EFFECTS OF SALT AND ALKALI STRESS ON DIFFERENTIAL EXPRESSION OF GENES IN RICE SEEDLINGS

Abstract:To characterize both individual and combinational effects of salt, alkaline concentration and different pH values on six salt and alkaline stress - related genes isolated via DDRT - PCR from rice by Liu et al (2000), fourteen - day - old seedlings of Nipponbare ( Oryza sativa L. subsp. japonica) were exposed to NaHCO3 (60 mmol/L, pH 8.50) in 6 h, 12 h, 24 h,48 h, as well as to NaCl (100 mmol/L), NaHCO3 (60 mmol/L) and Na2CO3 (30 mmol/L) in 24 h with pH values of 4.50,6.50, 8.50 and 10.50, followed by Northern blot analysis in the shoot. The results showed that, compared with that treated by H2O ( pH 6.50), mitochondrial ATPase 6 × 103 subunit was 2.73, 0.82, 0.01, 1.79 fold higher in 6 h, 12 h, 24 h, 48 h by NaHCO3 (60 mmol/L, pH 8.50). The transcription of HSP90 and CA were significantly repressed in 6 h later, transcript of UDPG - GT was enhanced in 6 h, but expression of thoredoxin was slightly repressed under stress treatment condition. Under 24 h and different pH value treatments, NaCl (60 mmol/L, pH 8.50), NaHCO3 and Na2 CO3 treatments between acidic (pH 4.50) and alkaline ( pH 8.50 and 10.50) obviously increased the expression of mitochondrial ATPase 6 × 103subunit, CA was coordinately influenced by salt, alkaline concentration and pH values, thioredoxin and cAPX were mainly affected by salt and pH, and UDPG - GT transcript reached the peak in Na2CO3 treatment. And low pH value increased HSP90 expression, however high pH could repress its expression. Fig 6, Tab 2, Ref 13
