Abstract:Total synthesis of 4d-deoxy Lewisx pentasaccharide is described. O-(2,3,6-Tri-O-benzoyl-4-deoxy-α-d-xylo-hexopyranosyl)trichloroacetimidate was condensed with a diol of glucosamine to give regioselectively a β1→4 linked disaccharide, which was further fucosylated to a protected deoxy Lewisx trisaccharide. A highly stereo- and regioselective glycosylation of this trisaccharide onto a lactoside diol was performed to provide a pentasaccharide in excellent yield. After deprotection, this pentasaccharide afforded the target oligosaccharide.

Abstract:The repeating GalpNAc-alpha-(1-->4)-GalpNAc unit is part of a series of essential structures that can be found in many important biomolecules such as the glycoproteins and the O-antigenic polysaccharides of clinically important bacterial strains. In this paper, we describe an exclusive alpha-selective glycosylation reaction, using a 4,6-di-O-tert-butyldimethylsilyl-N-acetyloxazolidinone-protected thioglycoside as the glycosyl donor, under pre-activation conditions, with only half amount of the promoter, providing the product GalpNAc-alpha-(1-->4)-GalpNAc in high isolated yield. This reaction can be also applied to increasing the length of the repeating structure, which is of significant use in further synthesis of branched or linear oligosaccharides.

Abstract:In the present study, an efficient preparative procedure was developed for the preparation of three main acylated pentasaccharides from Polygalae Radix using integrated extraction-adsorption method followed by semi-preparative high performance liquid chromatography. Firstly, the acylated pentasaccharides were simultaneously extracted and purified by column chromatographic extraction integrated with macroporous resin purification. Comparing this integrated process to conventional one, some obvious advantages were observed, namely more than 38.9% enhancement of the recovery of acylated pentasaccharides, about 49.4% and 50.4% reduction in water and ethanol consumption, respectively. And then, the obtained extracts were further purified by semi-preparative high performance liquid chromatography. Finally, three known acylated pentasaccharides were purified from 100 g of Polygalae Radix, including tenuifoliose I (13 mg), tenuifoliose H (15 mg), and tenuifoliose A (21 mg). The purities of the compounds were determined by high performance liquid chromatography and the chemical structures were confirmed by mass spectrometric and NMR analysis. (C) 2015 Elsevier B.V. Abstract:The synthesis of C-disaccharides of α-d-galactopyranosyl-(1 → 3)-d-galactopyranose (α-Gal), potential tools for studying the biology of α-Gal glycans, is described. The synthetic strategy, centers on the reaction of two easily available precursors 1,2-O-isopropylidene-d-glyceraldehyde and an α-C-glactosyl-E-crotylboronate, which affords a mixture of two diastereomeric anti-crotylation products. The stereoselectivity of this reaction was controlled with (R)- and (S)-TRIP catalysts, and the appropriate diastereomer was transformed to C-linked disaccharides of α-Gal, in which the aglycone segment comprised O-, C- and S-glycoside entities that can enable glycoconjugate synthesis.

Abstract:An expeditious three-component, one-pot sequential glycosylation protocol has been developed for the preparation of 3,6-branched unsymmetrical mannopentaose (Man5), employing a mannose trisaccharide donor, a mannose monosaccharide donor and a mannose monosaccharide acceptor. The high efficiency of this one-pot procedure was demonstrated by comparison study with a stepwise synthesis using the same three building blocks.

Abstract:This investigation describes a one‐pot reaction to prepare a series of building blocks for glycosylation reactions, such as 3‐alcohol glucosamines, fully protected glucosamines, O‐ 4 and O‐ 6 alcohol glucosamines. These reactions readily produce not only glycosyl donors and acceptors, but also different glycosyl units that can be changed based on the needs of the experiment. The synthesis of some molecules containing glucosamines, including saccharide chains of glycopeptide (GP) and precursors of lipid A disaccharide backbone, are also described. GP has good selectivity for tumor angiogenesis and this phenomenon makes the GP a potential target drug. Lipid A has recently been adopted as an adjuvant for human vaccines.

Abstract:A novel. chemoenzymatic approach for the synthesis of oligosacchrides containing free amino groups has been developed in which thermophilic glycosidases and a fusion enzyme containing catalytic domains of uridine-5'-diphospho-galactose 4-epimerase and alpha(1-->3) galactosyltransferase were used. This methodology, in conjunction with a convenient purification procedure employing ion exchange chromatography, facilitates the lengthy and high-cost process of carbohydrate synthesis. The prepared oligosaccharides range from disaccharide lactosamine (1a), trisaccharide alpha-Gal-(1-->3)-beta-Gal-(1-->4)-GlcNH(2) (2), tetrasaccharide beta-Gal-(1-->4)-beta-GlcNH(2)-(1-->3)-beta-Gal-(1-->4)-beta-Glc-N-3 (7), to pentasaccharide alpha-Gal-(1-->3)-beta-Gal-(1-->4)-beta-GlcNH(2)-(1-->3)-beta-Gal-(1-->4)-beta-Glc-N-3 (15). Compounds 2 and 15 are derivatives of natural alpha-Gal epitopes. Both of them have shown comparable activities with their natural parent compounds toward human anti-Gal IgG. This method provides a practical approach for the preparation and purification of oligosaccharides containing free amino groups, which can be further derivatized for the enhancement of biological activities.

Abstract:A tetrasaccharide, α-l-Rhap-(1→3)-β-d-GalpNAc-(1→6)-α-d-Galp-(1→2)-α-d-Galp, the carbohydrate moiety of clarhamnoside isolated from the marine sponge Agelas clathrodes, was synthesized as its propyl glycoside via a convergent approach. The key steps to the synthetic strategy were the stereoselective construction of the reducing-end disaccharide α-d-Galp-(1→2)-d-Galp (5) and efficient coupling with the terminal disaccharide α-l-Rhap-(1→3)-d-GalpNAc building block, in which the N-phthalimido-protected trifluoroacetimidate 13 was proved to be an effective donor.

Abstract:The total synthesis of 3′-deoxy Lewisx pentasaccharide is reported. 4-O-Acetyl-2,6-di-O-benzoyl-3-deoxy-β-d-xylo-hexopyranosyl trichloroacetimidate was condensed with a diol of glucosamine to give a disaccharide, which was further fucosylated to a Lewisx trisaccharide analogue. Glycosylation of a lactoside diol with this trisaccharide provided a pentasaccharide, which after deprotection, afforded the target pentasaccharide.

Abstract:A tetrasaccharide repeating unit of the O‐antigen from Escherichia coli O33 has been synthesized for the first time using a chemoselective [1+1+2] one‐pot glycan assembly strategy. The phosphoglycerol moiety was installed by using the phosphoramidite method with benzylidene‐protected glycerol 2‐phosphoramidite as the reagent. Overall, the target tetrasaccharide was synthesized from monosaccharide building blocks in 12 linear steps, resulting in a satisfactory yield of 28%.

Abstract:A new, robust one-pot four-enzyme synthetic method was developed for thymidinediphosphate-L-rhamnose starting from D-glucose-1-phosphate. The enzymes, Glc-1-P thymidylyltransferase, dTDP-Glc-4,6-dehydratase, dTDP-4-keto-6-deoxy-Glc-3,5-epimerase and dTDP-4-keto-Rha reductase were derived from Streptococcus pneumonia serotype 23F, expressed in Escherichia coli, and studied in detail to provide the first direct evidence for their functions.

Abstract:A practical and efficient synthesis of phosphatidylinositol pentamannoside (PIM5) was achieved based on a five-component one-pot sequential glycosylation protocol with exclusive regio- and stereo-selectivity. Two regioselective sequential glycosylations on inositol and p-tolyl thioglycosides as the sole type of building blocks made this protocol to avoid the tedious protective group manipulations. This synthetic strategy provides access to other important glycolipids with similar structures.

Abstract:The disaccharide of αGal Determinant Epitope in the circulation of human, Ga(αl → 3) Gal (α-OCH3), plays many important biological functions. Stereoselective synthesis of this disaccharide using thioglycoside, glycosyl bromide, and O-glycosyl trichloroacetate respectively as donors are described. The results indicate that the target compound is prepared by using the third compound as donor under mild condition with higher yield and higher stereoselectivity.

Abstract:The multifaceted biological importance of oligosaccharides and glycoconjugates has made them very popular synthetic targets in modem synthetic chemistry. Great progress has been made in the construction of complex oligosaccharides based on a variety of synthetic strategies. Among these strategies, the one-pot glycosylation is very attractive and shows great promise for assembly of oligosaccharide libraries and automated synthesis. Traditional one-pot glycosylation approach based on the chemoselective principle or orthogonal strategy needs extensive protecting group manipulations and/or aglycon adjustments for designing building blocks with suitable anomeric reactivities. The recently established pre-activation strategy has some advantages over the traditional one-pot glycosylation approach, since it does not require the tuning of the building blocks in an armed-disarmed fashion. The latest advances in one-pot oligosaccharide synthesis by pre-activation strategy are summarized in this review.

Abstract:An efficient route to orthogonally protected l-glucose, l-mannose, and l-galactose is described. Using the strategy to switch the functional groups at C1 and C5 of d-glucose, l-glucose is obtained via a key intermediate C-glycoside. Preparations of l-mannose and l-galactose derivatives from the orthogonally protected l-glucose were accomplished in good yields using the Lattrell–Dax method.
