Abstract:OBJECTIVE:To explore the role of hypoxia-inducible factor 1alpha (HIF-1α) in early lung fibrosis of rats with acute paraquat (PQ) poisoning.METHODS:Forty eight healthy SD rats were randomly divided into control group (6 rats) and paraquat poisoning group (42 rats). Control group was exposed to 1 ml normal solution by gastric gavage. The paraquat group was exposed to 1 ml paraquat solution (50 mg/kg) by gastric gavage for 2, 6, 12, 48, 72 and 120 h, respectively. The arterial blood gas analysis (PaO(2)) was detected. The pathological examinations of lung tissues were performed by HE and Mason staining. HIF-1α in lung tissues were measured by immunofluorescence. Western blot assay was used to detect the expression levels of HIF-1α protein in lung tissues.RESULTS:PaO2 of rats exposed to paraquat for 72 h was (62.33 ± 0.22) mm Hg, which was significantly lower than that (96.00 ± 5.20) of control group (P < 0.05). Pathological examination by HE staining indicated that the acute diffuse lesion appeared in the alveolar capillary endothelium, epithelia and interstitial tissues, and there was the inflammatory cell infiltration in the alveolar of rats exposed to paraquat at 2 h after exposure. At 12 h after exposure, the interstitial edema in lung tissues of rats decreased and the alveolar space became narrow. At 120 h after exposure, there were the alveolar structure derangement, abundant cicatrix, more fibroblasts and peripheral inflammation absorption. Pathological examination by Masson staining showed that there was obvious collagen deposition in the alveolar epithelia at 2h after exposure, the increased collagen fibrosis at 24 and 48 h after exposure and the obvious damage of alveolar tissues or much more fibrous connective tissue deposition at 120 h after exposure. The results of western blot and immunofluorescence assays exhibited that the expression levels of HIF-1α in lung tissues at 2, 24 and 48 h after exposure significantly increased, as compared with control group (P < 0.05), but there were no significant differences of HIF-1α expression among sub-groups at different time points after exposure.CONCLUSION:The results of present study shown that there were the pulmonary fibrosis and increased expression of HIF-1α in acute PQ poisoning rats at the early stage, and HIF-1α may be associated with pulmonary fibrosis.
Role of Epithelial-to-mesenchymal Transition in the Pulmonary Fibrosis Induced by Paraquat in Rats.

Abstract:BACKGROUND:This study aims to explore the characteristics of the epithelial-to-mesenchymal transition (EMT) process and its underlying molecular mechanisms in the period of paraquat (PQ)-induced pulmonary fibrosis (PF). METHODS:Picrosirius red staining and collagen volume fraction were utilized to evaluate the pathological changes of PQ-induced PF in rats. Immunohistochemistry, Western blot, and real-time reverse transcriptase-polymerase chain reaction (RT-PCR) were used to measure the protein and gene expression of EMT markers, EMT-associated transcription factors, and regulators of EMT-related pathways, respectively. RESULTS:The collagen deposition in the alveolar septum and increased PF markers were characteristics of pathological changes in PQ-induced PF, reached a peak on day 14 after PQ poisoning, and then decreased on day 21. The protein and gene expression of the fibrosis marker, EMT markers, transcription factors, and regulators of EMT-related signaling pathways significantly increased at different time points after PQ poisoning compared with corresponding controls (P<0.05), and most of them reached a peak on day 14, followed by a decrease on day 21. The gene expression of EMT markers was significantly correlated with PF markers, transcription factors, and regulators of EMT-related signaling pathways (P<0.05). The mRNA expression of transcription factors was significantly correlated with that of TGF-β1 and Smad2 (P<0.05 or P<0.01), instead of Wnt2 and β-catenin (P>0.05). CONCLUSIONS:EMT process plays a role in the PQ-induced PF, in which most PF and EMT markers have a peak phenomenon, and its underlying molecular mechanisms might be determined by further studies.
The Expression of HIF-1 Α in Liver Tissues in the Rat Model of Paraquat Poisoning

Abstract:Objective To observe the levels of HIF-α and TGF-β in the liver tissue,change of serum transaminase in different phases after paraquat (PQ) toxicity and liver histopathology change in PQinduced liver toxicity of rats models in order to analyze the relationship between HIF-1 α and hepatic toxicity induced by PQ.Methods A total of 48 healthy SD rats were randomly (random number) assigned into 2 groups:PQ poisoning group (n =42,20％ PQ solution,50 mg/kg in 1mL) and control group (n =6,normal saline 1mL).Rats were separately sacrificed and liver tissue samples were harvested at 2,6,12,24,48,72 h and 120 h after PQ administered by gastric lavage.Arterial blood gas (ABG) analysis and serum alanine transaminase were assayed.Liver tissue HE and Masson staining and immunochemistry changes were also investigated.HIF-1α and TGF-β levels were detected by Western blot technique.Results Lactic acid level was significantly higher in PQ group than that in control group at 6 h after PQ exposure (P ＜ 0.05).The level of serum alanine transaminase was increased and significantly higher if PQ group than that in control group at 2 h after PQ poisoning (P ＜ 0.05).The level of HIF-1 α and TGF-β protein in the liver tissue were up-regulated significantly and higher in PQ group than those in control group at 2 h after PQ exposure (P ＜ 0.05).The hepatocytes necrosis and inflammatory cells infiltration were observed around portal area 2 h after PQ poisoning.At 12 h after PQ exposure,the periportal area filled with necrosis of hepatocytes and the necrosis began to extend,and huge amounts of inflammatory cell infiltration were found.Cord-like fibroplasia was found.All the histological changes were around central portal area and were enlarged gradually.Conclusions The results show that there are increased level of HIF-1 α in the early stage of PQ poisoning rats.The liver fibroplasia may be associated with increasing the level of TGF-β promoted by HIF-1α.
HIF-1α Expression in Lung Tissue of Rats with Paraquat Poisoning

Abstract:Objective To study the HIF-1α expression in rats with paraquat poisoning (PQP).Methods Ninety-six healthy SD rats were randomly (random number) divided into 2 groups ( n =48 in each group) : PQP group and control group. The PQP group was further divided into six subgroups as per thedifferent intervals after Paraquat intoxication. Samples from 6 PQP subgroups and control group were taken at 0 h, and 2 h, 6 h, 12 h, 24 h, 48 h, 72 h and 120 h after paraquat administration. Aarterial blood wascollected for blood gas analysis, and the sections of lung tissue were stained with hematoxyhn-eosin and masson's trichrome to detect the trichrome positive area, and HIF-1α was detected by immunohistochemistryAll data were processed with one-factor analysis of variance and Pearson correlation analysis. Results Two hours after paraquat poisoning, the levels of HIF-1α protein and the collagen were both significantly upregulated in the lung tissue in rats with paraquat poisoning. However, the hypoxemia existed until 72h.There was correlation between levels of Masson's trichrome positive area and HIF-1α positive area (r =0. 819, P ＜0. 05 ). There was correlation between the levels of lactic acid in blood gas analysis and HIF-1α positive area (r=0. 761, P＜0. 05. But there was no correlation between levels ofPO2 and HIF-1α positive area (r=-5.24, P＞0. 05). Conclusions The level of HIF-1α was up-regulated in rats with paraquat poisoning and it was not associated with hypoxemla.
Expression and Significance of HIF-1α in Pulmonary Fibrosis Induced by Paraquat

Abstract:It is commonly accepted that epithelial-mesenchymal transition contributes to fibrotic remodeling, but the molecular pathways involved in paraquat (PQ)-induced epithelial-mesenchymal transition remain uncharacterized. The objective of this study was to evaluate the potential involvement of HIF-1 alpha in TGF-beta 1/beta-Catenin and Snail pathway after PQ poisoning. In our study, 86 Spragne-Dawley rats were randomly divided into control group and PQ group, which received intragastric infusion of 20% PQ solution 50 mg/kg. Rats in the PQ group were subsequently divided into eight subgroups (10 for each subgroup) and samples were collected at different predetermined time points (2, 6, 12, 24, 48, 72, 96 h and 7 d). Fibrosis markers, including beta-catenin, snail and alpha-SMA, were measured by western blot. The activity of HIF-1 alpha was determined by western blot and immunofluorescence. We found that in PQ-induced pulmonary fibrosis, the level of PaO2 was significantly reduced in the 6-h subgroup, when compared to the control group (P < 0.01). Interestingly, between 6 and 72 h, there was no significant difference in PaO2. On the other hand, the level of PaCO2 started to increase from 72-h subgroup (P < 0.01). Fibrosis markers including beta-catenin, snail and alpha-SMA, measured by western blot, were significantly increased at 2 h, while the level of p-GSK-3 beta was increased at 6 h. And the level of GSK-3 beta showed significant reduction beginning at 24 h. The activity of HIF-1 alpha measured by western blot assays was significantly increased starting from 2 h with sustained expression. The result of Pearson coefficient analysis showed that HIF-1 alpha was positively correlated with Snail (r = 0.935, P < 0.01) and beta-catenin (r = 0.761, P < 0.05). Meanwhile, immunofluorescent analysis of HIF-1 alpha revealed partial staining appearing from 2 h. Our data illustrated a positive correlation between Snail, beta-catenin signaling and HIF-1 alpha, suggesting a potential synergistic role of HIF-1 alpha in PQ-induced pulmonary fibrosis, which may be independent of GSK-3 beta. It might also represent a potential therapeutic window for treatment of paraquat poisoning.

Abstract:Paraquat (PQ) poisoning-induced pulmonary fibrosis is one of the primary causes of death in patients with PQ poisoning. Hypoxia-inducible factor-1 (HIF-1) and epithelial-mesenchymal transition (EMT) are involved in the progression of pulmonary fibrosis. Snail and -catenin are two other factors involved in promoting EMT. However, the relationship among HIF-1, Snail and -catenin in PQ poisoning-induced pulmonary fibrosis is not clear. Our research aimed to determine whether the regulation of HIF-1 in EMT occurs via the Snail and -catenin pathways in PQ poisoning-induced pulmonary fibrosis. Sixty-six Sprague-Dawley rats were randomly and evenly divided into a control group and a PQ group. The PQ group was treated with an intragastric infusion of a 20% PQ solution (50 mg/kg) for 2, 6, 12, 24, 48 and 72 hrs. A549 and RLE-6TN cell lines were transfected with HIF-1 siRNA for 48 hrs before being exposed to PQ. Western blotting, real-time quantitative PCR, immunofluorescence, immunohistochemistry and other assays were used in our research. In vivo, the protein levels of HIF-1 and -SMA were increased at 2 hrs and the level of ZO-1 (Zonula Occluden-1) was reduced at 12 hrs. In vitro, the transient transfection of HIF-1 siRNA resulted in a decrease in the degree of EMT. The expression levels of Snail and -catenin were significantly reduced when HIF- was silenced. These data demonstrate that EMT may be involved in PQ poisoning-induced pulmonary fibrosis and regulated by HIF-1 via the Snail and -catenin pathways. Hypoxia-inducible factor-1 may be a therapeutic target for the treatment of PQ poisoning-induced pulmonary fibrosis.
HIF-1α Promotes Paraquat Induced Acute Lung Injury and Implicates a Role NF-κB and Rac2 Activity

Abstract:Paraquat (PQ) is a bipyridine herbicide and oral exposure is the main way of PQ exposure with a very high mortality. At present, it is believed that large number of oxygen free radicals are generated and cause lipid peroxidation of tissue and organ cell membranes after PQ is absorbed. PQ exposure could cause multiple organ dysfunction, among which acute lung injury is the most common and most serious. However, its specific mechanism is still unclear. In this study, the C57BL/6J mouse (alveolar epithelial cell-specific knockout HIF-1α) model of acute lung injury (40 mg/kg PQ) at several time pointes and a model of acute type II alveolar epithelial cell (A549, 800 μM PQ) injury constructed. The oxidative stress (ROS, MDA) and inflammatory response (IL-1β, IL-6, TNF-α) were significantly inhibited in the alveolar epithelial cell-specific knockout of HIF-1α mice and siRNA technology to inhibit HIF-1α in alveolar epithelial cells. Further proteomic analysis showed that the expression of Rac2 protein, which is closely related to oxidative stress, was significantly increased after PQ exposure. And the inhibition of Rac2 expression in vitro significantly alleviated PQ-induced oxidative stress and inflammatory response. The expression of Rac2 protein was regulated by HIF-1α. The above suggests that HIF-1α may promote oxidative stress and inflammatory response in alveolar epithelial cells by regulating the expression of Rac2, and then participate in the promotion of PQ exposure-induced acute lung injury.
Effect of Hypoxia on Expression of AQP4 and HIF-1αin Protein in Prefrontal Cortex of Rats

Abstract:Objective To investigate the expression of AQP4 and HIF-1 αprotein in the prefrontal cortex of SD rats under hypoxic conditions for the basis of elucidating the mechanism of hypoxic brain edema. Methods Twelve SD rats were randomly divided into the normal oxygen group and the hypoxia group.Rats in hypoxia group were exposed to hypoxia (7.8% O2 ,equal to 7 000 m altitude)in a hypobaric chamber for 8 h.The expression of AQP4 and HIF-1 αprotein in rat prefrontal cortex were determined by Western blot. Results AQP4 and HIF-1 α protein were enhanced in the hypoxic rat prefrontal cortex after exposure to 7 000 m hypoxia for 8 h (P <0.05)by Western blot. Conclusion The expression of AQP4 and HIF-1 αprotein in the prefrontal cortex of SD rats were increased after 8 h hypoxia.It was showed that HIF-1 might contribute to hypoxic brain edema by increasing the expression of AQP4 protein.
Expression of Transforming Growth Factor-Β and Hypoxia Inducible Factor-1Α in Lung Tissues of Mice with Paraquat Poisoning

Abstract:Objective To investigate the expression of transforming growth factor-β(TGF-β) and hypoxia inducible factor-1α(HIF-1α) in mice with paraquat poisoning.Methods Thirty male C57BL/6J mice were randomly divided into control group(n=10) and paraquat group(n=20).Paraquat(20 mg/kg) was intraperitoneally administered to mice in paraquat group,while the same volume of normal saline was intraperitoneally administered to those in control group.Ten mice in paraquat group were sacrificed on the 7th day after treatment with paraquat,and the rest of mice were killed on the 28th day.The pathological examinations of lung tissues were performed with HE staining and Masson staining,and the expression of TGF-β and HIF-1α protein in lung tissues was detected by immunohistochemistry.The correlation of expression of TGF-β protein with expression of HIF-1α protein was explored by Pearson correlation analysis.Results HE staining and Masson staining indicated that there were presentations of lung fibrosis in paraquat group on the 7th day,and the severity of lung fibrosis increased on the 28th day.The expression of TGF-β protein in lung tissues in paraquat group was significantly higher than that in control group on the 7th day(P0.05),and the expression of TGF-β protein in lung tissues on the 28th day was significantly higher than that on the 7th day in paraquat group(P0.05).The expression of HIF-1α protein in lung tissues in paraquat group was significantly higher than that in control group on the 7th day and 28th day(P0.05).The expression of HIF-1α protein in lung tissues on the 28th day was higher than that on the 7th day in paraquat group,while there was no significant difference between them(P0.05).There was no correlation of expression of TGF-β protein with expression of HIF-1α protein on the 7th day and 28th day in paraquat group(r=0.295,P=0.630;r=0.218,P=0.725).Conclusion Paraquat poisoning can up-regulate TGF-β and HIF-1α protein in lung tissues of mice,and TGF-β and HIF-1α may be involved in the formation of lung fibrosis through different pathways.
Tripterygium wilfordii Hook.f. ameliorates paraquat-induced lung injury by reducing oxidative stress and ferroptosis via Nrf2/HO-1 pathway

Abstract:Paraquat (PQ) poisoning can induce acute lung injury and fibrosis and has an extremely high mortality rate. However, no effective treatments for PQ poisoning have been established. In this study, the potential efficacy of Tripterygium wilfordii Hook.f. (TwHF) in alleviating PQ-induced lung injury and fibrosis was investigated in a mouse model. Mice were randomly assigned to the control, PQ, PQ + TwHF1 (pretreatment before inducing poisoning), and PQ + TwHF2 (treatment after poisoning) groups. The mice in the PQ + TwHF1 group were pretreated with TwHF for 5 days before receiving one dose of PQ (120 mg/kg) and then received a daily oral gavage of the indicated dosages of TwHF until sacrifice. The mice in the PQ + TwHF2 group were treated with TwHF 2 h after PQ exposure until sacrifice. The pathological analysis and Fapi PET/CT showed that treatment with TwHF attenuated lung injury. And TwHF reduced pulmonary oxidative stress, as indicated by the reduction in, malondialdehyde (MDA), glutathione (GSH), and reactive oxygen species (ROS) levels, as well as by the increase in superoxide dismutase (SOD) levels. Accordingly, the Perls DAB staining showed increased iron concentrations and western blotting revealed a decreased GPX4 expression after PQ exposure, as well as the mitigation of the overexpression of Nrf2 and HO-1 induced by PQ. In conclusion, our study demonstrated the potential of TwHF as a treatment for PQ-induced lung injury and fibrosis. The protective mechanism of this medicinal herb may involve the regulation of ferroptosis.
Hypoxia-inducible Factor-1Α Regulates Epithelial-to-mesenchymal Transition in Paraquat-Induced Pulmonary Fibrosis by Activating Lysyl Oxidase

Abstract:Pulmonary fibrosis (PF) is one of the most prevalent causes of death following paraquat (PQ) poisoning. As demonstrated in previous studies by the present authors, epithelial-to-mesenchymal transition (EMT) is associated with PQ-induced PF. In addition, hypoxia-inducible factor-1α (HIF-1α) and lysyl oxidase (LOX) promote EMT following PQ poisoning. However, the association between HIF-1α- and LOX-mediated regulation of EMT remains unclear. The present study investigated the association between HIF-1α and LOX with regard to PQ-induced EMT. A549 and RLE-6TN cells were treated with PQ, and HIF-1α and LOX expression was silenced with short interfering RNAs. Changes in the expression of HIF-1α, LOX, β-catenin and EMT-related makers were detected using real-time quantitative polymerase chain reaction, immunofluorescence, and western blotting. HIF-1α and LOX were associated with PQ-induced EMT, and their expression levels were significantly increased (P<0.05). LOX expression was significantly decreased following PQ poisoning when HIF-1α expression was inhibited (P<0.05). However, the level of HIF-1α did not change significantly when LOX was silenced. The expression level of β-catenin and the degree of EMT were significantly decreased following HIF-1α and LOX silencing in both cell lines (P<0.05). The association between HIF-1α and LOX in regulating EMT during PQ-induced PF may be unidirectional. HIF-1α may regulate PQ-induced EMT through the LOX/β-catenin pathway.
[Expression of Inflammatory Factors in Lung Tissue of Acute Paraquat Poisoned Rats].

Abstract:OBJECTIVE:To investigate the expression of inflammatory factors in lung tissue of acute paraquat (PQ) poisoned rats.METHODS:Fifty male SD rats were randomized divided into two groups: the normal control group (NC group, n = 10) and the PQ group (n = 40). The 1 ml saline was administered once in normal control group. The PQ group was administered with 20 mg/kg 1% PQ by intraperitoneal injection to establish the model of PQ induced lung injury. At six hours, at the first, the third and the seventh day the PQ group were sacrificed, while at the first day the normal control group was sacrificed. The level of tumor necrosis factor alpha (TNF-alpha) mRNA, interleukin 10 (IL-10) mRNA, high mobility group box 1 (HMGB-1) mRNA in lung of rats were detected. Meanwhile, pathological changes of the lung were examined under optical microscope.RESULTS:Compared with that in normal control group, TNF-alpha mRNA expression in lung tissue of PQ group reached the peak at the six hour and decreased slowly at the first day [(0.740 +/- 0.100) and (0.584 +/- 0.049) respectively]. At the six hour and the first day in PQ group it was significantly higher than that in normal control group (P < 0.05 or P < 0.01). IL-10 mRNA expression in lung tissue of PQ group was elevated at the six hour, reached the peak at the first day, at the third day [(0.551 +/- 0.016) and (0.524 +/- 0.010) respectively] and the seventh day also higher than that in normal control group. At the first and the seventh day in the PQ group it was significantly higher than that in normal control group (P < 0.01). Meanwhile, HMGB-1 mRNA expression in lung tissue of PQ group was also elevated at the six hour, reached the peak at the first day, at the third [(0.695 +/- 0.060), (0.871 +/- 0.154) and (0.819 +/- 0.188) respectively] and the seventh day also higher than that in normal control group. At six hour, the first and the third day in the PQ group it was significantly higher than that in normal control group (P < 0.01). The histological changes such as alveolar edema, hemorrhage and inflammatory cell infiltration in the PQ group were more than those in the normal control group.CONCLUSION:In rats after PQ intoxication the levels of the inflammatory factors TNF-alpha, IL-10 and HMGB-1 are higher than normal rats, and inflammatory could play an important role in lung injury of poisoned rats.

Abstract:Irreversible pulmonary fibrosis induced by paraquat (PQ) poisoning is the major cause of death in patients with PQ poisoning. The epithelial-mesenchymal transition (EMT) is postulated to be one of the main mechanisms of pulmonary fibrosis. Here, we investigated the role of miR-210 in PQ-induced EMT and its relationship with hypoxia-inducible factor-1α (HIF-1α). Western blotting, immunofluorescence, immunoprecipitation and other methods were used in this study. We found that miR-210 expression was significantly increased after PQ poisoning, and it may be regulated by HIF-1α. Overexpression of miR-210 further increased the HIF-1α protein level and promoted EMT. Moreover, miR-210 knock-down reduced the HIF-1α protein level and decreased the degree of EMT. Runt-related transcription factor-3 (RUNX3), a direct target of miR-210, was inhibited by miR-210 in response to PQ poisoning. RUNX3 increased the hydroxylation ability of prolyl hydroxylase domain-containing protein 2 (PHD2), a key enzyme that promotes HIF-1α degradation. PHD2 immunoprecipitated with RUNX3 and its level changed similarly to that of RUNX3. The expression of the HIF-1α protein was significantly reduced when RUNX3 was overexpressed. HIF-1α protein levels were markedly increased when RUNX3 was silenced. Based on these results, a positive feedback loop may exist between miR-210 and HIF-1α. The mechanism may function through miR-210-mediated repression of RUNX3, which further decreases the hydroxylation activity of PHD2, enhances the stability of HIF-1α, and promotes PQ-induced EMT, aggravating the progression of pulmonary fibrosis. This study further elucidates the mechanism of PQ-induced pulmonary fibrosis and may provide a new perspective for the future development of therapies.
Transforming Growth Factor-Β1 / Smads Signaling Pathway Involves the Development of Paraquat-Induced Pulmonary Endothlial-Mesenchymal Transition and Fibrosis

Abstract:Objective To observe the endothlial-mesenchymal transition (EMT) phenomenon and related protein expressions in transforming growth factor-β1 (TGF-β1) / Smads signaling pathway in lung tissues of the rats with paraquat-induced pulmonary fibrosis. Methods Sixty male SD rats were randomly divided into paraquat group (n=30) and control group (n=30).The paraquat poisoning model was established by intraperitoneally injecting with 80 mg / kg of paraquat saline solution in the paraquat group, while rats in the control group were injected with 3 mL of sterile saline. The rats were sacrificed in batches at 1, 3, 7, 14, 21, 28 d after modeling. The wet / dry ratio of left lung tissue and the expression of hydroxyproline were calculated between these two groups. The pathology and pulmonary fibrosis in the inferior lobe of left lung were assessed by hematoxylin-eosin (HE) staining and Masson staining. The expressions of related proteins in EMT and TGF-β1/Smads signaling pathway in lung tissues on days 1, 3, 7, 14, 21, 28 after paraquat poisoning were analyzed by Western-blotting. Results The wet/dry ratio and hydroxyproline both showed significant differences between two groups at different time points after modeling (F=9.772, 22.541, both P<0.001). In the paraquat group, the wet/dry ratios at 3, 7, 14, 21, 28 d were much higher than those in the control group (all P<0.05), and the levels of hydroxyproline only increased at 14, 21, 28 d as compared with those in the control group (all P < 0.05). Pathological findings indicated that inflammatory cell infiltrated, alveolar interstitium thickened in the paraquat group at 1, 3, 7 d, and alveolar walls thickened gradually and a large amount of collagen fibers proliferated at 21, 28 d after modeling. There was no significant change in lung tissue of the control group. The expressions of TGF-β1, Smad2/3, p-Smad2/3, Smad4, p-Smad4, Smad7, p-Smad7, E-cadherin (E-cad), α-smooth muscle actin (α-SMA), Vimentin and fibroblast specific protein-1 (FSP-1) between the two groups at each time point all showed significant differences (F=10.685, 7.381, 7.878, 10.743, 14.575, 17.791, 33.200, 14.453, 10.849, 25.415, 26.263, all P<0.001). The expressions of TGF-β1, Smad2/3, p-Smad2/3, Smad4, p-Smad4, α-SMA, Vimentin and FSP-1 gradually increased and reached a peak at 21 d after paraquat poisoning, while the Smad7, p-Smad7 decreased and reached a bottom at 7 d in the paraquat group (all P<0.05). The expression of E-cad in the paraquat group reduced from 3 d to 21 d (all P < 0.05). Conclusion The TGF-β1 / Smads signaling pathway may affect the development of PQ-induced pulmonary fibrosis by regulating EMT process.
[Role of Connective Tissue Growth Factor and Α-Smooth Muscle Actin in Pulmonary Fibrosis among Acute Paraquat Poisoned Rats].

Abstract:OBJECTIVE:to observe the expression of the connective tissue growth (CTGF) and a smooth muscle actin (α-SMA) in acute paraquat (PQ) poisoned rats and investigate the mechanism of paraquat-induced pulmonary fibrosis.METHODS:sprague-Dawley rats were randomly divided into two experimental groups: control group (6 rats) and PQ group (56 rats). On the 3rd, the 7th, the 14th, the 28th and the 56th day after exposure, the expression of CTGF and α-SMA were evaluated by SABC Immunohistochemistry and Western blot; and the relationship of the expression with pathologic score, hydroxyproline were also analyzed, respectively. The lung pathological changes of rats were observed and pathological evaluation was made.RESULTS:it was similar that the expression pattern of CTGF, α-SMA detected by immunohistochemistry and Western blot. With the time passing, their expression in PQ group increased gradually compared with control group (P < 0.05 or P < 0.01). The increasing extent of CTGF, α-SMA were gentle on the 3rd, the 7th day. While their increasing extent was rapid from the 14th to the 56th day. CTGF was positively correlated with α-SMA, pathologic score and hydroxyproline respectively (r = 0.74, r = 0.87, r = 0.71, P < 0.05 or P < 0.01). Meanwhile, the histological changes such as lung fibroblast proliferation, disorganized collagen fibers were observed in PQ group.CONCLUSION:CTGF and α-SMA could play an important role in the development of pulmonary fibrosis caused by paraquat poisoning; CTGF may promote the transformation of fibroblasts to myofibroblasts and further strengthen the ability of synthesis collagen and extracellular matrix.

Abstract:Paraquat (PQ) is a highly effective herbicide with contact toxicity. PQ mainly accumulates in the lungs after absorption into the blood circulation. The respiratory function failure caused by PQ-induced lung injury, especially the irreversible pulmonary fibrosis in late phase, is the leading cause of death in patients with PQ poisoning. The mechanism of PQ poisoning is still unclear. Now it is speculated that oxidative stress and inflammation injury are the main pathogenic mechanisms, and abnormal gene expression, mitochondrial damage, loss of pulmonary surfactant, cytokine network and unbalanced matrix metalloproteinases/tissue inhibitors may be also involved in the pathogenesis. In addition to reducing poison absorption and increasing its removal, the current clinical treatment is mainly composed of antioxidant and anti-immune response, but has poor therapeutic effects. Although many novel methods of treatment have been proposed, most of them are still in the experimental stage. It is a hot spot to clarify the mechanism of PQ poisoning and to seek safe and effective treatment of pulmonary fibrosis. This article reviews the research progress on pathogenesis and treatment of PQ-induced pulmonary fibrosis.
The Effect on Inhibition of Wnt/β-catenin Signaling Pathway in Paraquat-Induced Pulmonary Fibrosis

Abstract:Objective Paraquat can lead to pulmonary fibrogenesis and its progression, but the underlying mechanisms are not yet unclear. This study aimed to explore the role of the Wnt/β-catenin signaling pathway in the occurrence and progression of pa-raquat-induced pulmonary fibrosis. Methods A total of 48 adult male Sprague-Dawley rats weighing 200-250 g were randomly divided into a paraquat ( PQ) poisoning and a control group of equal number, the former injected intraperitoneally with paraquat at 30 mg/kg to in-duce pulmonary fibrosis while the latter with an equivalent volume of sterile saline. At 14 and 28 days after modeling, all the rats were sac-rificed and their lung tissues collected for examination of the degreesof pulmonary fibrosis by HE and Masson staining and determination of the expression level of the key protein of the Wnt/β-catenin sig-naling pathway β-catenin by Western blot. Human pulmonary epithelial cells were cultured in vitro and divided into a blank control group, a PQ group, treated with 50 μmol/L of PQ for 3 days, and a DKK-1 group, treated with 50 μmol/L of PQ and 20 ng/ml of DKK-1 ( an antagonist of the Wnt/β-catenin signaling pathway) for 3 days) . Then the expression ofβ-catenin was detected by immuno-fluorescence staining and Western blot and those of epithelial and fibroblasts markers E-Cadherin, Occludin, α-SMA and Vimentin were determined by qRT-PCR. Results HE and Masson staining revealed wall thickening and structural derangement of the pulmo-nary alveoli, collagen deposition, and pulmonary fibrogenesis. Compared with the controls, the rats in the PQ group showed markedly increased expression ofβ-catenin at 14 days (0.38±0.04 vs 0.67±0.06, P<0.01), even more significantly at 28 days (1.05±0.08, P<0.01) . The levels of E-Cadherin and Occludin were remarkably down-regulated ( P<0.01) while those ofα-SMA and Vimentin up-reg-ulated in the animals of the PQ group as compared with the blank controls ( P<0.01) . On the contrary, the expressions of E-Cadherin and Occludin were significantly elevated (P<0.01) while those of α-SMA and Vimentin reduced in the DKK-1 group in comparison with the PQ group ( P<0.01) . Conclusion Paraquat poisoning can lead to pulmonary fibrogenesis and activate the Wnt/β-catenin signaling pathway while DKK-1 can block the Wnt/β-catenin signaling pathway and inhibit PQ-induced the epithelial-mesenchymal transition, which indicates that the Wnt/β-catenin signaling pathway plays an essential role in PQ-induced pulmonary fibrosis.
The expression of pulmonary inflammatory factor in paraquat poisoning rats

Abstract:Objective: To measure serum levels of inflammatory factors in paraquat poisoned rats.Methods: 96 rats were grouped in accordance with the randomly principle.They were divided into control group and high dose exposure group(120mg/kg),middle dose exposure group(60mg/kg) and low-dose exposure group(30mg/kg),n=24 rats.Experimental were divided into8,24,72 h points for observation.Used enzyme-linked immunosorbent assay(ELISA) measurement to measure serum TGF-β1,TNF-a,IL-10 concentration.The rats lung were used biopsy and Masson staining to observe pulmonary fibrosis.Results: With the exposure time,each dose group the levels of cytokines gradually increased.24,72 h compared with the control group,the differences were statistically significant(P<0.05 orP<0.01).The every target in 72h high dose group with the same point compared with low-dose group was significantly(P <0.05).72h high,medium and low-dose group compared with the 8h time group was statistically significant(P <0.05).Rat lung tissue infiltration appears inflammatory granulocytes,extensive diffuse interstitial fibrosis of the performance.Conclusion: In paraquat poisoned rats,theinflammatory cytokine of TGF-β1,TNF-a,IL-10 levels were significantly increased.
Paraquat induces pulmonary fibrosis through Wnt/β-catenin signaling pathway and myofibroblast differentiation

Abstract:Paraquat (PQ) poisoning-induced pulmonary fibrosis always results in fatal harm to patients. Our study aimed to investigate the functions of the Wnt/β-catenin pathway in PQ-induced pulmonary fibrosis. By comparing the proteomic profiles of rat lung tissues using protein array in the absence or presence of PQ, the Wnt/β-catenin signaling, as a fibrosis-related pathway, was discovered to be profoundly activated by PQ. The protein levels of Wnt/β-catenin signaling components including MMP-2, β-catenin, Wnt3a, Wnt10b, Cyclin D1, and WISP1 were increased in PQ-treated rat lung tissues. Surprisingly, PQ was found to be able to promote lung epithelial cells and fibroblasts differentiating into myofibroblasts by activating Wnt/β-catenin signaling pathway. Dickkopf-1 (DKK1), an antagonist of Wnt/β-catenin signaling pathway, could inhibit the myofibroblast differentiation and attenuate PQ-induced pulmonary fibrogenesis in vitro and in vivo. The expression levels of fibroblasts markers Vimentin, α-smooth muscle actin (α-SMA) and Collagen I was detected and found to be increased when PQ treated and restored with additional DKK1 treatment. In summary, these assays indicated that Wnt/β-catenin signaling pathway played a regulatory role in the differentiation of lung epithelial cells and fibroblasts, and the pathogenesis of pulmonary fibrosis related to PQ. Inhibition of the Wnt/β-catenin signaling pathway may be investigated further as a potential fibrosis suppressor for pulmonary fibrosis therapy.

The Relationship of Endoplasmic Reticulum Stress with Paraquat Induced Lung Fibrosis in Rats

Abstract:OBJECTIVE:To investigate the relationship between pulmonary fibrosis and endoplasmic reticulum stress (ERS) in rats with paraquat poisoning.METHODS:One hundred male Sprague-Dawley (SD) rats were randomly divided into control group (n=10) and paraquat poisoning group (n=90). The animals were sacrificed by exsanguination at 2, 6, 12, 24, 48, 72, 96, 168, 336 hours after administration with 20% parquat solution. The paraffin sections of lung tissue were stained with hematoxylin-eosin (HE) and Masson trichrome to observe the pathological changes. Glucose-regulated protein 78 (GRP78) was determined by immunohistochemistry, and malondialdehyde (MDA) of lung tissue was measured. The total protein of tissue was abstracted, and the α-smooth muscle actin (α-SMA) and GRP78 was detected by Western blotting.RESULTS:HE and Masson staining demonstrated inflammatory infiltration and collagen deposition in the lung after paraquat administration with a tendency of exaggeration with time, and finally resulted in fibrosis. The expressions of MDA, α-SMA and GRP78 in the lung tissue were significantly increased 2 hours after paraquat administration compared with those of control group (MDA: 1.38 ± 0.18 nmol/mg vs. 0.85 ± 0.05 nmol/mg, α-SMA: 0.23 ± 0.01 vs. 0.14 ± 0.03, GRP78: 0.72 ± 0.02 vs. 0.37 ± 0.06, P<0.05 or P<0.01), and the expressions of MDA and α-SMA were gradually increased with time. GRP78 protein expression was decreased at 72 hours after paraquat administration.CONCLUSIONS:The results reveal that the expressions of α-SMA and GRP78 in paraquat poisoning group are up-regulated, suggesting ERS plays an important role in paraquat induced-pulmonary fibrosis.
