Abstract:Objective: Epidermoid growth factor receptor (EGFR, HER1) is overexpressed in a majority of head-and-neck cancers, including nasopharyngeal carcinoma (NPC). Although EGFR inhibitors appear to be effective for some head-and-neck cancers, their efficacy in NPC remains unclear. Methods: The effect of EGFR-specific tyrosine kinase inhibitors, including PD153035 and ZD1839, were studied in NPC-TW01, NPC-TW04, and HONE1 cell lines. The effect of combining EGFR inhibitors with cytotoxic agents was evaluated in NPC-TW04 cells. Results: All three NPC cell lines expressed EGFR. PD153035 and ZD1839 inhibited the growth of NPC cells with IC(50)s around 10 and 20 mu M, respectively. These inhibitors, however, effectively suppressed ligand-stimulated EGFR activation in NPC cells with a much lower concentration (>= 0.1 mu M. The growth-suppression activity of EGFR inhibitors was closely associated with suppression of AKT phosphorylation. LY294002, a phosphatidylinositol-3 kinase (PI3K)/AKT inhibitor, did suppress the growth of NPC cells. Pretreatment of EGFR inhibitors by 24 h significantly enhanced the cytotoxic effect of doxorubicin, paclitaxel, cisplatin, and 5-fluororuacil in NPC-TW04 cells. Conclusions: Our data indicate that inhibition of EGFR activation is not sufficient to induce growth inhibition in NPC cells in vitro. EGFR inhibitors may be useful adjuncts in treating NPC when combined with conventional anticancer drugs. Copyright (C) 2005 S. Karger AG, Basel.
Correlation between inhibition effect of gefitinib and epidermal growth factor receptor in nasopharyngeal carcinoma cells

Abstract:OBJECTIVE: To investigate the correlation between the gefitinib sensitivity and EGFR mutation and gene copy number in nasopharyngeal carcinoma (NPC) cells. METHODS: The effect of gefitinib on NPC cells (CNE-1, CNE-2, HONE-1, SUNE-1, HNE-1 and HNE-2) was determined by MTT assay. Genomic DNA was isolated from cultured NPC cells. Intron-based PCR primers were used to amplify the seven exons (18-24)comprising the entire tyrosine kinase (TK) domain of the EGFR gene. Then, all PCR products were sequenced and analyzed from both sense and antisense directions. The gene copy number per cell was investigated by FISH using the LSI EGFR SpectrumOrange/CEP 7 Spectrum Green probe. RESULTS: The proliferation of NPC cells was inhibited by gefitinib and the effect was dose-dependent. The values of IC50 for the effect of gefitinib on NPC cells (CNE-1, CNE-2, HONE-1, SUNE-1, HNE-1 and HNE-2) were (25.2±3.2), (20.1±5.6), (25.5±4.5), (16.7±4.2), (23.5±7.8) and (23.0±6.5) μmol/L, respectively (P>0.05). There was no indication that EGFR mutation in the entire TK domain existed in the six tested NPC cell lines. Investigated by FISH, there was only an EGFR gene amplification in the HNE-2 cell line. CONCLUSIONS: Gefitinib can inhibit the proliferation of NPC cells. However, this effect does not correlate with EGFR gene amplication. The correlation between EGFR mutation and gefitinib effect on NPC cells is still unclear.

Abstract:Epidermal growth factor receptor (EGER) is one of the most promising targets for non-small cell lung cancer (NSCLC). Our study demonstrated the antitumor effects of icotinib hydrochloride, a highly selective epidermal growth factor receptor tyrosine kinase inhibitor (EGER TKI), in two EGFR-mutated lung cancer cell lines compared to A549, a cell line without EGFR mutations. We incubated PC-9 and HCC827 human lung cancer cell lines both with (E746-A750) mutations with various concentrations of icotinib and gefitinib for 48 h. Cell proliferation and migration were determined using a real-time cell invasion and migration assay and cytotoxicity assay. Apoptosis was assessed by measuring Annexin V staining using flow cytometry. The antitumor effects of icotinib compared to gefitinib were similar and were most effective in reducing the proliferation of EGFR-mutated cells compared to non-mutated controls. Our results suggest the possibility of icotinib as a new therapeutic agent of EGFR-mutated cancer cells, which has the potential to be used in the first-line treatment of EGFR-mutated NSCLC.

Abstract:Purpose: Epidermal growth factor receptor (EGFR) is usually overexpressed in nasopharyngeal carcinoma (NPC). We tested the antitumor effects of irreversible ErbB family inhibitor afatinib on human NPC using in vitro and in vivo models. Materials and methods: The effect of afatinib on NPC cells was evaluated using the Cell Counting Kit 8 (CCK8) assay, flow cytometry, and Western blot analyses. The effect of afatinib, as either a single agent or in combination with gemcitabine (GEM), on tumor growth was determined using NPC tumor xenografts in mice. Results: Afatinib inhibited cell growth in all three NPC cell lines tested in a dose-dependent manner. Afatinib promoted cell cycle arrest at the S and G2/M phases, and it significantly inhibited epidermal growth factor (EGF)-induced activation of EGFR and its downstream signaling factors. Co-treatment with afatinib and GEM more effectively inhibited tumor growth than either drug alone but was associated with increased toxicity. Conclusion: Afatinib induced cell cycle arrest and inhibited the proliferation of NPC cell lines. Afatinib in combination with GEM demonstrated significant antitumor effect in an NPC xenograft model. The administration of afatinib with GEM in NPC needs to be modified in order to be effective and tolerable.

Abstract:This study evaluated the effects of ZD1839, an orally active, selective epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, on nasopharyngeal carcinoma (NPC) both in vitro and in vivo. Influence of ZD1839 alone or combined with cisplatin on the NPC cell line CNE2 was detected by MTT assay with flow cytometry assessment of cell cycle distribution and apoptosis rates. Nude mice NPC xenografts were also used to evaluate the effects of ZD1839 alone or combined with cisplatin. The Student's t test evaluated statistical significance. ZD1839 alone or combined with cisplatin inhibited CNE2 cell line proliferation. ZD1839 induced CNE2 cell cycle arrest in the G1 phase, and higher concentrations induced apoptosis. Xenograft tumors were significantly smaller when treated with 200 mg/kg ZD1839, cisplatin, or cisplatin combined with 100 mg/kg ZD1839 than untreated controls. ZD1839 (200 mg/kg) alone showed good tumor inhibition effects, reduction of tumor weights, and smaller tumor volume without loss of body weight. ZD1839 (200 mg/kg) might provide a good and effective therapeutic reagent for NPC.
Lenvatinib for Effectively Treating Antiangiogenic Drug-Resistant Nasopharyngeal Carcinoma.

Abstract:Nasopharyngeal carcinoma (NPC) clinical trials show that antiangiogenic drugs (AADs) fail to achieve the expected efficacy, and combining AAD with chemoradiotherapy does not show superiority over chemoradiotherapy alone. Accumulating evidence suggests the intrinsic AAD resistance in NPC patients with poorly understood molecular mechanisms. Here, we describe NPC-specific FGF-2 expression-triggered, VEGF-independent angiogenesis as a mechanism of AAD resistance. Angiogenic factors screening between AAD-sensitive cancer type and AAD-resistant NPC showed high FGF-2 expression in NPC in both xenograft models and clinical samples. Mechanistically, the FGF-2-FGFR1-MYC axis drove endothelial cell survival and proliferation as an alternative to VEGF-VEGFR2-MYC signaling. Genetic knockdown of FGF-2 in NPC tumor cells reduced tumor angiogenesis, enhanced AAD sensitivity, and reduced pulmonary metastasis. Moreover, lenvatinib, an FDA recently approved multi-kinase inhibitor targeting both VEGFR2 and FGFR1, effectively inhibits the tumor vasculature, and exhibited robust anti-tumor effects in NPC-bearing nude mice and humanized mice compared with an agent equivalent to bevacizumab. These findings provide mechanistic insights on FGF-2 signaling in the modulation of VEGF pathway activation in the NPC microenvironment and propose an effective NPC-targeted therapy by using a clinically available drug.
Beta 1 Integrin Mediated Acquired Resistance to Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitor in Non-Small Lung Cancer Cells

Abstract:e13651 Background: With the general application of EGFR TKI in treatment for NSCLC, more and more adherent and acquired resistance to such agents happens. To further improve efficacy of this kind of agents, a better understanding of the mechanism of resistance is necessary. Methods: Gefitinib-resistant PC9/G cell lines was induced by exposure of PC9 cell lines to MNNG and gefitinib and maintained in the mediua containing 0.05 umol/L of gefitinib. The inhibitory effects of gefitinib and/or LY294002 and PD98059 on cellular proliferation were tested by MTT assay. Cell cycle and apoptosis were analyzed by flow cytometry. Western Blot was used to detect expression of EGFR, phospho-EGFR, Akt and phospho-Akt. The different profiles of gene expression between PC9 and PC9/G cells were obtained by DNA microarray. β1 integrin was knocked down by transiently transfected siRNA. A scrambled sequence was used as control. The sensitivity to gefitinib of transfected cells was determined by MTT assay. Sequencing products were amplified by nested PCR. Apoptosis of transfected cells was assayed by TUNEL methods. Results: Resistant index of PC9/G to gefitinib was about 140 to 241 fold compared with PC9.Overexpression of β1 integrin was observed in PC9/G. Depletion of β1 integrin with shRNA results in inhibition of proliferation and promotion of apoptosis of PC9/G. Phosphorylation of Akt was less inhibited by gefitinib in PC9/G than in PC9. The restoration of sensitivity of PC9/G to gefitinib by LY294002, PI3-K inhibitor, and the diminished level of phospho-Akt induced by knock down of β1 integrin with RNAi. Conclusions: These findings suggest that β1 integrin signaling through PI3-K may be an important mechanism of resistance to EGFR TKIs and β1 integrin may be a therapeutic target for treatment of NSCLC unresponsive to EGFR inhibitors. EGFR Tyrosine Kinase Inhibitor AG1478 Inhibits Cell Proliferation and Arrests Cell Cycle in Nasopharyngeal Carcinoma Cells

Abstract:Nasopharyngeal carcinoma (NPC), which occurs with a high incidence in southern China and southeast Asia, is of epithelial origin with overexpression of EGF receptor. To study the effect of inhibition of EGFR signaling on nasopharyngeal carcinoma cell proliferation and cell cycle distribution, EGFR tyrosine kinase inhibitor AG1478 was employed to treat Nasopharyngeal Carcinoma CNE2 cells. The results showed that AG1478 inhibited proliferation of CNE2 cells. Immunoblot showed that AG1478 inhibited EGFR phosphorylation in CNE2 cells without reduced expression of EGFR protein. The activation of Akt and MAPK which are downstream molecules of EGFR signaling pathway, were also inhibited by AG1478. AG1478 induced cell cycle arrest in G1 phase, and the levels of protein p27 were significantly up-regulated. We concluded that inhibition of the EGFR signaling induced cell cycle arrest in G1 phase in CNE2 cells and p27 up-regulation was involved in this process. The EGFR kinase specific inhibitor is of potential to be developed into drugs for NPC treatment.

Abstract:BACKGROUND & OBJECTIVE:ZD1839, an anilinoquinazoline, is an orally active, selective epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, which has been approved for the treatment of advanced non-small cell lung cancer by FDA. Nasopharyngeal carcinoma is of epithelial origin with overexpression of epidermal growth factor receptor and it remains unclear whether ZD1839 is useful in the treatment of nasopharyngeal carcinoma. The present study was designed to evaluate the effect of ZD1839 on nasopharyngeal carcinoma cell lines.METHODS:The effect of ZD1839 on nasopharyngeal carcinoma cells (CNE1, CNE2, SUNE1) was determined by MTT assay. The effect of ZD1839 combined with cisplatin (DDP) or 5-fluouracil (5-FU) on nasopharyngeal carcinoma cell CNE2 was also evaluated. Burgi formula was adopted to analyze combination effect. The distribution of cell cycle and cell apoptosis rate were analyzed by flow cytometry.RESULTS:The proliferation of nasopharyngeal carcinoma cell lines CNE1, CNE2, and SUNE1 was inhibited by ZD1839, and the effect was dose-dependent. The value of IC50 for the effect of ZD1839 on nasopharyngeal carcinoma cell lines CNE1, CNE2, and SUNE1 was 3.9 micromol/L, 5.6 micromol/L, and 5.5 micromol/L, respectively. ZD1839 could enhance the effect of DDP and 5-FU on CNE2 cells. The Q values by Burgi formula were 1.19+/-0.02 and 1.12+/-0.10, respectively. When CNE2 cells were treated with ZD1839 at the concentration of 0, 1.95, 3.9, 7.8, 15.6, 31.25 micromol/L, the percentages of G(0)/G(1) phase were (46.8+/-1.7)%, (48.8+/-1.6)%, (51.3+/-1.3)%, (54.0+/-1.3)%, (61.5+/-2.2)%, and (71.2+/-1.4)%, respectively; the percentages of S phase were (37.5+/-1.3)%, (35.8+/-1.6)%, (31.8+/-2.1)%, (34.3+/-2.7)%, (27.2+/-2.9)%, and (27.6+/-2.4)%, respectively;the percentages of G(2)/M phase were (15.7+/-0.4)%, (15.3+/-0.1)%, (16.9+/-0.9)%, (11.7+/-1.4)%, (11.3+/-0.7)%, and (1.1+/-0.8)%, respectively. When CNE2 cells were treated with ZD1839 at the concentration of 0, 1.95, 3.9, 7.8, 15.6, 31.25, 62.5 micromol/L, the rates of CNE2 cell apoptosis were (1.6+/-0.3)%, (1.7+/-0.3)%, (2.3+/-0.4)%, (3.3+/-0.4)%, (3.9+/-0.8)%,(8.0+/-1.1)%,and(14.3+/-2.3)%, respectively.CONCLUSION:ZD1839 inhibits the proliferation of nasopharyngeal carcinoma cells and enhances the effect of cytotoxic drugs.ZD1839 induces CNE2 cell cycle arrest in the G1 phase and higher concentration of ZD1839 could induce apoptosis.
Effect of Cell Growth Inhibition Through Simultaneously Targeted Blocking Epidermal Growth Factor Receptor Tyrosine Kinase and Mtor Signaling Pathway in Nasopharyngeal Carcinoma

Abstract:Objective The aim of the investigation was to verify that simultaneously blocking both EGFR and mTOR mediated pathways may be an efficient way to inhibit cell growth in nasopharyngeal carcinoma.Methods The effect of cell growth inhibition through a combination of EGFR-selective tyrosine kinase inhibitor Tarceva and mTOR inhibitor Rapamycin was evaluated.Cell growth assay and flow cytometric analysis were applied to assessing the cell cycle and apoptosis in both CNE-1 and CNE-2 cell lines.Results ① Tarceva and Rapamycin demonstrated a concentration dependent anti-proliferative effect on both CNE-1 and CNE-2 cell lines(P0.05).② The inhibition rate of cell growth in the group treated with a combination of two agents was higher than the sum of that in the two groups treated with only one agent(P0.05).③ The combination of Tarceva and Rapamycin significantly induced G1 cell arrest(P0.05)and increased apoptosis rate(P0.05)compared with the groups treated with single agent and the control group.Conclusion Results of the investigation suggest that a combination of Tarceva and Rapamycin induced cell growth inhibition in a synergistic way,which may be resulted from the increased rates of G1 cell arrest and apoptosis.
[The Effect of Simultaneously Blocking Target Epidermal Growth Factor Receptor Tyrosine Kinase and Cyclooxygenase-2 on the Growth of NPC Cell].

Abstract:OBJECTIVE:This study was designed to prove simultaneously blocking both EGFR and COX-2-mediated pathways may be an efficient means of inhibiting cancer cell growth in NPC. METHOD:A combination of tarceva (EGFR-selective tyrosine kinase inhibitors) with celecoxib (Cox-2 inhibitor) was studied on its effects on cell growth, cell cycle progression, apoptosis and protein expression in CNE-2 cell lines by cell growth assay, flow cytometric analysis assay and Western blotting. RESULT:(1) The inhibition rate of cell growth was higher in the group treated with a combination of two agents than that the sum of rates of the two groups treated with only one agent (P < 0.05). (2) The combination of tarceva with celecoxib significantly induced G1 arrest (P < 0.05), but did not increase apoptosis rate (P > 0.05). (3) The group of combination showed less expressions of p-EGFR and COX-2 than any other group. CONCLUSION:Simultaneously blocking EGFR and COX-2 mediated pathways would significantly inhibit the growth of CNE-2 cell line, increase G1 arrest and reduce the expression levels of p-EGFR and COX-2.
Effects of Gefitinib and Small Interfering RNAs Targeting EGFRin the Treatment of Prostate Eallcer

Abstract:Objective To probe into the inhibitory effects and mechanisms of Cefitinib and small interfering RNAs ( siRNA) targeting epidermal growth factor receptor ( EGFR) ( EGFR siRNA) against hormone independent prostate cancer (HIPC) in vitro and in vivo. Methods After PC-3 cells were trans-fected with lentivirus mediated EGFR siRNA recombinant or treated with Gefitinib (0-10 mg/L), MTT was used to measure cell growth inhibitory rate (IR), fluorescent real-time polymerase chain reaction (PCR) to detect EGFR mRNA level, and Western blotting to assay the expression of EGFR and its intracellular proteins, such as Akt, MAPK and PKC. Tumor growth was observed when EGFR siRNA or/and Gefitinib were used to treat mice with transplanted tumors. Results The IR of PC-3 cells was 40% -50% by using EGFR siRNA and transfection efficiency was 85%. Gefitnib inhibited the growth of PC-3 cells in a time-and concentration-dependent manner. The expression of EGFR mRNA and protein in PC-3 cells was down-regulated over 90% which was obviously higher than < 80% in Gefitinib group (P < 0.01) ; EGFR siRNA could significantly inhibit the expression of Akt and MAPK, but Gefitinib only significantly inhibit the expression of Akt. In the in vivo study, the tumor growth was inhibited significantly in Gefitinib group (53.95%) or Gefitinib + EGFR siRNA group (59. 28%) as compared with EGFR siRNA group (34. 83% ,P <0. 05). Conclusion EGFR pathways inhibitors could block PC cells growth effectively mainly via the down-regulation of the expression of EGFR and its intracellular protein Akt.
Cetuximab and Cisplatin Show Different Combination Effect in Nasopharyngeal Carcinoma Cells Lines via Inactivation of EGFR/AKT Signaling Pathway

Abstract:Nasopharyngeal carcinoma (NPC) is a common malignant cancer in South China. Cisplatin is a classical chemotherapeutic employed for NPC treatment. Despite the use of cisplatin-based concurrent chemoradiotherapy, distant failure still confuses clinicians and the outcome of metastatic NPC remains disappointing. Hence, a potent systemic therapy is needed for this cancer. Epidermal growth factor receptor (EGFR) represents a promising new therapeutic target in cancer. We predicted that combining the conventional cytotoxic drug cisplatin with the novel molecular-targeted agent cetuximab demonstrates a strong antitumor effect on NPC cells. In this study, we selected HNE1 and CNE2 cells, which have been proved to possess different EGFR expression levels, to validate our conjecture. The two-drug regimen showed a significant synergistic effect in HNE1 cells but an additive effect in CNE2 cells. Our results showed that cisplatin-induced apoptosis was significantly enhanced by cetuximab in the high EGFR-expressing HNE1 cells but not in CNE2 cells. Further molecular mechanism study indicated that the EGFR/AKT pathway may play an important role in cell apoptosis via the mitochondrial-mediated intrinsic pathway and lead to the different antitumor effects of this two-drug regimen between HNE1 and CNE2 cells. Thus, the regimen may be applied in personalized NPC treatments.
Erlotinib enhanced chemoradiotherapy sensitivity via inhibiting DNA damage repair in nasopharyngeal carcinoma CNE2 cells

Abstract:Background: Nasopharyngeal carcinoma (NPC), arising from nasopharynx epithelium, is a rare type of malignant carcinoma that has a specific geographical distribution and a high risk of distant metastases. For most of the diagnosed NPC patients, the total survival rate decreased significantly due to the high local recurrence rate and metastasis rate. Concurrent chemoradiotherapy (CCRT), as routine therapy strategy of NPC, usually accompanies with high-dosage cytotoxic agents and serious toxic side reaction. Therefore, there is an urgent need for update the existing therapy strategies. In this study, we sought to investigate the effects of a combined therapy strategy, erlotinib combined with cisplatin and radiotherapy, on biological characteristics of NPC CNE2 cells and the potential reasons. Methods: CNE2 cells in logarithmic phase seeding in 96-well plates received concentration gradients of erlotinib (at 0, 10, 20, 40, 80, 160, 320 mmol/L) or cisplatin (at 0, 0.25, 0.5, 1, 2, 4, 8 mg/L), in order to obtain the optimal working concentration of erlotinib and cisplatin via 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Then, cells were divided into control group and four treatment groups (Group I-IV). All treatment groups received irradiation of 4 Gy, moreover, Group II-IV respectively received erlotinib, cisplatin and erlotinib plus cisplatin at optimal working concentration. After 24 and 48 h of irradiation, growth inhibition rate was determined; invasion ability and migration ability was respectively detected by Boyden's chamber assay and cell scratch test; flow cytometry was performed for determining apoptosis rate and cell cycle distribution; the expression levels of epidermal growth factor receptor (EGFR) signal pathway proteins were semi-quantitatively analyzed by Western-blot. Results: Compared with Group I, all the other treatment groups showed better inhibition effect on cell viability, invasion and migration ability and higher apoptosis rate, while Group IV showed the strongest growth inhibition effect and highest apoptosis rate. In addition, EGFR signal pathway proteins of Group IV showed the lowest expression level. Conclusions: In combined therapy with radiotherapy/chemotherapy, erlotinib could enhance radiotherapy/chemotherapy sensitivity, probably because it could suppress DNA damage repair after radiotherapy/chemotherapy, thus weakening radiotherapy/chemotherapy resistance of tumor cells.
IGF-1R Inhibition Suppresses Cell Proliferation and Increases Radiosensitivity in Nasopharyngeal Carcinoma Cells

Abstract:Although ionizing radiation (IR) has provided considerable improvements in nasopharyngeal carcinoma (NPC) treatment, radioresistance is still a major threat for some subsets of patients. The insulin-like growth factor-1 receptor (IGF-1R) signaling pathway is tightly regulated and plays critical roles in mediating cell proliferation, growth, and survival. Thus, IGF-1R may be a potential therapeutic target for patients with different malignancies. However, its mechanism in NPC is not fully investigated. Linsitinib is an oral small molecule and is a tyrosine kinase inhibitor (TKI) of IGF-1R, which has been known for antitumor effects used widely. Here, we evaluated the proliferation and radiosensitivity of NPC cell lines (CNE-2 and SUNE-1) after linsitinib treatment. We found that linsitinib suppresses IGF-1-induced cell proliferation through inhibiting Akt and ERK phosphorylation. Moreover, linsitinib further boosted IR-induced DNA damage, G2-M cell cycle delay, and apoptosis in NPC cells. Finally, linsitinib reversed radioresistant NPC cells by decreasing the phosphorylation of IGF-1R. Our data indicated that the combination of linsitinib and IR and targeting IGF-1R by linsitinib could be a promising therapeutic strategy for NPC.
Antitumor Effect of Gefitinib, an Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitor, Combined with Cytotoxic Agent on Murine Hepatocellular Carcinoma

Abstract:AIM:To investigate the inhibitory effect of gefitinib combined with cytotoxic agent cisplatin (CDDP) on hepatocellular carcinoma (HCC).METHODS:Female Kunming mice and H22 hepatocarcinoma cells were used. Gefitinib at daily dose of 100 mg/kg body weight (BW) or lecithin liquid was given by gastrogavage once a day for 5 or 10 successive days. CDDP or normal saline (NS) was administered intraperitoneally (i.p.) once a day for 5 successive days. Mice were randomly divided into control group (lecithin, or NS, i.p.), CDDP group (daily dose, 1.2 mg/kg BW; d1-5, or d6-10), Gefitinib (d1-5, or d6-10, or d1-10), and Gefitinib combined with CDDP groups. The inhibitory rate (IR) of tumor, net BW, spleen index (SI), thymus index (TI) and the amount of peripheral blood cells of mice were detected on the 12th experiment day.RESULTS:The growth of HCC in mice was inhibited by Gefitinib alone (IR: 41% in d1-10 group and 30% in d1-5 group, respectively) or CDDP alone (IR: 32-54% in d1-5 group or d6-10 group). The highest inhibitory effect (IR: 56%) on HCC growth was observed in Gefitinib (d1-10) combined with CDDP (d1-5) group. Higher inhibition was also observed in CDDP (d1-5) followed by Gefitinib (d6-10) group than that in Gefitinib (d1-5) followed by CDDP (d6-10) group (IR: 61% vs 36%, P < 0.01) in the independent study. Net BW, SI, TI and the amount of blood cells of mice in Gefitinib alone group were not significantly different from those in control groups.CONCLUSION:Gefitinib can significantly inhibit the growth of murine H22 hepatocellular carcinoma. If Gefitinib is used after CDDP treatment in animal experiments, the inhibitory effect could be enhanced.
Inhibition of N-acetylglucosaminyltransferase V Enhances the Cetuximab-Induced Radiosensitivity of Nasopharyngeal Carcinoma Cells Likely Through EGFR N-glycan Alterations

Abstract:N-acetylglucosaminyltransferase V (GnT-V), an enzyme that catalyzes the formation of the N-linked β-1-6 branching of oligosaccharides, is related to the radiosensitivity of nasopharyngeal carcinoma (NPC). Cetuximab (C225) is an epidermal growth factor receptor (EGFR) inhibitor used as a radiosensitizer in the treatment of NPC. In this study, we used GnT-V as a molecular target to further sensitize cetuximab-treated NPC cells to radiation. The results from two NPC cell lines (CNE1 and CNE2) revealed that the silencing of GnT-V enhanced cetuximab-induced radiosensitivity by decreasing the β-1-6 branching of oligosaccharides on the EGFR. GnT-V down-regulation combined with cetuximab decreased the survival fraction, healing rate and cell viability and increased the apoptosis rate. Concomitantly, the combination of cetuximab and irradiation did not change the EGFR mRNA and protein levels and decreased the β-1-6 branching on the EGFR. Subsequently, we further explored the signaling downstream of EGF, particularly the PI3K/Akt signaling pathway, and discovered that treatment consisting of GnT-V down-regulation, irradiation and cetuximab was negatively correlated with phospho-Akt and phspho-PI3K. Finally, an in vivo experiment with radiotherapy revealed that the combination of GnT-V down-regulation and cetuximab decelerated tumor growth. In summary, our study demonstrated that the combination of decreased GnT-V activity and cetuximab enhanced NPC radiosensitivity, and the possible mechanism underlying this effect might involve the N-linked β1-6 branching of the EGFR.

Abstract:Purpose: Caffeic acid phenethyl ester (CAPE) is the main polyphenol extracted from honeybee propolis, which inhibits the growth of several kinds of tumor. This study aimed to assess the inhibitory effect of CAPE in nasopharyngeal carcinoma (NPC), evaluate the synergistic action of CAPE in radiotherapy sensitivity of NPC cell lines and further elucidate the possible molecular mechanism involved. Materials and methods: CCK-8 assay was used to analyze cell proliferation ability. Colony formation assay was used to evaluate the clonogenic ability and radio-sensitiveness of NPC cells by CAPE treatment. Wound-healing and transwell assay were used to assess the motility of cells. The expression of key molecules of the epithelial-mesenchymal transition (EMT) was determined by western blot analysis and changes in radiation sensitivity were measured by colony-formation assay. cDNA microarray analysis was used to determine differentially expressed genes with and without CAPE treatment, with Gene Ontology enrichment of gene function and KEGG pathways determined. Cell cycle and apoptosis were detected by flow cytometry and western blot analysis. Results: CAPE suppressed the viability of NPC cell lines time- and dose-dependently. It induced apoptosis in NPC cells along with decreased expression of Bcl-XL and increased cleavage of PARP and expression of Bax. G1 phase arrest was induced by CAPE with ower expression of CDK4, CDK6, Rb and p-Rb. The migratory and invasive ability of NPC cells was decreased by the EMT pathway. The irradiation sensitivity of NPC cells was enhanced with CAPE treatment. CAPE specifically inhibited nuclear factor κB (NF-κB) signaling pathway by suppressing p65 subunit translocation from cytoplasm to nucleus. CAPE treatment was synergistic with chemotherapy and radiotherapy. Conclusion: CAPE may inhibit the proliferation and metastasis of NPC cells but enhance radiosensitivity in NPC therapy by inhibiting the NF-κB pathway. CAPE could be a potential therapeutic compound for NPC therapy.

Abstract:A previous study reported that compound 5A, a caffeic acid phenethyl ester (CAPE) analog, exhibited obvious neuroprotective activity, in particular, compound 5A possessed higher stability and membrane permeability than CAPE. CAPE displays antitumour function; therefore, evaluating the antitumour effect of its analog with higher stability and membrane permeability is worthwhile. We first investigated the antitumour activity of compound 5A. We found that compound 5A significantly inhibited the proliferation of tumor cells and showed low cytotoxicity in normal cells. Furthermore, compound 5A was found to induce the cell cycle arrest and apoptosis of CNE2 cells. Through the prediction of SwissTargetPrediction and subsequent confirmation, epidermal growth factor receptor (EGFR) was identified as a target of compound 5A. Compound 5A also influenced the expression of genes downstream of EGFR in nasopharyngeal carcinoma (NPC) cells. Based on these findings, compound 5A inhibits the proliferation of NPC cells by targeting EGFR and may become a new candidate compound for NPC treatment.
Induction of Cell Cycle Arrest and Apoptosis in Human Nasopharyngeal Carcinoma Cells by ZD6474, an Inhibitor of VEGFR Tyrosine Kinase with Additional Activity Against EGFR Tyrosine Kinase

Abstract:ZD6474 is a vascular endothelial growth factor receptor (VEGFR) and epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. The present study was undertaken to investigate the direct antiproliferative effect of ZD6474 on human nasopharyngeal carcinoma (NPC) in vitro and the antitumor activity on NPC xenografts in vivo. Results indicated that ZD6474 treatment inhibited EGFR phosphorylation and led to a dose- and time-dependent decrease in NPC cell (CNE-1, CNE-2 and C666-1) proliferation. Further investigation demonstrated G0/G1 cell cycle arrest in all 3 cell lines, which was associated with an upregulation of p21 and/or p27, and downregulation of CDK4, CDK6 and CDK2. ZD6474 treatment also induced apoptosis in CNE-1 and CNE-2 cells. The apoptosis mechanisms involved reduction of Bcl-2 and/or Bcl-XL, induction of Bak and/or Bax, and activation of caspases-3, -9 and/or -8. The in vivo antitumor activity was evaluated in CNE-2 and C666-1 xenografted nude mice. Administration of ZD6474 (25-100 mg/kg/day, once-daily, p.o.) produced a dose-dependent inhibition of tumor growth and prolonged survival in both models. This study suggests that ZD6474 exerts direct antiproliferative effects on NPC cell lines in vitro by inducing G0/G1 arrest and apoptosis, and potent antitumor effects on NPC xenografts in vivo. It indicates that ZD6474 may offer a new and effective treatment for human NPC.
