Abstract:OBJECTIVE To investigate the therapeutic effect of Bcl-2 fusion protein on apoptosis in brain following traumatic brain injury. METHODS Bcl-2 gene was cloned by RT-PCR. Bcl-2 and EGFP genes were linked together and inserted into pAdeno-X vector. This recombinant vector was packaged into infectious adenovirus in HEK293 cells. Ninety Wistar rats were assigned randomly into experimental group (n=45) and control group (n=45). All rats were subjected to traumatic brain injury. Then recombinant adenovirus (for experimental group) or saline (for control group) was injected into the traumatic brain. The expression of Bcl-2 fusion protein was investigated by Western blotting, immunohistochemistry and fluorescence microscopy. Apoptosis in the injured brain was studied by TUNEL. Animals' behavior capacity was evaluated by tiltboard test. RESULTS In the experimental group, many fluorescent cells were found around the traumatic locus, which were also proven to be Bcl-2 positive by immunohistochemistry. On the contrary, few Bcl-2 positive cells and no fluorescent cell were detected in the control group. Bcl-2 expression of experimental group was much higher than that of control group, which was illustrated by Western blotting. The apoptosis index of experimental group was 0.027+/-0.005, and that of control group was 0.141+/-0.025 (P < 0.01). Two weeks after injury, animals of the experimental group behaved better than those of the control group. CONCLUSIONS A recombinant adenovirus vector expressing Bcl-2 fusion protein has been constructed. Bcl-2 fusion protein can suppress apoptosis and promote cell survival. Moreover, the behavior recovery of the injured animal is promoted. Bcl-2 fusion protein provides a way to track the target cells in vivo.

Abstract:Recovery after spinal cord injury (SCI) is rare in humans and experimental animals. Following SCI in adults, changes in gene expression and the regulation of these genes are associated with the pathological development of the injury. High levels of brain-derived neurotrophic factor (BDNF) in the injury area during the post-injury period contribute to enhanced neuroprotection and axonal regeneration. Intervention at the level of gene regulation has the potential to promote SCI repair. In this study, the injection of adenovirus-mediated BDNF in the lesion area (rostral spinal cord) up-regulated the expression of BDNF in the injury zone of a compression model in rat, thereby protecting neurons and enhancing behavioral function.
Cell Apoptosis and the Expression of Bcl-2 and Bax Genes in Neurons after Traumatic Brain Injury in Rats

Abstract:Objective To investigate the relationship between the expression of bcl-2,bax genes and posttraumatic cellular apoptosis in various degrees of head injury in rats.Methods In the study the improved Feeney's free falling body injury model was adopted in100rats.The cell apoptosis was examined with TUNEL(terminal-deoxynucleotidyl transferase mediated nick end labeling)method and the bcl-2and bax gene exˉpression was detected with Immunohistochemisty method.Results The apoptosis Labeling Index(LI)was sigˉnificantly higher in injured groups than that in controls(P0.05)and the expression rate of bcl-2and Bax was also higher in the injured rats(P0.05).And in injured groups the ratio of bax/bcl-2was higher than that in control group(P0.05).These parameters in mild and moderate groups presented significant differences compared with the severe group.Conclusion The expression of bcl-2and bax may play an important role in posttraumatic cellular apoptosis of brain and the levels of bcl-2and bax were proportional to the degree of head injury.
Brain-derived Neurotrophic Factor Combined with Neural Stem Cells Transplantation in Treatment of Severe Traumatic Brain Injury in Rats

Abstract:Objective To evaluate the efficacy of brain-derived neurotrophic factor (BDNF) combined with neural stem cell transplantation in treatment of severe traumatic brain injury in rate. Methods The rats were randomly divided into 4 groups; 3 experimental groups and 1 control group. After the sever traumatic brain injury was induced, rats in experimental groups were treated with BDNF and neural stem cell transplantation, animals in control groups were treated with transplantation only. The behavior of animals was observed and the blood nitrous oxide (NO) levels were measured. After the animals were sacrificed the brains were examined immunohistochemistry. Results The recovery of nerve function in experiment groups was better than that in control group(P0.05 or P0.01); the differentiation ratio of neural stem cells to neuron in experiment groups was higher than that in control group(P0.05)., the blood NO contents in experimental groups were lower than those in control group(P0.01). Conclusion The combined treatment of BDNF with neural stem cell transplantation may have better therapeutic effect on head injury than the transplantation alone.

Abstract:Functional recovery after transplantation of brain-derived neurotrophic factor (BDNF)-modified neural stem cells (NSCs) was evaluated in a rat model of cerebral ischaemia damage induced by temporary middle cerebral artery occlusion (tMCAO). Western blotting and enzyme-linked immunosorbent assay demonstrated up-regulated BDNF protein expression by rat embryonic NSCs transfected with the human BDNF gene (BDNF-NSCs). BDNF-NSCs stimulated neurite outgrowth in co-cultured dorsal root ganglion neurons, suggesting that BDNF increased neurogenesis in vitro. In vivo, BDNF promoted recovery of tMCAO. Phosphate-buffered saline, untransformed NSCs or BDNF-NSCs were introduced into the penumbra zone of the right striatum of tMCAO rats and neurological function deficit was assessed for up to 12 weeks using the neurological severity score (NSS). The NSS was significantly lower in the BDNF-NSC transfected transplant group than in all the other groups from week 10. BDNF-NSCs recovered 1 week after transplantation expressed BDNF protein. Transplanted NSCs had differentiated into mature neurons 12 weeks after transplantation. Transgenic NSCs have potential as a therapeutic agent for brain ischaemia.
The Expression of Bcl-2,Bax and Apoptosis after Severe Brain Injury

Abstract:Objective To discuss the relationship between expression of Bcl-2,Bax and apoptosis after severe brain injury. Methods 50 rats were experimented, which were injured in the brain. TUNEL (terminal-deoxynucleotidyl transferase mediated nick end labeling) and immunohistochemisty were used to detect apoptosis and Bcl-2, Bax expression in all the brain tissue.Results There were significant differences between the injury group and noninjury group with more TUNEL?Bcl-2 Bax positive cells in the injured rats. And in injured rats the value of Bax/Bcl-2 was larger than that in uninjured rats.Conclusion Disequilibrium between the Bcl-2 and Bax may be the reason of promoting apoptosis after severe brain injury.

Abstract:Objective To find out if transplanting neural stem cells with different quantity after traumatic brain injury(TBI) would make different outcome, hence , to decide the appropriate quantity for transplantation. Methods Transplanted neural stem cells (NSCs) with different quantity of 20 thousand, 1 million, and 2 million cells at 24 hours after traumatic brain injury of rats. The motor function of rats was assessed. The techniques of immunohistochemistry were applied to define the NSCs, differentiation and neuroprotection. Results Brain injured animals that have received NSCs transplants with the quantity of 1 million had marked and significant improved motor function scores compared with other groups. Conclusion Too much quantity of neurotransplantation was not always effective. The quantity of middle level would be appropriate.

Abstract:Objective　To demonstrate morphologic features of the cell suspension of the embryonic brain tissues which were transplanted into lesion c avities of brain injury models. 　Methods　In 46 SD rats, cerebral contusion models were made b y dropping weight method (impact energy: 20 g×30 cm)； of which 16 rats were sa crificed at 14, 20, 30, 40 days after injury and their lesion brains under HE staining, the other 30 rats transplanted by embryonic tissue homogenate at 14 days after injury. The grafted rats were sacrificed at 1, 5, 14, 30, 75 days after implant ation and their brains stained by HE dye. 　Results　All experimental rats survived and no hemiplegia see n. The concentration of fetal brain suspension was 4.48×107 cells/ml. The living cell was 85.3% and neurocyte 48.2%. After 14 days posterior to injury , the main features under microscope was glia proliferation and scar tissue. The immunocytes were found 5 days after transplantation; the grafts survived and t he capillary and the glia proliferation in host were noted 14 days later. Some n eurofibers were derived from cells within the grafts 30 days after transplan tation. The graft was well integrated into host at 75 days posterior to implanta tion. 　Conclusions　The donor cells can be survived and influence ho st and grafts itself.
Lymphocyte infiltration induced by transplantation of neural stem cells into traumatically injured brain

Abstract:Objective: The process of immune response to neural stem cell allograft has not been fully understood. This study was designed to investigate the lymphocyte infiltration induced by transplantation of neural stem cells into traumatically injured brain. Method: Sixty SD rats were assigned randomly to the transplantation group (n = 30) and the control (n = 30). All animals were subjected to focal brain contusion following the Feeney's method. Then the neural stem cells derived from newborn Wistar rats were transplanted into the traumatically injured brains. As for the rats of the control group, they were injected saline instead. Lymphocyte infiltration in the lesion was studied by means of HE staining, immunohistochemistry and flow cytometry. The immunogenicity of neural stem cells was evaluated by their expression of MHC- I molecules, as shown by flow cytometry. Results: About 6.57% ± 0.44% of neural stem cells expressed MHC-I molecules. Histological examination and immunohistochemistry revealed that many lymphocytes infiltrated in the adjacent area of the contusion. No similar lymphocyte infiltration was found in the control group. As shown by flow cytometry, in the traumatic brain tissues of the transplantation group, the cell number percentage of CD4 + lymphocytes was 13.28% ± 1.60%, and that in the control group was 0.41% ± 0.12%, P < 0.01. Likewise, the percentage of CD8 + lymphocytes of the transplantation group was 5.11% ± 1.03%, and that of the control was 0.57% ± 0.26%, P <0.01. Conclusions: Neural stem cells are inherent in immunogenicity. When transplanted into traumatically injured brain, they can induce lymphocyte infiltration.
Neuroprotective Effect of Erythropoiet in Treatment of Traumatic Brain Injury in Rats

Abstract:Objective To evaluate the neuroprotective effects of erythropoiet (EPO) for traumatic brain injury (TBI) in rats and investigate the possible role of endoplasmic reticullum stress response and Caspase-12-induced apoptosis.Methods According to the random number table,140 male Sprague-Dawley rats were divided into sham injury group,sham injury plus EPO group,TBI group and TBI plus EPO group,with 35 rats per group.TBI was induced by a fluid percussion device.EPO (5 000 U/kg in saline) was administered intraperitoneally at 6 hours after injury.The rate of TUNEL positive cells in injured cortex were measured to evaluate cell apoptosis status.Neurological function was assessed at days 1,4,7,21,28 and 35 after intervention using a modified neurological severity score (mNSS).At 24 hours after injury,the expressions of Caspase-12 in injured cortex and C/EBP-homologous protein (CHOP) which was the symbol of ERS response were measured by Western blot and immunofluorescent staining to assess the changes of ERS response after TBI and EPO treatment.Results TUNEL-positive staining cell density was significantly increased by (30.3 ± 2.3) ％ in the injured cortex 24 hours after injury (P ＜ 0.01).Compared with TBI group,TBI plus EPO group had a significant decrease of the positive rate of TUNEL cells [(14.6 ± 1.5) ％] (P ＜ 0.01).Compared with TBI group,mNSS score significantly was decreased in TBI plus EPO group at 7-35 days after injury (P ＜ 0.05).At 24 hours after injury,the results of Western blot showed that the expression levels of Caspase-12 and CHOP in the injured cortex in TBI group were higher than those in sham group,but that in TBI plus EPO group was lower than those in TBI group (P ＜ 0.01).At 24 hours after injury,the results of immunofluorescent staining showed the rates of Caspase-12 and CHOP positive cells in the injured cortex in TBI group were higher than sham group (P ＜ 0.01).But the rates of Caspase-12 and CHOP positive cells in TBI plus EPO group was lower than that in TBI Group (P ＜ 0.01).Conclusion Exogenous EPO has significant neuroprotective effects on TBI rats.EPO may exert its neuroproective effects through suppression of ERS response and inhibition of Caspase-12-induced apoptosis.

Abstract:Cytoskeletal proteins are involved in neuronal survival. Brain-derived neurotrophic factor can increase expression of cytoskeletal proteins during regeneration after axonal injury. However, the effect of neural stem cells genetically modified by brain-derived neurotrophic factor transplantation on neuronal survival in the injury site still remains unclear. To examine this, we established a rat model of traumatic brain injury by controlled cortical impact. At 72 hours after injury, 2 × 10<sup>7</sup> cells/mL neural stem cells overexpressing brain-derived neurotrophic factor or naive neural stem cells (3 mL) were injected into the injured cortex. At 1-3 weeks after transplantation, expression of neurofilament 200, microtubule-associated protein 2, actin, calmodulin, and beta-catenin were remarkably increased in the injury sites. These findings confirm that brain-derived neurotrophic factor-transfected neural stem cells contribute to neuronal survival, growth, and differentiation in the injury sites. The underlying mechanisms may be associated with increased expression of cytoskeletal proteins and the Wnt/β-catenin signaling pathway.

Abstract:BACKGROUND AIMS This study aimed to observe nine factors expressed in rat ischemic brain after transplantation of bone marrow stromal cells (BMSC) and/or endothelial progenitor cells (EPC). These factors were vascular endothelial growth factor (VEGF), stromal cell-derived factor-1 (SDF-1), basic fibroblast growth factor (bFGF), insulin-like growth factor (IGF-l), transforming growth factor-β (TGF-β), platelet-derived growth factor-BB (PDGF-BB), brain-derived neurotrophic factor (BDNF), glial cell line-derived neurotrophic factor (GDNF) and nerve growth factor (NGF). METHODS Adult Wistar rats were divided randomly into four groups: a vehicle group, BMSC group, EPC group and BMSC combined with EPC group. The rats were subjected to middle cerebral artery occlusion (MCAO) then implanted intravenously with 3 × 10(6) BMSC, EPC, BMSC/EPC or phosphate-buffered saline (PBS) 24 h after MCAO. Neurologic functional deficits were measured on days 1, 7, 14, 28 after transplantation. On day 7 after transplantation, quantitative reverse transcription (qRT)-polymerase chain reaction (PCR) and Western blot were employed to detect the expression of VEGF, SDF-1, bFGF, IGF-l, TGF-β, PDGF-BB, BDNF, GDNF and NGF. RESULTS The neurologic evaluation found that the neurologic severity scores were no different between the four groups on day 1, and the scores of rats in the BMSC/EPC group were significantly lower than those of rats in the other groups on days 7, 14 and 28 after transplantation. The expressions of bFGF, VEGF and BNDF were significantly higher in the BMSC/EPC group compared with the other groups. CONCLUSIONS The intravenous transplantation of BMSC combined with EPC could promote the functional rehabilitation of rats with focal cerebral ischemia, and the mechanism may be related to the enhanced expression of factors.
Bcl-2 Enhances the Formation of Newborn Striatal Long-Projection Neurons in Adult Rat Brain after a Transient Ischemic Stroke

Abstract:OBJECTIVE:It has been reported that B-cell lymphoma 2 (Bcl-2) enhances neurogenesis as well as supporting axonal growth after injury. In the present study, we investigated whether Bcl-2 overexpression plays a role in the formation of newborn striatonigral projection neurons in the adult rat brain after transient middle cerebral artery occlusion (MCAO).METHODS:We infused human Bcl-2-expressing plasmid (pBcl-2) into the lateral ventricle immediately after 30 min of MCAO, injected 5'-bromodeoxyuridine (BrdU) intraperitoneally to label proliferative cells, and microinjected fluorogold (FG) into the substantia nigra at 11 weeks of reperfusion followed by multiple immunostaining of striatonigral projection neurons at 12 weeks.RESULTS:We found that pBcl-2 treatment significantly increased the number of newborn neurons (BrdU(+)-NeuN(+)) in the striatum ipsilateral to the MCAO. We further detected newborn striatonigral projection neurons (BrdU(+)-FG(+)-NeuN(+)) in the ipsilateral striatum at 12 weeks. More interestingly, the number of newborn striatonigral projection neurons (BrdU(+)-FG(+)) was significantly increased by pBcl-2 treatment compared to that by pEGFP, a control plasmid.CONCLUSION:Taken together, we found that Bcl-2 overexpression in the brain enhanced the generation of newborn striatonigral projection neurons. This provides a potential strategy for promoting the reestablishment of neural networks and brain repair after ischemic injury.
THE STUDY OF NEUROPROTECTION OF Bcl-2 GENE BY TRANSFECTING IT INTO PC12 CELL LINE

Abstract:Objective To investigate the biological effects of bcl-2 gene on neurons. Methods The recombinant expression plasmid pc DNA3-bcl-2 was constructed from pSFFV-bcl-2,then it was introduced into PC12 cell line by liposome method.Western blotting and immunohistochemistry in situ were applied to exam the exogenous gene expression.The two groups of cells(Group A,PC12 transfected by pcDNA3-bcl-2 and Group B,PC12 transfected by pcDNA3) were exposed to cisplatin with the concentration of 10*!μmol/L,50*!μmol/L, and 100*!μmol/L 72 hours later,the survival cells were estimated.Cell cycle indexes between these two groups were also studied by FCM. Results The recombinant expression plasmid pcDNA 3-bcl-2 was constructed successfully and PC12 cell line transfected by the plasmid could express Bcl-2 protein effectively.Compared with the control(25 79%),there was a significant decrease of cells from the S phase in PC12 with bcl-2 gene(8.81%).After exposed with 10*!μmol/L,50*!μmol/L,and 100*!μmol/L cisplatin,the surviving cells in group A were 276±13,185±11 and 108±10 respectively,which increased much more than in group B while they were 100±9,12±3 and 2±2 accordingly.Conclusions bcl-2 can protect PC12 cells against cytotoxic insults of cisplatin,and it suggested that it might act via cell cycle controlling.
The expression and distribution of Nrf2-ARE in rats models of traumatic brain injuries

Abstract:Objective To observe the dynamic expression and distribution of Nrf2-ARE after traumatic brain injuries,and to explore the significant correlations between the Nrf2-ARE pathway and traumatic brain injuries.Methods Controlled cortical impact (CCI) injury was performed in Sprague-Dawley rats.Animals were randomly divided into 6 groups according to different times:1 h,6 h,12 h,24 h,48 h and 72 h.RT-PCR and Western blotting analysis of Nrf2 were performed according to different times after TBI.The expression and distribution of Nrf2 after TBI was examined by immunofluorescence.Results RT-PCR shows that there were no significant differences in Nrf2 mRNA levels after TBI (P ＞0.05).The Western blotting shows that there was a statistically significant difference between the sham group and each TBI group (P ＜ 0.01,respectively).After TBI,Nrf2 was induced in the ipsilateral cortex,which was the most vulnerable region to TBI.Little reactivity was observed in the sham group,whereas strong Nrf2 immunostaining was detected in these regions 24 h after TBI.Both nuclear and cytoplasmic staining was observed in glial cells and neurons.Conclusion The Nrf2-ARE pathway is activated in the brain in the early stage after TBI,which means that the Nrf2/ARE signaling pathway may be an endogenous antioxidant mechanism in the cellular defense against oxidative stress after TBI.

Abstract:Objective:To study the effects of transplanted induced neural stem cells (iNSCs) receiving CR2-Crry pre-treatment on neuroprotection following brain injury.Methods:Healthy adult male C57BL/6 mouse brain injury models were established and mice having an neurological severity scores (NSS) of 4-8 were enrolled in brain injury group (n=18). These brain injury animals were randomly divided into 3 groups: the CR2-Crry-iNSC group (n=6), the phosphate buffered solution (PBS)-iNSC group (n=6) and the PBS group (n=6). At 12 h after brain injury, CR2-Crry-iNSCs, PBS-iNSCs or PBS were separately injected into the brains of brain injury mice via stereotactic method. On day 14 after brain injury, mice were sacrificed for morphological analysis. Immunofluorescent staining and TdT-mediated dUTP nick and labeling (TUNEL) staining were utilized to determine the effects of CR2-Crry-iNSC and PBS-iNSC grafts on Crry+/NeuN+ and NeuN+/TUNEL+ cells in the brains of brain injury mice.Results:Double-labeling experiments revealed that the level of Crry+/NeuN+ neurons in the brains of the PBS group was significantly lower than that in CR2-Crry-iNSC and PBS-iNSC groups (P<0.05). Furthermore, the level of Crry+/NeuN+ neurons was substantially higher in the CR2-Crry-iNSC group than in the PBS-iNSC group (P<0.05). In contrast, TUNEL staining showed that the level of NeuN+/TUNEL+ neurons in the brains of the PBS group was significantly higher than that in CR2-Crry-iNSC and PBS-iNSC groups (P<0.05). Moreover, the level of NeuN+/TUNEL+ neurons was substantially lower in the CR2-Crry-iNSC group than in the PBS-iNSC group (P<0.05).Conclusion:Transplantation of iNSCs receiving CR2-Crry pre-treatment could increase the levels of Crry expression in neurons and alleviate complement-mediated neuronal damage following brain injury.
Brain-Derived Neurotrophic Factor Increases Synaptic Protein Levels via the MAPK/Erk Signaling Pathway and Nrf2/Trx Axis Following the Transplantation of Neural Stem Cells in a Rat Model of Traumatic Brain Injury

Abstract:Brain-derived neurotrophic factor (BDNF) plays an important role in promoting the growth, differentiation, survival and synaptic stability of neurons. Presently, the transplantation of neural stem cells (NSCs) is known to induce neural repair to some extent after injury or disease. In this study, to investigate whether NSCs genetically modified to encode the BDNF gene (BDNF/NSCs) would further enhance synaptogenesis, BDNF/NSCs or naive NSCs were directly engrafted into lesions in a rat model of traumatic brain injury (TBI). Immunohistochemistry, western blotting and RT-PCR were performed to detect synaptic proteins, BDNF-TrkB and its downstream signaling pathways, at 1, 2, 3 or 4 weeks after transplantation. Our results showed that BDNF significantly increased the expression levels of the TrkB receptor gene and the phosphorylation of the TrkB protein in the lesions. The expression levels of Ras, phosphorylated Erk1/2 and postsynaptic density protein-95 were elevated in the BDNF/NSCs-transplanted groups compared with those in the NSCs-transplanted groups throughout the experimental period. Moreover, the nuclear factor (erythroid-derived 2)-like 2/Thioredoxin (Nrf2/Trx) axis, which is a specific therapeutic target for the treatment of injury or cell death, was upregulated by BDNF overexpression. Therefore, we determined that the increased synaptic proteins level implicated in synaptogenesis might be associated with the activation of the MAPK/Erk1/2 signaling pathway and the upregulation of the antioxidant agent Trx modified by BDNF-TrkB following the BDNF/NSCs transplantation after TBI.

Abstract:Neuroprotection is a major therapeutic approach for ischemic brain injury. We investigated the neuroprotective effects induced by transplantation of human embryonic neural stem cells (NSCs) into the cortical penumbra 24 h after focal cerebral ischemia.NSCs were prepared from human embryonic brains obtained at 8 weeks of gestation. Focal cerebral ischemia was induced in adult rats by permanent occlusion of the middle cerebral artery. Animals were randomly divided into two groups: NSCs-grafted group and medium-grafted group (control). Infarct size was assessed 28 days after transplantation by hematoxylin and eosin staining. Neurological severity scores were evaluated before ischemia and at 1, 7, 14, and 28 days after transplantation. The terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay and immunohistochemical analysis of Bcl-2 and Bax were performed at 7, 14, and 28 days after transplantation.Physiological parameters of the two groups were comparable, but not significantly different. NSC transplantation significantly improved neurological function (P<0.05) but did not reduce the infarct size significantly (P>0.05). Compared with the control, NSC transplantation significantly reduced the number of TUNEL- and Bax-positive cells in the penumbra at 7 days. Interestingly, the number of Bcl-2-positive cells in the penumbra after NSC transplantation was significantly higher than that after medium transplantation (P<0.05).The results indicate that NSC transplantation has anti-apoptotic activity and can improve the neurological function; these effects are mediated by the up-regulation of Bcl-2 expression in the penumbra.
Effect of Brain Derived Neurotrophic Factor on TrkB Mrna and Protein Expression in Embryonic Rat Brain Suffered from Intrauterine Hypoxic-Ischemic Injury

Abstract:Objective:To investigate the effect of brain derived neurotrophic factor on TrkB mRNA and Protein expression in embryonic rat brain suffered from intrauterine hypoxic-ischemic injury.Methods:The uterine arteries of pregnant rats were clamped for 30 min,and saline or BDNF was injected to the rat through caudal veins for 5 days.Rats with sham surgery were used as controls.The expression of TrkB mRNA and protein was measured using RT-PCR and Western blotting.Results:TrkB mRNA and levels of protein expression were higher in embryonic rat brain after clamping the uterine arteries of pregnant rats for 30 min than those in control group.BDNF injection could up-regulate the TrkB expression significantly.Conclusion:BDNF injection through rat caudal veins could up-regulate the levels of the TrkB mRNA and protein expression in embryonic rat brain suffered from intrauterine hypoxic-ischemic injury and may enhance its neuroprotection effect on cortical neurons.

Abstract:To determine whether Bcl-2 could influence adult neurogenesis and prevent apoptosis of newborn neurons, we injected Bcl-2 expressing plasmid into the lateral ventricle of rat brain immediately following a 30-min occlusion of the middle cerebral artery (MCAO). We found that Bcl-2 increased neural progenitor cells (BrdU+-DCX+) in the ipsilateral striatum, newborn immature neurons (BrdU+-Tuj-1+) and newborn mature neurons (BrdU+-MAP-2+) in the ipsilateral striatum and frontal cortex at 1 to 4 weeks following MCAO. Bcl-2 overexpression promoted development of newborn neurons into GABAergic and cholinergic neurons in the ipsilateral striatum. Moreover, Bcl-2 significantly decreased the apoptosis of newborn neurons, determined by double staining of Tuj-1 and activated caspase-3 (Tuj-1+-Casp+). These results indicate that overexpression of Bcl-2 in adult rat brain enhances neurogenesis and survival of newborn neurons. Increasing neurogenesis and preventing the death of newborn neuron may be a strategy to aid in the repair of adult brain after stroke.
