Abstract:A series of chiral tridentate ligands derived from readily available enantiopure l-proline were designed and synthesized. The ligands together with Cu(OAc)2 were successfully used in asymmetric Henry reactions. Various structurally divergent aldehydes and nitromethane were converted into versatile β-nitro alcohols in MeOH at room temperature with very good yields (up to 85%) and enantioselectivities (up to 86%).

Abstract:Highly diastereoselective Michael addition reactions of chiral Ni(II)-complex of glycine with alpha,beta-unsaturated ketones in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) and MeOH at ambient temperature were achieved. The operationally convenient procedure for preparation of the target products renders that this method is an attractive strategy for practical synthesis of 3,5-disubstituted prolines.

Abstract:N-acylation of l-prolinol (2) with enoyl chloride provided l-cinnamoylprolinol (3a) and l-crotonylprolinol (3b), respectively. The attachment of (3a) or (3b) to Merrifield resin and the utilization of such a system for asymmetric Michael additions of Grignard reagents to α,β-unsaturated carbonyl compounds were studied. The stereoselectivities have been increased compared with those observed for the same reactions in solution. On the other hand, it has been proved by the FT-IR difference spectrum of the Merrifield resin and polymer-supported chiral auxiliary that l-prolinol derivatives have been coupled to the resin.

Abstract:An efficient and Brønsted acid free one-pot protocol to directly generate structurally sophisticated α-formylpyrrole derivatives in moderate to good yields has been demonstrated, involving an iron-mediated domino oxidation/formylation reaction of readily available 2,3-dihydro-1H-pyrroles in dimethyl sulfoxide and air atmosphere, in which dimethyl sulfoxide acts as the formyl donor. A possible mechanism is presented.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:A highly effective protocol for the synthesis of C-3-substituted prolines has been developed. Pd-catalyzed C(sp3)–H activation is used for the straightforward functionalization of prolines. The use of an 8-aminoquinolinecarboxamide directing group allows direct arylation, alkenylation, and alkylation at the C-3 position of prolines in moderate to high yields with diverse iodo- or bromo precursors. The resulting 3-substituted proline derivatives are important units in many biologically active compounds and are useful promoters for a variety of functional group transformations.

Abstract:A novel proline derivative, (4R)-4-(beta-Naphthalenyl)methoxy-(S)-proline, was conveniently prepared from the naturally occurring (4S)-hydroxy-(S)-proline; 5 mol % of this compound efficiently catalyzes the asymmetric aldol reactions of various benzaldehydes with acetone in excess of acetone as the solvent, giving the aldol adducts in good yields with ee up to 89.8%.

Abstract:The effect of L-proline as a promoter on the condensation reaction of salicylaldehyde or its derivatives with ethyl acetoacetate in neutral ionic liquid [emim]BF4 was studied. All reactions were carried out under mild reaction conditions and achieved high yields. Moreover, the ionic liquid containing L-proline could be recycled and reused for several times without noticeably decreasing in productivity. The results show that the L-proline-[emim]BF4 system has a potential in contribution to the development of environmentally friendly and inexpensive processes in organic syntheses.

Abstract:Sulfur-containing compounds are a class of important motifs extensively applied in pharmaceutical and pesticidal industries as well as in electrochemistry, toxicant separation, and organic syntheses. Herein, we describe a novel and efficient metal-free catalytic strategy for the rapid synthesis of β-thioketones from sustainable enone derivatives and thiols via thia-Michael addition enabled by heterogeneous prolinamide. At room temperature, up to 98% yield of β-thioketones could be obtained over the solid UiO-66-NH-proline catalyst facilely prepared by the covalent immobilization of proline onto UiO-66-NH2 (a well-known metal–organic framework) via a stable amido linkage. A cooperative effect of proline (amino group) and UiO-66-NH2 (in situ-derived amide species) was observed to play a promotional role in the proceeding of thia-Michael addition, resulting in a high TOF value of 1124.3 h–1. A three-component "iminium" intermediate was illustrated to the key species approaching the product β-thioketone. Moreover, the UiO-66-NH-proline could be easily recovered from the reaction mixture and recycled for at least five times with a slight loss of activity.

Abstract:Sulfonamides and lipids are widely found in natural products, bioactive substances, and pharmaceuticals. Here, we report N-sulfonylation and esterification of carboxylic acids in an environment-friendly one-pot tandem protocol involving 1,2-dichloroethane (DCE). Moreover, 1,8-diazabicyclo (5.4.0) undec-7-ene was necessary for this reaction as a strong base, which drives the reaction to completion. Although DCE is a very low activity reagent, it acts not only as a solvent but also as a reactant in the reaction. The β-chloroester contained in the reaction product can be easily dissociated to react with N, S, and O atoms, increasing the possibility for subsequent synthesis.

Abstract:A new efficient synthesis of (2S,3R)-3-hydroxy-3-methylproline (3) is reported. During the course of a recent study on the Lewis acid promoted intramolecular opening of an epoxide by a carbamate NH, a highly concerted rearrangement was unexpectedly observed. Further investigations of substrate generality show that delta-carbamate-alpha,beta-epoxide esters commonly underwent similar rearrangements with the aid of Lewis acids. Retrosynthetic analysis of such a C-2-N disconnection can lead to an efficient synthesis of (2S,3R)-3-hydroxy-3-methylproline (3) in high enantio purity. Stereochemistries were established by a Sharpless asymmetric dihydroxylation and a diastereoselective reductive amination.

Abstract:In asymmetric Michael addition between ketones and nitroolefins catalyzed by L-proline, we observed that it was benzoic acid or its derivatives rather than other proton acid that could accelerate the reaction greatly, and different benzoic acid derivatives brought different yields. To explain the experimental phenomena, a density functional theory study was performed to elucidate the mechanism of proline-catalyzed asymmetric Michael addition with benzoic acid. The results of the theoretical calculation at the level of B3LYP/6-311+G(2df,p)//B3LYP/6-31G(d) demonstrated that benzoic acid played two major roles in the formation of nitroalkane: assisting proton transfer and activating the nitro group. In the stage of enamine formation from imine, the energy profiles of benzoic acid derivatives were also calculated to investigate the reasons why different benzoic acid derivatives caused different yields. The results demonstrated that the pKa value was the major factor for p-substituted benzoic acid derivatives to improve the yields, whereas for m/o-substituted benzoic acid derivatives, both pKa value and electronic and steric effects could significantly increase the yields. The calculated results would be very helpful for understanding the reaction mechanism of Michael addition and provide some insights into the selection of efficient additives for similar experiments. Abstract:Attachment of the proline-derived ligand 5 to two types of polymers, i.e. aminomethylated polystyrene resin and respectively MeO-PEG, was achieved via a succinyl linkage from the 4-hydroxy group. Both of the supported ligands (7 and 8) were proved to be highly active with good enantioselectivity (up to 90% ee) for the catalyzed asymmetric diethylzinc addition to aldehydes. The insoluble polymer-supported catalyst could be easily recovered and was reusable for several consecutive catalytic runs without significant loss in enantioselectivity.

Abstract:Direct asymmetric aldol addition of methyl ketones to 2,2,2-trifluoro-1-phenylethanone and its ring-substituted derivatives was achieved using L-proline as a chiral promoter. Various optically active beta-trifluoromethyl-beta-hydroxy ketones were obtained in almost quantitative yields with moderate enantioselectivities up to 64 % ee.

Abstract:An asymmetric allylation and allenylation of isatins with facile organoboron reagents was developed under the catalysis of a Lewis acid. A series of optically pure 3-allyl-3-hydroxyoxindoles and 3-allenyl-3-hydroxyoxindoles can be obtained in excellent yields (up to 99% yield) and high enantioselectivities (up to 97% ee). The possible transition state was supported by DFT calculation and the corresponding mechanism was proposed. A gram scale experiment and further functionalization of these chiral 3-hydroxyoxindoles are established.
