Carvacrol alleviates cerebral edema by modulating AQP4 expression after intracerebral hemorrhage in mice.

Abstract:AIM: To discuss the change of astrocytic connexin 43 (Cx43) expression in cerebral edema and the potential roles of Cx43 in the development of cerebral edema. METHODS:Cryoinjury-induced cerebral edema model was established in the right parietal lobe cortex in the rats. The animals were divided into sham group, cryoinjury-induced cerebral edema model group, and cryoinjury-induced cerebral edema model injected with carbenoxolone ( or octanol ) group. Dry and wet weight method was used to measure the content of brain water. Methanamide method was used to evalu-ate the permeability of blood-brain barrier ( BBB) after cryoinjury. HE staining showed the physiological changes in injured brain cortex. The protein expression of Cx43 was detected by Western blot and immunohistochemical staining. RESULTS:The brain water content in the injured cortex of rats was increased and cerebral edema developed to the peak at 24 h after cryoinjury. The permeability of BBB was increased in the injured cortex of the rats, and the area where BBB was damaged was larger than injured area. HE staining showed that many cells were dead in the central area of cryoinjury, and at the same time cerebral edema was appeared obviously in the surrounding area of injured brain cortex. The protein expression of Cx43 was up-regulated in the surrounding area of injured brain cortex, while Cx43 expression decreased in the injured cor-tex at different periods after cryoinjury. Both the content of brain water in the injured cortex and the permeability of BBB at 24 h after cryoinjury were reduced after injecting carbenoxolone or octanol intraperitoneally to block Cx43. CONCLU-SION:The cerebral edema up-regulates the expression of Cx43 and also strengthens its function. Once Cx43 is blocked, the degree of cerebral edema in rats is attenuated.
Propofol Pretreatment Attenuates Aquaporin-4 Over-Expression and Alleviates Cerebral Edema after Transient Focal Brain Ischemia Reperfusion in Rats

Abstract:BACKGROUND: Cerebral edema is a major threat for stroke victims. Most studies have focused on the neuroprotective activities of propofol, addressing infarct volume rather than cerebral edema. Aquaporin-4 (AQP4) plays an important role in maintaining brain water homeostasis under various neurological insults. We explored the effect of propofol pretreatment on cerebral edema in a rat model of brain ischemia reperfusion and assessed the involvement of AQP4.METHODS: To induce brain ischemia reperfusion, we introduced a silicone-coated monofilament nylon suture into the origin of the middle cerebral artery, withdrawing it after 90 min. Treatment groups (n = 32), received propofol (0.1 mL . kg(-1) . min(-1)) infusion for 30 min before occlusion; the vehicle group (n = 32) and the sham-operated group (n = 28), which received the intralipid vehicle at the same time and rate. To assess cerebral infarct volume, we used 2, 3, 5-triphenyltetrazolium chloride staining; wet-dry weight ratio was the basis for cerebral edema estimation, and we used immunohistochemistry and Western blot to detect AQP4 expression.RESULTS: The wet-dry weight ratio decreased from 86.89% +/- 0.71%, in the vehicle group (n = 6) to 72.42% +/- 0.74% in the propofol group (n = 6), corresponding to an average decrease of 16%. In parallel and based on immunohistochemical semi-quantification, the propofol group exhibited remarkable attenuation of AQP4 over-expression in the ischemic border zone compared with the vehicle group: 1.28 +/- 0.03 vs 1.40 +/- 0.05, n = 7, respectively; P < 0.05. Values derived from Western blot quantification were similarly decreased ill the propofol group compared to the vehicle group: 20.85% +/- 4.18% vs 31.67% +/- 3.23%, 11 = 4, respectively; P < 0.05. However, infarct volume and neurologic deficit in postischemic rats in the propofol group were not statistically different from values in the vehicle group.CONCLUSIONS: We conclude that prestroke treatment with propofol reduces postischemic cerebral edema in rats, possibly through inhibiting AQP4 over-expression in the boundary zone of ischemia.

Abstract:Brain edema leading to an expansion of brain volume has a crucial impact on morbidity and mortality following traumatic brain injury as it increases intracranial pressure, impairs cerebral perfusion and oxygenation, and contributes to additional ischemic injuries. Classically, two major types of traumatic brain edema exist: "vasogenic" and "cytotoxic/cellular". However, the cellular and molecular mechanisms contributing to the development/resolution of traumatic brain edema are poorly understood and no effective drugs can be used now. Aquaporin-4 (AQP4) is a water-channel protein expressed strongly in the brain, predominantly in astrocyte foot processes at the borders between the brain parenchyma and major fluid compartments, including cerebrospinal fluid and blood. This distribution suggests that AQP4 controls water fluxes into and out of the brain parenchyma. In cytotoxic edema, AQP4 deletion slows the rate of water entry into brain, whereas in vasogenic edema, AQP4 deletion reduces the rate of water outflow from brain parenchyma. AQP4 has been proposed as a novel drug target in brain edema. These findings suggest that modulation of AQP4 expression or function may be beneficial in traumatic brain edema.
Research about Relationship between AQP4 and Brain Edema Following Intracerebral Hemorrhage

Abstract:The purpose of this study was to examine the role of AQP4 in edema formation after intracerebral hemorrhage(ICH) by using AQP4-/- mice. Methods: ICH was induced by microinjecting 5 μL autologous whole blood into the striatum of AQP4+/+ and AQP4-/- mice. Neurological deficits, brain edema contents of whole hemorrhagic ipsilateral hemisphere, specific gravity of brain tissue surrounding hematoma, Evans blue leakage and ultrastructure of brain microvessels were compared between AQP4+/+ and AQP4-/- mice following ICH. Results: Our experiments showed a significant increase of AQP4 expression following ICH in AQP4+/+ mice. AQP4 deletion aggravated neurological deficits and brain edema contents of whole hemorrhagic ipsilateral hemisphere. Besides, it also reduced the specific gravity of brain tissue surrounding hematoma. Moreover, it enhanced Evans blue leakage and ultrastructure of brain microvessel damage. Conclusion: These results suggest that AQP4 deletion increases ICH damage, including edema formation, blood-brain barrier damage. Further studies on the protective role of activated AQP4 expression following ICH may provide useful therapeutic target for ICH-induced brain injury.
Curcumin Attenuates Brain Edema in Mice with Intracerebral Hemorrhage Through Inhibition of Aqp4 and Aqp9 Expression

Abstract:Aim: Aquaporins (AQPs) are the water-channels that play important roles in brain water homeostasis and in cerebral edema induced by brain injury. In this study we investigated the relationship between AQPs and a neuroprotective agent curcumin that was effective in the treatment of brain edema in mice with intracerebral hemorrhage (ICH). Methods: ICH was induced in mice by autologous blood infusion. The mice immediately received curcumin (75, 150, 300 mg/kg, ip). The Rotarod test scores, brain water content and brain expression of AQPs were measured post ICH. Cultured primary mouse astrocytes were used for in vitro experiments. The expression of AQP1, AQP4 and AQP9 and NF-κB p65 were detected using Western blotting or immunochemistry staining. Results: Curcumin administration dose-dependently reduced the cerebral edema at d 3 post ICH, and significantly attenuated the neurological deficits at d 5 post ICH. Furthermore, curcumin dose-dependently decreased the gene and protein expression of AQP4 and AQP9, but not AQP1 post ICH. Treatment of the cultured astrocytes with Fe 2+ (10–100 μmol/L) dose-dependently increased the expression and nuclear translocation of NF-κB p65 and the expression of AQP4 and AQP9, which were partly blocked by co-treatment with curcumin (20 μmol/L) or the NF-κB inhibitor PDTC (10 μmol/L). Conclusion: Curcumin effectively attenuates brain edema in mice with ICH through inhibition of the NF-κB pathway and subsequently the expression of AQP4 and AQP9. Curcumin may serve as a potential therapeutic agent for ICH.

Abstract:Vascular endothelial growth factor (VEGF) has protective effects on many neurological diseases. However, whether VEGF acts on brain edema following intracerebral hemorrhage (ICH) is largely unknown. Our previous study has shown aquaporin-4 (AQP4) plays an important role in brain edema elimination following ICH. Meanwhile, there is close relationship between VEGF and AQP4. In this study, we aimed to test effects of VEGF on brain edema following ICH and examine whether they were AQP4 dependent. Recombinant human VEGF165 (rhVEGF165) was injected intracerebroventricularly 1 d after ICH induced by microinjecting autologous whole blood into striatum. We detected perihemotomal AQP4 protein expression, then examined the effects of rhVEGF165 on perihemotomal brain edema at 1 d, 3 d, and 7 d after injection in wild type (AQP4(+/+)) and AQP4 knock-out (AQP4(-/-)) mice. Furthermore, we assessed the possible signal transduction pathways activated by VEGF to regulate AQP4 expression via astrocyte cultures. We found perihemotomal AQP4 protein expression was highly increased by rhVEGF165. RhVEGF165 alleviated perihemotomal brain edema in AQP4(+/+) mice at each time point, but had no effect on AQP4(-/-) mice. Perihemotomal EB extravasation was increased by rhVEGF165 in AQP4(-/-) mice, but not AQP4(+/+) mice. RhVEGF165 reduced neurological deficits and increased Nissl's staining cells surrounding hemotoma in both types of mice and these effects were related to AQP4. RhVEGF165 up-regulated phospharylation of C-Jun amino-terminal kinase (p-JNK) and extracellular signal-regulated kinase (p-ERK) and AQP4 protein in cultured astrocytes. The latter was inhibited by JNK and ERK inhibitors. In conclusion, VEGF reduces neurological deficits, brain edema, and neuronal death surrounding hemotoma but has no influence on BBB permeability. These effects are closely related to AQP4 up-regulation, possibly through activating JNK and ERK pathways. The current study may present new insights to treatment of brain edema following ICH.
Carvacrol decreases blood-brain barrier permeability post-diffuse traumatic brain injury in rats

Abstract:Previously, we showed that Satureja Khuzestanica Jamzad essential oil (SKEO) and its major component, carvacrol (CAR), 5-isopropyl-2-methylphenol, has anti-inflammatory, anti-apoptotic, and anti-edematous properties after experimental traumatic brain injury (TBI) in rats. CAR, predominantly found in Lamiaceae family (Satureja and Oregano), is lipophilic, allowing diffusion across the blood-brain barrier (BBB). These experiments test the hypothesis that acute treatment with CAR after TBI can attenuate oxidative stress and BBB permeability associated with CAR's anti-edematous traits. Rats were divided into six groups and injured using Marmarou weight drop: Sham, TBI, TBI + Vehicle, TBI + CAR (100 and 200 mg/kg) and CAR200-naive treated rats. Intraperitoneal injection of vehicle or CAR was administered thirty minutes after TBI induction. 24 h post-injury, brain edema, BBB permeability, BBB-related protein levels, and oxidative capacity were measured. Data showed CAR 200 mg/kg treatment decreased brain edema and prevented BBB permeability. CAR200 decreased malondialdehyde (MDA) and reactive oxygen species (ROS) and increased superoxide dismutase (SOD) and total antioxidative capacity (T-AOC), indicating the mechanism of BBB protection is, in part, through antioxidant activity. Also, CAR 200 mg/kg treatment suppressed matrix metalloproteinase-9 (MMP-9) expression and increased ZO-1, occludin, and claudin-5 levels. These data indicate that CAR can promote antioxidant activity and decrease post-injury BBB permeability, further supporting CAR as a potential early therapeutic intervention that is inexpensive and more readily available worldwide. However, more experiments are required to determine CAR's long-term impact on TBI pathophysiology.
Curcumin Alleviates Brain Edema by Lowering AQP4 Expression Levels in a Rat Model of Hypoxia-Hypercapnia-induced Brain Damage.

Abstract:The present study aimed to investigate the therapeutic effects of curcumin (CU) against brain edema in a rat model of hypoxia-hypercapnia (HH)-induced brain damage (HHBD). Male Sprague-Dawley rats were divided into five groups, including a control group and four treatment groups. The rats in the control group were raised under normal laboratory conditions and were injected with water, whereas the rats in the treatment groups were exposed to a low O2/high CO2 environment simulating HH conditions, and were injected with water, CU, dimethyl sulfoxide (solvent control) or monosialoganglioside GM1. After 2 weeks, the morphological characteristics of the brain tissues were analyzed using optical and electron microscopy. In addition, aquaporin (AQP)-4 protein expression levels in brain tissue samples were analyzed using streptavidin-biotin complex immunohistochemistry and western blotting, and mRNA expression levels were detected using reverse transcription-quantitative polymerase chain reaction. Severe brain edema, tissue structure disruption and increased AQP4 expression levels were detected in the brain tissues of the HH rats. Conversely, the rats treated with CU or GM1 exhibited attenuated HHBD-induced brain edema and tissue structure disruption, and decreased mRNA and protein expression levels of AQP4. The results of the present study suggested that CU treatment was able to attenuate HHBD-induced brain edema by downregulating the expression levels of AQP4 in a rat model. Therefore, CU may be considered a potential therapeutic drug for the treatment of patients with brain edema.
Effects of hyperbaric oxygen on brain edema and expression of aquaporin-4 surrounding brain tissue of hemorrhagic focus following experimental intracerebral hemorrhage in two kinds of rat models

Abstract:Objective:To study the effects of hyperbaric oxygen(HBO) on brain edema and expression of aquaporin-4(AQP-4) surrounding brain tissue of hemorrhagic focus following experimental intracerebral hemorrhage(ICH) in two kinds of rat models.Method:A total of 126 rats were randomly divided into five groups:sham-operated group(SHG,6 rats);control-1 group(autologous blood induced intracerebral hemorrhage model,A-ICH,30 rats),control-2 group(collagenase-induced intracerebral hemorrhage model,C-ICH,30 rats),trial-1 group(A-ICH+HBO,30 rats),trial-2 group(C-ICH+ HBO,30 rats).HBO therapy was intervened at 24h after operation,once a day.All rats were sacrificed at 24h,48h,72h,5d,7d post operation(each time point 6 rats).Brain edema and expression of AQP-4 were tested.Result:The success rates of A-ICH model and C-ICH model were 65% and 75% respectively.Brain edema content of A-ICH,C-ICH,A-ICH+HBO and C-ICH+HBO groups were significant higher than that of SHG group(P 0.05).The cerebral edema of A-ICH+HBO and C-ICH+HBO groups alleviated obviously compared with A-ICH and C-ICH groups,and the difference was significant respectively at 48h,72h,5d,7d and 48h,5d post operation(P 0.05).The brain edema of A-ICH+HBO group alleviated more early and obviously than that of C-ICH+HBO group,especially at 72h post operation(P0.05).The expression of AQP-4 appeared in all rats brain tissue,but expression of AQP-4 in SHG group was lower.That of A-ICH and C-ICH groups started rising at 24h post operation,the peak appeared at 48h,then dropped at 72h,but there were still significant different compared with SHG group at 7d post operation(P0.05).The difference between A-ICH group and C-ICH group was significant at 72h post operation(P0.05),and the difference between A-ICH+HBO group and A-ICH group was significant at all time points post operation(P0.05).but the difference between C-ICH+HBO group and C-ICH group was significant only at 48h and 72h post operation(P0.05).Compared with C-ICH +HBO group,expression of AQP-4 in A-ICH+HBO group was more obvious and early,especially at 48h and 72h post operation(P0.05).Conclusion:HBO might play neuroprotection role by relieving brain edema and down-regulating AQP-4 expression.A-ICH model was more fit for brain edema study of ICH rat.

Abstract:Cerebrovascular diseases are conditions caused by problems with brain vasculature, which have a high morbidity and mortality. Aquaporin-4 (AQP4) is the most abundant water channel in the brain and crucial for the formation and resolution of brain edema. Considering brain edema is an important pathophysiological change after stoke, AQP4 is destined to have close relation with cerebrovascular diseases. However, this relation is not limited to brain edema due to other biological effects elicited by AQP4. Till now, multiple studies have investigated roles of AQP4 in cerebrovascular diseases. This review focuses on expression of AQP4 and the effects of AQP4 on brain edema and neural cells injuries in cerebrovascular diseases including cerebral ischemia, intracerebral hemorrhage and subarachnoid hemorrhage. In the current review, we pay more attention to the studies of recent years directly from cerebrovascular diseases animal models or patients, especially those using AQP4 gene knockout mice. This review also elucidates the potential of AQP4as an excellent therapeutic target.
The cornel Iridoid glycoside attenuated brain edema of the cerebral ischemia/reperfusion rats by modulating the polarized aquaporin 4

Abstract:Brain edema after ischemic stroke could worsen cerebral injury in patients who received intravenous thrombolysis. Cornus officinalis Sieb. et Zucc., a long-established traditional Chinese medicine, is beneficial to the treatment of neurodegenerative diseases including ischemic stroke. In particular, its major component, cornel iridoid glycoside (CIG), was evidenced to exhibit neuroprotective effects against cerebral ischemic/reperfusion injury (CIR/I). Aimed to explore the effects of the CIG on brain edema of the CIR/I rats, the CIG was analyzed with the main constituents by using HPLC. The molecular docking analysis was performed between the CIG constituents and AQP4-M23. TGN-020, an AQP4 inhibitor, was used as a comparison. In the in vivo experiments, the rats were pre-treated with the CIG and were injured by performing middle cerebral artery occlusion/reperfusion (MCAO/R). After 24 h, the rats were examined for neurological function, pathological changes, brain edema, and polarized Aqp4 expressions in the brain. The HPLC analysis indicated that the CIG was composed of morroniside and loganin. The molecular docking analysis showed that both morroniside and loganin displayed lower binding energies to AQP4-M23 than TGN-020. The CIG pre-treated rats exhibited fewer neurological function deficits, minimized brain swelling, and reduced lesion volumes compared to the MCAO/R rats. In the peri-infarct and infarct regions, the CIG pre-treatment restored the polarized Aqp4 expression which was lost in the MCAO/R rats. The results suggested that the CIG could attenuate brain edema of the cerebral ischemia/reperfusion rats by modulating the polarized Aqp4 through the interaction of AQP4-M23 with morroniside and loganin.

Abstract:Aquaporin4(AQP4),a member of the aquaporin family,is mainly expressed inastrocytes end-feet in the central nervous system. A large number of experimental studies have show n that AQP4 expression plays an important role in the occurrence, development, and regression of brain edema after different types of stroke. In addition, the AQP4 expression can affect the development process of cerebrovascular disease through the mechanisms such as affecting the integrity of the blood-brain barrier and promoting astrocyte migration, nerve regeneration, and neuroinflammatory response. Investigation of regulation mechanisms of AQP4 in transmembrane transport of substance in brain and intracel ular and extracel ular environmental balance and its expression in model of cerebrovascular disease have an important significance for understanding the occurrence, development, protection, and treatment of clinical cerebrovascular disease.
Effect of Deferoxamine on Aquaporin-4 Expression in a Rat Model of Intracerebral Haemorrhage

Abstract:Objective To study the dynamic changes of AQP4 expression between Intracerebral Haemorrhage(ICH)models and Deferoxamine(DFO) intervention models.To explore the mechanism and role of AQP4 and DFO in edema formation after ICH.Methods ICH was established by infusing autologous blood into rat putamina;Immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR) method were respectively used to evaluate AQP4 protein and mRNA expression in different group and distinct period of time.Results AQP4 positive cells mostly located in astrocyte in the perivascular glial feet processes in the brain.As compared with the sham operation groups,there appeared a significant increase of AQP4 protein expression in all ICH models and DFO intervention models;At 7 day,the AQP4 protein expression reached its maximum in ICH models and DFO intervention models;At 7 and 14 day,in DFO groups AQP4 protein expression decreased remarkably than ICH groups.There showed a significant enhanced expression of AQP4 mRNA in all ICH models and DFO intervention models at 3,7 day compared with sham operation controls;At 3 day,AQP4 mRNA expression reached its maximum in ICH models and DFO intervention models;AQP4 mRNA expression decreased significantly in DFO models than ICH groups;There were descending trend in DFO models with the time extended;there were no differences between DFO models and controls at 14 day.There were positive correlations with AQP4 mRNA and AQP4 protein expression.Conclusions AQP4 may play a key role in edema formation and absorption after ICH,DFO was likely to be a new effective agent to intervene brain edema formation after ICH.

Abstract:Objective: To explore the protective effects of Yangyin Yiqi Huoxue infusion (YYYQHXI，养阴益气活血冲剂) on the cerebral ischemiareperfusion injury (CIRI).Methods:The rat models with CIRI were prepared and randomly divided into shamopera tive group,CIRI group,ischemia group,treated group with different dosage of YYYQ HXI,and venoruton control group.The changes in following parameters,including the contents of prostacyclin I2 (PGI２) and thromboxin A2 (TXA2) in brain tissue,the activites of superoxi de dismutase (SOD) and the contents of malonyldialdehyde (MDA) in even pulp of br ain and the activities of tissue plasminogen activator (tPA) and its inhib itor (PAI) were observed.Results:YYYQHXI was able to increase the contents of PGI2 and decrease the contents of TXA2 in brain tissue obviously,to regulate the activities of plasma tP A and PAI,to reduce the content of MDA obviously,and to heighten the activity of SOD in brain tissue.Conclusions:YYYQHXI possesses anticoagulative,increasing fibrolytic,promoting anti oxidative,and additional effects so that it has significantly protective actions on CIRI.
Edaravone Attenuates Brain Edema and Injury by Down-Regulating Expressions of P38 Mitogen-Activated Protein Kinase and Aquaporin-4 after Focal Cerebral Ischemia and Reperfusion in Mice

Abstract:Objective To investigate the effects on brain tissue p38 mitogen-activated protein kinase (MAPK) and aquaporin 4 (AQP4) and neuroprotective effect of edarvone after focal cerebral ischemia and reperfusion in mice.Methods A total of 196 healthy male Kunming mice were randomly divided into four groups:a sham operation group,an ischemia-reperfusion group,a saline control group,and an edaravone group (n =49 in each group).A middle cerebral artery occlusion (MCAO) mothod was used to induce a cerebral ischemia-reperfusion model.At 2 h after ischemia,immediately after reperfusion in the edaravone group and the saline control group,edaravone (5 mg/kg) and the same volume of saline were injected intraperitoneally in mice,then repeated once every 24 h.At 2 h after MCAO,the brain water content and infarct volume at different time points after reperfusion (12 h,24 h,48 h,and 3 d) were measured respectively.At 24 h after MCAO,the expressions of AQP4 and p38 MAPK in the brain tissue of ischemic peripheral cortex were measured by Western blotting.Results The volumes of cerebral infarction (all P ＜ 0.01) and the brain water contents (all P ＜0.05) in the edaravone group were decreased than those in the ischemia-reperfusion group and saline control group at different time points,and they were most significant at 48 h.After 24-h reperfusion,the expression levels of AQP4 (0.985 ± 0.129,1.024 ± 0.117,0.713 ± 0.231) and phospho-p38 MAPK (1.123 ± 0.142,1.214 ± 0.096,0.986 ± 0.087) in the brain tissue of ischemic peripheral cortex in the ischemia-reperfusion group,the saline control group,and the edaravone group were upregulated significantly than those in the sham operation group (AQP4:0.265 ± 0.123;phospho-p38 MAPK:0.465 ±0.023;all P ＜0.01),but edaravone group were significantly lower than the ischemia-reperfusion group and the saline control group (all P ＜ 0.05).Conclusions Edaravone can downregulate the expression level of AQP4 and effectively protect cerebral ischemia reperfusion injury in mice,Its mechanism may be associated with the inhibition of p38 MAPK pathway.

Abstract:Iron overload and AQP4 may play a critical role in the formation of brain edema after ICH. In addition, AQP4 expression was affected by iron concentration. Importantly, treatment with DFO significantly reduced brain edema in rats and inhibited the AQP4 upregulation after ICH. Deferoxamine may be a potential therapeutic agent for treating ICH.
Effects of Chemical Synthesized Curcumin Preconditioning on the Expression of AQP-4 and Cerebral Edema after Focal Cerebral Ischemia/reperfusion Damage in Rats

Abstract:Aim To explore the effect of chemical synthesized curcumin(Cur) preconditioning on the expression of aquaporin 4(AQP-4) and cerebral edema after focal cerebral ischemia-reperfusion damage in rats.Methods Local ischemia/reperfusion model of middle cerebral artery occlusion(MCAO) was made by inserting a nylon thread into internal carotid artery in rats.72 healthy SD rats weighing 290~310 g were randomly divided into 3 groups: sham operated group(S)(n=18),ischemia/reperfusion group(I)(n=18),Cur group(C1 and C2)(n=18) which were preconditioned with cur 50 or 100 mg·kg-1 intraperitoneally 30 min before focal cerebral ischemia-reperfusion.In I group,NS was given instead of Cur and in S group,the carotid arteries were exposed without performing MCAO.The rats in I group and C1,C2 group were killed 24 hours after cerebral ischemia/reperfusion.Neurological scores were studied,method of dry/wet weight was used to observe water content,and Evans Blue(EB) was collected to observe the changes of blood-brain barrier(BBB) permeability.Expression of AQP-4 and activation of c-Jun N-terminal kinase(JNK) in rat brain were determined by Western blot.Results Sham operated group presented no neurological changes.The water content and EB in C1 and C2 group were significantly lower than those of Ⅰ group(P0.05).Curcumin could also alleviate the expression of AQP-4 and the phosphorylation of JNK(P0.05).The neurological deficiency scores in C1 and C2 groups were obviously higher than those of I group(P0.05).Conclusions Chemical synthesized curcumin can alleviate the expression of AQP-4 and the phosphorylation of JNK,therefore protect the structure of BBB and alleviate the cerebral edema which can exert the neuroprotective effect after focal cerebral ischemia and reperfusion.

Abstract:Objective: Cerebral ischemia is the most common type of neuronal injury and is characterized by a reduction in the function and number of hippocampal neurons. Carvacrol has a significant neuroprotective effect in cerebral ischemia. However, the mechanisms by which carvacrol affects cerebral ischemia, especially with respect to the regulation of neuronal damage by iron levels, have never been systematically studied. This study aimed to reveal the mechanisms by which carvacrol protects against hippocampal neuron impairment after ischemic stroke in gerbils. Materials and methods: The Morris water maze test was performed to evaluate learning and memory impairments. Iron ion content and oxidative stress index were detected by the kit. MTT assay was performed to assess the cell viability. The morphology and molecular characteristics were detected by electron micrographs and western blot. Results: In the present study, we demonstrated the neuroprotective effects of carvacrol in vivo and in vitro. The Morris water maze test showed that the learning and memory abilities of the gerbils treated with carvacrol were significantly improved. Lipid peroxide injury was evaluated by measuring the levels of lipid peroxide bio-markers; the results indicated that carvacrol decreased the level of lipid peroxide in ischemic gerbil brain tissue. Histopathological examinations and western blotting were performed to evaluate injury in neurons, and carvacrol reduced cell death. Moreover, ferroptosis in the hippocampus was evaluated by measuring the levels of proteins involved in this iron-dependent form of regulated cell death. These results indicated that carvacrol reduced cell death and that carvacrol inhibited ferroptosis by increasing the expression of glutathione peroxidase 4(GPx4). This study showed that carvacrol may be a valuable drug for treating cerebral ischemia. Conclusion: Carvacrol provides protection for hippocampal neurons against I/R in gerbils by inhibiting ferroptosis through increasing the expression of GPx4.
Relationship between aquaporin-4 expression and blood-brain barrier damage after acute experimental intracerebral hemorrhage

Abstract:Objective: To investigate the relationship between Aquaporin-4 (AQP-4) expression and blood-brain barrier damage experimentally after acute intracerebral hemorrhage. Methods: The intracerebral hemorrhage (ICH) was established by infusing autoblood into rat caudate nucleus. Brain water content and Evan blue were used to confirm edema, BBB damage. RT-PCR and Western blot were respectively used to evaluate AQP-4 mRNA and protein expression and to observe the correlation between AQP-4 and tight junction protein ZO-1, occludin. Electron microscope was used to explore the level of BBB tight junction damage and astrocyte foot process swelling. Results: Electron microscope showed astrocyte foot process swollen without BBB tight junction damage at the 6th hour after hemorrhage. As compared with the sham group, there appeared a significant increase in AQP-4 mRNA and protein expression following ICH (P < 0.05): expression of AQP-4 mRNA appeared increased at the 3rd hour (1.05 ± 0.13) and protein appeared increased (0.62 ± 0.05) at the 6th hour following the hemorrhage. Expression of AQP4 mRNA and protein reached its maximum at the 72th hour (1.46 ± 0.02). Evan blue and electron microscope revealed BBB tight junction began to damage at the 12th hour following the hemorrhage. At the same time, expression of ZO-1, occludin (0.71 ± 0.05 and 0.72 ± 0.04, respectively) was decreased and shown a negative correlation with expression of AQP-4. Conclusions: BBB damage might have negative correlation with expression of AQP-4 following hemorrhage. Increased expression of AQP-4 and astrocyte foot process swollen might be one of the early cause of BBB damage.

Abstract:Cerebral edema contributes significantly to morbidity and death associated with many common neurological disorders. However, current treatment options are limited to hyperosmolar agents and surgical decompression, therapies introduced more than 70 years ago. Here we show that mice deficient in aquaporin-4 (AQP4), a glial membrane water channel, have much better survival than wild-type mice in a model of brain edema caused by acute water intoxication. Brain tissue water content and swelling of pericapillary astrocytic foot processes in AQP4-deficient mice were significantly reduced. In another model of brain edema, focal ischemic stroke produced by middle cerebral artery occlusion, AQP4-deficient mice had improved neurological outcome. Cerebral edema, as measured by percentage of hemispheric enlargement at 24 h, was decreased by 35% in AQP4-deficient mice. These results implicate a key role for AQP4 in modulating brain water transport, and suggest that AQP4 inhibition may provide a new therapeutic option for reducing brain edema in a wide variety of cerebral disorders.
