Abstract:This study was conducted to investigate the importance of direct contact between Kupffer cells (KCs) and hepatocytes (HCs) during the hepatic inflammatory responses, and the effect of leflunomide's active metabolite, A771726, on cytokines in KCs and HCs (DC cocultures) and KC cultures using an in vitro approach. Lipopolysaccharide (LPS)-induced inflammatory response in monocultures of rats HCs and KCs were compared with DC cocultures. Tumor necrosis factor-α (TNF-α) and interleukin-1 (IL-1) concentrations of different culture supernatants were measured with ELISA. TNF-α and IL-1 mRNA in KCs of inflammatory liver injury was analyzed with reverse transcription polymerase chain reaction (RT-PCR). Our data showed that DC cocultures exhibited the highest production of TNF-α and IL-1 compared with other cultures, and these cytokines were mainly produced by KCs, in particular activated KCs. Time course studies revealed an increased production of TNF-α preceding the IL-1 production, suggesting that increased TNF-α levels could be involved in the increased IL-1 production. Leflunomide's active metabolite, A771726, has significantly inhibitory effect on TNF-α and IL-1 at protein and transcription levels, and reduced production of IL-1 by A771726 was associated with inhibitory action of A771726 on TNF-α. These results provided evidence that leflunomide significantly inhibited TNF-α and IL-1 from KCs.

Abstract:Objective To investigate the effect of A771726 on the immune function of peritoneal macrophages(PMФ ) in collagen-induced arthritis (CIA) and its potential mechanism.Methods Chicken type Ⅱcollagen (CⅡ) was used to induce CIA in rats.Peritoneal macrophages suspensions of CIA rats were prepared in asepsis and incubated with A771726 (0.1,1,10 μmol/L) in vitro.Interleukin-1 (IL-1) was measured by mouse thymocyte proliferation assay.Oxidative stress was assessed by chemical methods.The expression of TLR4 mRNA of PMФ in CIA rats were detected by reverse transcription polymerase chain reaction(RT-PCR) analysis. Results A771726 (1,10 μmol/L)significantly inhibited the secretion function of PMФ in CIA rats.A771726 (1,10 μmol/L) markedly ameliorate oxidative stress in PMΦ of CIA rats. In addition,A771726(1,10 μmol/L) significantly inhibited the expression of TLR4 mRNA of PMФ in CIA rats.Conclusion A771726 (1,10 μmol/L) can regulate the secretion function and oxidative stress,and advanced investigation. Iits mechanism whether or not be related to inhibitory level of TLR4mRNA of PMФ in CIA rats would be performed in future.

Abstract:AIM: To study the effect of leflunomide on CCl4-induced hepatic fibrosis in rats. METHODS: Hepatic fibrosis was induced by subcutaneous injection with 50 % CCl4, in Sprague-Dawley rats. The amount of CCl4 administered was 1 mg/kg. The alanine aminotransferase (ALT), aspartate aminotransferase (AST), nitric oxide (NO) levels in plasma and hydroxyproline (Hyp) contents in liver tissue were assayed by spectrophotometry. The hyaluronic acid (HA) and procollagen III (PC 111) were assessed by radioimmunoassay. The transforming growth factor-beta1 (TGF-beta1) in serum was determined by ELISA. The nuclear factor-kappa B (NF-kappaB) in liver tissue was examined by immunohistochemistry. Liver samples collected after 12 weeks of CCl4 treatment were stained with hematoxylin and eosin. RESULTS: Leflunomide (1, 3, and 9 mg/kg) significantly decreased indices of liver and spleen, the serum transaminase (AST, ALT) activities, HA and PC III levels, and Hyp contents in liver tissue in rats of hepatic fibrosis. Histopathological examination showed leflunomide had inhibitory effect on fibrogenesis and formation of pseudolobulus. Furthermore, leflunomide significantly inhibited NF-kappaB expression in liver tissue, and reduced elevated serum TGF-beta1 and NO levels in rats of hepatic fibrosis. CONCLUSION: Leflunomide showed inhibitory action on hepatic fibrosis induced by CCl4 in rats.
In vitro effect of A771726 on the secretion of nitric oxide of peritoneal macrophages from collagen-induced arthritis rats

Abstract:AIM: To study the effect and mechanism of A771726,the active metabolite of leflunomide,on the production of nitric oxide(NO) of peritoneal macrophages(PMФ) from collagen-induced arthritis(CIA) rats.METHODS: Collagen-induced arthritis(CIA) model was induced in rats and the PMФ were collected.A771726 were used in vitro.The dose-dependent and time-dependent effects of A771726 on the secretion of NO of PMФ from CIA rats were checked by nitric acid reduction synthase method;MTT reagent was used to detect the effect of A771726 on the cell viability of PMФ from CIA rats;RT-PCR was used to detect the expression of inducible NO synthase(iNOS) mRNA.Western blot was used to detect the expression of iNOS protein.RESULTS: A771726 could inhibit the secretion of NO of PMФ from CIA rats in a dose-dependent and time-dependent manner.The cytotoxicity of A771726 was not detected during the time course and dose range.A771726(0.1,1,10 mol/L) significantly inhibited the expression of iNOS mRNA and iNOS protein in PMФ from CIA rats.CONCLUSION: A771726(0.1,1,10 mol/L) can inhibit the transcription of iNOS mRNA in PMФ from CIA rats,and reduce the translation of iNOS protein,which leads to suppressing the secretion of NO.And the suppression is not related to the cytotoxicity of A771726.
Fluorofenidone Attenuates Hepatic Fibrosis By Suppressing The Proliferation And Activation Of Hepatic Stellate Cells

Abstract:Fluorofenidone (AKF-PD) is a novel pyridone agent. The purpose of this study is to investigate the inhibitory effects of AKF-PD on liver fibrosis in rats and the involved molecular mechanism related to hepatic stellate cells (HSCs). Rats treated with dimethylnitrosamine or CCl4 were randomly divided into normal, model, AKF-PD treatment, and pirfenidone (PFD) treatment groups. The isolated primary rat HSCs were treated with AKF-PD and PFD respectively. Cell proliferation and cell cycle distribution were analyzed by bromodeoxyuridine and flow cytometry, respectively. The expression of collagen I and alpha-smooth muscle actin (alpha-SMA) were determined by Western blot, immunohistochemical staining, and real-time RT-PCR. The expression of cyclin D1, cyclin E, and p27(kip1) and phosphorylation of MEK, ERK, Akt, and 70-kDa ribosomal S6 kinase (p70S6K) were detected by Western blot. AKF-PD significantly inhibited PDGF-BB-induced HSC proliferation and activation by attenuating the expression of collagen I and alpha-SMA, causing G0/G1 phase cell cycle arrest, reducing expression of cyclin D-1 and cyclin E, and promoting expression of p27(kip1). AKF-PD also downregulated PDGF-BB-induced MEK, ERK, Akt, and p70S6K phosphorylation in HSCs. In rat liver fibrosis, AKF-PD alleviated hepatic fibrosis by decreasing necroinflammatory score and semiquantitative score, and reducing expression of collagen I and alpha-SMA. AKF-PD attenuated the progression of hepatic fibrosis by suppressing HSCs proliferation and activation via the ERK/MAPK and PI3K/Akt signaling pathways. AKF-PD may be used as a potential novel therapeutic agent against liver fibrosis.

Abstract:Objective:To evaluate the Turtle Carapace Decoction and drug containing serum on hepatic,stellate cells(HSC).Methods;Double- antibody sandwich ELISA assay was used to detect the effect of Turtle Carapace Decoction and drug containing serum on TGF- Bl- induced collagen and regulatory factors expression in LX-1 cells;MTS assay was used to detect the effect of Turtle Carapace Decoction and drug containing serum on TGF- Bl- induced LX- 1 cell proliferation;Western blotting analysis was used to detect the effect of Turtle Carapace Decoction and drug containing serum on TGF- Bl- induced LX-1 cell activation,collagen synthesis and extracellular matrix expression.Results;Turtle Carapace Decoction and drug containing serum can significantly inhibit HSC proliferation and down- regulate TGF- Bl- induced type I,type III and type IV collagen,α- SMA and HA expressions in LX- 1 cells,and for the TGF- Bl downstream signal pathway proteins,it can inhibit the expressions of Smad 3,P- smad3 and other cytokines,which act on the TGF- Bl feedback loop in all stages of blocking TGF- Bl.Moreover,Turtle Carapace Decoction and drug containing serum can significantly strengthen the expressions of MMP- 1 and MMP-3 proteins.By inhibiting TIMP- 1 protein expression,it was conducive to improve collagen enzyme activity,and to promote ECM degradation.Within the experimental concentrations,the effect of Turtle Carapace Decoction and drug containing serum on hepatic stellate cells was in a dose-dependent manner,and this effect was significantly better than those of-γ- interferon,and was comparable to those of Fuzheng Huayu capsule,and generally accepted effective anti- liver fibrosis drugs or cytokine.Conclusion;Turtle Carapace Decoction and drug containing serum can inhibit hepatic stellate cells activated multiplication and extra cellular matrix(ECM) synthesis or secretion,promote ECM degradation,regulate cell factor level and regulate signaling pathway,and significantly block or treat liver fibrosis.This experimental result is verifying the validity of turtle carapace for anti- liver fibrosis,providing scientific evidences and research ideas for basic investigation on material basis for its anti-liver fibrosis effect as well as development and clinical application of its active components.
Inhibiting Effects of Leflunomide Metabolite on Overexpression of CD147, MMP-2 and MMP-9 in PMA Differentiated THP-1 Cells.

Abstract:Recent studies have reported elevated expression of cluster of differentiation (CD) 147 on CD14(+) monocytes of the peripheral blood of patients with active rheumatoid arthritis and a correlation of CD147 expression with Disease Activity Score. Thus, CD147 may be a new target for treatment of rheumatoid arthritis. Leflunomide is a disease-modifying antirheumatic drug that is commonly used to treat rheumatoid arthritis. The effect of leflunomide in blocking the up-regulation of CD147 and in blocking the down-regulation of metalloproteinases (MMP)-2 and MMP-9 in active macrophages has not yet been established. In this study we investigated the effect of A771726, the active metabolite of leflunomide, on expression of CD147 and on the gelatinolytic activity of MMP-2 and MMP-9 in phorbol myristate acetate (PMA) differentiated THP-1 cells. The expression of CD147, MMP-2, and MMP-9 mRNAs were determined by real-time quantitative reverse transcription PCR, the levels of cellular surface expression of CD147 were determined by flow cytometry, and the gelatinolytic activity of MMP-2 and MMP-9 were determined by zymography. Our results showed that A771726 significantly inhibited the expression of CD147 on the cell surface of activated THP-1 cells in a dose-dependent manner (P<0.01), inhibited the expression of MMP-2 and MMP-9 mRNAs in a dose-dependent manner (P<0.01), and inhibited the gelatinolytic activity of MMP-2 and MMP-9 at concentration of 15 μg/ml and 45 μg/ml (P<0.01). Our results indicate that A771726, the active metabolite of leflunomide, inhibited CD147 expression at the protein level and inhibited gelatinolytic activity of MMP-2 and MMP-9 in PMA-differentiated THP-1 cells.
The Inhibitory Effects of Hjc0416 on Liver Fibrogenesis in Activated Hepatic Stellate Cells

Abstract:Introduction The STAT3 signaling pathway regulates activation of hepatic stellate cells (HSCs) and promotes its fibrogenic behavior. Using a fragment‐based drug design approach, our group has developed HJC0416, a novel orally bioavailable small‐molecule inhibitor of STAT3. HJC0416 was found to induce apoptosis and decreased tumor growth in breast cancer models. In the present study, we studied the antifibrogenic properties of HJC0416 in activated hepatic stellate cells (HSC), which are the major effector cells of liver fibrosis. Methods Human (LX‐2) and rat (HSC‐T6) hepatic stellate cell lines were used. Cell proliferation was measured by Alamar Blue assay. Cellular protein expression levels were determined by Western blotting and immunofluorescence. Cell cycle was assessed by flow cytometry. Yo‐pro‐1 staining was used to measure apoptosis. Results HJC0416 treatment significantly inhibited LX‐2 and HSC‐T6 cell proliferation in a dose‐dependent fashion, and induced apoptosis and S‐phase cell cycle arrest. HSC activation marker α‐smooth muscle actin was also attenuated. HJC0416 suppressed Tyr705‐phosphorylated STAT3 (pSTAT3) and impaired pSTAT3 nuclear translocation and transcriptional activity. STAT3 target genes cyclin D1 and c‐myc were down‐regulated by HJC0416. Static, a well‐known STAT3 inhibitor, confirmed that HSC proliferation was suppressed by STAT3 inhibition. Extracellular matrix (ECM) proteins collagen type I (Coll.I) and fibronectin (FN) are major components of hepatic scar, with TGFβ being the most potent stimulator for ECM production. Our data revealed that endogenous Coll.I and FN expression were suppressed by HJC0416 in a dose‐dependent manner; TGFβ‐stimulated Coll.I and FN production were prevented by pretreatment with HJC0416. Conclusion HJC0416 inhibits HSC activation via the STAT3 signaling pathway. HJC0416 represents a promising anti‐hepatic fibrogenic agent. Opioid-like compound exerts anti-fibrotic activity via decreased hepatic stellate cell activation and inflammation

Abstract:Hepatic fibrosis is characterized by excess type I collagen deposition and exacerbated inflammatory response. Naltrexone, an opioid receptor antagonist used for treating alcohol abuse, attenuates hepatocellular injury in fibrotic animal models, which can be accompanied by deleterious side effects. Additionally, opioid neurotransmission is upregulated in patients with inflammatory liver disease. Several derivatives of Naltrexone, Nalmefene (Nal) and JKB-119, exert immunomodulatory activity; however, unlike Nal, JKB-119 does not show significant opioid receptor antagonism. To delineate the potential hepatoprotective effects of these compounds, we investigated if JKB-119 and Nal could modulate activation of hepatic stellate cells (HSCs), primary effector cells that secrete type I collagen and inflammatory mediators during liver injury. Our results demonstrated that Nal or JKB-119 treatment decreased smooth muscle α-actin, a marker of HSC activation, mRNA and protein expression. Despite decreased collagen mRNA expression, both compounds increased intracellular collagen protein expression; however, inhibition of collagen secretion was observed. To address a possible mechanism for suppressed collagen secretion or retention of intracellular collagen, endoplasmic (ER) protein expression and matrix metalloproteinase (MMP) activity were examined. While no change in ER protein expression (Grp78, PDI, Hsp47) was observed, MMP13 mRNA expression was dramatically increased. In an acute LPS inflammatory injury animal model, JKB-119 treatment decreased liver injury (ALT), plasma TNFα and PMN liver infiltration. Overall, these results suggest that JKB-119 can directly inhibit HSC activation attributed to anti-inflammatory activity and may, therefore, attenuate inflammation associated with HSC activation and liver disease.
Targeting Transforming Growth Factor Βrii Expression Inhibits the Activation of Hepatic Stellate Cells and Reduces Collagen Synthesis

Abstract:Abnormal production of extracellular matrix (ECM) components significantly contributes to the development of liver fibrosis. This study aimed at examining the effects of short-hairpin RNA (shRNA)-mediated transient knockdown of transforming growth factor βRII (TGF βRII) expression on the proliferation and activation of hepatic stellate cells (HSCs) and synthesis of fibrogenic ECM components in HSC cells. Three different shRNA-expressing plasmids were constructed for the expression of shRNA-(a, b, c) targeting to the rat TGF βRII mRNA beginning at nucleotide position 339, 444 and 528 and they were transfected into a rat stellate cell line, HSC-T6 cells, respectively. The levels of TGF βRII, α-smooth muscle actin ( α-SMA), and type I and III collagen expressions were characterized by reverse transcription polymerase chain reaction and Western blot assays. The concentrations of hyaluronic acid (HA) and type IV collagen in the supernatants of cultured cells were measured by enzyme-linked immunosorbent assay. Transfection with the TGF βRII-specific shRNAs resulted in varying levels of inhibition in the expression of TGF βRII in HSC-T6 cells, and transfection with the potent shRNA-c inhibited the expression of TGF βRII in a dose-dependent manner. Knockdown of TGF βRII expression significantly reduced the levels of α-SMA, type I, III and IV collagen, and HA expression in HSC-T6 cells ( P < 0.01). In conclusion, our data indicated that knockdown of TGF βRII expression inhibited the activation of HSCs and the production of fibrogenic ECM components in HSC-T6 cells. Therefore, our findings support the notion that TGF βRII is an important factor of the pathogenic process of liver fibrosis.
[Effects of Leflunomide and Its Active Metabolite on the Production and Mrna Expression of TNF-alpha in Peritoneal Macrophages and Synovial Cells with Adjuvant Arthritis in Rats].

Abstract:AIM:To observe the effects of leflunomide (LEF), an isoxazole immunomodulatory agent and its active metabolite, A771726, on the production and mRNA expression of TNF-alpha in peritoneal macrophages and synovial cells with adjuvant arthritis rats and to further investigate the immunosuppression effects of leflunomide and its mechanisms.METHODS:ELISA methods were used for assaying the levels of TNF-alpha. The RT-PCR methods were used for measuring the expression of TNF-alpha.RESULTS:The production of TNF-alpha was increased in the supernatant of PM psi in adjuvant arthritis (AA) model rat. LEF (5, 10, 25 mg.kg-1, ig) was shown to inhibit the release of TNF-alpha from peritoneal macrophages induced by LPS and the inhibitory effects were in a dose-effect relevance manner. A parallel investigation of cytokine mRNA expression was undertaken using semi-quantitative reverse transcribed polymerase chain reaction (RT-PCR) to follow the kinetics of cytokine appearance in PM psi and synovial membrane tissue cells obtained from AA/normal rats treated with A771726. The results of RT-PCR from macrophages and synovial membrane tissue cells of AA rats at the peak of inflammatory phase showed that TNF-alpha mRNA expression levels were higher than those of normal rats, while the expression of TNF-alpha mRNA was reduced by treating with A771726 in vitro. On the other hand, the TNF-alpha mRNA expression showed kinetics very similar to those obtained by ELISA technique which measured protein expression.CONCLUSION:Leflunomide and its active metabolite, A771726, was found to inhibit the production and mRNA expression of TNF-alpha in peritoneal macrophages and synovial cells with adjuvant arthritis rats model. It might be involved in the mechanisms of its anti-inflammation and immunosupression.
Leflunomide Active Metabolite Inhibites the Expression of Phorbol-12-myristate-13-acetate-induced CD147,matrix Metallo-Proteinase-2 and Matrix Metallo-Proteinase-9 on THP-1 Cells

Abstract:Objective To investigate the effects of the leflunomide active metabolite (A771726) on the expression of phorbol-12-myristate-13-acetate (PMA) -induced CD147, matrix metallo-proteinase (MMP)-2 and MMP-9 on THP-1 cells. Methods THP-1 cells were cultured in RPMI-1640 medium supplemented with 10％ fetal bovine serum. For all experiments, THP-1 cells were cultured at an initial density of 5×105/ml. Before A771726 treatment, cells were cultured with serum-free RPMI-1640 medium for 12 h, THP-1cells were co-cultured with PMA at three different concentrations of A771726 (5, 15 , 45 μg/ml) for 24 h.The mRNA expression of CD147, MMP-2 and MMP-9 was measured by real-time PCR. CD147 expression on the cells were evaluated by flow cytometric analysis. The activity of MMP-2 and MMP-9 were evaluated by gelatin zymography. Statistical differences among the groups were tested by one-way ANOVA or KruskalWallis test. Results The expression of CD147, MMP-2 and MMP-9 were upgraded by the PMA. The expression of CD147 on THP-1 cells was inhibited significantly by A771726 in a dose-dependent pattern (P＜0.01). The mean fluorescence intensity (MFI) of CD147 in positive control group was 109.5±3.8, the MFI in A771726 (5, 15, 45 μg/ml) group were 73.3±2.5, 64.5±2.3, 40.9±2.7, respectively. The expression of MMP-2, MMP-9 mRNA and the activity of MMP-2, MMP-9 in the supernatant was inhibited significantly by A771726 (P＜0.01). The expression of CD147 mRNA was not inhibited significantly by A771726 (P＞0.05).Conclusion Leflunomide active metabolite (A771726) can inhibit the expression of PMA-induced CD147,MMP-2 and MMP-9 on THP-1 Cells.
Leflunomide Induces Immunosuppression in Collagen-Induced Arthritis Rats by Upregulating CD4+CD25+ Regulatory T Cells.

Abstract:This study was to investigate the effect of leflunomide on the immunosuppressive CD4+CD25+ regulatory T cells (CD4+CD25+ Tregs) in collagen-induced arthritis (CIA) rats. CIA was induced by collagen type II in Wistar rats. Immunofluorescence flow cytometry and RT-PCR were used to determine the proportion of CD4+CD25+ Tregs and the expression of Foxp3 mRNA, respectively. Proliferation of T lymphocytes was assayed with MTT reagent, and the level of transforming growth factor beta1 (TGF-beta1) in the supernatant of concanavalin A (Con A)-induced T lymphocytes was determined by ELISA kit. Our investigations demonstrated that inhibition of arthritis by leflunomide was related to changes in CD4+CD25+ Tregs. In addition, A771726, which is the active metabolite of leflunomide, promoted the differentiation of spleen lymphocytes into CD4+CD25+ Tregs, increased antiinflammatory cytokine TGF-beta1 secretion, and adjusted the activity of Con A-induced lymphocytes in vitro.

Abstract:The natural compound oridonin has been proven to possess anti‐neoplastic and anti‐inflammatory properties. Previously, we described that oridonin caused apoptosis, inhibition of cell proliferation and down‐regulation of extracellular matrix proteins (ECM) in activated hepatic stellate cells (HSC). In this study, we investigated the potentially more potent derivative CYD0625 in vitro . Methods Cell proliferation was measured by Alamar Blue assay. Cell apoptosis was detected by staining with Yo‐Pro‐1 and propidium iodide. Immunoblot and immunofluorescence staining were performed for cellular protein expression. Results Treatment of CYD0625 significantly inhibited LX‐2 cells proliferation in a dose‐ and time‐dependent manner with ~20–fold greater potency than oridonin. Similar results were observed in HSC‐T6 cells, whereas 1 μM of CYD0625 treatment had no effect on the human hepatocyte cell line C3A. CYD0625 induced LX‐2 cells apoptosis and S‐phase cell cycle arrest, and it was associated with activation of cell cycle inhibitory proteins p21, p16. The myofibroblast marker protein α‐smooth muscle actin and major ECM proteins collagen type I and fibronectin were markedly down‐regulated by CYD0625. Interestingly, treatment of HSC with CYD0625 at a low dosage (0.1 μM), significantly suppressed collagen type I expression in LX‐2 cells. Furthermore, pre‐treatment with CYD0625 blocked PDGF‐BB‐induced collagen type I and fibronectin expression in LX‐2 cells. image Conclusion In comparison with oridonin, its novel derivative CYD0625 could be a more potent and safer hepatic anti‐fibrosis agent.
Effects of Artesunate on Cell Proliferation, Collaging Generating and KLF6 Expression in Hepatic Stellate Cells

Abstract:Objective:To explore the effect of artesunate on the proliferation of LX-2 cell and to understand the initiatory anti-fibrogenic mechanisms of artesunate. Methods:Hepatic stellate cells(HSCs) LX-2 were divided into control group and artesunate groups with different concentrations. The inhibition of cell proliferation was determined by MTT colorimetric assay. The content of LDH was determined by using colorimetric method while the content of hydroxyproline(Hyp) was determined by colorimetric method. The Western blotting was used for the detection of the expression of KLF-6 protein in HSCs LX-2. Results:The artesunate had an inhibition effect on cell proliferation(P0.01) with dose and time-effect relationships;the activity of LDH in supernatant had no statistical significance compared with control group(P0.05). The content of Hyp was gradually decreased compared with control group(P0.01) with dose-effect relationship. The expression of KLF6 protein was decreased(P0.01) in dose-dependent manner. Conclusion:Artesunate could markedly inhibit the proliferation of HSCs LX-2;The mechanism may be associated with suppressing the expression of collagen and thus decreasing the expression of KLF6 protein.
[Effect of anti-fibrosis compound contained serum on procollagen Type I and IV, matrix metalloproteinase and its tissue inhibitor-1 gene expression in HSC-LI90 cell line]

Abstract:Objective: To study the effects of Anti-fibrosis Compound contained serum (AFCS) on procollagen type I and IV (ProC-I and ProC-IV), matrix metalloproteinase (MMP) and its tissue inhibitor (TIMP-1) gene expression in hepatic stellate cell line LI90 (HSC-LI90). Methods: AFCS was prepared by gastric infusing different dosage (0.5 g/kg, 2.0 g/kg and 4.0 g/kg) of Anti-fibrosis Compound Recipe to rats. After HSC-LI90 cells were exposed to AFCS for 48 hrs, levels of ProC-I, ProC-IV, gene expression of MMP-2, MMP membrane type 1 (MT1-MMP) and TIMP-1 in the cells were detected by Northern blot, and gelatinase activity of MMP-2 was measured by zymography. Results: AFCS of different concentrations could inhibit ProC-I and ProC-IV and TIMP-1 gene expression (P < 0.05 or P < 0.01), increase MT1-MMP gene expression (P < 0.01), but it showed no effect on gene expression and activity of MMP-2 (P > 0.05). Conclusion: Anti-fibrosis Compound Recipe has anti-liver fibrosis action, its effects in inhibiting TIMP-1 gene expression of HSC-LI90 cells and promoting degradation of collagen might be one of the mechanisms of the action.

Abstract:Liver fibrosis may lead to portal hypertension, liver failure or hepatocellular carcinoma, and predominantly results from the proliferation and activation of hepatic stellate cells. OSU‑03012, a non‑cyclooxygenase‑inhibiting celecoxib derivative, has been previously demonstrated to promote apoptosis in certain cell types, however, its function in hepatic fibrosis remains unclear. In the current study, the inhibitory effect of OSU‑03012 on the proliferation of the LX2 human hepatic stellate cell line was evaluated by cell counting kit‑8 assay. Reverse transcription‑quantitative polymerase chain reaction was performed in order to examine the expression of α‑smooth muscle actin and type I collagen, which are representative of LX2 cell activation. The senescence of LX2 cells was measured by senescence‑associated β‑galactosidase staining, and the cell cycle and apoptosis levels were assessed by flow cytometry. The impact of senescence‑associated signaling on protein expression was assessed by western blot analysis. OSU‑03012 was observed to inhibit cell proliferation and prevent the secretion of profibrotic factors in LX2 cells in a dose‑dependent manner. Furthermore, the results demonstrated that OSU‑03012 inhibited the proliferation and activation of LX2 via the induction of cell senescence at the G1 phase, rather than via cell apoptosis. The induction of senescence may be via the upregulation of p16, p21 and p27. In conclusion, the current study provided insight into the pharmacological mechanisms of OSU‑03012 in preventing the proliferation and activation of hepatic stellate cells through cell senescence. The current study supports the theory that OSU‑03012 is a novel agent for potential use against liver fibrosis.

Abstract:Liver fibrosis is a chronic liver disease caused by prolonged liver injuries. Excessive accumulation of extracellular matrix replaces the damaged hepatocytes, leading to fibrous scar formation and fibrosis induction. Lactoferrin (LF) is a glycoprotein with a conserved, monomeric signal polypeptide chain, exhibiting diverse physiological functions, including antioxidant, anti-inflammatory, antibacterial, antifungal, antiviral, and antitumoral activities. Previous study has shown LF's protective role against chemically-induced liver fibrosis in rats. However, the mechanisms of LF in liver fibrosis are still unclear. In this study, we investigated LF's mechanisms in thioacetamide (TAA)-induced liver fibrosis in rats and TGF-β1-treated HSC-T6 cells. Using ultrasonic imaging, H&E, Masson's, and Sirius Red staining, we demonstrated LF's ability to improve liver tissue damage and fibrosis induced by TAA. LF reduced the levels of ALT, AST, and hydroxyproline in TAA-treated liver tissues, while increasing catalase levels. Additionally, LF treatment decreased mRNA expression of inflammatory factors such as Il-1β and Icam-1 , as well as fibrogenic factors including α-Sma , Collagen I , and Ctgf in TAA-treated liver tissues. Furthermore, LF reduced TAA-induced ROS production and cell death in FL83B cells, and decreased α-SMA, Collagen I, and p-Smad2/3 productions in TGF-β1-treated HSC-T6 cells. Our study highlights LF's ability to ameliorate TAA-induced hepatocyte damage, oxidative stress, and liver fibrosis in rats, potentially through its inhibitory effect on HSC activation. These findings suggest LF's potential as a therapeutic agent for protecting against liver injuries and fibrosis.

Astilbin Suppresses Collagen-Induced Arthritis Via the Dysfunction of Lymphocytes

Abstract:OBJECTIVE AND DESIGN:To examine the therapeutic effects of astilbin, a flavanoid isolated from Rhizoma Smilacis Glabrae, on arthritis and to compare it with cyclosporine A (CsA).MATERIALS AND METHODS:Type II collagen-induced arthritis in mice and its in vitro assays for proliferation, matrix metalloproteinase (MMP) and NO production were performed.RESULTS:Astilbin dose-dependently inhibited the footpad swelling, arthritic incidence, and clinical scores without influencing the body weights, while CsA showed strong inhibition with a significant weight loss. Histological examination revealed marked inflammatory damage in arthritic mice including joint swelling, synovial hyperplasia, and cartilage destruction. Against these, an intact joint structure was maintained in astilbin-treated or CsA-treated mice. In isolated spleen cells from arthritic mice, increased potentials in proliferation, NO production, and MMP-2 and 9 activities were suppressed dose-dependently by the oral administration of astilbin. Additionally, astilbin showed neither any cytotoxicity to nor influence on Con A-induced proliferation of spleen cells from naive mice, while CsA showed a dose-dependent cytotoxicity and inhibition of the proliferation.CONCLUSIONS:Astilbin may act as an efficient therapeutic agent for arthritis like CsA but with less toxicity. Its mechanism includes a selective suppression on lymphocyte functions via reducing MMP and NO production.
