Abstract:In Heck reactions of cyclic olefins, the products usually have aryl groups that end up at the allylic and/or homoallylic position. We herein report new selectivity that adds aryl groups to the vinylic position. Cyclic olefins of various ring size worked well. The desired isomers were produced by palladium-hydride-catalyzed isomerization of the initial products. Thus, a specific catalyst must be used so that it can perform two jobs under one set of reaction conditions.

Abstract:In intermolecular Heck reactions of styrene and vinylarenes, the aryl and vinyl groups routinely insert at the position. However, selective insertion at the position has been very rare. Herein, we provide a missing piece in the palette of Heck reaction, which gave >20:1 selectivity. The key to our success is a new ferrocene 1,1-bisphosphane (dnpf) that carries 1-naphthyl groups. Our mechanistic studies revealed that the high selectivity is partly attributable to the steric effect of dnpf. The rigid and bulky 1-naphthyl groups of dnpf sterically disfavor insertion.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:The mechanisms and chemo- and regioselectivities of Ru(II)-catalyzed decarboxylative C-H alkenylation of aryl carboxylic acids with alkynes were investigated with density functional theory (DFT) calculations. The catalytic cycle involves sequential carboxylate-directed C-H activation, alkyne insertion, decarboxylation and protonation. The facile tether-assisted decarboxylation step directs the intermediate toward the desired decarboxylative alkenylation, instead of typical annulation and double alkenylation pathways. The decarboxylation barrier is very sensitive to the tether length, and only the seven-membered ring intermediate can selectively undergo the designed decarboxylation, suggesting a tether-dependent chemoselectivity. This tether-dependent chemoselectivity also applies to the alkyl tethers. In addition, the polarity of solvent is found to control the chemoselectivity between the decarboxylative alkenylation and [4 + 2] annulation. Solvent with low polarity (toluene) favors the decarboxylation pathway, leading to the decarboxylative alkenylation. Solvent with high polarity (methanol) favors the ionic stepwise C-O reductive elimination pathway, leading to the [4 + 2] annulation. To understand the origins of regioselectivity with asymmetric alkynes, the distortion/interaction analysis was applied to the alkyne insertion transition states, and led to a predictive frontier molecular orbital model. The asymmetric alkynes selectively use the terminal with the larger HOMO orbital coefficient to form the C-C bond in the insertion step.

Abstract:A palladium-catalyzed Heck diarylation of terminal olefins under ligand-free conditions in acetic acid is described. This procedure allows double arylation of terminal olefins affording trisubstituted olefins in good to excellent yields. The methodology is applicable to the coupling of both electron-deficient and electron-rich aryl iodides leading to symmetrical and unsymmetrical β,β-diarylated alkenes.

Abstract:Axially chiral tetrasubstituted alkenes are of increasing value and interest in chemistry‐related areas. However, their catalytic asymmetric synthesis remains elusive, due to the high steric repulsion and relatively low conformational stability. Herein, we disclose the straightforward construction of atropisomeric tetrasubstituted alkenes via an effective enantiocontrol of vinyl cations. This copper‐catalyzed enantioselective C(sp2)–H functionalization of sterically hindered (hetero)arenes with vinyl cations enables the efficient and atom‐economical preparation of axially chiral acyclic tetrasubstituted styrenes and pyrrolyl ethylenes with high atroposelectivities. Importantly, this reaction represents the first example for the assembly of axially chiral alkenes through vinyl cation approach. Computational mechanistic studies reveal the reaction mechanism, origin of regioselectivity, Z/E selectivity and enantioselectivity. The synthetic utility has been demonstrated by diverse product derivatizations, chiral organocatalyst synthesis, as well as further applications in asymmetric catalysis.

Abstract:Chiral mono-N-protected aminomethyl oxazoline (MPAO) ligands are found to promote enantioselective C-H arylation and vinylation of the cyclobutyl carboxylic acid derivatives via Pd(II)/Pd(IV) redox catalysis. This ligand scaffold overcame two important limitations of the previous MPAHA (mono-N-protected alpha-amino-O-methylhydroxamic acid) ligand-enabled asymmetric C-H activation/C-C coupling reactions of cyclic carboxylic amides through Pd(II)/Pd(0) catalysis: substrates containing alpha-hydrogen atoms are not compatible, and vinylation has not been developed. Sequential C-H arylation and vinylation of cyclobutanes are also performed to construct three contiguous chiral centers on the crowded cyclobutane rings, rendering this reaction highly versatile for the preparation of chiral cyclobutanes.

Abstract:Abstract Since the landmark work of Heck, Negishi and Suzuki on Pd-catalyzed crossing coupling reactions, innovative discovery of new reactions forming C-C bonds and constructing functional olefins via nonmetal catalysts remains an imperative area in organic chemistry. Herein, we report a transition-metal-free arylation method of vinyl pinacolboronates with diaryliodonium salts to form C(sp 2 )-C(sp 2 ) bond and provide trans-arylvinylboronates. The resulting vinylboronates can further react with the remaining aryl iodides (generated from diaryliodonium salts) via Suzuki coupling to afford functional olefins, offering an efficient use of aryliodonium salts. Computational mechanistic studies suggest radical-pair pathway of the diaryliodonium salts promoted by the multi-functional wet carbonate.

Abstract:We describe herein a new method that allows selective production of Catellani-Heck isomers from various aryl halides, including ones without ortho-groups. Under previous conditions, unhindered aryl halides were plagued with the formation of simple Heck isomers and multiple arylation and norbornene insertion. The use of a bulky PtBu3 ligand accelerates C-C reductive elimination from the key palladacycle.

Abstract:The Mizoroki-Heck reaction, which couples aryl halides with olefins, has been widely used to stitch together the carbogenic cores of numerous complex organic molecules. Given that the position-selective introduction of a halide onto an arene is not always straightforward, direct olefination of aryl carbon-hydrogen (C-H) bonds would obviate the inefficiencies associated with generating halide precursors or their equivalents. However, methods for carrying out such a reaction have suffered from narrow substrate scope and low positional selectivity. We report an operationally simple, atom-economical, carboxylate-directed Pd(II)-catalyzed C-H olefination reaction with phenylacetic acid and 3-phenylpropionic acid substrates, using oxygen at atmospheric pressure as the oxidant. The positional selectivity can be tuned by introducing amino acid derivatives as ligands. We demonstrate the versatility of the method through direct elaboration of commercial drug scaffolds and efficient syntheses of 2-tetralone and naphthoic acid natural product cores.

Abstract:Directed C–H arylations have proven to be some of the most advantageous strategies for the synthesis of biaryls. However, their sensitivity toward steric hindrance is a key limitation. Couplings of 2,6-disubstituted aryl halides with arenes have so far been elusive. This weakness is overcome by a ruthenium 3,4,7,8-tetramethyl-1,10-phenanthroline catalyst. It allows the selective ortho-C–H arylation of widely available (hetero)aromatic acids with bulky aryl halides in up to 95% isolated yield. 46 examples of tri-substituted (hetero)biaryls, all outside the scope of established catalyst systems, demonstrate the efficiency of the protocol. Computational and experimental studies illustrate how this unique combination of carboxylate directing group and chelating N-ligand facilitates the selectivity determining C–H activation step. The preference for oxidative addition of the aryl halide over competing benzoic acid coordination is decisive to suppress unwanted dehydrogenative homocoupling.

Abstract:An effective method has been developed for Rh(III)-catalyzed direct vinylation of arenes to give functionalized styrenes, using vinyltriethoxysilane as a convenient and inexpensive vinyl source. A wide variety of substrates, including 1-aryl-2-pyrrolidinones, anilines, benzamides and ketones, were compatible with this reaction. Moreover, this method can be applied to the two-step synthesis of functionalized indoles. Mechanistic investigation reveals that the reaction probably proceeds through an oxidative Heck/desilylation pathway.

Abstract:A palladium-catalyzed aryl-to-alkyl radical relay Heck reaction of amides at α-C(sp3)-H sites with vinyl arenes is described. This process displays a broad substrate scope with respect to both amide and alkene components and provides access to a diverse class of more complex molecules. The reaction is proposed to proceed via a hybrid palladium-radical mechanism. The core of the strategy is the fast oxidative addition of aryl iodide and fast 1,5-HAT overcome the slow oxidative addition of alkyl halides, and photoexcitation effect suppress the undesired β-H elimination. It is anticipated that this approach would inspire the discovery of new palladium-catalyzed alkyl-Heck methods.

Abstract:Olefins play a vital role in the fields of bioscience, medicine, and chemistry. Stereo‐defined olefins are important resource in both chemical laboratory and industry. Using a palladium‐catalyzed decarboxylative transformation of vinyl cyclic carbonates, we have developed an efficient and reliable method for the direct construction of highly substituted olefins with the concomitant formation of a new Csp3‐Csp3 bond. The desired olefins could be generated in reasonable yields with good stereoselectivities, in contrast to the existing methods, which are usually non‐stereoselective. The palladium‐catalyzed decarboxylative transformation will undoubtedly provide new synthetic routes for the formation of multi‐substituted allylic scaffolds.

Abstract:Palladium-catalyzed asymmetric allylic alkylation (AAA) of vinyl benzoxazinanones has become an important strategy for the synthesis of chiral nitrogen-containing heterocycle compounds. However, the asymmetric synthesis of linear-selective products has rarely been reported. The simultaneous control of regio-, E/Z- and enantioselectivities constitutes a major challenge and inhibits the advancement of this chemistry. Herein, we present a palladium-catalyzed AAA of vinyl benzoxazinanones with α-thiocyanato ketones, affording various chiral thiocyanates characterized with high linear-, E- and stereoselectivities. The reaction has a broad substrate scope and the chiral thiocyanates can be transformed to useful heterocycles. Experimental and computational studies suggest an inner-sphere mechanism for AAA process, which results from the acidic and coordination effect of the nucleophilic substrates with palladium catalyst.A unique inner-sphere mechanism for the palladium-catalyzed asymmetric allylic alkylation of vinyl benzoxazinanones characterized with extremely high linear-selectivity, E-selectivity and stereoselectivity.
