047440 gene which can be effectively silenced in RNA inference was confirmed in our previous study.The cDNA encoding for sense and antisense Oligo DNA fragments of the targeting sequence was designed,synthesized and cloned into the pFU-GW-iRNA vector which had been double digested by the HpaⅠ and XhoⅠ,and vector then the results confirmed by PCR and DNA sequencing.293T cells were cotransfected with lentiviral vector pFU-GW-shBC047440,pHelper 1.0 and pHelper 2.0 by Lipofectamine 2000,to produce lentiviral vector which can infect other cell lines.The virus titer and the fittest multiplicity of infection(MOI) in HepG2 were measured by the expression level of green fluorescent protein(GFP).The infected HepG2 was divided into BC047440-shRNA group,control-shRNA group and HepG2 group.The changes of BC047440 mRNA and protein level were detected by RT-PCR and Western blot analysis respectively.Results The data of PCR demonstrated that the lentivirus RNAi vector of BC047440 was constructed successfully.The titer of virus was 5×108 TU/ml,and the fittest MOI in HepG2 was 20.BC047440 and BC047440 mRNA expression of BC047440-shRNA group was obvious lower than the control-shRNA group and HepG2 group,but there was no visible different between the control-shRNA group and HepG2 group.Conclusion The lentivirus RNAi vector of BC047440 is constructed successfully.After this lentivirus vector infects HepG2 cells,the target of silencing the expression on BC047440 is achieved.
Construction and identification of vector against Kir6.2 gene expression in hepatoma cells by short interfering RNAs

Abstract:Objective:To construct the expression vector pSlience-3-siRNA that can express the short hairpin RNAs(shRNA) against Kir6.2 gene and to screen the effective target sequence of shRNA.Methods:Two shRNA against Kir6.2 gene were transcript synthesized intracellularly by expressed templates of plasmid vector pSilence-3,and the target sequence of Kir6.2 gene was inserted into the upstream of the reporter gene in order to construct the recombinant plasmid vector pSlience-3.The plasmids containing 2 different sequences of human Kir6.2 mRNA coding region were constructed and transfected into Sk-Hep1 cells with Lipofectamine 2000 methods,respectively. The cells were divided into 4 groups: SK(normal),HK(transfected the recombinant plasmid HK),K1(transfected the recombinant plasmid K1) and K2(transfected the recombinant plasmid K2) group.The selected single clone was cultured after screening by G418.The expression of Kir6.2 gene was detected by semiquantitative reverse transcription-polymerase chain reaction(RT-PCR) and Western blotting.The effective siRNA against the expression of Kir6.2 was screened out.Results:The relative quantity of the Kir6.2 gene expression of SK,HK,K1,K2 detected by RT-PCR was 0.936±0.042,0.848±0.058,0.225 ±0.036,0.171 ±0.029,respectively.Compared with SK and HK groups,the expression of Kir6.2 mRNA in K1 and K2 groups decreased significantly(P<0.05).The relative quantity of the Kir6.2 protein of SK,HK,K1,and K2 was 0.925±0.045,0.901±0.067,0.269±0.024,0.318±0.031,respectively.The quantity of Kir6.2 protein in K1and K2 groups reduced more than in SK and HK groups(P<0.05).Conclusion:The shRNA can inhibit the expression of Kir6.2 gene in hepatoma cells.

Underexpression of Kir6.2 gene significantly decreased the proliferation and invasion of the HepG2 cells.CONCLUSION:shRNA can inhibit the expression of Kir6.2 gene in the HepG2 cells, and underexpression of Kir6.2 gene decreased the proliferation and invasion of the HepG2 cells.
Efficient Attenuation of NK Cell–Mediated Liver Injury Through Genetically Manipulating Multiple Immunogenes by Using a Liver-Directed Vector

Abstract:Adenovirus or adenoviral vectors were reported to induce serious liver inflammation in an NK cell–dependent manner, which limits its clinical applicability for liver gene therapy. We tried to develop an efficient liver-directed therapeutic approach to control hepatic NK cell function via simultaneously manipulating multiple immune genes. Based on our previous study, we found that CCL5 knockdown synergistically enhanced the attenuating effect of silencing CX3CL1 (fractalkine [FKN]) in adenovirus-induced acute liver injury. In addition, the combined treatment of human IL-10 expression with FKN knockdown would further strengthen the protective effect of silencing FKN. We used a hepatocyte-specific promoter to construct a hepatocyte-specific multiple function vector, which could simultaneously overexpress human IL-10 and knock down CCL5 and FKN expression. This vector could attenuate adenovirus-induced acute hepatitis highly efficiently by reducing liver NK cell recruitment and serum IFN-γ and TNF-α. The multiple function vectors could be delivered by nonviral (hydrodynamic injection) and viral (adenovirus) approaches, and maintained long-term function (more than 1 month in mice). Our results suggest a possible strategy to ameliorate the acute liver injury induced by adenovirus by modulating multiple immune genes. The novel multifunction vector has an extensive and practical use for polygenic and complex liver diseases such as malignancies and hepatitis, which correlate with multiple gene disorders.
BC047440 Antisense Eukaryotic Expression Vectors Inhibited HepG2 Cell Proliferation and Suppressed Xenograft Tumorigenicity.

Abstract:The biological functions of the BC047440 gene highly expressed by hepatocellular carcinoma (HCC) are unknown. The objective of this study was to reconstruct antisense eukaryotic expression vectors of the gene for inhibiting HepG(2) cell proliferation and suppressing their xenograft tumorigenicity. The full-length BC047440 cDNA was cloned from human primary HCC by RT-PCR. BC047440 gene fragments were ligated with pMD18-T simple vectors and subsequent pcDNA3.1(+) plasmids to construct the recombinant antisense eukaryotic vector pcDNA3.1(+)BC047440AS. The endogenous BC047440 mRNA abundance in target gene-transfected, vector-transfected and naive HepG(2) cells was semiquantitatively analyzed by RT-PCR and cell proliferation was measured by the MTT assay. Cell cycle distribution and apoptosis were profiled by flow cytometry. The in vivo xenograft experiment was performed on nude mice to examine the effects of antisense vector on tumorigenicity. BC047440 cDNA fragments were reversely inserted into pcDNA3.1(+) plasmids. The antisense vector significantly reduced the endogenous BC047440 mRNA abundance by 41% in HepG(2) cells and inhibited their proliferation in vitro (P < 0.01). More cells were arrested by the antisense vector at the G(1) phase in an apoptosis-independent manner (P = 0.014). Additionally, transfection with pcDNA3.1(+)BC047440AS significantly reduced the xenograft tumorigenicity in nude mice. As a novel cell cycle regulator associated with HCC, the BC047440 gene was involved in cell proliferation in vitro and xenograft tumorigenicity in vivo through apoptosis-independent mechanisms.
Construction of hsa-miR-152lentiviral vector and screening of cell strain for stable expression

Abstract:Objective To construct a lentiviral vector of homo sapiens-miR-152(hsa-miR-152) and screen the cell strain for its stable expression.Methods The fragments of pre-miR-152 were cloned from genomic DNA of human adipose tissue and inserted into lentiviral vector pGIPZ.The recombinant plasmid pGIPZ-miR-152 was confirmed by restriction endonuclease analysis and DNA sequencing.293T cells were transfected with the recombinant plasmid pGIPZ-miR-152 and the supernatant containing the lentivirus particles was harvested to determine the virus titer and used to infect HepG2 cells.Puromycin was used to screen the infected cells,so as to obtain a cell strain for stable expression.The expression of miR-152 was determined using real-time PCR.Results The restriction enzyme digestion and DNA sequencing demonstrated the lentiviral vector pGIPZ-miR-152 was constructed.The lentiviral vector was transfected into 293T cells and the viral titer reached 7.5×107 TU/mL.HepG2 cells were infected by the lentivirus and the infection efficiency was over 70%.The expression of miR-152 was increased nearly 10 folds in the overexpression group following screening with puromycin for 10 days.Conclusion The lentiviral vector expressing hsp-miR-152 was constructed successfully.The cell strain with stable overexpression of miR-152 was also obtained,which provided the material for the further study of molecular function of miR-152 in nonalcoholic fatty liver disease.

Abstract:Successful RNAi applications depend on strategies allowing robust and persistent expression of minimal gene silencing triggers without perturbing endogenous gene expression. Here, we propose a novel avenue which is integration of a promoterless shmiRNA, i.e. a shRNA embedded in a micro-RNA (miRNA) scaffold, into an engineered genomic miRNA locus. For proof-of-concept, we used TALE or CRISPR/Cas9 nucleases to site-specifically integrate an anti-hepatitis C virus (HCV) shmiRNA into the liver-specific miR-122/hcr locus in hepatoma cells, with the aim to obtain cellular clones that are genetically protected against HCV infection. Using reporter assays, Northern blotting and qRT-PCR, we confirmed anti-HCV shmiRNA expression as well as miR-122 integrity and functionality in selected cellular progeny. Moreover, we employed a comprehensive battery of PCR, cDNA/miRNA profiling and whole genome sequencing analyses to validate targeted integration of a single shmiRNA molecule at the expected position, and to rule out deleterious effects on the genomes or transcriptomes of the engineered cells. Importantly, a subgenomic HCV replicon and a full-length reporter virus, but not a Dengue virus control, were significantly impaired in the modified cells. Our original combination of DNA engineering and RNAi expression technologies benefits numerous applications, from miRNA, genome and transgenesis research, to human gene therapy.

Abstract:Lentiviral vectors (LVs) offer several advantages over traditional oncoretroviral vectors. LVs efficiently transduce slowly dividing cells, including hematopoietic stem-progenitor cells (HSCs), resulting in stable gene transfer and expression. Additionally, recently developed self-inactivating (SIN) LVs allow promoter-specific transgene expression. For many gene transfer applications, transduction of more than one gene is needed. We obtained inconsistent results in our attempts to coexpress two transgenes linked by an internal ribosomal entry site (IRES) element in a single bicistronic LV transcript. In more than six bicistronic LVs we constructed containing a gene of interest followed by an IRES and the GFP reporter gene, GFP fluorescence was undetectable in transduced cells. We therefore investigated how to achieve consistent and efficient coexpression of two transgenes by LVs. In a SIN LV containing the elongation factor 1α promoter, we included a second promoter from cytomegalovirus, the phosphoglycerate kinase gene, or the HLA-DRα gene. Using a single LV containing two constitutive promoters, we achieved strong and sustained expression of both transgenes in transduced engrafting CD34+ HSCs and their progeny, as well as in other human cell types. Thus, such dual-promoter LVs can coexpress multiple transgenes efficiently in a single target cell and will enable many gene transfer applications.
Genetic Modification of Hepatocytes towards Hepatocyte Transplantation and Liver Tissue Engineering

Abstract:Cell-based therapies, including liver tissue engineering following hepatocyte transplantation, have therapeutic potential for several types of liver diseases. Modifications in the methodology to manipulate the donor hepatocytes in a more simple and timely manner prior to transplantation would enhance the therapeutic efficacy of this procedure. Conventional approach for vector-mediated gene transduction to the isolated hepatocytes has been performed under primary culture conditions that routinely require several days to complete. In our study, we have established a clinically feasible approach that requires only 1 h of infection time with an adenoviral vector system that results in an extremely efficient transduction efficiency (>80%). To optimize transduction efficiency and sustain normal cellular function, we determined that the isolated hepatocytes should be maintained in UW solution as a suspension medium and infected with adenoviral vectors (Ad) for no more than 1 h at a MOI of 1. To establish if the isolated hepatocytes could be used as a source for cell-based therapies, we transplanted the Ad-transduced hepatocytes into the liver or under the kidney capsule. When the cells were transplanted into the liver, Ad-transduced hepatocytes cultured in suspension conditions were found to have a significantly higher survival rate (p < 0.01) than Ad-transduced hepatocytes cultured under standard conditions. We also confirmed that these Ad-transduced hepatocytes have ability to survive long term and were able to engineer a biologically active hepatic tissue under the kidney capsule. Finally, we obtained high level of transduction into canine, porcine, and human isolated hepatocytes in a suspension solution mixed with Ad. In conclusion, the present studies demonstrate that isolated hepatocytes could be genetically modified using Ad when kept in a suspension solution. For this reason, this cell-modified technique could be used for the treatment of liver-targeted diseases and/or disorders.

[Construction of All-in-one CRISPR/Cas9 Vector System Targeting Mir-101A Gene in Mouse Hepatic Cell Line AML12].

Abstract:OBJECTIVE To develop an all-in-one CRISPR/Cas9 vector system that can efficiently knockdown miR-101a expression in mice. METHODS Three sgRNAs targeting mouse miR-101a gene and a small guide (sgRNA) targeting green fluorescent protein gene were designed and constructed into an all-in-one vector system (pENTRY-U6-sgRNA-WT Cas9). Moreover, sgRNA1 and sgRNA3 were selected and constructed into a double-nicking Cas9 vector (pENTRY-U6-sgRNA-U6-sgRNA-Cas9 D10A). The constructed plasmids were transfected into mouse liver AML12 cells for validation by T7 EndoⅠ(T7EⅠ) 72 h after transfection. The pAD vectors were cloned via the Gateway system, and the recombinant adenovirus vectors were packaged in 293A cells. The virus particles were used to infect AML12 cells and the expression levels of mature miR-101a were analyzed to monitor the knockout efficiency after 72 h. RESULTS The constructed pENTRY all-in-one vectors were validated by gene sequencing and T7EⅠ assay, which showed CRISPR/Cas9-mediated mismatches at target sites of miR-101a gene. The adenovirus vectors were constructed successfully. The CRISPR/Cas9 containing adenovirus was introduced to AML12 cells and the quantitative real-time PCR assays indicated that the expression level of mature miR-101a was significantly decreased compared with that of the control (all P<0.01). CONCLUSIONS We have successfully constructed two "all-in-one" CRISPR/Cas9 vector systems targeting miR-101a gene in mouse liver AML12 cells with high efficiency. It provides experimental basis for research of microRNA, and a reference method for knockout of other miRNAs.
Novel Recombinant Hepatitis B Virus Vectors Efficiently Deliver Protein and RNA Encoding Genes into Primary Hepatocytes

Abstract:ABSTRACT Hepatitis B virus (HBV) has extremely restricted host and hepatocyte tropism. HBV-based vectors could form the basis of novel therapies for chronic hepatitis B and other liver diseases and would also be invaluable for the study of HBV infection. Previous attempts at developing HBV-based vectors encountered low yields of recombinant viruses and/or lack of sufficient infectivity/cargo gene expression in primary hepatocytes, which hampered follow-up applications. In this work, we constructed a novel vector based on a naturally occurring, highly replicative HBV mutant with a 207-bp deletion in the preS1/polymerase spacer region. By applying a novel insertion strategy that preserves the continuity of the polymerase open reading frame (ORF), recombinant HBV (rHBV) carrying protein or small interfering RNA (siRNA) genes were obtained that replicated and were packaged efficiently in cultured hepatocytes. We demonstrated that rHBV expressing a fluorescent reporter (DsRed) is highly infective in primary tree shrew hepatocytes, and rHBV expressing HBV-targeting siRNA successfully inhibited antigen expression from coinfected wild-type HBV. This novel HBV vector will be a powerful tool for hepatocyte-targeting gene delivery, as well as the study of HBV infection.
Re-Designed Recombinant Hepatitis B Virus Vectors Enable Efficient Delivery of Versatile Cargo Genes to Hepatocytes with Improved Safety.

Abstract:Hepatitis B virus (HBV) takes humans as its sole natural host, and productive infection in vivo is restricted exclusively to hepatocytes in the liver. Consequently, HBV-derived viral vectors are attractive candidates for liver-targeting gene therapies. Previously, we developed a novel recombinant HBV vector, designated 5c3c, from a highly replicative clinical isolate. 5c3c was demonstrated to be capable of efficiently delivering protein or RNA expression into infected primary tupaia hepatocytes (PTH), but the design of 5c3c imposes stringent restrictions on inserted sequences, which have limited its wider adoption. In this work, we addressed issues with 5c3c by re-designing the insertion strategy. The resultant vector, designated 5dCG, was more replicative than parental 5c3c, imposed no specific restrictions on inserted sequences, and allowed insertion of a variety of cargo genes without significant loss of replication efficiency. 5dCG-based recombinant HBV effectively delivered protein and RNA expression into infected PTH. Furthermore, due to the loss of functional core ORF, 5dCG vectors depend on co-infecting wild type HBV for replication and efficient expression of cargo genes. Development of the improved 5dCG vector makes wider applications of recombinant HBV possible, while dependence on co-infecting wild type HBV results in improved safety for certain in vivo applications.

Abstract:The application of RNA interference (RNAi) as a research and therapeutic tool depends on its ability to silence genes in a sequence-specific manner. Recent studies have reported that the effective knockdown of genes can be achieved by multiple short hairpin RNAs (shRNAs) in a single vector. Moreover, this approach can depress several genes simultaneously. However, current methods for the construction of multiple shRNA vectors often suffer from vector instability and are time-consuming. Here, we describe a simple, quick and low-cost approach to construct a single vector expressing four shRNA sequences driven by four different promoters. Using this vector, we were able to improve the gene silencing efficiency and make it possible to silence four different genes simultaneously, further expanding the application spectrum of RNAi, both in functional studies and therapeutic strategies. Construction of a regulable gene therapy vector targeting for hepatocellular carcinoma

Abstract:Aim: To construct a gene modified hepatocellular carcinoma (HCC) specific EGFP expression vector regulated by abbreviated cis-acting element of AFP gene. Methods: The minimal essential DNA segments of AFP gene enhancer and promoter were synthesized through PCR from Genome DNA of HepG2 cells. Gene fragments were then cloned into the multiple cloning site of non-promoter EGFP vector pEGFP-1. Recombinant plasmid was transferred into positive or negative AFP cell lines by means of lipofectamine. The expression of EGFP was tested by fluorescence microscope and flow cytometry. The effect of all-trans retinoic acid (ATRA) on the expression of EGFP was tested in different concentrations. Results: By the methods of restriction digestion and sequence analyses we confirmed that the length, position and orientation of inserted genes of cis-acting element of AFP were all correct. The transcription of EGFP was under the control of AFP cis-acting element. The expressing EGFP can only been detected in AFP producing hepatoma cells. The expression rate of EGFP in G418 screened cell line was 34.9+/-4.1 %. 48 h after adding 1X10(-7)M retinoic acid, EGFP expression rate was 14.7+/-3.5 %. The activity of AFP gene promoter was significantly suppressed by addition of 1 x 10(-7)M retinoic acid (P<0.05, P=0.003, t=6.488). Conclusion: This recombinant expression vector can be used as a gene therapy vector for HCC. The expression of tumor killing gene will be confined within the site of tumor and the activity of which can be regulated by retinoic acid.

Abstract:Objective To construct lentiviral vector mediated SiRNA targeting IKKα and IKKβ gene and explore its effects on the growth of hepatocellular carcinoma (HCC) cells.Methods Two recombinant vectors were generated by subcloning a target DNA into lentiviral vector.After being infected with the virus,the expression of IKKα and IKKβ in HCC cells was examined.The transfected cells were also injected into mice subcutaneously,and the growth of tumor was observed. Results The expression of IKKα and IKKβ protein in transfected cells decreased,and tumor growth inhibition was observed obviously in mice.Conclusion Lentiviral vector transduced small interference RNA targeting IKKα and IKKβ gene can suppress the growth of HCC cells.
Gene Manipulation in Liver Ductal Organoids by Optimized Recombinant Adeno-Associated Virus Vectors.

Abstract:Understanding the mechanism of how liver ductal cells (cholangiocytes) differentiate into hepatocytes would permit liver-regenerative medicine. Emerging liver ductal organoids provide an ex vivo system to investigate cholangiocyte-to-hepatocyte differentiation. However, as current gene manipulation methods require organoid dissociation into single cells and have only low efficiency, it is difficult to dissect specific gene functions in these organoids. Here we developed the adeno-associated virus (AAV) vector AAV-DJ as a powerful tool to transduce mouse and human liver ductal organoids. Via AAV-DJ-mediated up- or down-regulation of target genes, we successfully manipulated cholangiocyte-to-hepatocyte differentiation. We induced differentiation by overexpressing the hepatocyte-specifying regulator hepatocyte nuclear factor 4 alpha (HNF4 alpha) and blocked differentiation by stimulating Notch signaling or interfering with Smad signaling. Further screening for transcriptional factors critical for cholangiocyte-to-hepatocyte differentiation identified HOP homeobox (HOPX), T-box 15 (TBX15), and transcription factor CP2-like 1 (TFCP2L1) as master regulators. We conclude that this highly efficient and convenient gene manipulation system we developed could facilitate investigation into genes involved in cell lineage transitions and enable application of engineered organoids in regenerative medicine.

Abstract:Hydrodynamic tail vein injection (HTV) is the “gold standard” for delivering naked DNA vectors to mouse liver, thereby transfecting predominately perivenous hepatocytes. While HTV corrects metabolic liver defects such as phenylketonuria or cystathionine β-synthase deficiency, correction of spfash mice with ornithine transcarbamylase (OTC) deficiency was not possible despite overexpression in the liver, as the OTC enzyme is primarily expressed in periportal hepatocytes. To target periportal hepatocytes, we established hydrodynamic retrograde intrabiliary injection (HRII) in mice and optimized minicircle (MC) vector delivery using luciferase as a marker gene. HRII resulted in a transfection efficiency below 1%, 100-fold lower than HTV. While HRII induced minimal liver toxicity compared with HTV, overexpression of luciferase by both methods, but not of a natural liver-specific enzyme, elicited an immune response that led to the elimination of luciferase expression. Further testing of MC vectors delivered via HRII in spfash mice did not result in sufficient therapeutic efficacy and needs further optimization and/or selection of the corrected cells. This study reveals that luciferase expression is toxic for the liver. Furthermore, physical delivery of MC vectors via the bile duct has the potential to treat defects restricted to periportal hepatocytes, which opens new doors for non-viral liver-directed gene therapy.

Abstract:Being relatively simple and practical, Drosophila transgenic RNAi is the technique of top priority choice to quickly study genes with pleiotropic functions. However, drawbacks have emerged over time, such as high level of false positive and negative results. To overcome these shortcomings and increase efficiency, specificity and versatility, we develop a next generation transgenic RNAi system. With this system, the leaky expression of the basal promoter is significantly reduced, as well as the heterozygous ratio of transgenic RNAi flies. In addition, it has been first achieved to precisely and efficiently modulate highly expressed genes. Furthermore, we increase versatility which can simultaneously knock down multiple genes in one step. A case illustration is provided of how this system can be used to study the synthetic developmental effect of histone acetyltransferases. Finally, we have generated a collection of transgenic RNAi lines for those genes that are highly homologous to human disease genes.
