Abstract:Vascular endothelial growth factor (VEGF) may cause functional deficiency in dendritic cells (DCs) in vitro. The roles of peripheral blood dendritic cells (PBDCs) and VEGF in patients with oral squamous cell carcinoma (OSCC) are not well understood. The authors analysed the correlation between VEGF and PBDC in 81 OSCC patients. They assessed the effect of VEGF on DC function in vitro. VEGF levels were significantly increased in OSCC patients compared with control subjects (P < 0.05), but PBDC levels were significantly lower (P < 0.05). VEGF expression in TNM I-II (67%) and T1-T2 (74%) was significantly lower, compared with TNM III-IV (88%, P < 0.05) and T3-T4 (89%, P < 0.05). Increased VEGF expression in primary tumours was significantly correlated with elevated serum VEGF levels, but reduced PBDC levels. In vitro cultured DC exposed to VEGF showed significantly decreased expression of functional proteins, enhanced endocytosis activity, and elicited weaker proliferation of T cells, compared with that of free VEGF (P < 0.01). These findings suggest that decreased PBDC and elevated VEGF occur in OSCC patients. Higher VEGF levels may affect precursor cells, resulting in decreased numbers of functional DC.

Abstract:Dendritic cells(DC) are the most powerful professional antigen presenting cells.Lots of studies have demonstrated that the DC vaccines play an important role in antitumor immunity.With the developments of molecular biology,immunology and genetic engineering,rapid progress has been made in DC vaccines and application of DC-based immunotherapy.In this review,the DC biological property,the relationship between the development of cancer and dendritic cell,the production of dendritic cell tumor vaccines and their use in clinical trials were discussed.It may provide valuable information with respect to DC primary study and clinical trials.

Abstract:OBJECTIVE:To explore the role of vascular endothelial growth factor C (VEGF-C), VEGFR-3 and nm23-H(1) in lymphatic metastasis of oral squamous cell carcinoma (OSCC).METHODS:The expression of VEGF-C, VEGFR-3 and nm23-H(1) were examined by immunohistochemical SP method. The lymphatic vessels density (LVD) in tumors was counted and analyzed with clinicopathologic parameters.RESULTS:VEGF-C, LVD and nm23-H(1) expressions were significantly different between OSCC and benign and precancerous lesions (P < 0.05). VEGF-C was correlated with lymph node metastasis and LVD (P < 0.01). nm23-H(1) was related to clinical stage, histological grade and lymphatic metastasis (P < 0.05).CONCLUSIONS:The high expressions of VEGF-C/VEGFR-3 and inactivation of nm23-H(1) may play an important role in lymphatic metastasis in OSCC.
Effect of siRNA-mediated downregulation of VEGF in Tca8113 cells on the activity of monocyte-derived dendritic cells.

Abstract:Vascular endothelial growth factor (VEGF) is a tumor angiogenesis factor that is important in immune regulation. In our previous study, we found that VEGF expression in the peripheral blood and neoplasm nest from patients with oral squamous cell carcinoma (OSCC) was positively correlated with the course of disease, while an inverse correlation between VEGF expression and dendritic cells (DCs) was identified in the peripheral blood. Therefore, in the present study, we investigated whether inhibition of human VEGF in the human tongue carcinoma cell line Tca8113 had effects on the activity of monocyte-derived DCs. We knocked down the expression of human VEGF in Tca8113 cells using the small interfering RNA (siRNA) technique. Tca8113 cells pre-transfected with si RNA targeting VEGF were co-cultured with monocyte-derived immature and mature DCs. Cell proliferation was evaluated by a WST-8 assay. Cell apoptosis, cell cycle and cell phenotypes were determined by flow cytometry. The data revealed that downregulation of the human VEGF significantly inhibited the proliferation of Tca8113 cells and increased apoptosis. Inhibition of human VEGF arrested the cell cycle of Tca8113 cells at the G0/G1 phase. Our results showed that the co-culture of DCs with Tca8113 cells markedly inhibited the expression of the mature markers of DCs including HLA-DR, CD80, CD86,CD40 and CDIa, as well as the immature marker CD83, while inhibition of human VEGF in Tca8113 cells significantly reversed these effects. Therefore, human VEGF in Tca8113 cells may not only regulate the cell proliferation and apoptosis of oral squamous cell carcinoma cells, but may also inhibit DC maturation.
[Tumor-Infiltrating Dendritic Cells in Epithelial Ovarian Carcinoma and Correlation with the Expression of Vascular Endothelial Growth Factor].

Abstract:OBJECTIVE:To investigate the density and activation status of tumor infiltrating dendritic cells (TIDC) in epithelial ovarian carcinoma (EOC) and correlation with the expression of vascular endothelial growth factor (VEGF).METHODS:Streptavidin-peroxidase (SP) and Picture two-step immunohistochemistry methods were used to detect S-100(+), CD(83)(+) TIDC and the expression of VEGF in 57 primary EOCs, 32 benign ovarian tumors (benign control) and 16 normal ovarian tissues (normal control).RESULTS:(1) Two types of heterogeneous distribution pattern of TIDC in EOC were observed under the microscope. The number of S-100(+) TIDC in EOC [median 4.3 cells/high power field (HPF)], was significantly higher than that in benign controls (median 1.8 cells/HPF) and normal controls (median 2.0 cells/HPF, P = 0.000 and 0.015). The number of S-100(+)DC in early stage was significantly higher than that in advanced stage (median 6.0 and 3.8 cells/HPF, P = 0.026). Few CD(83)(+) TIDCs infiltrated tumor stromal tissue in EOC (median 0). (2) The expression of VEGF was significantly higher in EOC than in controls (P = 0.000). (3) The number of S-100(+) DC in EOC was negatively correlated to the expression of VEGF in tumor cells (P = 0.001).CONCLUSIONS:(1) The number of S-100(+) TIDC increases significantly in EOC. Ovarian carcinoma cells may stimulate recruitment of TIDC in EOC, but TIDC can be suppressed by VEGF. (2) Maturation of TIDC in EOC is severely inhibited.
Relation between VEGF and tumor infiltrated dendritic cells in esophageal squamous cell carcinoma

Abstract:Purpose To investigate the relation between VEGF and tumor infiltrated dendritic cells in esophageal squamous cell carcinoma.Methods The expression of VEGF and infiltration of dendritic cells in forty-six specimens of esophageal squamous cell carcinoma were examined by immunohistochemical staining(S-P method) and in situ hybridization,using anti-VEGF monoclonal antibody,and anti-S-100 polyclonal antibody and labeled VEGF mRNA probes.Results There was remarkable difference of VEGF protein expression in esophageal squamous cell carcinoma in different situation of invasion and metastasis.The mean density of S-100 positive dendritic cells in esophageal squamous cell carcinoma was 19.83±13.91 per HPF.The densities of S-100 positive dendritic cells in different grade tumor were 18.50±17.27,28.08±14.05 and 15.740±10.38 per HPF,while the densities in different invasion situation were 32.78±16.60,15.20±11.96,19.83±13.92 per HPF,respectively.There were a significant difference in the density of dendritic cells in tumor with different invasion and metastasis.The densities of dendritic cells of VEGF positive expression tumors(14.93±12.83/HPF) were much lower than the tumor of negative expression(26.79±12.61/HPF).There was a close negative relation between the expression of VEGF and the density of dendritic cells in esophageal squamous cell carcinoma(r=-0.462).Conclusions VEGF protein is associated with the invasion and metastasis of esophageal squamous cell carcinoma.It is firstly detected the relation of TIDC and VEGF protein,suggesting that VEGF protein affects the density of TIDC and might affect the immune system and the invasion and metastasis of esophageal squamous cell carcinoma.

Abstract:Abstract Background Immunotherapy is a crucial therapeutic approach in oncology. However, most patients with head and neck squamous cell carcinoma (HNSCC) do not derive benefit from immunotherapy. Vascular endothelial growth factor (VEGF)/VEGF Receptor 2 (VEGFR2) signaling pathway is one of the most important pathways regulating angiogenesis in tumor. The combination of immunotherapy and anti-angiogenic therapy is considered to improve efficacy of immunotherapy. The correlation between VEGF signaling pathway and tumor immune microenvironment in HNSCC patients is unclear. Methods We utilized RNA sequencing and clinical data of HNSCC patients from the TCGA database to study the correlation between VEGF signaling pathway and tumor immune microenvironment, on aspect of immune cell infiltration, immune-related gene expression profiling and immune-related biological pathways. Results We observed that VEGF signaling pathway is positively correlated with immune cell infiltration, immune-related gene expression profiles, and the prognosis of HNSCC patients. The functional enrichment analysis of differentially expressed genes between different VEGF score subtypes detected multiple immune-related biological processes. Conclusion Our findings suggested that combining anti-VEGF signaling pathway agents with immunotherapy, such as immune checkpoint inhibitors (ICI) therapy, may exhibit encouraging benefits in HNSCC.

The infiltration of DC with different phenotype in head and neck squamous cell carcinoma(HNSCC) patients correlates to tumor prognosis closely. This review will discuss the correlation of tumor prognosis with different subtypes of DC and their infiltration in tumor tissue. Followed by the decreased amount,abnormal maturity and dysfunction of DC in HNSCC patients,including DC in tumor tissue,lymph node and peripheral blood. And then discuss reasons for DC dysfunction and the present research on dendritic cell vaccine.
Immunohistochemical Analysis of Characteristic Phenotype of Dendritic Cell in Draining Lymph Node of Patients with Oral Squamous Cell Carcinoma

Abstract:Objective: To evaluate the functional state of dendritic cell(DC) in the draining lymph nodes of patients with oral squamous cell carcinoma.Methods: Immunohistochemistry was used to detect CD1a and CD83 molecules expressed by DC in paraffin embedded tissues from 34-case draining lymph nodes of OSCC and 10-case tonsils.DC counts,representing the density of DC infiltrating in the tissues,was performed under high-power field.The densities of DC expressing CD1a and CD83 molecules,and the ratio of CD83~+DC/CD1a~+DC were compared between two groups.Results: There were CD1a~+DCs in all cases.The density of CD1a~+DCs in the group of draining lymph node was significantly higher than the one of tonsil(P0.01),but it was lower in draining lymph nodes with metastasis than without metastasis.The density of CD83~+DC was significantly lower in draining nodes than tonsils(P0.01),but there was no significant difference between the nodes with metastasis and without metastasis(P0.05).The ratio of CD83~+DC/CD1a~+DC was significantly higher in draining lymph nodes(P0.05) than tonsils.Conclusion: Immature DC was predominant in the draining lymph nodes of OSCC,and the functional mature of DC was blocked so that DC could have difficulty in activating T lymph cells.

Abstract:Oral squamous cell carcinoma (OSCC) is the most aggressive oral and maxillofacial malignancy with a high incidence and low survival rate. OSCC is mainly diagnosed by tissue biopsy, which is a highly traumatic procedure with poor timeliness. Although there are various options for treating OSCC, most of them are invasive and have unpredictable therapeutic outcomes. Generally, early diagnosis and noninvasive treatment cannot be always satisfied simultaneously in OSCC. Extracellular vesicles (EVs) are involved in intercellular communication. EVs facilitate disease progression and reflect the location and status of the lesions. Therefore, EVs are relatively less invasive diagnostic tools for OSCC. Furthermore, the mechanisms by which EVs are involved in tumorigenesis and tumor treatment have been well studied. This article dissects the involvement of EVs in the diagnosis, development, and treatment of OSCC, providing new insight into the treatment of OSCC by EVs. Different mechanisms, such as inhibiting EV internalization by OSCC cells and constructing engineered vesicles, with potential applications for treating OSCC will be discussed in this review article.

Abstract:Vascular endothelial growth factor (VEGF) is an important proangiogenic factor and has been considered as a key target of antiangiogenetic therapy in oral squamous cell carcinoma (OSCC). However, clinical application of bevacizumab, a specific VEGF antibody, didn't improve the survival rate of OSCC patients. One possible explanation is that VEGF gene expresses diverse isoforms, which associate with extracellular vesicles (EVs), and EVs potentially contribute to VEGF resistance to bevacizumab. However, clear solution is lacking in addressing this issue.

Abstract:OBJECTIVE To investigate the effects of vascular endothelial growth factor (VEGF) on differentiation and function of dendritic cells derived from CD34+ hematopoietic progenitor cells. METHODS After isolation from umbilical cord blood with a high-gradient magnetic cell sorting system (MACS), the CD34+ cells were cultured with a cocktail cytokines for differentiating into dendritic cells (DC). The cells were stimulated by VEGF (25 ng/ml) either at the beginning or at day 9 of culture. Kinetics analysis of cell proliferation was performed during the process of cell culture, and the expression of DC differentiation antigens including CD1alpha, CD83, CD80, CD54 and HLA-DR was examined by flow cytometry. DC function was evaluated by the ability to induce proliferation of allogeneic T cells in mixed lymphocyte reaction (MLR) assay, and the production of IL-12 by ELISA. RESULTS VEGF added at day 1 of culture induced an increase of total cell numbers by (1.51 +/- 0.23)-folds (P = 0.001). VEGF added at the initial but not the late stage of culture could dramatically down-regulate the expression of CD1a [(33.00 +/- 2.12)% vs (81.20 +/- 6.93)%], CD83 [(42.23 +/- 1.15)% vs (87.98 +/- 7.97)%], CD80 (42.93 +/- 1.32)% vs (94.53 +/- 0.87)%], and HLA-DR [(37.93 +/- 5.30)% vs (74.15 +/- 3.74)%], while obviously up-regulate the expression of CD14. Moreover, the inhibitory effect of VEGF on DC function was confirmed by a reduced ability to induce proliferation of allogeneic T cells and production of IL-12 (P < 0.01). CONCLUSIONS VEGF could induce the expansion of hematopoietic progenitor cells and inhibit at the early stage their differentiation into mature DC.

Abstract:We investigated the expression of Notch1 in human oral squamous cell carcinoma (SCC) and explored its potential correlation with epidermal growth factor receptor (EGFR) signalling in oral SCC. Paraffin sections of primary SCC of the tongue and normal mucosa were screened immunohistochemically for Notch1 and EGFR proteins. Human SCC of the tongue Tca8113 cells were treated with AG1478 to block EGFR signalling, and were transfected with the vector that encodes the specific short hairpin RNA (shRNA) that targets EGFR. In SCC of the tongue expression of Notch1 was cancelled except in sites of squamous metaplasia where it was raised, while expression of EGFR was found in the peripheral cells of carcinomas, but not in sites of squamous metaplasia. In normal tongue mucosa, Notch1 was expressed mainly in the stratum corneum, but not in the stratum basale, while EGFR was expressed mainly in the stratum basale, but not in the stratum granulosum or stratum corneum. The blocking of EGFR signalling or the silencing of the EGFR gene resulted in upregulation of Notch1 at mRNA and protein levels in Tca8113 cells. These observations suggest that downregulation of Notch1 in oral SCC may be associated with upregulation of EGFR signalling.
Expression and clinical study of vascular endothelial growth factor and its receptors FLT-1, FLK-1/KDR in non-small cell lung cancer

Abstract:Objective To investigate the relationship between the expression of vascular endothelial growth factor (VEGF), its receptors FLT-1, FLK-1/KDR and tumor angiogenesis, clinical staging, lymph node metastasis in non-small cell lung cancer (NSCLC). Methods Immunohistochemical staining and enzyme-linked immunosorbent assay (ELISA) were used to detect the levels of VEGF and its receptors FLT-1,FLK-1/KDR in the tissue and serum of 112 patients with NSCLC, and analysed their clinical significances. Results The expression of VEGF and its receptors FLT-1, FLK/KDR in lung tissue of NSCLC were significantly higher than that in normal alveolar tissue and paracancerous tissue (P<0.05) and they increased significantly in proportion with clinical stages Ⅰ, Ⅱ, Ⅲ-Ⅳ (P<0.05). The positive rates of VEGF and its receptors FLT-1, FLK-1/KDR in lymph node metastasis group were significantly higher than those without lymph node metastasis group (P<0.05). Se- rum VEGF in NSCLC was significantly higher than that in the control group and also increased according to clinical staging (Ⅰ, Ⅱ, Ⅲ-Ⅳ; P<0.05). Serum VEGF was significantly higher in the lymph node metastasis group than that of the absence group (P<0.05) . Conclusions The expression of VEGF and its receptors FLT-1, FLK/KDR in NSCLC increased significantly, and positively correlated with tumor development, progression and lymph node metastasis. Serum VEGF is an important marker of hyperplasia and metastasis in NSCLC.
Microvesicles releasing by oral cancer cells enhance endothelial cell angiogenesis via Shh/RhoA signaling pathway

Abstract:The present study aimed to investigate the significance of hedgehog signaling pathway in association with clinicopathology parameters and its effect on angiogenesis in oral squamous cell carcinoma (OSCC). The expression of Sonic Hh (Shh) and Gli1 were done on primary tumors and metastatic lymph nodes in OSCC samples from 80 patients by immunohistochemical analysis. The western blot was used to examine the expression of Shh in OSCC cell lines and OSCC-derived microvesicles (MVs). The role of Shh carried by MVs to induce endothelial cell angiogenesis was further investigated by matrigel assay. Our results indicated that the expression of Shh was positive associated with microvesseldentisty(MVD), TNM stage, tumor recurrence and lymph node metastasis. Moreover, Shh and Gli1 expression were higher in paired metastatic lymph nodes compared with expression of their primary tumors. The expression of Shh was abundant in Cal27, and present in SCC4, SCC9, and the amount of Shh protein in Cal27 targeting MVs was increased significantly than Cal27 cell group, up to ∼ fifth-fold. The Cal27 derived MVs increased significantly angiogenesis of HUVECs in vitro, and this effect was blocked with exoenzyme C3 transferase (C3) and shRNA targeting RhoA by suppressing RhoA expression and activation. The data suggested that OSCC derived Shh carried by MVs may facilitate the tumor growth and modulate the preparation of a vascular network in primary tumor and/or premetastatic niche.

Abstract:Loco-regional dissemination of ovarian carcinoma is associated with immunosuppression of the peritoneal cavity. One marked characteristic of the peritoneal immunity in this disease is the defective function of dendritic cells (DCs). In this study, the affect of ovarian carcinoma cells on DCs derived from hematopoetic progenitor cells was observed. The study demonstrated that the expansion, phenotype, and function of DCs generated from CD34+ precursors were significantly altered by the supernatant secreted by ovarian carcinoma cells, and this effect could be partly explained by tumoral overproduction of Vascular Endothelial Growth Factor (VEGF). The results indicated that a role of ovarian carcinoma cells in the differentiation and function of DCs could be associated with the immunosuppression and development of ovarian carcinoma.

Abstract:OBJECTIVETo elucidate the functional status of dendritic cells (DC) in the tissue of oral squamous cell carcinoma by analyzing characteristic phenotype of them.METHODS34 specimens from oral squamous cell carcinoma cases primarily treated with surgery were selected as test group. In addition, 30 specimens of normal mucosa from oral mucocele cases were used as control. Distribution of DC expressing CD1a+, HLA-DR+ and CD83+ in tumor tissue and normal mucous membrane was observed by immunohistochemistry. The number of DC expressing the antigens, which represented the density of DC infiltrating into tissue, was counted by microscope. The density of DC and the rate of DC expressing HLA-DR in oral carcinoma group and control were statistically compared.RESULTSThere was no CD83+ DC in all cases, but CD1a+ DC was found in all samples. The density of CD1a+ DC in tumor tissue was significantly lower than that in normal mucous membrane (P < 0.05). HLA-DR antigen expressed on the surface of DC in tumoral epithelium of 27-case carcinoma specimens and in normal mucous epithelium of 23 cases. The rate of HLA-DR positive expression of TIDC had no statistic significance between the two groups.CONCLUSIONThe lower density of DC infiltrating in tumor tissue might reflect the microenviromental immunodeficiency of hosts with oral squamous cell carcinoma, and the functional mature of DC might be inhibited by the immunosuppressive action of oral squamous cell carcinoma.
Serum IL-17F Combined with VEGF As Potential Diagnostic Biomarkers for Oral Squamous Cell Carcinoma

Abstract:Although interleukin (IL) 17A can promote angiogenesis in several tumors, there are limited clinical evidences on cancer about the correlation between serum vascular endothelial growth factor (VEGF) and IL-17F, which is the most homologous to IL-17A. In this study, serum concentration of IL-17F and VEGF from healthy (n = 28), leukoplakia (n = 15), and oral squamous cell carcinoma (OSCC) groups (n = 85) were assessed and showed that IL-17F level was remarkably downregulated from healthy group (394.3 pg/ml) to OSCC group (82.96 pg/ml). Conversely, the OSCC group had a highest level of VEGF (P < 0.05) in whole groups, and there was a negative correlation between IL-17F and VEGF in serum or in the peripheral blood mononuclear cells (PBMCs) at mRNA level. Moreover, the lowest ratio of IL-17F/VEGF was found in OSCC patients (P < 0.05) and lower ratio of IL-17F/VEGF correlated to higher tumor stage and lymph node metastasis. Furthermore, the serum level of IL-17F and the ratio of IL-17F/VEGF were positively associated with the numbers of CD3+ CD4+ T cells, which indicated that serum IL-17F could originate from PBMCs during the development of OSCC, and could be used for the diagnosis by effectively distinguishing OSCC patients from healthy individuals.
Relationship Between Human Papillomavirus Infection and Dendritic Cell Distribution in Oral Squamous Cell Carcinoma

Abstract:Objective:To investigate the relationship between human papillomavirus(HPV) infection and dendritic cell(DC) distribution in oral squamous cell carcinoma(OSCC).Methods:In situ hybridization(ISH) technology was used to detect the infection of HPV16/18 and immunohistochemistry EnVision technique was applied to observe the distribution of DC in 56 cases of OSCC,26 cases of oral leukoplakia(OLK) and 10 cases of normal oral mucosa.Results:The infection rates of HPV16/18 in OSCC and OLK were obviously higher than that in normal tissues(P0.05).The infection of HPV16/18 was significantly related with smoking(P0.05).The expression of CDla was low in OSCC infected with HPV16/18(P0.01).Conclusion:ISH is an ideal method to detect HPV.HPV infection is an etiology factor of some OSCC.The dysfunction of local immunity of the patients infected with HPV may result in immune escape of the tumor.

Abstract:Dendritic cells (DCs), the most potent antigen-presenting cells, play a central role in the initiation, regulation, and maintenance of the immune responses. Vascular endothelial growth factor (VEGF) is one of the important cytokines in the tumor microenvironment (TME) and can inhibit the differentiation and functional maturation of DCs. To elucidate the potential mechanisms of DC dysfunction induced by VEGF, the effects of VEGF on the biophysical characteristics and motility of human mature DCs (mDCs) were investigated. The results showed that VEGF had a negative influence on the biophysical properties, including electrophoretic mobility, osmotic fragility, viscoelasticity, and transmigration. Further cytoskeleton structure analysis by confocal microscope and gene expression profile analyses by gene microarray and real-time PCR indicated that the abnormal remodeling of F-actin cytoskeleton may be the main reason for the deterioration of biophysical properties, motility, and stimulatory capability of VEGF-treated mDCs. This is significant for understanding the biological behavior of DCs and the immune escape mechanism of tumors. Simultaneously, the therapeutic efficacies may be improved by blocking the signaling pathway of VEGF in an appropriate manner before the deployment of DC-based vaccinations against tumors.
