Abstract:AIM: To study the effect of wild type (wt) p53 gene transfection on drug resistant human hepatocellular carcinoma (HCC) cells induced by 5-Fluorouracil (5-FU). METHODS: The cytotoxicity of anticancer drugs on Bel7402 and Bel7402/5-FU cells was assessed using SRB assay. p53 expression was detected at its mRNA level by RT-PCR assay and at its protein level Western blot or immunocytochemistry assay in Bel7402/5-FU cells transfected with either control vector or wt p53. AnnexinV-FITC/PI double labeled assay was performed to detect apoptosis. The chemosensitivity of Bel7402/5-FU cells transfected with wt p53 was assessed using SRB assay. RESULTS: Bel7402/5-FU cells exhibited cross-resistance to vincristine, doxorubicin, paclitaxel, and so on. wt p53 gene transfection upregulated the expression of p53 in Bel7402/5-FU cells. wt p53 was able to greatly inhibit cell proliferation and significantly induce apoptosis in Bel7402/5-FU cells. Moreover, wt p53 gene transfection increased the chemosensitivity of Bel7402/5-FU cells to some anticancer drugs. CONCLUSION: These results indicated that the wt p53 gene transfection not only induced suppression of cell growth, but also increased the sensitivity of Bel7402/5-FU cells to 5-FU, vincristine, and doxorubicin.
Change From Promotion To Inhibition On Apoptosis In Hepatoma Cells Transferred Wild-Type P53 Gene

Abstract:In order to study the relationship between p53 gene and hepatoma the wild-type p53 gene(wt-p53) was transferred into HCC-9204 hepatoma cell line, in which p53 proteins were overexpression immunohistochemically, and the mutation was found in exon 7, detected by SSCP. Human wt-p53 of 1.8 Kb was inserted at BamHI in the retrovirus pLXSN vector. Then PA 317 cells were transfected by the p63-pLXSN viruses.The medium containing p53pLXSN particles was used to culture the hepatoma cells, which were further screened by G418. Based on the model of apoptosis induced by adriamycin at 20 uM/L for 3 hours, the sensitivity of apoptosis was increased by wt-p53 in transferred cells, in which the apoptotic cell account was increased from 30.7% to 72.1%, detected by flow cytometry analysis. However, the apoptosis could not be induced by the same treatment, even more than for 24 hours, in the 15th generation transferred cells. The results were repeatable. The mechanisms of this change from promotion to inhibition on apoptosis in hepatoma cells transferred wtp53 in stabe is unknown.

Abstract:Objective:To investigate the effect of exogenous wild type p53 gene transfection on the sensitivity of T lymphocytic lymphoma cell line Jurkat to chemotherapy.Methods:p53-gene was transfected into Jurkat cells by liposome mediation.Jurkat cells with stable expression of exogenous p53 gene were screened by G418.The cell viability was detected by trypan blue exclusion test.Then the cells were exposed to various concentrations of doxorubicin(ADM,0.1-100.0 μg/mL) and etoposide(VP16) for 24 h,and the cell viability was measured by MTT assay.Results:The inhibitory ratio on the proliferation of Jurkat cells with stable expression of exo-genous p53 gene was 50% after treatment with 0.47 μg/mL ADM or 2.5 μg/mL VP16.Compared with the control cells transfected with empty vector or without any transfection,the value of half inhibitory concentration(IC50) was significantly decreased(P<0.05).Conclusion:Expression of p53 could increase the sensitivity of T lymphoma Jurkat cells to ADM and VP16.p53 gene combined with chemotherapy regimen may become a novel approach to treat drug-resistant T lymphoma.

Abstract:e22018 Background: Several studies demonstrated that p53 gene transfection enhanced the anti-tumor effects of radiotherapy. This study is to investigate whether p53 gene transfection can increase the radiosensitivity of liver tumor cells. Methods: Liver tumor cell lines, HepG2 (with wild type p53 genes) and PLC/PRF/5 (with mutant p53 genes), were separately transfected with recombinant adenoviral human p53 gene (rAdp53) and adenoviral enhance green fluorescent protein gene (AdEGFP), forming 4 types of cells: HepG2 transfected with rAdp53, HepG2 with AdEGFP, PLC/PRF/5 with rAdp53, and PLC/PRF/5 with AdEGFP. The transfected and untransfected HepG2 and PLC/PRF/5 cells were irradiated with 6MV-X at doses of 0, A2, A4, A6, A8, A and 10 Gy. After exposition to radiation, cell survival, clonogenic capacity, and apoptosis were analyzed. The effect differences between cell types were analyzed using statistical methods of pairwise group comparisons and mixed effect model. Results: After exposing irradiation, all the cells’ survival rate and clonogenic capacity decreased, and the proportion of apoptotic cell increased. These effects become stronger with increaseing dose of radiation. Between untransfected cells, radiation had a stronger effect on HepG2 cells than PLC/PRF/5 and the differences were statistically significant for all the 3 measures. The rAdp53 transfection, not the AdEGFP, significantly enhanced radiation effects on both cell lines. The enhanced effects on PLC/PRF/5 cells were significantly stronger then the enhanced effects on HepG2 cells. Conclusions: PLC/PRF/5 cells with mutant p53 genes were more resistant to radiation then HepG2 with wide type of p53 genes. The rAd-P53 transfection could enhance radiosensitivity of both cell lines, but the enhanced effect on PLC/PRF/5 cells with mutant p53 gene was stronger than HepG2 with wide type of p53 genes.

Abstract:Purpose:To investigate the effects of tumor suppressor gene p53 on the activity of telomerase/hTERT in human HCC-9204 cell line.Methods:Lipofection-mediated gene transfection method was used to transfect wild-type p53(wt-p53) gene into HCC 9204 cell which expresses telomerse /hTERT and carries mutant-type p53 gene. The expression of p53 was confirmed by Western blot. Both the telomerase activity and hTERT mRNA expression were detected by PCR-ELISA,TRAP-silver staining,in-situ hybridization and RT-PCR methods. The apoptotic appearance was examined by FCM.Results:Higher telomerase activity and hTERT mRNA level were detected in HCC-9204,and they were markedly inhibited after transfection with wt-p53. Meanwhile,decreasing level of bcl-2 protein and appearance of apoptosis were also shown in the transfected cells.Conclusions:Over expression of the exogenous wt-p53 gene does suppress both telomrease activity and hTERT mRNA expression in HCC cell line. There is a p53-dependent regulatory pathway for activation and expression of telomerase/hTERT in HCC.

Abstract:OBJECTIVE:To investigate the effect of wt p53 gene transfection on the sensitivity of hyperthermia in C6 glioma.METHODS:The recombinant eukaryotic expressive vector pCMV-p53 plasmids were extracted, identified and transmitted to C6 glioma cells by stable transfection. The biological characteristics were compared among C6/p53 (+) cells, thermo-treated C6/p53 (+) cells, thermo-treated C6 cells, thermotolerance C6 cells and control C6 cells. Transplanted glioma was treated by laser interstitial thermotherapy (LITT) in tumor-bearing nude mice.RESULT:The wt p53 gene segments were extracted and correctly identified by restriction enzyme (Hind and EcoR). The optimal transfection concentration of plasmid to liposome was 1:6, neo gene expressed stably in C6 cells transfected with positive and blank plasmid. Thermotolerance C6 cell growth was normal in vitro, growth of C6/p53 (+) cells was inhibited. Growth of hyperthermia treated C6 cells was inhibited significantly 12 h after thermal treatment, cell proliferation activity of C6/p53 (+) cells was suppressed significantly 6 h after hyperthermia treatment. Apoptosis was observed in C6/p53 (+), thermo-treat C6 and thermo-treat C6/p53 (+) cells. The apoptosis ratio of hyperthermia treated C6/p53(+) cells was increased by 30 folds compared with that of control groups. Study in nude mouse model demonstrated lower positive inoculation rate in groups of C6/p53 (+) cells (57 %), hyperthermia treated C6 cells (75 %) and hyperthermia treated C6/p53(+) cells (20 %). C6/p53(+) LITT group showed the highest antitumor effect, compared with C6 LITT group(P<0.05). However, thermotolerance C6 LITT group had the lowest effect.CONCLUSION:Transfection of wt p53 gene can increase the sensitivity of hyperthermia in glioma cells.

Abstract:Purpose : The p53 gene is considered one of the most important in the control of apoptosis, and its mutations have a close relationship with chemosensitivity. The aim of this work was to investigate the role of p53 in the apoptosis of colorectal cancer cells in vitro, induced by 5-fluorouracil (5-FU) and hydroxy-camptothecin (HCPT). Methods : A total of 39 colorectal cancer samples from patients were treated in vitro with 5-FU (10 μg/ml), 5-FU (10 μg/ml) + leucovorin (5 μg/ml), HCPT (0.1 μg/ml) and HCPT (0.1 μg/ml) + Salvia mitorrhiza (6 μl), using an in situ terminal deoxynucleotidyltransferase assay to detect chemosensitivity. p53 gene mutations from tumor DNA were detected, after amplification by the polymerase chain reaction of exons 5–8, by non-radioactive single-strand conformation polymorphism. Results : p53 gene mutations were observed in 43.6% (17/39) of colorectal carcinomas, when the terminal deoxynucleotidyltransferase assay was used to detect the tumor apoptotic rate. Cells with mutated p53 had lower chemosensitivity than those without ( p < 0.01). Conclusion : Routine assessment of p53 status may be helpful in selecting patients with the wild-type p53 gene, who have a predictably better response to chemotherapy.

Abstract:Objective To evaluate the effects of wild-type p53 gene on the growth of human colorectal carcinoma SW1116 cells. Methods Recombinant retroviral vector (Dol-p53) carrying wild-type p53 gene was transfered into SW1116 cells, the cell growth curve, soft agar colony formation and tumorigenesis in nude mice of transfected SW1116 cells were evaluated and the change of their biologic characteristics was also analyzed. Results The transfection of wild-type p53 gene into SW1116 cells can suppress the phenotype of SW1116 cells, including inhibition of proliferation in culture, anchorage-independent growth and tumorigenecity. Conclusion Wild-type p53 gene can inhibit the growth of human colorectal carcinoma cells.

Abstract:The p53 null HL-60 cell line was transfected with plasmids coding for either the wild-type p53 or mutant p53 gene. The stable expression of wild-type p53 resulted in a significant increase in sensitivity to the topoisomerase II poisons etoposide and doxorubicin, but not to the topoisomerase II inhibitors razoxane and ADR-529. HL-60 cells expressing wild-type p53 demonstrated 8- to 10-fold more VP-16 induced DNA breaks by the alkaline elution assay. The effect of inducible expression of wild-type p53 was also studied in the p53 null erythroblastoid cell line K562 and in the human squamous carcinoma cell line SqCC. The inducible expression of wild-type p53 in the K562 cell line resulted in a 3-fold increase in sensitivity to VP-16. The quantity of topoisomerase IIalpha was not altered by the transfection as determined by immunoblotting, while the amount of the beta isoform was increased 2.5-fold in HL-60 cells. The topo II catalytic activity present in nuclear extracts was measured as the decatenation of kinetoplast DNA, and found to be unaltered by p53 expression. Immunostaining for topoisomerase IIalpha was substantially diminished in both stable and inducible wild-type p53 expressing cells when three different antibodies were used (two polyclonal and one monoclonal). However, the addition of VP-16 resulted in a rapid appearance of nuclear fluorescence for topoisomerase IIalpha. No changes in topoisomerase IIbeta immunostaining were observed. These results suggest that an epitope for topoisomerase IIalpha is concealed in cells expressing wild-type p53 and that a complex between topoisomerase IIalpha and p53 may be disrupted by the addition of antitumor drugs.
Effect of Adenovirus-mediated Wild-Type P53 Gene Transfection on the Growth and Radiosensitivity of Human Lung Adenocarcinoma Cells

Abstract:Objective: To evaluate the effects of wild-type p53 gene on the growth and radiosensitivity of human lung adenocarcinoma cells.Methods: Recombinant adenovirus carrying wild-type p53 gene(Adp53) was transferred into human lung adenocarcinoma cell A549,and p53 protein expression was detected by immunocytochemistry.A549 cell was divided into 4 groups: control group,transfection group,radiation group and combined treatment group.The cell growth inhibition and apoptosisin were assessed by MTT and flowcytometry. Results: The transfection of p53 gene into A549 cells was confirmed by immunocytochemistry.MTT showed that the inhibition rate(IR) on the growth of A549 cells transferred into by Ad-p53 gene was(50.60±5.69)%.The IR of radiation(2,4,6 Gy) on A549 cells was respectively(16.06±(4.35))%,(16.39±1.67)%,(17.73±4.68)%.When combined treatment of wild-type p53 gene transfection and radiation was used,their IR was(80.60±5.35)%,(89.30±1.67)%,(90.30±2.01)%,significantly increased(P0.05).The apoptotic rate(AR) of A549 cells induced by p53 gene transfection was(10.38±1.68)%.Their AR by radiation(2,4,6 Gy) was(3.98±0.72)%,(4.84±0.60)%,((5.40)±0.70)%.The apoptotic rate was also significantly increased to(17.80±1.30)%,(19.03±(0.54))%,(21.34±2.51)% respectively after combined treatment of p53 and radiation(2,4,6 Gy) withdifferent doses(P0.05). Conclusion: Wild-type p53 gene and radiation could result in synergistic inhibition effect on the growth of human lung adenocarcinoma cells and increase radiosensitivity.

Abstract:AIM: To observe the reversal effects of wide-type p53 gene on multi-drug resistance to 5-FU (LOVO/5-FU).METHODS: After treatment with Ad-p53, LOVO/5-FU sensitivity to 5-Fu was investigated using tetrazolium dye assay. Multidrug resistance gene-1 (MDR1) gene expression was assayed by semi-quantitative reverse transcription-polymerase chain reaction and the expression of p53 protein was examined by Western blotting.RESULTS: The reversal activity after treatment with wide-type p53 gene was increased up to 4.982 fold at 48 h. The expression of MDR1 gene decreased significantly after treatment with wide-type p53 gene, and the expression of p53 protein lasted for about 5 d, with a peak at 48 h, and began to decrease at 72 h.CONCLUSION: Wide-type p53 gene has a remarkable reversal activity for the high expression of MDR1 gene in colorectal cancers. The reversal effects seem to be in a time dependent manner. It might have good prospects in clinical application.

Abstract:The tumor suppressor p53 is somatically mutated in half of all human cancers. Paradoxically, the wild-type p53 (WTp53) is often retained in certain human cancers, such as hepatocarcinoma (HCC). We discovered a physiological and oncogenic role of WTp53 in suppressing pyruvate-driven oxidative phosphorylation by inducing PUMA. PUMA inhibits mitochondrial pyruvate uptake by disrupting the oligomerization and function of mitochondrial pyruvate carrier (MPC) through PUMA-MPC interaction, which depends on IκB kinase-mediated phosphorylation of PUMA at Ser96/106. High expression levels of PUMA are correlated with decreased mitochondrial pyruvate uptake and increased glycolysis in HCCs and poor prognosis of HCC patients. These findings are instrumental for cancer drug discovery aiming at activating WTp53 or restoring WTp53 activity to p53 mutants.

Abstract:BACKGROUND:p53 is the most frequently mutated tumor-suppressor gene in human cancers. It has been reported that mutations in p53 result not only in the loss of its ability as a tumor suppressor, but also in the gain of novel cancer-related functions that contribute to oncogenesis. The present study evaluated the potential of silencing of mutant p53 by small interfering RNA in the treatment of bladder cancer cells in vitro.METHODS:We used the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay to assess cell viability and flow cytometry to detect cell cycle alterations and apoptosis. The related molecular mechanisms were assessed by western blotting. We also used the MTT assay and flow cytometry to investigate if silencing of mutant p53 by knockdown with small interfering (si)RNA would change the sensitivity to cisplatin treatment.RESULTS:Using 5637 and T24 human bladder cancer cell lines characterized by mutations in p53, we found that silencing of the mutant p53 by RNA interference induced evident inhibition of cell proliferation and viability, which was related to the induction of G2 phase cell cycle arrest and apoptosis. Moreover, our study also showed that the p53-targeting siRNA cooperated with cisplatin in the inhibition of bladder cancer cells.CONCLUSIONS:These findings suggest that RNA interference targeting mutant p53 may be a promising therapeutic strategy for the treatment of bladder cancer.
Exogenous Wild-Type P53 Gene Improved Survival Of Nude Mice Injected With Murine Erythroleukemia Cell Line Through Amelioration Of Hemorheological Changes

Abstract:Objectives: Previous investigations have shown that human wild-type p53 gene (WTp53) inhibits the growth of leukemia and tumor cells in vitro. In the present: study; the authors used nude mice and examined the therapeutic role of p53 gene for erythroleukemia ill vivo in the absence of MHC effects.Methods: The dude mice Were injected with murine erythroleukemia cells (MEL), MFL cells transfected with wild-type p53 gene (MEL-W), and MEL cells transfected with mutated p53 gene (MEL-M). Abnormalities were found in the hemorheological and biophysical properties of red blood cells in all :3 groups of animals; but: the abnormalities were lesser ill degree and later in appearance in MEL-W group than in MEL and MEL-M groups. Futhermore, the nude mice in MEL-W group lived longer than those ill MEL and MEL-M groups.Results: The results showed that: WTp53 restrained the growth of erythroleukemia cells in vivo and reduced the erythroleukemia tumorigenesis in the microcirculation by improving the hemorheological and biophysical properties of MEL cells; which helped to prolong the life span of nude mice suffering from erythroleukemia.Conclusion: These results contribute to our knowledge on the use of wild-type p53 gene for the treatment of erythroleukemia disease.

Abstract:Mutant p53 could have either a dominant negative effect or a gain of function to interfere with p53's ability to maintain genomic stability. In the present study, we screened for TP53 mutations in hepatocellular carcinoma (HCC) samples from 202 Chinese patients, followed by analysis of transcriptional and apoptotic activities of 21 p53 mutants with or without wild-type p53 present. We identified a new point mutation p.P72A, and a mutation (p.E294SfsX51) with a record long frameshift. We found TP53 mutations in HCC bear mutants without a dominant wild-type p53 inhibition on p21 transcription at a higher frequency. We found an anti-correlation for p53 WT/mutant heterotetramer to activate p21 and BAX transcription, i.e., at given p53 WT/mutant concentration, the fold increase p21 transcription is proportional to the fold of decreasing BAX transcription. Our kinetic model reproduced the trend in the experimental observation and confirmed that the p53 WT-dimer/mutant-heterotetramer is the major species to confer the differential activation of p21 and BAX transcription. p53 may have different binding modes on p21 and BAX, most likely resulting from the combinational effects of core domain binding and C-terminal mediation. Our study demonstrated that p53 mutants interfere with the ability of WT p53 to maintain genomic stability.

Abstract:Objective To identify the role of p53 in the induction of GADD45β in HCC cells. Methods Hep3B~(+p53) clone was established by the transfection of the full-length p53 sequence to Hep3B. Following S-adenesylmethionine administration, quantitative real-time PCR were employed to validate the expression changes of GADD45β. pGL3 basic lucfferase plasmids including promoter fragments were synthesized in vitro and transfected into cells. The effects on promoter activity, cell growth and the expression of Caspase were further focused on. Results Hep3B~(+p53) could express p53 protein stably. The transfection of p53 could enhance the induction of GADD45β in Hep3B by S-adenosylmethionine. The promoter activity of fragments constructing NF-κB binding sites could be induced by transfection of p53. The colony formation and DNA syntheses were inhibited apparently in Hep3B~(+p53) with p53 by S-adenosylmethionine. However, the p53 transfection could not influence the cleavage of Caspases. Conclusion p53 may play a role in the induction of GADD45β in Hep3B, which may result from the regulation of transcription factors in proximal promoter.

Abstract:Objective To identify the role of p53 in the induction of growth arrest DNA damage-inducible gene 45β (GADD45β) in HCC cells by Oxaliplatin.Methods A Hep3B+p53 clone was established by transfection of the full-length p53 sequence to Hep3B.Following oxaliplatin administration,quantitative real-time PCR was employed to validate the expression changes of GADD45β.pGL3 basic luciferase plasmids including promoter fragments were synthesized in vitro and transfected into cells.The effects on promoter activity,cell growth and the cleavage of Caspase-3 were further focused on.Results Hep3B+p53 expressed p53 protein stably.The transfection of p553 enhanced the induction of GADD45β in Hep3B by Oxaliplatin.The promoter activity of fragments constructed NF-κB and E2F-1 binding sites was induced about 1.5 and 0.8 folds by transfection of p53.The colony formation and DNA syntheses were inhibited apparently in Hep3B+p53 with p53 by Oxaliplatin (30.41％ and 75.60％ by 100 μmol/L Oxaliplatin,respectively).Moreover,p53 transfection triggered cleavage of Caspase-3 more rapidly.Conclusion p53 played a role in the induction of GADD45β in Hep3B by Oxaliplatin.
Deficient or R273H and R248W Mutations of p53 Promote Chemoresistance to 5-FU <i>via</i> TCF21/CD44 Axis-Mediated Enhanced Stemness in Colorectal Carcinoma

Abstract:Background: p53 mutations are highly frequent in various human cancers and are reported to contribute to tumor malignance and chemoresistance. In this study, we explored the mechanism by which mutant p53 promotes carcinogenesis and chemoresistance and provided novel insights into cancer therapy.Materials and methods: A total of 409 patients with colorectal carcinoma from TCGA database were subdivided into two groups according to the p53 status, namely, mutant p53 and wild-type p53, following with GSEA analysis. The differences of the clinicopathologic index were also analyzed. Two HCT116 cell lines containing hot spots at codons R273H and R248W of p53 were constructed based on HCT116 with knockout p53, respectively. Cell viability, mobility, clonogenesis, and stemness were detected by CCK8, transwell migration and invasion, colonogenic, and sphere formation assays. Resistance to 5-FU was examined by live-dead staining and flow cytometry. qPCR, Western blot, and luciferase reporter assay were performed to identify that deficient or mutant p53 promoted chemoresistance of the colorectal carcinoma cell line HCT116 through the TCF21/CD44 signaling pathway, with the following rescue assays by overexpression of TCF21 and knockdown of CD44.Results: Patients with recurrence harbor a higher frequency of mutant p53 than those without recurrence (p < 0.05). The mutant p53 group developed a larger tumor than the wild-type one. GSEA analysis showed that oncogenic signatures were enriched in the mutant p53 group. Extracellular assays showed that cancer cells with deficient or mutant p53 (R273H and R248W, respectively) promoted colon cancer cell growth, migration, invasion, and stemness. The mutant cancer cells were also observed to be significantly resistant to 5-FU. Xenografts also confirmed that HCT116 cells harboring deficient or mutant p53 promoted cancer growth and 5-FU tolerance. Luciferase reporter assay showed that deficient or mutant p53 R237H and R248W endowed cancer cells with chemoresistance by activating CD44 via repressing the nuclear transcription factor TCF21 expression. Overexpression of TCF21 or knockdown of CD44 could rescue the sensitivity to 5-FU in deficient and mutant p53 HCT116 cell lines.Conclusion: Our results, for the first time, reveal a novel deficient or mutant p53/TCF21/CD44 signaling pathway which promotes chemoresistance in colorectal carcinoma. The axis could be an effective therapeutic strategy against deficient- or mutant p53-driven chemoresistance.
